Bisphenol A (BPA) is an endocrine-disrupting chemical with estrogen-like activity, known to impair immune function. BPA may act as a pro-inflammatory agent, reducing immune response efficacy, increasing bacterial load in E. coli infections, and altering immune responses in parasitic infections (Leishmania major, Nippostrongylus brasiliensis, Toxocara canis) through cytokine and regulatory T-cell modulation. Following its ban in food contact materials in Europe, several analogs have been introduced. This study assessed the immunotoxicity of BPA and six analogs, namely BPAP, BPE, BPP, BPS-MAE, BPZ, and TCBPA, by evaluating in vitro the antibody production. Peripheral blood mononuclear cells from healthy male and female donors were exposed to increasing concentrations of each compound for 24 h. After stimulation with rhIL-2 and ODN2006, IgM and IgG secretion were measured on day six. All compounds suppressed antibody production in a concentration-dependent manner, with some sex-related differences. IC50 values showed BPP as the most potent suppressor, and BPE as the weakest. Similarly, IC20 values confirmed these differences in potency, except for BPA being the weakest for IgM in males. Overall, te results do not support the idea that BPA analogs are safer than BPA. This research focuses on medium-term load forecasting in a tropical region post-pandemic. This study presents one of the first attempts to analyze medium-term forecasting using half-hourly resolution in the Java-Bali power system post-COVID-19 period. The dataset comprises load measurements recorded every 30 min (48 data points per day) from 2014 to 2022. Three distinct methods, namely polynomial regression, split polynomial regression, and Long Short-Term Memory (LSTM) networks, were employed and compared to predict the electricity load demand. The analysis found that LSTM outperformed the other methods, exhibiting the lowest error rates with Mean Absolute Percentage Error (MAPE) at 3.86% and Root Mean Squared Error (RMSE) at 1247.93. Additionally, a consistent observation emerged, showing that all methods performed better in predicting load demand during nighttime hours (6 p.m. to 6 a.m.). The hypothesis is that data stability during nighttime, with fewer significant fluctuations, contributed to the improved prediction accuracy. These findings provide valuable insights for improving load forecasting in the post-pandemic tropical region and offer opportunities for enhancing power grid efficiency and reliability. Full article

Background/Objectives: Single-dose pre-operative antibiotic prophylaxis for cardiac-device implantation is recommended but extending antibiotic prophylaxis is common. Locally, 50–60% of patients had extended prophylaxis after pacemaker insertion or generator change. Our antimicrobial stewardship programme (ASP) incorporated behavioural change strategies in implementing a multi-pronged intervention bundle to address this and evaluated its effectiveness and safety. Methods: This single-centre, retrospective cohort study included patients aged 21 years old or older, undergoing uncomplicated pacemaker insertion or generator change at Singapore General Hospital (SGH) from October 2022 to March 2025. To improve antibiotic use, ASP interventions incorporating behaviour change strategies were implemented, namely (1) data-driven feedback, (2) targeted education, (3) identification and engagement of ASP champion, and (4) clinical pathway revision. Results: There were 779 patients evaluated; 380 (48.8%) received standard prophylaxis while 399 (51.2%) received extended prophylaxis with oral antibiotics (mean duration, 3.3 ± 0.8 days). Following ASP interventions, the practice of extended prophylaxis declined significantly from 43.8% to 24.0% (p < 0.01). The incidence of surgical site infections was low and similar in both groups (0.8%, p = 1.000); all infections were superficial. There was also significant reduction in the proportion of patients on all antibiotics from 20.7% to 16.3% (p < 0.01). Identification and engagement of ASP champion proved pivotal in changing prescribing behaviour through peer influence and credibility. Conclusions: The bundled ASP interventions, incorporating behavioural change strategies, have effectively and safely reduced the use of extended prophylaxis post-cardiac device implantation. Behavioural change interventions are essential to achieve sustained stewardship success. Full article

