This study evaluated the effects of dietary hydroxytyrosol (HT) addition on piglets during the nursery phase across two experiments. In the first, 72 weaned male piglets (~26 days old, 7.3 ± 0.5 kg) were assigned to one of four diets containing 0, 5, 10, or 50 mg HT/kg feed. Growth performance, serum biochemistry, histological and behavioral parameters, and meat lipid profiles were assessed. In the second study, the apparent digestibility of diets containing 0, 25, or 50 mg HT/kg feed was evaluated using 15 male piglets (21.5 ± 1.5 kg) through total excreta collection. Results revealed that HT influenced serum glucose and gamma-glutamyl transferase, histological inflammation, and active behaviors. HT modified lipid profiles, reduced capric, lauric, linolenic, arachidonic, cis-5,8,11,14,17-eicosapentaenoic fatty acid concentrations, and increased the nervonic acid profile. The digestibility of dry matter, organic matter, energy, and protein increased with HT use up to 50 mg/kg of feed. These findings demonstrate that HT positively impacts piglet efficiency, changing the fatty acid profile with increased nervonic acid, highlighting its potential as a dietary additive for improving nursery pig production. This study assesses the quality of fat in conventional and specialist infant formulas (IFs) available in Poland. The IFs studied were characterized in terms of fatty acid profiles and lipid quality indices. The study material consisted of eight types of conventional and specialist IFs. The determination of fatty acids was carried out using gas chromatography (GC). Lipid quality indices were estimated based on established formulas. Goat milk-based formulas showed significantly higher levels of caproic acid (C6:0) and capric acid (C10:0) than cow milk-based formulas of the same category (initial or follow-on) (p < 0.05). In addition, these IFs stood out in terms of conjugated linoleic acid (CLA) content (0.30%) compared to cow and specialist formulas (about 0.20%). It was shown that the average ratio of n6/n3 fatty acids was significantly lower in conventional IFs (6.07:1) compared to specialist IFs (8.10:1). The goat’s milk-based IFs had the most favorable values for individual lipid quality indices (index of desirable fatty acids (DFAs) = 62.46; index of hypercholesterolemic fatty acids (OFAs) = 25.94; index of atherogenicity (AI) = 0.71; index of thrombogenicity (TI) = 0.88; hypocholesterolemic/hypercholesterolemic ratio (H/H) = 2.05), while the specialist S-PH formula was characterized by the lowest DFA value (49.17) and the highest AI and TI indices (1.48 and 1.68). Multivariate analysis clearly classified the division of formulas into two groups—conventional and specialist—based on lipid quality indices. The results obtained provide new information on the variation in the lipid profile of IFs depending on the intended use of the formula and may serve as a basis for further research in this area. Comparison of Food Compound Intake Between Food-Allergic Individuals and the General Population

Background: Individuals with food allergies typically need to avoid specific allergens, leading to distinct dietary choices. Their food product intake may therefore vary from that of the general population, potentially leading to differences in their intake of nutrients and other food compounds. Methods: We compared food compound and nutrient group intakes between the general Dutch adult population (n = 415) and food allergic Dutch adult patients with either milk and/or egg allergies (n = 16), peanut and/or tree nut allergies (n = 35) or a combination of milk/egg and peanut/tree nut allergies (n = 22). We translated 24-hour dietary recall data into food compound intake values. We used a mixed effects ANOVA model and considered compound intakes statistically significantly different at FDR-corrected p < 0.05. Additionally, compounds with uncorrected p < 0.01 were explored for potential relevance. Results: A total of 489 compounds or nutrient groups were included in the statistical analysis. Milk/egg and mixed allergic patients had significantly lower intakes of beta-lactose, butyric acid, caproic acid, caprylic acid, capric acid, lauric acid, myristic acid, myristoleic acid, conjugated linoleic acid, and remainder saturated fatty acids (p < 0.05, FDR corrected), with mean intake factors of 1.6–3.2 and 1.3–2.9 lower, respectively, than the general population. In addition, 36 other compounds showed intake differences with a p < 0.01 without FDR correction. There were no statistically significant differences between the peanut/tree nut allergy group and the general population. Conclusions: Our study shows significantly lower