In the context of critical challenges in curcumin-modified polyurethane synthesis—including limited curcumin bioavailability and suboptimal biodegradability/biocompatibility—a novel polyurethane material (Cur-PU) with good mechanical, shape memory, pH-responsive, and biocompatibility was synthesized via a one-pot, two-step synthetic protocol in which HO-PCL-OH served as the soft segment and curcumin was employed as the chain extender. The experimental results demonstrate that with the increase in Cur units, the crystallinity of the Cur-PU material decreases from 32.6% to 5.3% and that the intensities of the diffraction peaks at 2θ = 21.36°, 21.97°, and 23.72° in the XRD pattern gradually diminish. Concomitantly, tensile strength decreased from 35.5 MPa to 19.3 MPa, and Shore A hardness declined from 88 HA to 65 HA. These observations indicate that the sterically hindered benzene ring structure of Cur imposes restrictions on HO-PCL-OH crystallization, leading to lower crystallinity and retarded crystallization kinetics in Cur-PU. As a consequence, the material’s tensile strength and hardness are diminished. Except for the Cur-PU-3 sample, all other variants exhibited exceptional shape-memory functionality, with Rf and Rr exceeding 95%, as determined by three-point bending method. Analogous to pure curcumin solutions, Cur-PU solutions demonstrated pH-responsive chromatic transitions: upon addition of hydroxide ion (OH) solutions at increasing concentrations, the solutions shifted from yellow-green to dark green and finally to orange-yellow, enabling sensitive pH detection across alkaline gradients. Hydrolytic degradation studies conducted over 15 weeks in air, UPW, and pH 6.0/8.0 phosphate buffer solutions revealed mass loss <2% for Cur-PU films. Surface morphological analysis showed progressive etching with the formation of micro-to-nano-scale pores, indicative of a surface-erosion degradation mechanism consistent with pure PCL. Biocompatibility assessments via L929 mouse fibroblast co-culture experiments demonstrated ≥90% cell viability after 72 h, while relative red blood cell hemolysis rates remained below 5%. Collectively, these findings establish Cur-PU as a biocompatible material with tunable mechanical properties, and pH responsiveness, underscoring its translational potential for biomedical applications such as drug delivery systems and tissue engineering scaffolds. This study evaluated the antibacterial activity and residual functional compounds of blood orange (Citrus sinensis L. Osbeck) juice by-product extract (BJBE). The effects of dietary BJBE on growth performance, digestive enzyme activity, antioxidant status, immune parameters, and disease resistance against Vibrio harveyi were examined in juvenile black rockfish (Sebastes schlegelii). In total, 630 juvenile rockfish were randomly assigned to 21 rectangular tanks (50 L) for a feeding trial, with 30 fish per tank in triplicate. Seven isonitrogenous and isolipidic experimental diets were formulated with BJBE at 0 (control, BJBE0), 0.1 (BJBE0.1), 0.2 (BJBE0.2), 0.3 (BJBE0.3), 0.5 (BJBE0.5), 0.7 (BJBE0.7), and 1.0 (BJBE1) g kg−1. A disk diffusion assay confirmed BJBE’s strong antibacterial efficacy against V. harveyi. After an 8-week feeding trial, fish fed BJBE0.7 and BJBE1 exhibited significantly a greater final weight, weight gain, and specific growth rate compared with those fed BJBE0. Feed efficiency was significantly higher in fish fed BJBE0.7 than in those fed BJBE0. The protein efficiency ratio was significantly higher in fish fed BJBE0.3, BJBE0.5, BJBE0.7, and BJBE1 relative to those fed BJBE0. Intestinal amylase activity was significantly higher in fish fed BJBE0.7 and BJBE1 compared with those fed BJBE0, and trypsin activity was significantly higher in BJBE0.7-fed fish than in BJBE0-fed fish. In comparison to the BJBE0 diet, the plasma superoxide dismutase, catalase, and glutathione levels of fish fed BJBE0.7 and BJBE1 diets were significantly higher. Lysozyme activity and immunoglobulin M level in fish fed BJBE0.7 and BJBE1 were significantly higher than that in fish fed BJBE0. After a challenge with V. harveyi, disease resistance was significantly higher in fish fed BJBE0.5, BJBE0.7, and BJBE1 compared with those fed BJBE0. Overall, 0.7–1.0 g kg−1 is proposed as the optimal dietary BJBE inclusion level for enhancing growth performance, digestive enzyme activity, antioxidant status, immune parameters, and disease resistance against V. harveyi infection in juvenile black rockfish. Impact of Ultrasound Pretreatment and Temperature on Drying Kinetics and Quality Characteristics of Blood Orange Slices: Comparison with Different Drying Methods

