Allyl isothiocyanate (AITC) is a naturally occurring, pungent compound abundant in cruciferous vegetables and functions as a repellent for various organisms. The antibacterial effect of AITC against various bacteria has been reported, but there are no reports on the effect on Streptococcus mutans, a major bacterium contributing to dental caries. In this study, we investigated the inhibitory effect and mechanism of AITC on the survival and growth of S. mutans. AITC showed an antibacterial effect in a time- and concentration-dependent manner. In addition, bacterial growth was delayed in the presence of AITC, and there were almost no bacteria in the presence of 0.1% AITC. In a biofilm assay, the amount of biofilm formation with 0.1% AITC was significantly decreased compared to the control. RNA sequencing analysis showed that the expression of 39 genes (27 up-regulation and 12 down-regulation) and 38 genes (24 up-regulation and 14 down-regulation) of S. mutans was changed during the survival and the growth, respectively, in the presence of AITC compared with the absence of AITC. Protein–protein interaction analysis revealed that AITC mainly interacted with genes of unknown function in S. mutans. These results suggest that AITC may inhibit cariogenicity of S. mutans through a novel mechanism. Full article

Cruciferous vegetables are popular in Asian dishes. However, striped flea beetles prefer to feed on leaves, which can damage the appearance of crops and reduce their economic value. Due to the lack of pest monitoring, the occurrence of pests is often irregular and unpredictable. Regular and quantitative spraying of pesticides for pest control is an alternative method. Nevertheless, this requires manual execution and is inefficient. This paper presents a system powered by solar energy, utilizing batteries and supercapacitors for energy storage to support the implementation of edge AI devices in outdoor environments. Raspberry Pi is utilized for artificial intelligence image recognition and the Internet of Things (IoT). YOLOv5 is implemented on the edge device, Raspberry Pi, for detecting striped flea beetles, and StyleGAN3 is also utilized for data augmentation in the proposed system. The recognition accuracy reaches 85.4%, and the results are transmitted to the server through a 4G network. The experimental results indicate that the system can operate effectively for an extended period. This system enhances sustainability and reliability and greatly improves the practicality of deploying smart pest detection technology in remote or resource-limited agricultural areas. In subsequent applications, drones can plan routes for pesticide spraying based on the distribution of pests. Full article

We investigated the genetic and environmental variables determining the glucosinolate (GSL) content of cruciferous vegetables and the implications for cancer prevention. The enzyme myrosinase hydrolyzes GSLs, which are sulfur-containing chemicals found mostly in cruciferous vegetables, producing isothiocyanates (ITCs), which are physiologically active molecules. GSL breakdown products have considerable anti-carcinogenic, antioxidant, and anti-inflammatory capabilities, making them vital to human health. The review dives into genetic heterogeneity among cruciferous species, the importance of individual genes in GSL manufacturing, and breeding techniques for increasing GSL content. It also examines how environmental variables like soil type, pH, plant, nutrient availability, and temperature affect GSL levels. This report also covers the function of GSLs in plant defense, their bioavailability in humans, and their mechanisms in cancer prevention, emphasizing the chemicals’ potential for lowering cancer risk through cruciferous vegetable consumption. The findings highlight the necessity of optimizing both genetic and environmental variables required to increase the nutritional content and medicinal potential of cruciferous vegetables. Pancreatic ductal adenocarcinoma (PDA) is a highly challenging malignancy to treat, with a high mortality rate and limited therapeutic options. Despite advances in cancer research, the prognosis for patients diagnosed with PDA is often poor due to late-stage detection and resistance to conventional therapies. Consequently, there is growing interest in the potential of bioactive compounds as alternative or adjuvant treatments, given their ability to target multiple aspects of cancer biology, offering a more holistic approach to treatment. In the context of PDA, certain bioactive compounds, such as polyphenols (found in fruits, vegetables, and tea), flavonoids, carotenoids and compounds in cruciferous vegetables, have shown potential in inhibiting cancer cell growth, reducing inflammation, and promoting cancer cell apoptosis. This review aims to elucidate the mechanisms, by which these bioactive compounds exert their effects, modulating the oxidative stress, influencing inflammatory pathways and regulating cell survival and death. It also highlights current clinical trials that are paving the way toward incorporating these natural agents into mainstream treatment strategies, with the goal of boosting the efficacy of conventional therapies for PDA. Conventional pesticides and fertilizers are frequently linked to high resource consumption, environmental damage, and poor nutrient usage efficiency in the production of broccoli. Nanofertilizers (e.g., iron, copper, zinc oxide, and boron NMs) and pesticide nanoparticles (NPs) are examples of nanotechnology that is mainly related to broccoli production. These technologies can increase the efficiency of nutrient uptake and utilization as well as broccoli’s resistance to drought, heavy metal stress, saline and alkaline stress, and other conditions. Through accurate fertilization and dosing, nanotechnology can reduce environmental contamination and the need for traditional chemical pesticides and fertilizers. Crops with nanomaterials have higher micronutrient content and better nutritional quality. This study examines the use of nanotechnology in the production of broccoli, which could improve crop yield and quality. However, much research is still required to determine how nanomaterials affect the environment and whether or not they might be hazardous to broccoli because of their minuscule particle size and unique physical and chemical characteristics. Researchers and agricultural professionals both within and outside the field of nanobiotechnology will be able to choose the right nanoparticles for broccoli production with the help of the information presented in this paper. The use of nanotechnology can reduce production costs and support sustainable agricultural growth. Additionally, it opens up new possibilities for the future production, transportation, and storage of cruciferous vegetables. Full article