intakes of 10 mainly dairy-derived compounds by the milk/egg and mixed-allergic patients, presenting the potential for long-term health consequences and the need for supplementation a relevant consideration, warranting further research. Sixteen long-ripened, high-quality Cabrales cheeses from independent producers underwent a comprehensive biochemical and microbiological characterisation. Significant variations in total microbial counts and specific microbial groups were observed among the cheeses. A metataxonomic analysis identified 249 prokaryotic amplicon sequence variants (ASVs) and 99 eukaryotic ASVs, respectively, which were classified into 52 prokaryotic and 43 eukaryotic species. The predominant species included bacteria of the genera Tetragenococcus, Lactococcus (of which Lactococcus lactis was used as a starter), and Staphylococcus, followed by Brevibacterium and Corynebacterium species. The starter mould Penicillium roqueforti was highly abundant in all cheeses; Debaryomyces hansenii, Geotrichum candidum, and Kluyveromyces spp. constituted the subdominant fungal populations. Glutamic acid (≈20 mg g−1) was the most abundant free amino acid in all samples, followed by lysine, leucine, and valine (≈10–13 mg g−1). Moderate-to-high amounts of the biogenic amines tyramine and ornithine were detected. A large variation between cheeses of the main organic acids (lactic, acetic, or butyric) was detected. Differences between samples were also observed for the majority volatile compounds, which included organic acids, alcohols, esters, and ketones. Positive and negative correlations between bacterial and fungal species were detected, as well as between microbial populations and key biochemical markers. Among the latter, Tetragenococcus halophilus correlated positively with ethyl caprylate and hexanoic acid, and Loigolactobacillus rennini correlated positively with γ-aminobutyric acid. Conversely, Staphylococcus equorum showed a strong negative correlation with ethyl caprylate and capric acid. These microbial and biochemical insights enabled us to propose a microbiota-based starter culture comprising prokaryotic and eukaryotic components to enhance Cabrales cheese quality. Full article

In this investigation, a comprehensive validation framework for an integrated electrochemical-thermal model that addresses critical thermal management challenges in lithium-ion batteries (LIBs) is presented. The two-dimensional numerical model combines the Newman–Tiedemann–Gu–Kim (NTGK) electrochemical-thermal battery framework with the enthalpy-porosity approach for phase change material (PCM) battery thermal management systems (BTMSs). Rigorous validation against benchmarks demonstrates the model’s exceptional predictive capability across a wide range of operating conditions. Simulated temperature distribution and voltage capacity profiles at multiple discharge rates show excellent agreement with the experimental data, accurately capturing the underlying electrochemical-thermal mechanisms. Incorporating Capric acid (with a phase transition range of 302–305 K) as the PCM, the thermal management model demonstrates significantly improved accuracy over existing models in the literature. Notable error reductions include a 78.3% decrease in the Mean Squared Error (0.477 vs. 2.202), a 53.4% reduction in the Root Mean Squared Error (0.619 vs. 1.483), and a 55.5% drop in the Mean Absolute Percentage Error. Statistical analysis further confirms the model’s robustness, with a high coefficient of determination (R2 = 0.968858) and well-distributed residuals. Liquid fraction evolution analysis highlights the PCM’s ability to absorb thermal energy effectively during high-discharge operations, enhancing thermal regulation. This validated model provides a reliable foundation for the design of next-generation BTMS, aiming to improve the safety, performance, and lifespan of LIBs in advanced energy storage applications where thermal stability is critical. Cannabidiol (CBD), a phytocannabinoid commonly isolated from chemotype III Cannabis sativa plants, is known for its therapeutic potential. However, comprehensive information on its bioavailability is still lacking. The key objective of this study was to investigate the impact of specific formulations on CBD delivery to the site of action and, in particular, the brain of experimental animals. As brain tissue is an extremely complex matrix, a highly sensitive method employing liquid chromatography–tandem mass spectrometry (LC-MS/MS) had to be implemented. To make it applicable for multiple analytes, the method was validated for 17 other phytocannabinoids and selected metabolites. Using this