The Effects of Inulin on the Growth, Oxidative Stress, and Immune Function of Weaned Kids

Goats are important producers of meat, milk, hair, and leather. Early-weaned kids may encounter issues such as weaning stress and stress from high-density farming, which can hinder their growth and development. Therefore, exploring feed additives capable of alleviating stress in kids to enhance their growth performance is of particular importance. This experiment aims to investigate the effects of inulin on the growth performance, immune function, and intestinal health of weaned kids. Thirty healthy 60-day-old Leizhou black goat weaned kids with similar body weights (9.00 ± 0.05 kg) were selected and evenly divided into five groups by weight, with six kids in each group. The control group was fed the basal diet, while the AM HCl group received 0.05‰ aureomycin hydrochloride. The inulin groups were supplemented with 0.1%, 0.3%, and 0.5% inulin, respectively. By comparison, the results of the 0.3% inulin group in this experiment were most consistent with those of the AM HCl group, and the final weight of the kids was the highest. The 0.3% and 0.5% inulin groups had significantly enhanced immune-related indicator (such as sIgA, IgA, and IgG) concentrations and antioxidant activities (p < 0.05) in kids. Inulin significantly increased cecal total volatile fatty acids (TVFA) and the activities of jejunal α-amylase, chymotrypsin, and trypsin (p < 0.05); the activities of intestinal antioxidant enzymes, including GSH-PX, SOD, and T-AOC, were significantly increased (p < 0.05); and the intestinal anti-inflammatory factor IL-10 was significantly elevated (p < 0.05). Inulin significantly increased the mRNA expression levels of the tight junction protein genes TJP1 (ZO-1) and OCLN in the jejunum (p < 0.05). It also significantly upregulated the mRNA expression levels of anti-inflammatory factors TGF-β1 and IL-10 while significantly reducing the mRNA level of pro-inflammatory factor IL-6 in the jejunum (p < 0.05). Therefore, inulin enhanced the growth performance and antioxidant capacity of weaned kids, improved the body’s immune response, and reduced inflammatory responses. Specifically, when comparing the growth status, antioxidant enzyme activity, and tight junction protein mRNA level among the groups of kids, the 0.3% inulin supplementation yielded the best results in this experiment. The influence of bakery waste (BAWA) in the diet of black goat kids on nutrient intake, health, growth potential, and carcass features was assessed. Three treatment diets, consisting of 0 BAWA (CON), 50 g/kg BAWA (BAWA50), and 100 BAWA (BAWA100) of dietary dry matter (DM), were administered to 27 male kids (initial body weight = 17.43 ± kg; age = 105 ± 3.5 days) at random (9/treatment). Bakery waste was included in diets BAWA50 and BAWA100, replacing some of the barley grain. The study lasted for 70 days (7 days used for adaptation and 63 days used for data collection). Kids in the BAWA100 group consumed considerably more (p < 0.01) DM, crude protein (CP), and metabolizable energy than those in the BAWA50 and CON groups. However, intakes of neutral detergent fiber (NDF), acid detergent fiber (ADF), and ether extract (EE) were similar (p ≥ 0.08) among the treatment diets. The kids’ initial and final body weights, total growth, and average daily weight were not substantially impacted (p ≥ 0.13) by the addition of BAWA. All diets had similar feed efficiency when it came to converting feed into body weight increases (p ≥ 0.11). Nevertheless, the BAWA diet yielded a considerably lower cost gain (p = 0.04) than the CON diet. The three treatment groups had no differences (p ≥ 0.15) in DM, ADF, NDF, and EE digestibility. However, compared to the CON group, the digestibility of CP tended to improve (p = 0.07) in the BAWA100 and BAWA50 groups. The three experimental groups’ nitrogen (N) intakes did not differ significantly from one another (p > 0.33). Additionally, the three diets’ N retention percentage and grams/d were equivalent (p > 0.33). Several carcass metrics, such as dressing percentage, hot and cold carcass weights, fasting live weight, and non-carcass components, were not significantly affected (p > 0.23) by the addition of BAWA. Additionally, no differences in carcass cut weights were found. The inclusion of BAWA100 and BAWA50 increased loin cut weight (p = 0.043) and total lean muscle weight (p = 0.001) compared to the CON group. Other components, such as intermuscular fat, subcutaneous fat, total fat, total bone, meat-to-bone ratio, and meat-to-fat ratio, were similar among the three diet groups. Concerning carcass linear dimension measurements, only rib fat depth was greater (p = 0.008) in kids fed the BAWA100 diet compared to the BAWA50 and CON diets. No significant variation was observed among the treatments concerning various physicochemical properties of the longissimus dorsi muscle in the black goat kids. All measured serum metabolites and hematological parameters were not affected by the incorporation of BAWA into the diet. In summary, the inclusion of 50 and 100 g/kg BAWA in the diet had comparable effects on the black goat kids’ feed intake, digestibility, growth performance, carcass characteristics, meat quality, and overall health. Importantly, feed costs can be reduced by using up to 100 g/kg of BAWA on a DM basis. Full article