This study aimed to investigate the impact of ultrasonic pretreatment vacuum drying (UAVD) and temperature on drying kinetics and qualitative attributes of blood oranges in comparison to several drying methods: hot air drying (HAD), vacuum drying (VD), and freeze drying (FD). The drying kinetics and modeling, total phenolic content (TPC), antioxidant capability (assessed using DPPH and ABTS tests), individual phenolic profiles, vitamin C concentration, and color factors were meticulously examined. Drying times were recorded as 22.5 h, 12.5 h, and 9 h for HAD; 11.5 h, 9.5 h, and 8.5 h for VD; and 10 h, 8.5 h, and 7.5 h for UAVD at 50, 60, and 70 °C, respectively. The HAD, VD, and UAVD procedures were conducted at 50, 60, and 70 °C, resulting in reduced drying periods with increasing temperature. The integration of ultrasound markedly lowered drying durations. Eleven thin-layer drying models were utilized to recreate the drying process precisely. The Deff values of the HAD, VD, and UAVD dried samples varied from 9.08 × 10−6 to 2.82 × 10−5 m2/s, from 2.60 × 10−5 to 2.96 × 10−5 m2/s, and from 2.20 × 10−5 to 2.99 × 10−5, respectively. Among the desiccated blood orange slices, the greatest total phenolic content (TPC) was observed in freeze-dried samples (131.27 mg GAE/100 g), followed by those dried using ultrasonic-assisted vacuum drying (UAVD) at 50 °C (128.77 mg GAE/g DM). Dried blood orange slices had a vitamin C content of 29.79 to 49.01 mg/100. The drying process substantially impacted the color parameters L*, a*, and b*. These findings highlight the efficacy of ultrasound-assisted drying in decreasing drying duration while improving the retention of bioactive components in blood orange slices. Full article

Blood oranges grown in subtropical and tropical regions have low anthocyanin levels and a pale internal color at the point of commercial maturity. Since blood oranges are cold-dependent and can enhance anthocyanin during postharvest storage, the effects of various non-chilling storage temperatures (10, 15, and 20 °C) on ‘Moro’ blood orange fruit quality and biochemical changes over a period of up to 42 days were investigated for total anthocyanin concentration (TAC), total phenolic content (TPC), total antioxidant activity (TAA), juice attributes, and physical qualities. Fruit weight and firmness losses increased during storage, with the lowest losses occurring at 10 °C. Titratable acidity (TA) decreased, with the lowest values recorded at 20 °C. Total soluble solids (TSSs) and the TSS/TA ratio increased, with the highest values observed at 20 °C. The sucrose content showed slight changes, while glucose and fructose levels increased during storage, with the highest concentrations of glucose and fructose noted at 20 °C. TAC, TPC, and TAA significantly increased during storage, with the highest values recorded at 10 °C, suggesting upregulation of the fruit antioxidant system and associated bioactive components in response to the lower temperature. Throughout the storage period, peel color parameters, such as L*, b*, C*, and h°, decreased at all temperatures, whereas a*, citrus color index (CCI), and total color difference (ΔE) increased. This study concludes that a storage temperature of 10 °C proved effective in enhancing TAC, TPC, and TAA during storage, as well as in maintaining the other physicochemical attributes. Full article

Blood orange peel flour (BO-pf)—a by-product of the citrus supply chain—still contains bioactive molecules with known health benefits, such as antiradical scavenging activity or an antiproliferative activity regarding tumors. In vitro studies have demonstrated that orange polyphenols showed potential involvement in necroptosis. In addition to previous research, we tested BO-pf on two colorectal cancer cell lines. Using HT29 and Caco2 cells, our experiments confirmed the regulation of inflammasome expression. They provided valuable insights into how BO-pf influences the cancer cell features (i.e., viability, proliferation, and pro- and anti-inflammatory activity). Notably, BO-pf extract is a rich source of polyphenolic compounds with antioxidant properties. Western blot and real-time PCR analyses showed that treatment with BO-pf extract demonstrated beneficial effects by influencing the expression of both pro-inflammatory