Inscribed trays, plates, mortars, and pestles made of beautiful green chert bearing formulaic administrative textual formulae were found during excavations at the Persepolis Treasury in the 1930s. These implements and the enigmatic formulae inscribed upon them present scholars with a complex and unique challenge whose correct interpretation holds important implications for the study of Achaemenid history, imperial administration, and relations between ancient Arachosia (roughly modern-day Afghanistan) and the centers of power, as well as—as I argue in this article—for the symbiosis between administration and cult in antiquity. They continue to be hotly debated ever since their inauspicious initial publication by Bowman in 1970, yet they have thus far remained obscure. By comparing these finds with material and textual data from across the Achaemenid empire and early Parthian sources, this article offers a new comprehensive study of these objects. My analysis suggests that these objects are to be considered as a more systematized and tightly controlled Arachosian form of “informal taxation”—namely, regulated gifts—which are comparable to similar imperial donations found in the Treasury at Persepolis. Specifically, they take part in an “economy of fealty” demonstrating loyalty to king and empire through the adherence to the era’s Mazdean ritual orthopraxy. Full article

The need for renewable and eco-friendly materials is driving the increasing demand for biobased polymers for food applications, with cellulose emerging as a promising option due to its degradability and environmental sustainability. Therefore, in the present study, a strategy to obtain cellulose-based materials with antimicrobial properties was explored by using a selected antimicrobial peptide named RKT1, which was stably and efficiently tethered to cellulose films via physical adsorption, harnessing the high number of functional groups on the polymeric surface. Firstly, the peptide, identified among the previous or new projected compounds, was structurally and functionally characterized, evidencing high conformational stability under a wide range of environmental conditions and efficient antibacterial activity against the foodborne pathogens Escherichia coli, Salmonella Typhimurium, and Listeria monocytogenes and the spoilage bacteria Enterococcus and Pseudomonas koreensis, all isolated from meat products. Moreover, in an extended application, the RKT1-activated cellulose films were tested in vivo on beef carpaccio. The results supported their effectiveness in increasing the shelf life of carpaccio by least two days without affecting its organoleptic properties. Therefore, RKT1, physically adsorbed on cellulose, still retains its activity, and the newly generated biopolymers show potential for use as a green food packaging material. Full article

The comprehensive analysis of microbial communities reveals the unique microbial identity of different olive varieties, paving the way for new strategies in their development and commercial exploitation. In this context, the present study aimed to explore the microbial diversity and functional characteristics of Tsounati variety olives from the Monemvasia region of Peloponnese, Greece, that were naturally fermented for three months. The bacterial and fungal microbiota of both olives and brines were fingerprinted throughout the fermentation through classical microbiological analysis combined with molecular techniques. Among the 148 isolated bacteria, 85 were lactic acid bacteria (LAB), and 63 belonged to the Enterobacteriaceae family, while the 178 fungal isolates comprised 136 yeasts and 42 non-yeast or yeast-like fungi. Metataxonomic analysis confirmed the dominance of the bacterial genera Lactiplantibacillus, Leuconostoc, along with the Enterobacteriaceae family, and it revealed the presence of Coleofasciculaceae cyanobacteria mostly in olives. The dominant fungal genera were yeasts, namely Saccharomyces, Nakazawaea, and Cyberlindnera. Using the Folin–Ciocalteu assay, the average total polyphenol content of Tsounati fermented olive samples was 761.80 ± 128.87 mg gallic acid equivalents kg−1 after 90 days of fermentation. The concentrations of the triterpenic, maslinic, and oleanolic acids, as determined by HPLC, remained stable throughout fermentation, with average values of 4764 and 1807 mg kg−1, respectively. Finally, sensory analysis revealed the rich aromatic character of Tsounati variety, highlighting its potential to be used for Greek-style table olive production. Full article