Sulforaphane (SFN) is a natural isothiocyanate compound with multiple bioactive effects, abundantly found in cruciferous vegetables. SFN and cadmium (Cd) were limited in their application as chemotherapeutic agents due to insufficient cellular uptake, low bioavailability, and high systemic toxicity, respectively. In this study, mPEG-PLGA nanoparticles were used as a carrier to load Cd-γ-PGA conjugates and SFN, enabling favorable drug release under acidic microenvironments with excellent pH responsiveness. The NP-Cd-SFN nanoparticles exhibited a particle size of 102.1 nm, a zeta potential of -14.48 mV, and a PDI value of 0.257. These characteristics contribute to the nanoparticles’ prolonged circulation in the bloodstream and their ability to passively target tumors. Compared to the single-dose groups and the combined Cd + SFN group, the NP-Cd-SFN group significantly reduced the viability of HepG2 cells and increased their apoptosis rate by inducing mitochondrial oxidative stress and promoting cell apoptosis. Overall, the addition of SFN and the encapsulation of mPEG-PLGA enhanced the therapeutic effects of Cd on HepG2 cells. Full article

Glucosinolates (GSLs) are sulfur-containing compounds predominantly found in cruciferous vegetables such as broccoli, kale, and Brussels sprouts, and are recognized for their health-promoting properties. Upon consumption, GSLs undergo hydrolysis by the enzyme myrosinase, resulting in bioactive compounds like isothiocyanates and specific indole glucosinolate degradation products, such as indole-3-carbinol (I3C) and 3,3′-diindolylmethane (DIM), which contribute to a range of health benefits, including anti-cancer, anti-inflammatory, and cardioprotective effects. This review explores the structure, metabolism, and bioavailability of GSLs. Recent evidence supports the protective role of GSLs in chronic diseases, with mechanisms including the modulation of oxidative stress, inflammation, and detoxification pathways. Furthermore, the innovative strategies to enhance GSL bioactivity, such as biofortification, genetic introgression, and optimized food processing methods, have been examined. These approaches seek to increase GSL content in edible plants, thereby maximizing their health benefits. This comprehensive review provides insights into dietary recommendations, the impact of food preparation, and recent advances in GSL bioavailability enhancement, highlighting the significant potential of these bioactive compounds in promoting human health and preventing chronic diseases. Cruciferae family vegetables are remarkably high in phytochemicals such as Indole-3-carbinol (I3C) and Diindolylmethane (DIM), which are widely known as nutritional supplements. I3C and DIM have been studied extensively in different types of cancers like breast, prostate, endometrial, colorectal, gallbladder, hepatic, and cervical, as well as cancers in other tissues. In this review, we summarized the protective effects of I3C and DIM against cardiovascular, neurological, reproductive, metabolic, bone, respiratory, liver, and immune diseases, infections, and drug- and radiation-induced toxicities. Experimental evidence suggests that I3C and DIM offer protection due to their antioxidant, anti-inflammatory, antiapoptotic, immunomodulatory, and xenobiotic properties. Apart from the beneficial effects, the present review also discusses the possible toxicities of I3C and DIM that are reported in various preclinical investigations. So far, most of the reports about I3C and DIM protective effects against various diseases are only from preclinical studies; this emphasizes the dire need for large-scale clinical trials on these phytochemicals against human diseases. Further, in-depth research is required to improve the bioavailability of these two phytochemicals to achieve the desirable protective effects. Overall, our review emphasizes that I3C and DIM may become potential drug candidates for combating dreadful human diseases. Full article