cytokines (IL-1β, IL-6) through the modulation of the TLR4/NF-kB/NLRP3 inflammasome signaling. Moreover, the results of this study demonstrate that BO-pf extracts can enhance the expression of anti-inflammatory cytokines, such as IL-10 and TGFβ, suggesting that BO-pf extracts may represent a promising functional ingredient to counteract the intestinal inflammatory responses involved in IBD. The distribution of oral pathogens is influenced by genetic background, diet, socioeconomic status, and racial factors. This study aimed to assess the distribution and characteristics of oral pathogens based on blood glucose levels in a South Korean population. This cross-sectional, retrospective study included subjects from 17 health promotion centers in 13 South Korean cities between November 2021 and December 2022. Real-time multiplex PCR was used to detect 10 periodontitis-related pathogens, 6 dental caries-related pathogens, and 1 dental caries-protective bacterium. The most prevalent periodontitis-related pathogens were Parvimonas micra (97.6%), Porphyromonas endodontalis (96.8%), and Treponema socranskii (95.0%). Among dental caries-related pathogens, Streptococcus sanguinis and Veillonella parvula were found in all subjects. The prevalence of periodontitis-related pathogens was higher in males, while pathogens related to periodontitis and dental caries were more prevalent in older individuals. In the diabetes group, Aggregatibacter actinomycetemcomitans, red and orange complexes, and Streptococcus mutans were more prevalent. The relative amount of S. sanguinis was lower, while V. parvula was higher in individuals with diabetes mellitus. The prevalence and composition of oral pathogens vary by sex, age, and blood glucose levels. Diabetic individuals showed a pathogenic community structure linked to increased risks of periodontitis and dental caries. Full article

Background/Objectives: Minimal hepatic encephalopathy (MHE) is a clinical condition characterized by neurological impairments, including brain inflammation, arising from the accumulation of toxic metabolites associated with liver dysfunction and leaky gut. This study investigated the pharmacological activity of a new phytocomplex extracted from red orange by-products (AL0042) using hydrodynamic cavitation and consisting of a mixture of pectin, polyphenols, and essential oils. Methods: Preliminary in vitro studies evaluated the impact on the epithelial integrity (TEER) of enterocytes challenged by a pro-inflammatory cocktail. The effect of AL0042 was then evaluated in a model of thioacetamide (TAA)-treated mice that mimics MHE. A group of 8–10-week-old male C57BL/6 mice was intraperitoneally injected with TAA to establish the MHE model. The intervention group received TAA along with AL0042 (20 mg/kg, administered orally once daily for 7 days). At the end of the treatment, the rotarod test was conducted to evaluate motor ability, along with the evaluation of blood biochemical, liver, and brain parameters. Results: In vitro, AL0042 (250 μg/mL) partially recovered the TEER values, although anti-inflammatory mechanisms played a negligible role. In vivo, compared with the control group, the test group showed significant behavioral differences, together with alterations in plasma ammonia, serum TNF-α, ALT, AST, corticosterone levels, and SOD activity. Moreover, histological data confirmed the anti-inflammatory effect at liver and brain level. Conclusions: AL0042 treatment revealed a significant therapeutic effect on the TAA-induced MHE mouse model, curbing oxidative stress and peripheral and central inflammation, thus suggesting that its pharmacological activity deserves to be further investigated in clinical studies. The choice of time/temperature combination is critical for ensuring microbiological stability and retaining the characteristic taste of dried blood orange slices. The aim of this study was to investigate the capability of hot air-drying technique to maintain the quality characteristic of dried blood orange slices stored in modified atmosphere packaging (MAP). Hot air-drying at 70 °C for 12 h preserved shrinkage without altering the longitudinal diameter, though thickness was significantly reduced, especially in samples with passive MAP. Increased hardness and masticability were noted due to water removal, with active MAP maintaining high hardness and colour integrity up to 100 days of storage (D100). Sensory analysis revealed differences in colour intensity and flavour between