Background/Objectives: The consumption of refined carbohydrates has increased globally. It is associated with inflammation and oxidative stress, both recognized as risk factors for cardiovascular disease. This study investigated the effects of a refined carbohydrate-rich diet on the vascular reactivity of rat aorta. Methods: We acclimatized adult male Wistar rats for two weeks and then randomly assigned them to two experimental groups: a control (CT) group and a high-carbohydrate diet (HCD) group. The CT group received standard laboratory chow for 15 days, while the HCD group received a diet composed of 45% sweetened condensed milk, 10% refined sugar, and 45% standard chow. After the dietary exposure period, we evaluated the vascular reactivity of aortic rings, gene expression related to inflammation, superoxide dismutase activity, and biochemical parameters, including cholesterol, triglycerides, fasting glucose, and glucose and insulin tolerance. Results: The results demonstrate a reduction in vascular reactivity caused by endothelial alterations, including increased NO production, which was observed as higher vasoconstriction in the presence of L-NAME and aminoguanidine and upregulation of iNOS gene expression. In addition, increased production of free radicals, such as O2-, was observed, as well as immune markers like MCP-1 and CD86 in the HCD group. Additionally, the HCD group showed an increase in the TyG index, suggesting early metabolic impairment. GTT and ITT results revealed higher glycemic levels, indicating early signs of insulin resistance. Conclusions: These findings indicate that short-term consumption of a refined carbohydrate-rich diet may trigger oxidative stress and endothelial dysfunction, thereby increasing the risk of cardiovascular complications. Background: Conventional practices for the cultivation of “Cipollotto Nocerino” spring onion are mainly based on growers’ experience, and up to 250 kg/ha for N is commonly furnished among growing cycles. Facing the issue of reduced availability of natural resources for crop production (for example mineral resources), we investigated the optimization of the productivity. Methods: In our research, we tested the use of depletion nutrient supply strategy (CAL-FERT®) to enhance fertilization in accordance with the principle of sustainable agriculture included in the Farm to Fork strategy. In our study, besides the common initial fertilization, three different strategies for cover fertilizations have been elaborated with the support of CAL-FERT® software. The treatments were as follows: (i) commercial standard fertilization as control (named CF); (ii) fertilization equivalent to 50% of the N applied in the control (named F-50); (iii) fertilization corresponding to 25% of the N applied in the control (named F-25); and (iv) strongly reduced fertilization compared to the control (named F-0). The parameters investigated included the following: plant height, yield, SPAD index, nitrogen use efficiency, dry matter, soluble solid content, and pyruvate contents in bulbs and leaves. Nitrogen content was also analyzed for both hypogeous and epigeous apparatuses. Results: Among the most interesting vegetative results, plant height and SPAD readings were reduced only by the extreme treatment F-0 compared with the other treatments at 104 days after planting. Regarding qualitative and productive parameters, the treatments F-50 and F-25 showed the highest yield without prejudging Soluble Solid Content and reducing pungency. Conclusion: In nutritional experiments, onion could be considered as a crop model to investigate quality in vegetables due to its consumption as fresh product and for its particular response, in terms of yield and quality, to fertilization. The use of simulation software can support the identification of strategies to reduce the nutrient supply without any detrimental effect on yield and other vegetative and qualitative parameters in onion crops. Oenological residues may cause environmental pollution when processing does not significantly reduce volume and/or harmful conditions. The lack of proper valorisation alternatives entails high disposal costs and resource inefficiency that jeopardise the sustainability and competitiveness of the industry. Interestingly, wine by-products are underappreciated sources of multipurpose bioactive compounds, such as anthocyanins, associated with health benefits. Alternatively, transforming oenological by-products into valuable co-products will promote sustainability and thus, create new business opportunities. In this context, the present study has assessed the applicability of winery by-products (grape pomace and wine lees) as ingredients to develop new functional kombucha-analogous beverages “3S” (safe, salubrious, and sustainable) by the Symbiotic Culture of Bacteria and Yeast (SCOBY). Concerning the main results, during the kombucha’s development, the fermentation reactions modified the physicochemical parameters of the beverages, namely pH, total soluble solids, acetic acid, ethanol, and sugars, which remained stable throughout the monitored shelf-life period considered (21 days). The fermented beverages obtained exhibited high anthocyanin concentration, especially when using wine lees as an ingredient (up to 5.60 mg/L at the end of the aerobic fermentation period (10 days)) compared with the alternative beverages produced using grape pomace (1.69 mg/L). These findings demonstrated that using winery by-products for the development of new “3S” fermented beverages would provide a dietary source of bioactive compounds (mainly anthocyanins), further supporting new valorisation chances and thus contributing to the competitiveness and sustainability of the winery industries. This study opens a new avenue for cross-industry innovation, merging fermentation traditions with a new eco-friendly production of functional beverages that contribute to transforming oenological residues into valuable co-products. The ability to study mature neuronal cells ex vivo is complicated by their non-dividing nature and difficulty in obtaining large numbers of primary cells from organisms. Thus, numerous transformed progenitor models have been developed that can be routinely cultured, then scaled, and differentiated to mature neurons. In this paper, we present a new method for differentiating one such model, the Lund human mesencephalic (LUHMES) dopaminergic neurons. This method is two days faster than some established protocols, results in nearly five times greater numbers of mature neurons, and involves fewer handling steps that could introduce technical variability. Moreover, it overcomes the problem of cell aggregate formation that commonly impedes high-resolution imaging, cell dissociation, and downstream analysis. While recently established for herpes simplex virus type 1, we demonstrate that LUHMES neurons can facilitate studies of other herpesviruses, as well as RNA viruses associated with childhood encephalitis and hemorrhagic fever. This protocol provides an improvement in the generation of large-scale neuronal cultures, which may be readily applicable to other neuronal 2D cell culture models and provides a system for studying neurotrophic viruses. We named this method the Streamlined Protocol for Enhanced Expansion and Differentiation Yield, or SPEEDY, method. Full article