method, a pharmacokinetic study was conducted on 200 brain samples collected from rats that had been administered various CBD formulations (carriers) via oral gavage. The peak concentration in brain occurred within 1–2 h; notably, the highest was reached with carriers containing triacylglycerols with the shortest fatty acid chains (caprylic/capric). In addition to the parent compound, 7-hydroxy-cannabidiol and 7-carboxy-cannabidiol were detected, confirming rapid post-administration metabolism. Overall, this research enhances understanding of CBD distribution in the brain and underscores the impact of specific formulations on its bioavailability, offering insights into optimizing CBD-based therapies to be both effective and ‘patient-friendly’. Medium chain triglycerides (MCTs) are regarded as an important ingredient for functional foods and nutraceuticals. Cinnamomum camphora seed kernel oil (CCSKO) contains more than 95% medium chain fatty acids (MCFAs), which is a significantly higher level than palm kernel oil (62%) and coconut oil (55%). However, the safety assessment of CCSKO, as the only natural MCT oil rich in capric acid and lauric acid found so far in the world, has not been fully verified. The study aimed to investigate the 90-day sub-chronic oral toxicity and teratogenicity of CCSKO. In the sub-chronic oral toxicity study, no clinically significant adverse events occurred in male or female Sprague–Dawley (SD) rats with CCSKO daily administration for 13 weeks. Moreover, there were no dose–response relationships between CCSKO and body-weight gain, food intake and food utilization in male or female SD rats. No significant differences (p > 0.05) were found in the hematological properties or organ weights between the male and female SD rats. In the teratogenicity test, no toxicological signs were observed in either Wister pregnant rats or fetuses. The no-observed-adverse-effect level of CCSKO was determined to be more than 4 mL/kg body weight. These results suggested that CCSKO may be an excellent edible oil with high oral safety. Full article

Background: Compared to soybean oil intravenous fat emulsion (SO-IFE), use of mixed-oil IFE (MO-IFE) is associated with reduced rates of catheter-related bloodstream infections caused by coagulase-negative Staphylococcus species (CoNS) in pediatric patients receiving parenteral nutrition. Methods: Using an in vitro biofilm model, this study aimed to assess the impact of IFEs on biofilm formation among Staphylococcus species. S. aureus, S. capitis, S. epidermidis, S. haemolyticus, S. hominis, and S. lugdunensis were cultivated as biofilms in media supplemented with SO-IFE, MO-IFE, or fish oil IFE (IFE). Biomass was quantified by the crystal violet method, and follow-up planktonic growth assays assessed antimicrobial effects of IFEs. Results: Compared to SO-IFE, MO-IFE and FO-IFE significantly inhibited biofilm formation of S. aureus but did not impact planktonic growth. Contrary to clinical data, CoNS biofilm formation was not impacted by any of the IFEs tested. S. aureus biofilm inhibition in IFEs was further investigated by comparing differences following growth in SO-IFE supplemented with capric acid, docosahexaenoic acid (DHA), or eicosapenaenoic acid (EPA) to concentrations matching those of MO-IFE. Capric acid supplementation was associated with significant reduction in biofilm formation compared to SO-IFE alone. However, this was attributed to a bactericidal effect based on follow-up planktonic growth assays. Conclusions: These results suggest that biofilm formation in S. aureus is variably impacted by fatty acid composition in clinically relevant IFEs, with capric acid exhibiting bactericidal activity against tested isolates. Background/Objectives: Diabetic mouth ulcers are a pathological condition of the oral mucosa leading to increases in susceptibility to infection and prolonged wound healing time. Still, there is a lack of natural formulations for treating this condition. Our principal objective was to formulate solid lipid nanoparticles (SLNs) that contained Althaea officinalis (marshmallow) (M.) extract with clove oil (CO.), subsequently integrated into a collagen sponge for enhancing stability, solubility, sustained release, antimicrobial efficacy, and healing power when targeting diabetic oral ulcers. Methods: A factorial design of 34 trials was established to evaluate the influence of lipid concentration (A), SAA concentration (B), lipid type (C), and SAA type (D). The optimized M-CO-SLNs was selected using Design Expert®, the based Poly dispersibility index (Y2), zeta potential (MV) (Y3), and encapsulation efficiency (%) (Y4). The optimized SLNs