Extracts from 58 species of corticioid fungi (phylum Basidiomycota), mainly belonging to the orders Hymenochaetales, Polyporales and Russulales, were tested for their inhibitory activity against five species of bacteria: Corynebacterium striatum, Haemophilus influenzae, Klebsiella pneumoniae, Pseudomonas aeruginosa, and Staphylococcus aureus. Twenty-four of the species we analyzed in this study were tested for antibacterial activity for the first time. The fruiting bodies of the fungi were collected from dead wood in the forests of north-eastern Poland, and macerated in methanol. Dried extracts were redissolved in dimethyl sulfoxide and applied to broth cultures of the bacteria, which were then inoculated on agar plates. Noblesia crocea demonstrated moderate inhibitory activity against all five tested bacteria; Amylocorticium subincarnatum, Laxitextum bicolor, Peniophora laeta, P. rufomarginata, Phanerochaete sordida, and Xylobolus frustulatus inhibited four bacterial species. The extracts from 14 fungal species tested were moderately active against only two bacteria, P. aeruginosa and C. striatum; 17 species were active against C. striatum only. Cyphellaceae, a small and under-studied family of Agaricales, includes mostly saprophytic taxa with varied basidiomes. In this study, we focus on wood-decay species with corticioid or stereoid basidiomes. Phylogenetic analyses of concatenated ITS-nrLSU sequences uncovered seven generic lineages of corticioid or stereoid fungi—Acanthocorticium, Cericium, Chondrostereum, Cunninghammyces, Gloeostereum, Granulobasidium, and Stratocorticium gen. nov. The genus Cericium is shown to be in the Cyphellaceae family, and two new species, Cericium gloeocystidiatum and Stratocorticium sinensis, are described from East Asia. Morphologically, Ce. gloeocystidiatum is characterized by resupinate basidiomes with smooth hymenophores, a dimitic hyphal system with clamped generative hyphae and micro-binding hyphae, cystidia with resinous-like or golden yellow contents, and ellipsoid basidiospores. Stratocorticium is monotypic, differing from Cericium by a trimitic hyphal system of clamped generative, micro-binding, and brown, thick-walled skeletal-like hyphae, clavate to cylindrical cystidia with homogenous, colorless contents, and hyphidia. Descriptions and illustrations are provided for the new taxa and Cericium luteoincrustatum, and a key to corticioid or stereoid genera in Cyphellaceae is included. Full article

Hymenochaetales, belonging to Agaricomycetes, Basidiomycota, comprises most polypores and corticioid fungi and, also, a few agarics. The latest taxonomic framework accepts 14 families in this order. When further exploring species diversity of Hymenochaetales, two corticioid specimens collected from China producing cystidia with large umbrella-shaped crystalline heads attracted our attention. This kind of cystidia was reported only in three unsequenced species, viz. Tubulicrinis corneri, T. hamatus and T. umbraculus, which were accepted in Tubulicrinaceae, Hymenochaetales. The current multilocus-based phylogeny supports that the two Chinese specimens formed an independent lineage from Tubulicrinaceae as well as the additional 13 families and all sampled genera in Hymenochaetales. Therefore, a monotypic family, Umbellaceae, is newly described with the new genus Umbellus as the type genus to represent this lineage. The two Chinese specimens are newly described as U. sinensis, which differs from T. corneri, T. hamatus, and T. umbraculus in a combination of a smooth to grandinioid hymenophoral surface, not flattened, broadly ellipsoid basidiospores with a tiny apiculus, and growth on angiosperm wood. Due to the presence of the unique cystidia, the three species of Tubulicrinis, even though they lack available molecular sequences, are transferred to Umbellus as U. corneri, U. hamatus, and U. umbraculus. Hereafter, all known species with large umbrella-shaped crystalline-headed cystidia are in a single genus. In summary, the current study provides a supplement to the latest taxonomic framework of Hymenochaetales and will help to further explore species diversity and the evolution of this fungal order. Over the last decade, fungal conservation has become an increasingly important topic, especially for species tied to forest ecosystems. Among these, wood decay fungi are a group of interesting species from ecological and applicative points of view. Culture collections represent an important tool for the conservation of species and research material. The aim of this study was to establish the first research culture collection of wood decay fungal strains, mainly corticioid and polyporoid species, at Salamanca University (Spain). From two areas of the Iberian Peninsula, a total of 120 basidiomata were collected and morphologically identified. From these, 55 strains were successfully isolated in pure culture and their identity was confirmed by DNA molecular analysis. The average growth rate of each strain was recorded and mycelium characteristics, such as colony morphology and microscopic features, were described. Notable strains in the collection included: (1) Botryobasidium asperulum and Phlebia rufa for taxonomical studies; (2) Hericium erinaceus, Grifola frondosa and Pleurotus species for medicinal properties; (3) Irpex lacteus, Phanerochaete sordida and Trametes versicolor for their degradation capabilities; (4) Stereum gausapatum and Stereum hirsutum for their applicative and enzymatic potential. The new fungal strain culture collection represents a valuable tool for the ex situ conservation of Mediterranean wood decay fungi. Full article