Accurate forecasting of river water flow helps to warn of floods and droughts in advance, provides a basis for the rational allocation of water resources, and at the same time, offers important support for the safe operation of hydropower stations and water conservancy projects. Water flow is characterized by time series, but the existing models focus on the positive series when LSTM is applied, without considering the different contributions of the water flow series to the model at different moments. In order to solve this problem, this study proposes a river water flow prediction model, named AT-BiLSTM, which mainly consists of a bidirectional layer and an attention layer. The bidirectional layer is able to better capture the long-distance dependencies in the sequential data by combining the forward and backward information processing capabilities. In addition, the attention layer focuses on key parts and ignores irrelevant information when processing water flow data series. The effectiveness of the proposed method was validated against an actual dataset from the Shizuishan monitoring station on the Yellow River in China. The results confirmed that compared with the RNN model, the proposed model significantly reduced the MAE, MSE, and RMSE on the dataset by 27.16%, 42.01%, and 23.85%, respectively, providing the best predictive performance among the six compared models. Moreover, this attention mechanism enables the model to show good performance in 72 h (3 days) forecast, keeping the average prediction error below 6%. This implies that the proposed hybrid model could provide a decision base for river flow flood control and resource allocation. Full article

Genetic and Biological Properties of an Epidemic Feline Panleukopenia Virus Strain (Ala91Ser) in China