were integrated into a collagen sponge matrix and tested for their antibacterial and antifungal efficacy against Pseudomonas aeruginosa, Escherichia coli, and Candida albicans, respectively. Moreover, they were tested for their wound healing power in a diabetic mouth ulcer model. Results: The optimized formula (Run 16: 5% lipid concentration, 4% SAA concentration, capric acid) demonstrated P.S (110 ± 0.76 nm), ZP (−24 ± 0.32 mV), PDI (0.18 ± 0.05), and EE% (90 ± 0.65%.). The optimized M-CO-SLNs formula was incorporated into a cross-linked collagen sponge and showed superior antimicrobial efficacy, an increased swelling ratio, and was effective in an in vivo oral ulcer study, as evidenced by ELISA biomarkers, gene expression analysis, and histological analysis. Conclusions: M-CO-SLNs embedded in collagen sponges is a promising therapeutic formula for clinical application against diabetic mouth ulcers. Full article

The sustainable production of healthy structured lipids (SLs) using oils extracted from agro-industry by-products or non-conventional lipid sources is of utmost importance in the framework of a circular bioeconomy, toward a zero-waste goal. In this study, low-calorie triacylglycerols (TAGs) containing a long-chain (L) fatty acid (FA) at position sn-2 and medium-chain (M) FAs at positions sn-1,3 (MLM type SL) were obtained from virgin cold-pressed milk thistle (51.55% linoleic acid; C18:2), grapeseed (66.62% C18:2), and apricot kernel (68.61% oleic acid; C18:1) oils. Lipase-catalyzed acidolysis with capric acid (C10:0) or interesterification with ethyl caprate (C10 Ethyl) in solvent-free media were performed. In batch reactions, immobilized Rhizomucor miehei lipase (Lipozyme RM) was used as a biocatalyst. For all tested oils, new TAG (SL) yields, varying from 61 to 63%, were obtained after 6 h of interesterification. Maximum new TAG yields were reached after 6, 24, and 30 h of acidolysis with grapeseed (64.7%), milk thistle (56.1%), or apricot kernel (69.7%) oils, respectively. Continuous acidolysis and interesterification of grapeseed oil were implemented in a packed-bed bioreactor, catalyzed by immobilized Thermomyces lanuginosus lipase (Lipozyme TL IM). Throughout 150 h of continuous operation, no lipase deactivation was observed, with average SL yields of 79.2% ± 4.1 by interesterification and 61.5% ± 5.91 by acidolysis. Full article

Serpa cheese, a Portuguese Protected of Denomination Origin (PDO) cheese, known for its unique sensory attributes, is made from the raw milk of native sheep. In this preliminary work, ten samples of Serpa cheese were submitted for a sensory evaluation performed by an expert panel in a sensory laboratory accredited according to ISO 17025 for Serpa cheese parameters, and the panelists classified the cheeses based on texture, taste and odor scores, in accordance with the specifications for the classification of this type of cheese. All cheeses were analyzed by SPME-GC-MS. Following an exploratory unsupervised multivariate analysis, the supervised multivariate analysis by partial least squares—discriminant analysis (PLS-DA), associated the relative percent area of the identified volatiles with the classification of cheeses attributed by the sensory panel. Among the 144 compounds putatively identified, there was a pattern of compound distribution of some of them, such as acetoin, diacetyl, and 2,3-butanediol, leaning toward the cheese samples with high taste and odor scores, while other compounds, such as ethyl caprate, capric acid, and 3-methylindole, were more associated with the cheese samples rated with a low score. Despite the reduced number of samples that may have imposed some restrictions on the conclusions drawn, there was a clear trend in the volatiles’ distribution, allowing us to identify, based on the higher correlation loadings, potential candidates for the Serpa cheese sensory quality. This preliminary study presents, for the first time, an overview of the volatiles that are present in Serpa PDO cheese that may be responsible for the positive or negative sensory evaluation of this PDO cheese. Using fatty acids has generated significant interest in the building sector for improving energy storage in the form of latent heat. In this work, using vacuum impregnation, we analyzed the properties of a capric acid and myristic acid eutectic (83-17%) as a bio-based phase change material in Pinus radiata. The delignification of Pinus radiata samples facilitated the impregnation process, which was carried out using the Kraft pulping method. Morphological, chemical, mechanical, thermal, and acoustic impedance analyses were performed. The results revealed that impregnating PCM in Pinus radiata samples increases the thermal inertia of the impregnated samples, which is comparable to that of delignified samples. Additionally, the analyses showed no significant difference between natural and delignified samples after treatment with PCM. Full article