active and passive MAP-stored slices. Headspace solid-phase microextraction (HS-SPME) chromatography identified key chemical compounds contributing to aroma and flavour, highlighting the complex interplay between temperature, storage conditions, and volatile organic compounds production. The study demonstrates that drying combined with MAP storage enhances organoleptic qualities and nutritional value, offering a method to produce a healthy, tasty, and visually appealing snack. Background/Objectives: Mandarin orange peel (MOP) is recognized for its traditional medicinal properties due to its high flavonoid content. This study aimed to analyze MOP harvested in Japan for specific bioactive compounds and to explore its health applications in dogs, including effects on gut microbiota and cognitive symptoms. Methods: Flavonoid content (particularly hesperidin and nobiletin) of MOP was measured. High-flavonoid MOP was then incorporated into gelatin cubes. In the time–course blood concentration study, experimental beagle dogs received a single oral dose (4 g). For intestinal microbiota analysis and blood biochemical tests, beagle dogs were administered 1 g twice daily. For the Demonstration Test, older dogs (a 14-year-old female Shiba Inu, a 14-year-old female Miniature Dachshund, and a 19-year-old male Miniature Dachshund) were administered 1 g twice daily. Analysis included microbiota profiling via 16S rDNA sequencing and observational assessment of cognitive indicators in a pilot study involving senior dogs with Alzheimer’s disease. Results: The MOP powder contained 9.3% hesperidin in early-ripening varieties (October harvest) and 6.9% in ripe varieties (December harvest). Nobiletin content was 41 mg/100 g (0.041%) and 35 mg/100 g (0.035%) for the early and late harvests, respectively. Administration of MOP-enriched gelatin cubes reduced Fusobacteriaceae and increased Eggerthellaceae. Cognitive symptoms like howling and counterclockwise turning showed improvement in senior dogs (n = 3). Conclusions: This study provides preliminary support for the potential health benefits of MOP in canine dietary applications, particularly for gut health and cognitive function. Improvement in cognitive symptoms may be due to the anxiolytic effects of mandarin peel. Full article

Background: This experiment evaluated whether the combination of essential oils (thyme, rosemary, orange) with extracts of Cinnamomum and Quillaja saponaria (enriched with tocopherols) in steers’ diet has positive effects on the health, digestibility, growth, oxidative stress, and fatty acid profile of meat. Methods: The experiment included 24 Holstein steers, divided into two groups, randomly divided (treated = phytogenic mixture (n = 12); control = monensin (n = 12)), undergoing a growth phase of 120 days and 90 days of finishing in individual pens, with a monthly collection of blood, rumen fluid, feces, and feed. Results: In the finishing phase (days 150 to 210), there was a tendency (p = 0.07) toward more significant average daily weight gain when cattle consumed a phytogenic mixture than monensin. The use of the phytogenic product showed greater feed efficiency in the period from 150 to 240 days (p < 0.05), with higher levels of short-chain fatty acids (SCFAs), without differences in the composition of these SCFAs between the control and treated groups, with a reduction in apparent digestibility in the treated group. In blood parameters, the treated group showed higher erythrocyte counts (p < 0.05) without changes in leukocytes, lymphocytes, granulocytes, monocytes, and platelets. The phytogenic treatment demonstrated higher total protein and globulin levels, with increased glucose levels on days 30 and 210 (p < 0.05). Ceruloplasmin and haptoglobin levels increased in the treated group without changes in ferritin, transferrin, C-reactive protein, IgA, and heavy-chain immunoglobulins (p < 0.05). A reduction in oxidative stress was observed using the phytogenic agent, with a marked decrease in the levels of reactive oxygen species (ROS) and carbonyl proteins, with a tendency to increase glutathione S-transferase (p < 0.05). In meat, the phytogenic reduced saturated fatty acids with increased polyunsaturated fatty acids and increased the omega 6/omega 3 ratio. Lower levels of thiobarbituric acid reactive substances and ROS were detected in the meat combined with a higher concentration of total thiols in the treated group than in the control group. Conclusions: The use of the