Allyl isothiocyanate (AITC) is a low-molecular-weight natural chemical predominantly obtained from the autolysis of sinigrin, a glucosinolate found in cruciferous vegetables like mustard, horseradish, and wasabi. AITC has sparked widespread interest due to its various biological actions, which include strong antioxidant, anti-inflammatory, antibacterial, and anticancer capabilities. This compound offers promising potential in several fields, particularly in food preservation, medicine, and enhancing food quality through natural means. AITC’s effectiveness against a broad spectrum of microorganisms, including foodborne pathogens and spoilage agents, makes it an attractive natural alternative to synthetic preservatives. The potential to extend the shelf life of perishable foods makes AITC an important tool for food production, meeting rising customer demand for natural additives. In addition to its antimicrobial effects, AITC demonstrates significant anti-inflammatory activity, reducing levels of pro-inflammatory cytokines and modulating key signaling pathways, which could make it valuable in managing chronic inflammatory conditions. Furthermore, emerging research highlights its potential in cancer prevention and treatment, as AITC has been demonstrated to induce apoptosis and inhibit cell increase in several cancer cell lines, offering a natural approach to chemoprevention. This review delves into the chemical structure, metabolism, and bioavailability of freshly produced AITC, providing a comprehensive overview of its beneficial properties. Challenges related to AITC’s volatility, dosage optimization, and regulatory considerations are also discussed, alongside future research directions to enhance the stability and efficacy of AITC-based formulations. The findings underscore AITC’s role as a versatile bioactive compound with known potential to support human health and the sustainable food industry. Full article

Cytoplasmic male sterility (CMS) and dominant genic male sterility (DGMS) both result in the inability to produce or release functional pollen, making them pivotal systems in the hybridization breeding programs of Brassica crops such as cabbage (B. oleracea var. capitata). However, the underling molecular mechanisms are still largely unexplored. This study integrated transcriptomic and metabolomic analyses of cabbage DGMS line, Ogura CMS line, and the maintainer line to uncover the molecular mechanisms underlying these sterility types. The joint analysis predominantly identified significantly enriched pathways, including carbohydrate metabolism, flavonoid biosynthesis, and phenylpropanoid pathways between the MS lines and the maintainer. Especially, the CMS line exhibited a broader range of metabolic perturbations, with a total of 3556 significantly differentially expressed genes (DEGs) and 439 differentially accumulated metabolites (DAMs) detected, particularly in the vitamin B6 metabolism pathway, which showed significant alterations. Given the differences in the inactivation period of microspores in CMS and DGMS lines, we found that DEGs unique to DGMS and maintainer line, such as BoGRPs and BoLTPs, primarily regulate fertility development before the unicellular stage. The DEGs shared between CMS_vs_maintainer and DGMS_vs_maintainer mainly govern microspore development after release from the tetrad, such as BoHXK1 and BoIDH. Additionally, the DEGs unique to CMS_vs_maintainer may contribute to other damage in floral organs beyond male fertility, potentially leading to severe bud abortion, such as BoPNPO. These findings provide a comprehensive framework for understanding the molecular mechanisms of male sterility and offer valuable insights into future breeding strategies in cruciferous vegetables. Cruciferous vegetables of the plant order Brassicales are an attractive dietary component and a valuable source of fiber. However, the nutritional–physiological properties are different when comparing soluble and insoluble fibers. Another significant impact is the transformation of fibers by different influencing factors during food preparation. Cruciferous vegetables, especially, are dominantly processed to soften the matrix. As a result, during cooking, the polysaccharides are dissolved, swelled, or degraded to a certain extent, influencing the composition and the nutritional–physiological properties. The aim of the present study was to analyze the impact of different cooking procedures on changes in the dietary fiber content profile of three different plants: white cauliflower (Brassica oleracea L. var. botrytis), broccoli (B. oleracea L. var. italica), and Brussels sprouts (B. oleracea L. var. gemmifera). The sample material was subjected to direct (“in the water”) and steam cooking. The dietary fiber content and the content of its fractions were determined using an enzymatic analysis method. The results of the research show that the cooking process had a significant influence on the content of dietary fiber fractions in cruciferous vegetables. The concentration of insoluble dietary fiber decreased, whereas the content of soluble dietary fiber increased. When considering the average influence of each process, both steam cooking and direct cooking had a similar impact on changes in the concentrations of dietary fiber fractions. It can therefore be concluded that, when considering dietary fiber content, both processes can be equally well chosen as a thermal treatment for cruciferous vegetables. Full article

The worldwide obesity epidemic has led to a drastic increase in diabetes and cardiovascular disease in younger generations. Further, maintaining metabolic health during aging is frequently a challenge due to poor diets and decreased mobility. In this setting, bioactive nutrients that are naturally occurring antioxidants, such as sulforaphane (SFN), are of high nutritional interest. SFN, a bioactive compound that is present in cruciferous vegetables, is a molecule that protects cells from cytotoxic damage and mitigates oxidative stress, protecting against disease. It exerts its action through the activation of the transcription factor nuclear factor erythroid 2-related factor 2 (Nrf2). Many studies have been performed in animals and humans to evaluate its effects on cancer, brain health, and neurodegenerative disorders. However, fewer clinical studies have been performed to evaluate its effects on insulin resistance and the development of type 2 diabetes mellitus (T2DM) across the lifespan. Given that, in some parts of the world, particularly in Europe, the population is growing older at a significant rate, it is crucial to promote healthy habits (healthy foods, dietary pattern, precision nutrition, and physical activity) from an early stage in life and across the lifespan to avoid debilitating health conditions occurring during adulthood and aging. The Effect of the Glucosinolate Sinigrin on Alterations in Molecular Biomarkers of the Myocardium in Swiss Mice