Lüliang black goat (LBG) is a unique livestock genetic resource of great significance for the local agriculture and economic development of Shanxi, China. However, the kidding rate of LBG is relatively low, which limits efforts to improve the reproductive performance and economic benefits of this breed. Therefore, improving the kidding characteristics of LBG is crucial for increasing its economic benefits. In this study, 20 LBG does were selected for whole-genome resequencing and divided into two groups: 10 in the T group (does with three consecutive kiddings of twin kids) and 10 in the S group (does with three consecutive kiddings of single kids). Based on whole-genome resequencing data, this study comprehensively assessed the population structure and genetic diversity of LBG and explored the related genes that affect reproductive performance. Three selection signal analysis methods—Fst (Fixation Index), π (nucleotide diversity), and XP-CLR (Cross Population Composite Likelihood Ratio)—were applied to screen a total of 838 genes, and enrichment analysis was performed to identify genes closely related to the reproductive performance of LBG, including ENPP3, APC, and GLI2. A generalized linear model was used to conduct a correlation analysis between non-synonymous mutations in the three genes and the number of kids produced. Two loci that were significantly correlated with kidding number were identified (p < 0.05): GLI2 g.63400363 C>T and GLI2 g.63417538 C>T. In general, the LBG population has high genetic diversity and good prospects for genetic improvement. The findings revealed that mining high-fecundity selection characteristics provides a basis for research on goat reproductive mechanisms. Full article

Natural selection and artificial breeding are crucial methods for developing new animal groups. The Baiyu black goats and Chuanzhong black goats are indigenous goat breeds from distinct ecological regions in Sichuan Province, with dramatically different growth and reproductivity. This study aimed to systematically elucidate the differences in production performance and genetic traits between Baiyu black goats and Chuanzhong black goats. We quantified growth and reproductive attributes for both breeds. Furthermore, we conducted a comprehensive analysis of genetic diversity, population structure, and selection signatures using whole-genome resequencing data. This dataset included 30 individuals from the Baiyu black goat breed, 41 from the Chuanzhong black goat breed, and an additional 59 individuals representing Chengdu grey goats, Tibetan cashmere goats, and Jianchang black goats, totaling 130 individuals across five goat breeds. The comparative analysis of production performance revealed that the weight and body size of Chuanzhong black goats were significantly higher than those of Baiyu black goats (p < 0.01). At the same time, the average kidding rate and kid-weaning survival rate of Chuanzhong black goats were also notably superior to those of Baiyu black goats (p < 0.01). The Baiyu black goats exhibited a more abundant genetic diversity and distinct genetic differences compared to the Chuanzhong black goat, according to an analysis grounded on genomic variation. The Baiyu black goats are more closely related to Tibetan cashmere goats, whereas Chuanzhong black goats share a closer genetic relationship with Chengdu grey goats. Additionally, we employed the π, Fst, and XP-EHH methodologies to identify genes related to immunity (TRIM10, TRIM15, TRIM26, and TRIM5), neurodevelopment (FOXD4L1, PCDHB14, PCDHB4, PCDHB5, PCDHB6, and PCDHB7), reproduction (BTNL2 and GABBR1), body size (NCAPG, IBSP, and MKNK1), and meat quality traits (SUCLG2 and PGM5). These results provide a theoretical basis for further resource conservation and breeding improvement of the Baiyu black goat and Chuanzhong black goat. Full article