Deadwood is involved in several important ecological roles, being the fundamental habitat of wood-decay fungi. At the same time, this polyphyletic group of fungi is the principal agent of wood decomposition, regulating the carbon cycle and the food resource for many other organisms. It is known that the diversity and community composition of wood-decay fungi are related to the tree species, decay stage and size of the woody debris in which they are living. Nevertheless, there is a lack of information on Mediterranean Quercus cerris L. forests. In response, we explored how wood-decay fungi colonize different woody types and how the productivity, richness and community composition of these fungi is influenced by the decay stage and size of the deadwood. Our results indicate that the studied groups, i.e., Ascomycetes, Corticioids, Polyporoids and Heterobasidiomycetes responded differently to the woody debris classes. Moreover, we note the high importance of smaller and soft-decayed woody debris for community composition and richness, hosting a great number of species, in addition to the positive effect of the heterogeneity of the woody debris size for wood-decay fungi productivity. The Auriculariaceae accounts for most of the species in the Auriculariales, and all species in the family are wood-decaying fungi with gelatinous, crustaceous, or woody basidiomes. Many new taxa were published recently, but the taxonomy and phylogeny of the corticioid species in the Auriculariaceae are far from resolved. We undertook a comprehensive taxonomic and phylogenetic study of the family with emphasis on corticioid specimens collected from East and Southeast Asia. Phylogenetic analyses on concatenated ITS and 28S rDNA sequences of representative taxa of the Auriculariaceae and the genera Eichleriella and Heteroradulum were carried out that resolved five new lineages. Heterocorticium gen. nov. is established for two species with resupinate coriaceous basidiomes with smooth, pigmented hymenophores. Five new species, H. bambusicola (generic type), H. latisporum, Eichleriella alpina, E. bambusicola, and Heteroradulum maolanense, are described and illustrated. In addition, Heterochaete delicata, H. discolor, and H. sinensis are transferred to Eichleriella, whereas H. roseola is regarded as a synonym of Kneiffia discolor (= H. discolor). Eichleriella aculeobasidiata is treated as a synonym of Heterochaete sinensis (= E. sinensis). Heterochaete mussooriensis is transferred to Heteroradulum with Heteroradulum semis as a heterotypic synonym. The present study contributes to the understanding of species diversity, taxonomy, and phylogeny of corticioid fungi in Asia. Full article

Peniophora is an old corticioid genus, from which two small satellite genera, Dendrophora and Duportella, were derived based on morphological differences. Molecular systematic studies showed that they belong to Peniophoraceae, Russulales, but the inter- and intra-generic phylogenetic relationships are still unclear. Moreover, the species diversity of this group in subtropical and tropical Asia has not been sufficiently investigated and studied. In this study, we carried out an intensive taxonomic and phylogenetic study on Peniophora sensu lato based on analyses of concatenated ITS1-5.8S-ITS2 (ITS, Internal Transcribed Spacer) and 28S (D1–D2 domains of nuc 28S rDNA) sequence data of all available species worldwide. In the phylogenetic trees, species of Peniophora s. l. (sensu lato) including types of Peniophora s.s. (sensu stricto), Dendrophora and Duportella were interspersed within a strongly supported clade. It means that the morphological delimitations of the three genera are not reliable, and they should be regarded as a large genus. As a result, eight species of Duportella were transferred to Peniophora, although five of them have not been sequenced. Four new distinct lineages, corresponding to Peniophora cremicolor, P. major, P. shenghuae and P. vietnamensis spp. nov., were recovered in the trees. Peniophora taiwanensis is treated as a later synonym of P. malaiensis based on morphological and molecular evidence. Duportella renispora is excluded from Peniophoraceae, because BLAST results of the ITS and 28S sequences of the holotype showed that it is closely related to Amylostereum. Descriptions and illustrations are provided for the four new species, and an identification key is given for all 25 species of Peniophora in China. Our results indicated that the species diversity of the corticioid fungi in Asia is rich and deserves further study. Full article