phytogenic in steers’ diets has shown to be a viable candidate for replacing monensin, presenting equal or superior performance results with systemic modulating effects on oxidative stress, meat quality, and serum and biochemical parameters that contribute to more substantial health and efficiency. This study evaluated bioactive compounds in blood orange (Citrus sinensis (L.) Osbeck) peel (BOP) as dietary additives. An 8-week feeding trial was conducted to investigate the effects of dietary supplementation on the growth performance, body composition, digestive enzyme activity, antioxidant capacity, and immune response of juvenile black rockfish. A total of 1260 juvenile rockfish (1.4 ± 0.01 g) were randomly distributed into seven treatment groups, each with 50 fish per circular tank. The groups were fed seven different diets containing graded levels of 0 (control, BOP0), 1 (BOP1), 2 (BOP2), 3 (BOP3), 5 (BOP5), 7 (BOP7), and 10 (BOP10) g kg−1, respectively. The BOP10 diet significantly enhanced the final weight, weight gain, specific growth rate, protein efficiency ratio, and protein retention in fish. The BOP treatments notably affected the fishes’ whole-body crude protein and lipid contents. Plasma total cholesterol levels of fish fed the BOP0 and BOP1 diets were significantly higher than those fed the BOP7 and BOP10 diets. The activities of trypsin and lipase were significantly affected by dietary BOP levels. The antioxidant enzyme activity in the plasma of fish fed the BOP10 diet was significantly higher than those fed the BOP0 diet. The lysozyme activity and levels of immunoglobulin M and G in fish fed the BOP0 diet were significantly lower than those in fish fed the BOP10 diet. In conclusion, dietary supplementation of BOP at 10 g kg−1 improved the growth performance and overall health of juvenile black rockfish. Full article

Blood oranges are increasingly cultivated worldwide as consumers become more aware of the health benefits of their nutraceutical properties and natural antioxidants, specifically polyphenols and anthocyanins. The amounts of these compounds in the fruit mostly depend on the cultivar, rootstock, maturity stage, and environmental conditions. This work focused on the study of the qualitative features of numerous blood orange cultivars grown in three different environments in Spain and Italy. The aim of the work was to investigate the accumulation of primary and secondary metabolites, including bioactive compounds, and to characterize fruit qualitative traits at the time of harvest. Simple sugars were identified and quantified by liquid chromatography and organic acids, polyphenols, and flavonoids by spectrophotometric analysis. The antioxidant potential of the juice was assessed by ABTS, DPPH, and FRAP assays. Cultivation area affected juice color, with Moro and T. Ippolito being the varieties with the highest pigmentation. The cultivation area also determined the pattern of primary and secondary metabolite accumulation in the Tarocco lines. Furthermore, the antioxidant potential was influenced by the diverse environments. Principal Component Analysis highlighted three clusters, two overlapping clusters for the varieties grown in the two Spanish plots and a third clearly separated cluster for the genotypes grown in Italy. This study provides novel knowledge on primary and secondary metabolite accumulation in blood oranges, elucidating the role of genotype and environmental conditions on fruit quality. Full article

Background: Consumption of flavonoid-rich orange juice has been shown to reduce adiposity and liver steatosis in murine models of diet-induced obesity. However, little is known about the effects of whole orange intake, independent of body weight changes, on liver function and steatosis in individuals with metabolic dysfunction-associated steatotic liver disease (MASLD). The goal is to understand the direct impact of orange consumption on metabolic health. Methods: Sixty-two men and women aged 30–65 with MASLD (Controlled Attenuation Parameter, (CAP) > 275 dB/m) were randomly assigned to consume either 400 g of whole oranges or non-citrus fruits daily for 4 weeks. Baseline evaluations included medical assessments, blood tests, and body composition. Liver health was assessed using transient elastography (FibroScan®) for steatosis and fibrosis, conducted by blinded personnel. This clinical trial was registered at ClinicalTrials.gov (NCT05558592). Results: After 4 weeks of orange supplementation, liver steatosis