To genetically characterise an epidemic isolate of feline panleukopenia virus (FPLV) harbouring the Ala91Ser mutation in China, a clinical strain (accession number: OR921195.1), named FPLV-CC19-02, was isolated from a PCR-positive faecal swab sample. Phylogenetic analysis revealed that it is far removed from all current commercial vaccine strains and differs from the FPLV prototype strain Cu-4 (M38246.1), specifically the vaccine strain of Fel-O-Vax® PCT, at positions 91 (Ala91Ser) and 101 (Ile101Thr) within the VP2 protein. This virus can induce the typical cytopathic effect seen in parvovirus infection in feline kidney cells, resulting in severe clinical symptoms in cats, including haematochezia and hyperthermia. Furthermore, infected cats died of virus infection within 5–10 days post-infection (dpi) (100% morbidity and 83% mortality), indicating that FPLV-CC19-02 is a strain with increased virulence. Additionally, it demonstrated good immunogenicity in cats. Overall, these findings may help us to better understand the molecular prevalence of feline panleukopenia virus in cats and provide valuable basic data for the development of effective, locally adapted feline panleukopenia virus vaccines in China. Burnout syndrome is characterized mainly by three criteria (emotional exhaustion, depersonalization, and low personal accomplishment), and further exacerbated by night shift work, with profound implications for individual and societal well-being. The Maslach Burnout Inventory survey applied to 97 medical care professionals (with day and night work) revealed different scores for these criteria. Blood metabolic profiles were obtained by UHPLC-QTOF-ESI+-MS untargeted metabolomics and multivariate statistics using the Metaboanalyst 6.0 platform. The Partial Least Squares Discrimination scores and VIP values, Random Forest graphs, and Heatmaps, based on 99 identified metabolites, were complemented with Biomarker Analysis (AUC ranking) and Pathway Analysis of metabolic networks. The data obtained reflected the biochemical implications of night shift work and correlated with each criterion’s burnout scores. Four main metabolic pathways with important consequences in burnout were affected, namely lipid metabolism, especially steroid hormone synthesis and cortisol, the energetic mitochondrial metabolism involving acylated carnitines, fatty acids, and phospholipids as well polar metabolites’ metabolism, e.g., catecholamines (noradrenaline, acetyl serotonin), and some amino acids (tryptophan, tyrosine, aspartate, arginine, valine, lysine). These metabolic profiles suggest potential strategies for managing burnout levels in healthcare professionals, based on validated criteria, including night shift work management. Full article

The study of traffic parameters in cities is the basis for making adequate decisions related to the organization and regulation of traffic. This publication presents a study of one of the main parameters of transport flows, namely, its intensity. The study covers one of the busiest streets in the city of Sofia, which is part of the radial connection in the radial circular street network of the city, for the evening peak period of the day. Data analysis presents the influence of the intensity of transport flows at the intersections, which are formed by the intersection with other streets, on the load of the studied street. The share of the load of each transport flow at the individual intersections on the total load of the studied section was recorded for the subsequent assessment of the existing traffic management. The results have been provided to the relevant directorates in the structure of Sofia Municipality for information and use. Full article

Watermeal (Wolffia globosa) is an emerging sustainable feed ingredient, valued for its rapid growth, high protein content, and rich nutrient profile, offering great potential to improve the efficiency and sustainability of edible cricket production systems through efficient resource use and reduced environmental impact. This study aimed to evaluate the effects of replacing commercial cricket feed with dried watermeal and to determine the optimal substitution level for the production performance of two-spotted crickets (Gryllus bimaculatus). Seven dietary treatments were tested, namely 0% (control; T1), 10% (T2), 25% (T3), 50% (T4), 75% (T5), 90% (T6), and 100% (T7). Crickets were reared for four weeks in an open system with 6 replicates per treatment, with each containing 120 nymphs. Body weight, feed intake, and survival rate were recorded weekly, and a production index was calculated at the end of the trial. Results showed average growth rates ranging from 7.40 to 28.20 mg/day, feed conversion ratios (FCR) between 1.03 and 1.68, survival rates of 29.28% to 69.73%, and production index values from 24.00 to 128.30. A significant decline in production efficiency (p < 0.05) was observed at substitution levels of 75% and above, with the 100% watermeal group showing the lowest production efficiency (p < 0.05), likely due to nutrient imbalances and indigestible compounds. Segmented regression analysis identified 36.7% as the optimal level of watermeal supplementation. In terms of body composition, crickets fed diets containing up to 50% watermeal (10%, 25%, and 50%) showed crude protein, crude fat, crude fiber, and ash contents comparable to those fed the commercial diet. This finding suggests that dried watermeal can replace up to 50% of the commercial cricket feed without negatively affecting growth performance, survival, or nutritional quality, with 36.7% identified as the optimal replacement level for maximizing production efficiency.