This study evaluates Natural Deep Eutectic Solvents (NADES) for extracting antioxidant compounds from Portulaca oleracea dried leaves, compared to traditional ethanol extraction. NADES were synthesized using terpenoids (menthol and β-citronellol) and organic acids (lactic and capric acid), characterized by favorable viscosity, density, and pH, ensuring liquid stability at ambient temperature. NADES extraction outperformed ethanol, with NADES 1 yielding the highest bioactive contents: 83.66 Eq GA/mg, 786.55 Eq Q/mg, and 0.78 Eq C/mg versus ethanol’s 58.49 Eq GA/mg, 363.23 Eq Q/mg, and 0.44 Eq C/mg. HPLC-DAD analysis identified higher levels of phenolic acids (caffeic and syringic acid) and flavonoids (rutin and quercetin) in NADES extracts, compounds absent in ethanol. Antioxidant potential, assessed via IC50 values, confirmed superior activity for NADES extracts (NADES 1-Ext: IC50 28.10 ± 1.73 µg/µL) compared to ethanol (IC50 1615.97 ± 5.34 µg/µL), and the Trolox method has confirmed extensively this superiority. Additionally, NADES demonstrated improved antimicrobial effects, varying with microorganisms. Despite their high viscosity potentially limiting extraction efficiency, adjusting temperature offers a promising approach to enhance mass transfer. These findings emphasize NADES as a sustainable alternative for bioactive compound extraction, paving the way for optimizing extraction techniques through viscosity reduction strategies. Full article

Crude glycerol (CG), a major biodiesel production by-product, is the focus of ongoing research to convert it into polyhydroxyalkanoate (PHA). However, few bacterial strains are capable of efficiently achieving this conversion. Here, 10 PHA-producing strains were isolated from various media. Among them, Burkholderia sp. Oh_219 exhibited the highest polyhydroxybutyrate (PHB) production from glycerol and was therefore characterized further. Burkholderia sp. Oh_219 demonstrated significant tolerance to major growth inhibitors in CG and metabolized the fatty acids present as impurities in CG. Furthermore, the Oh_219 strain was genetically engineered using phaCBP-M-CPF4 and phaJPa to enable the fatty acid-based production of poly(3-hydroxybutyrate-co-3-hydroxyhexanoate) (PHBHHx), a component of CG. The resulting strain produced PHBHHx containing 1.0–1.3 mol% of 3HHx from CG. Further supplementation with capric and lauric acids increased the 3HHx molar fraction to 9.7% and 18%, respectively. In a 5 L fermenter, the Oh_219 strain produced 15.3 g/L PHB from 29.6 g/L biomass using a two-stage fermentation system. This is the highest yield reported for PHA production from glycerol by Burkholderia spp. Additionally, PHB produced from CG had a lower melting point than that from pure glycerol and fructose. Taken together, Burkholderia sp. Oh_219 is a promising new candidate strain for producing PHA from CG. Full article

Background/Objectives: Laurocapram (Azone) attracted attention 40 years ago as a compound with the highest skin-penetration-enhancing effect at that time; however, its development was shelved due to strong skin irritation. We had already prepared and tested an ante-enhancer (IL-Azone), an ionic liquid (IL) with a similar structure to Azone, consisting of ε-caprolactam and myristic acid, as an enhancer candidate that maintains the high skin-penetration-enhancing effect of Azone with low skin irritation. In the present study, fatty acids with different carbon numbers (caprylic acid: C8, capric acid: C10, lauric acid: C12, myristic acid: C14, and oleic acid: C18:1) were selected and used with ε-caprolactam to prepare various IL-Azones in the search for a more effective IL-Azone. Methods: Excised porcine skin was pretreated with each IL-Azone to assess the in vitro skin permeability of antipyrine (ANP) as a model penetrant. In addition, 1,3-butanediol was selected for the skin permeation test to confirm whether the effect of IL-Azone was due to fatty acids and if this effect differed depending on the concentration of IL-Azone applied. Results: The results obtained showed that C12 IL-Azone exerted the highest skin-penetration-enhancing effect, which was higher than Azone. On the other hand, many of the IL-Azones tested had a lower skin-penetration-enhancing effect. Conclusions: These results suggest the potential of C12 IL-Azone as a strong and useful penetration enhancer. Full article