decreased in the treatment group, with 70.9% showing steatosis compared to 100% in controls (p < 0.004), indicating a 30% reduction in liver disease prevalence. There were no significant changes in fibrosis or plasma liver enzymes, though plasma gamma glutaril transferase (GGT) levels decreased significantly. Body weight, waist circumference, body composition, lipid profile, fasting glucose, insulin, and C-reactive protein levels remained unchanged. Dietary analysis revealed no change in caloric intake, but vitamins C, A, thiamine, and riboflavin increased in the orange group. Conclusions: Our findings suggest that phytochemical-rich foods, especially whole fruits like oranges, may enhance liver function as an adjunct treatment for MASLD. The notable reduction in liver steatosis prevalence occurred independently of body weight changes. Further studies are needed to investigate the long-term effects of orange supplementation on steatosis and fibrosis progression and to identify the specific bioactive compounds and mechanisms involved. In modern orchard systems, the tree canopy is designed to ensure homogeneity in fruit quality. However, even in those crops there are some variables that affect the fruit maturation process and fruit quality properties. The aim of this work was to determine if canopy layer (upper vs. lower), fruit shoot position (grouped vs. individual) and orientation (west vs. east) affect fruit quality attributes of ‘Sanguinelli’ blood oranges. Thus, different quality traits, such as weight, internal colour (IC), external colour (EC), total soluble solids (TSS), titratable acidity (TA) and maturity index (MI) were determined for this purpose. Results showed that fruit weight, internal colour, TA and MI were influenced by the number of fruits per shoot. In this sense, the highest values of weight, IC and MI were found in the grouped fruits, while the highest values in TA were in the individual fruits. Regarding the EC and TSS, they were strongly related to the canopy layer, since the highest values were found in fruit located at the upper parts of the canopy. On the contrary, the orientation did not have a significant effect on fruit quality properties. Therefore, consistent differences in quality traits of ‘Sanguinelli’ blood oranges fruits were observed depending on canopy layer and number of fruits per shoot. Full article

The agri-food processing industry predominantly relies on fossil fuels, contributing significantly to greenhouse gas emissions and extensive water use. Climate change requires a conversion of food processing technologies towards sustainability. Our research focuses on testing and validating pulsed electric field (PEF) technology as a mild processing method for stabilizing freshly squeezed, not from concentrate, blood orange juice. Experiments were carried out on a continuous pilot plant endowed with a patented treatment chamber that ensures a constant flow of product without “hot spots” for a homogeneous treatment. Once the operative conditions of the process in terms of energy density were optimized, PEF-treated blood orange juice was tested in order to evaluate the effects on physico-chemical parameters (total soluble solids, total acidity, pH, CIE L*, a*, b*, C*, h color indices, cloud, bioactive compounds (ascorbic acid, total anthocyanins, total and individual flavanones), antioxidant activity (ORAC units, total phenolic content), microbial communities (aerobic mesophilic viable count, yeasts and molds, acid-tolerant microorganisms), residual enzymatic pectinmethylesterase activity, and sensory attributes (flavor, off-flavor, off-odor, color, intensity of odor, acidity, sweetness, bitter, freshness, cooked flavor). Then, in order to simulate the commercial refrigerated distribution of PEF-treated blood orange juice and define its refrigerated shelf-life, the PEF-treated freshly squeezed, not from concentrate, blood orange juice was subjected to physico-chemical, antioxidant, microbiological, and sensory evaluations over twenty days of refrigerated storage at 4 ± 1 °C. The PEF treatment effectively ensured excellent microbial inactivation and enhanced the nutritional and health characteristics of the juice, thereby extending its shelf-life. This study demonstrates the significant potential of the PEF treatment to produce blood orange juice suitable for a new retail segment—freshly squeezed juices with superior health quality, fresh-like characteristics, and extended refrigerated shelf-life.