Glucosinolates are chemically stable compounds that exhibit biological activity in the body following hydrolysis catalyzed by the enzyme myrosinase. While existing in vitro and in vivo studies suggest that the hydrolysis products of glucosinolates predominantly exert beneficial effects in both human and animal organisms, some studies have found that the excessive consumption of glucosinolates may lead to toxic and anti-nutritional effects. Given that glucosinolates are primarily ingested in the human diet through dietary supplements and commercially available cruciferous vegetables, we investigated the in vivo effects of the glucosinolate sinigrin on molecular markers in the myocardia of healthy Swiss mice. This study aims to elucidate whether sinigrin induces positive or negative physiological effects in mammals following consumption. The alterations in myocardial parameters were assessed by measuring metabolic, inflammatory, structural, and antioxidant markers. Our findings revealed that subchronic exposure to sinigrin in the myocardia of female mice resulted in a significant increase (p ≤ 0.05) in the levels of the myokine irisin, matrix metalloproteinases (MMP-2, MMP-9), catalase (CAT), and total glutathione (tGSH), alongside a marked decrease (p ≤ 0.05) in the levels of atrial natriuretic peptide (ANP), compared to the control group consisting of both female and male mice. These results suggest that the hydrolysis products of sinigrin may exert a potentially toxic effect on the myocardial tissue of female mice and possess the capability to modulate transcription factors in vivo in a sex-dependent manner. This observation calls for further investigation into the mechanisms regulating the actions of glucosinolate hydrolysis products, their interactions with sex hormones, and the determination of permissible intake levels associated with both beneficial and adverse outcomes. While a balanced diet can fulfill most nutritional needs, optimizing the composition of specific foods like broccoli can amplify their health benefits. Background/Objectives: Broccoli (Brassica oleracea L. Italica group) is a widely consumed cruciferous vegetable valued for its gastrointestinal and immune health benefits. However, the individual contributions and interactions of broccoli glucosinolates, as they hydrolyze into bioactive isothiocyanates, remain poorly understood. Methods: This study investigated mixtures of four major aliphatic glucosinolates—glucoraphanin, gluconapin, progoitrin, and sinigrin—in individual and combinational models to assess their effects on human colorectal cell proliferation. Results: Combination index analysis revealed moderate to strong antagonistic interactions among these glucosinolates, with the most significant antagonism observed during enzymatic hydrolysis by myrosinase. Mixture analysis identified an optimal glucosinolate ratio including glucoraphanin (81–84%), gluconapin (9–19%), and others (0–7%) to maximize their antiproliferative effects (adjusted R2 > 0.80). This optimal profile was achievable within the target broccoli mapping population. Testing the near-optimal VB067 isogenic broccoli line showed a 44% increase in antiproliferative activity compared to the initial breeding parent or an average sister line. Conclusions: This study highlights the potential of leveraging nutrient–nutrient interactions to guide molecular breeding and produce functional varieties of cruciferous vegetables with optimized health benefits. Isothiocyanates (ITCs), found in edible plants such as cruciferous vegetables, are a group of reactive organo-sulfur phytochemicals produced by the hydrolysis of precursors known as glucosinolates. ITCs have been studied extensively both in vivo and in vitro to define their therapeutic potential for the treatment of chronic health conditions. Therapeutically, they have shown an intrinsic ability to inhibit oxidative and inflammatory phenotypes to support enhanced health. This review summarizes the current evidence supporting the observation that the antioxidant and anti-inflammatory activities of ITCs temper the pathogenic effects of a group of reactive metabolites called advanced glycation end products (AGEs). AGE exposure has significantly increased across the lifespan due to health risk factors that include dietary intake, a sedentary lifestyle, and comorbid conditions. By contributing to a chronic cycle of inflammatory stress through the aberrant activation of the transmembrane receptor for AGE (RAGE), increased AGE bioavailability is associated with chronic disease onset, progression, and severity. This review debates the potential molecular mechanisms by which ITCs may inhibit AGE bioavailability to reduce RAGE-mediated pro-oxidant and pro-inflammatory phenotypes. Bringing to light the molecular impact that ITCs may have on AGE biogenesis may stimulate novel intervention strategies for reversing or preventing the impact of lifestyle factors on chronic disease risk.