Bacterial resistance to antibiotics is a serious public health problem that needs new antibacterial compounds for control. Fungi, including resupinated fungi, are a potential source to discover new bioactive compounds efficient again to bacteria resistant to antibiotics. The inhibitory capacity against the bacterial species was statistically evaluated. All the species (basidiomata and strains) were molecularly characterized with the ITS1-5.8S-ITS2 barcoding marker. The strains Ceraceomyces sp., Fuscoporia sp., Gloeocystidiellum sp., Oliveonia sp., Phanerochaete sp., and Xenasmatella sp. correspond to resupinate Basidiomycetes, and only the strain Hypocrea sp. is an Ascomycete, suggesting contamination to the basidiome of Tulasnella sp. According to the antagonistic test, only the Gloeocystidiellum sp. strain had antibacterial activity against the bacterial species Escherichia coli of clinical interest. Statistically, Gloeocystidiellum sp. was significantly (<0.001) active against two E. coli pathotypes (O157:H7 and ATCC 25922). Contrarily, the antibacterial activity of fungi against other pathotypes of E. coli and other strains such as Serratia sp. was not significant. The antibacterial activity between 48 and 72 h increased according to the measurement of the inhibition halos. Because of this antibacterial activity, Gloeocystidiellum sp. was taxonomically studied in deep combined morphological and molecular characterization (ITS1-5.8S-ITS2; partial LSU D1/D2 of nrDNA). A new species Gloeocystidiellum lojanense, a resupinate and corticioid fungus from a tropical montane rainforest of southern Ecuador, with antibacterial potential against E. coli, is proposed to the science. This paper aims to understand the species diversity, taxonomy, and phylogeny of Cystostereaceae (Agaricales), which is based primarily on material from East and Southeast Asia. Cystostereaceae is a small, understudied family of saprobes of woody plants with a worldwide distribution. Phylogenetic analyses of the LSU and ITS sequences revealed four distinct clades in the Cystostereaceae, representing the genera Crustomyces, Cystostereum, Effusomyces gen. nov., and Parvodontia. In addition, phylogenetic analyses showed that Cystidiodontia and Rigidotubus are synonyms of Crustomyces for their type of species nested within the Crustomyces clade. The new monotypic genus Effusomyces, based on specimens from Thailand, lacks any distinctive morphological features. Parvodontia, originally erected for a species from South America, is reported for the first time from Asia. The widely distributed and morphologically well-characterized Cystostereum is represented in East Asia by two new species: Cystostereum crassisporum and C. submurrayi. In addition, three new species, viz., Crustomyces albidus, Effusomyces thailandicus, and Parvodontia austrosinensis, are described and illustrated. Finally, three new combinations are proposed: Crustomyces isabellinus, C. laminiferus, and C. tephroleucus. A key to the genera and species of Cystostereaceae is provided. Our results proved that the species diversity of wood-decaying fungi in East and Southeast Asia is rich and suggested that more investigations and studies should be carried out in the future. Auriculariales is diverse, embracing a number of corticioid, poroid, and hydnoid genera. The present study covers a new wood-inhabiting fungal species of Heteroradulum niveum sp. nov that is proposed on the basis of a combination of morphological features and molecular evidence. The species is characterized by the resupinate basidiomata, a monomitic hyphal system with generative hyphae with clamp connections; tubular cystidia; two- to four-celled basidia; and allantoid, colorless, thin-walled, smooth, IKI–, CB–, basidiospores (6.5–13.5 × 2.7–5.5 µm). Sequences of ITS and nLSU rRNA gene regions of the specimens were generated, and phylogenetic analyses were carried out with methods of maximum parsimony, maximum likelihood, and Bayesian inference. These phylogenetic analyses inferred from ITS+nLSU indicated that H. niveum is nested in Heteroradulum within Auriculariales. Further study within Heteroradulum on the basis of ITS+nLSU dataset revealed that it formed a monophyletic lineage with a strong support (100% BS, 100% BP, 1.00 BPP) and then grouped with H. yunnanensis. Two new corticioid fungal species, Phanerochaete pruinosa and P. rhizomorpha spp. nov. are proposed based on a combination of morphological features and molecular evidence. Phanerochaete pruinosa is characterized by the resupinate basidiomata with the pruinose hymenial surface, a monomitic hyphal system with simple-septate generative hyphae and subcylindrical basidiospores measuring as 3.5–6.7 × 1.5–2.7 µm. Phanerochaete rhizomorpha is characterized by having a smooth hymenophore covered by orange hymenial surface, the presence of rhizomorphs, subulate cystidia, and narrower ellipsoid to ellipsoid basidiospores. Sequences of ITS+nLSU nrRNA gene regions of the studied specimens were generated and phylogenetic analyses were performed with maximum likelihood, maximum parsimony, and Bayesian inference methods. These phylogenetic analyses showed that two new species clustered into genus Phanerochaete, in which P. pruinosa was sister to P. yunnanensis with high supports (100% BS, 100% BT, 1.00 BPP); morphologically differing by a pale orange to greyish orange and densely cracked hymenial surface. Another species P. rhizomorpha was closely grouped with P. citrinosanguinea with lower supports; morphologically having yellow to reddish yellow hymenial surface, and smaller cystidia measuring as 31–48 × 2.3–4.8 µm. Thelephora grandinioides is characterized by laterally stipitate basidiocarps with a grandinoid hymenial surface, a monomitic hyphal system with clamped generative hyphae, and the presence of tubular and septated cystidia and subglobose to globose basidiospores measuring as 5.3–7.4 × 4–6.5 µm. Thelephora wuliangshanensis is characterized by infundibuliform basidiocarps, radially black striate on the pileus, a smooth, umber to coffee hymenial surface, a monomitic hyphal system with thick-walled generative hyphae, and basidiospores that turn greenish grey to buff in 5% KOH. Phylogenetic analyses of rDNA internal transcribed spacer region (ITS) and nuclear large subunit region (nrLSU) showed that the two new Thelephora are phylogenetically distinct: T. grandinioides is sister to T. aurantiotincta and T. sikkimensis, while T. wuliangshanensis is sister to a clade comprising T. austrosinensis and T. aurantiotincta with high support as well. Fibrodontia is a genus of wood-inhabiting fungi consisting of four species so far, including F. gossypina as generic type. Two new species, Fibrodontia austrosinensis and F. subalba, are described and illustrated from China. Fibrodontia austrosinensis from southwestern China is characterized by a grandinioid to odontioid hymenophore with numerous small aculei, a dimitic hyphal system with scattered, smooth skeletal hyphae and ellipsoid basidiospores measuring 4.2–5.2 × 3.5–4.5 μm. Fibrodontia subalba from the West Tianshan Mountain in northwestern China is distinguished by an odontioid to hydnoid hymenophore, a dimitic hyphal system, and ellipsoid basidiospores measuring 3.7–4.4 × 2.8–3.4 μm. The phylogenies inferred from the data set of nuc rDNA ITS1-5.8S-ITS2 (ITS) and D1–D2 domains of nuc 28S rDNA (28S), and that of ITS, 28S, translation elongation factor (tef1α), and RNA polymerase II second largest subunit (rpb2) supported Fibrodontia as a monophyletic genus in the Trechisporales, and F. austrosinensis and F. subalba as separate lineages within Fibrodontia. Multi-rate Poisson Tree Processes, Automatic Barcode Gap Discovery and genetic distance methods based on ITS sequences of Fibrodontia also supported F. austrosinensis and F. subalba as distinct species. The taxonomic status of F. fimbriata that was recently transferred from Cystidiodendron, is briefly discussed. A key to all six known species of Fibrodontia is provided. Full article

The diversity of sporocarp-inhabiting fungi (SCIF) was examined using six samples of xylarialean fungi from two different forests in Ibaraki Prefecture, Japan: a moist forest in the Sakuragawa area and an urban dry forest in the Tsukuba area. These fungi were enumerated using direct observation and dilution plate methods. We obtained 44 isolates, and careful morphological and molecular phylogenetic studies of these isolates revealed that approximately 30% of the operating taxonomic units were undescribed or cryptic species related to known fungi. Although typical mycoparasitic fungi, such as helotialean fungi and Trichoderma spp., were not isolated, the genera Acremonium, Acrodontium, and Simplicillium were detected. Comparisons of SCIF communities between the two forests suggested that the number of isolated species in the Sakuragawa area was lower than that in the Tsukuba area. Soil-borne fungi, such as Aspergillus, Beauveria, Penicillium, and Talaromyces, or polypores/corticioid mushrooms, are frequently detected in the Tsukuba area. Factors affecting SCIF communities in the two forests are discussed. Some noteworthy fungi are briefly described with notes on taxonomy, ecology, and molecular phylogeny. Full article

Wood-decaying fungi play crucial roles as decomposers in forest ecosystems. In this study, two new corticioid fungi, Rhizochaete fissurata and R. grandinosa spp. nov., are proposed based on a combination of morphological features and molecular evidence. Rhizochaete fissurata is characterized by resupinate basidiomata with a cracking hymenial surface, a monomitic hyphal system with simple-septa generative hyphae, presence of subfusiform to conical cystidia encrusted at the apex or coarse on the upper half, and ellipsoid basidiospores. Rhizochaete grandinosa differs in its resupinate basidiomata with a smooth hymenial surface, presence of two types of cystidia, and ellipsoid basidiospores. Sequences of ITS and nLSU rRNA markers of the studied samples were employed, and phylogenetic analyses were performed with maximum likelihood, maximum parsimony, and Bayesian inference methods on two datasets (ITS+nLSU and ITS). Both dataset analyses showed that two new species clustered into the genus Rhizochaete, in which, based on the ITS+nLSU dataset, R. fissurata was sister to R. belizensis, and R. grandinosa grouped with R. radicata; the phylogram inferred from ITS sequences inside Rhizochaete indicated that R. fissurata formed a monophyletic lineage with a lower support; R. grandinosa grouped closely with R. radicata. In addition, an identification key to all Rhizochaete species worldwide is provided. Full article