The present study aimed to elucidate the effect of different levels of dietary protein during late pregnancy on the performance of Black Bengal does and their kids. Twelve does were divided into three groups, with four in each, and three diets, i.e., high protein (18% CP), medium protein (14% CP), and low protein (10% CP) were supplied for 50 days, commencing from 100 days post-coitum to parturition. During the first 100 days of pregnancy, uniform rations with similar ingredients were provided to fulfill the nutrient requirements depending on the live weight of does. All three diets were isocaloric (10.0 MJ/kg DM). Data were subjected to one-way ANOVA, and the significance of the difference among means was determined by Duncan’s Multiple Range Test (DMRT). The main effects of diet and sex, as well as their interaction, were analyzed by two-way ANOVA by using the GLM procedure. The relative expression values of qPCR were calculated by using the 2−ΔΔCt analysis method. Live weight gain was significantly (p < 0.05) higher in high-protein-fed dams than other groups during the experimental period. The milk yield of does was significantly (p < 0.05) higher in high-protein-fed goats than in the low-protein group. The lactation length of does was significantly (p < 0.05) higher in the high- and medium-protein-fed does than in the low-protein-fed does. The duration of post-partum anestrus of does was significantly (p < 0.05) higher in the low-protein-fed dams than in the high-protein group. The birth weight of kids tended to be higher in the high-protein group but did not differ significantly among the treatment groups. In male kids, weaning weight, final weight, live weight gain, and average daily gain were significantly (p < 0.05) higher than in female kids. Weaning weight was higher (p < 0.05) in kids of the high-protein-fed does than the low-protein group. Final weight and live weight gain were significantly (p < 0.05) higher in kids of the high-protein-fed does than in the low-protein-fed group. On the other hand, average daily gain was significantly (p < 0.05) higher in kids of the high- and medium-protein-fed does than the low-protein group. The average body length and wither height of kids at the 32nd week was significantly (p < 0.05) higher in kids of high-protein-fed does than those of the low-protein-fed group. The average heart girth of kids at the 32nd week was significantly (p < 0.05) higher in kids of high-protein-fed does than the medium- and low-protein groups. The survival rate of kids was higher in the medium- and high-protein-fed does than in low-protein group. Hot carcass weight and ether extract content of meat were significantly (p < 0.05) higher in the high-protein group than in the other groups. The dressing percentage was significantly (p < 0.05) higher in the kids of high-protein-fed does than low-protein-fed goats. The expression of the H-FABP gene was significantly (p < 0.05) higher in kids of high-protein-fed does than those of the medium- and low-protein groups. In conclusion, maternal dietary protein levels positively influences the production performance of Black Bengal does and their kids. Full article

Korean Native Black Goats deliver mainly during the cold season. However, in winter, there is a high risk of stunted growth and mortality for their newborns. Therefore, we conducted this study to develop a KNBG parturition detection system that detects and provides managers with early notification of the signs of parturition. The KNBG parturition detection system consists of triaxial accelerometers, gateways, a server, and parturition detection alarm terminals. Then, two different data, the labor and non-labor data, were acquired and a Decision Tree algorithm was used to classify them. After classifying the labor and non-labor states, the sum of the labor status data was multiplied by the activity count value to enhance the classification accuracy. Finally, the Labor Pain Index (LPI) was derived. Based on the LPI, the optimal processing time window was determined to be 10 min, and the threshold value for labor classification was determined to be 14 240.92. The parturition detection rate was 82.4%, with 14 out of 17 parturitions successfully detected, and the average parturition detection time was 90.6 min before the actual parturition time of the first kid. The KNBG parturition detection system is expected to reduce the risk of stunted growth and mortality due to hypothermia in KNBG kids by detecting parturition 90.6 min before the parturition of the first kid, with a success rate of 82.4%, enabling parturition nursing. This study investigated the dietary effect of incorporating different levels of olive cake (OC) on the metabolic responses, growth performance, and carcass characteristics of black goat male kids. Thirty kids (body weight = 17.3 ± 0.40 kg) were randomly distributed into one of three equally sized dietary groups: a control diet (CON), OC at 75 g/kg (OC75), and OC at 150 g/kg (OC150) of dietary dry matter (DM). The results revealed that the intake of DM, crude protein (CP), neutral detergent fiber (NDF), and acid detergent fiber (ADF) was similar (p ≥ 0.11) among the three treatment groups. However, the OC150 group had the greatest (p < 0.0001) ether extract (EE) intake compared to the OC75 and CON groups. The growth measurements were similar (p ≥ 0.13) among the three groups. Feed efficiency was not affected by the inclusion of OC. In contrast to the CON diet, the cost of gain was, however, reduced (p = 0.004) in the OC diets. All three treatment groups’ digestibility of DM, CP, and ADF was similar. However, when compared to the OC75 and CON groups, the digestibility of NDF was better (p < 0.05) in the OC150 group. The N intake did not differ among the three experimental groups. Nitrogen retained as g/d was higher (p = 0.04) in the OC150 and OC75 groups compared to the CON group, while retention as a percentage (g/100 g) was similar among the three groups. Except for intermuscular fat, total fat, leg fat depth, and tissue depth, the inclusion of OC did not result in any discernible treatment effects on the carcass and meat quality parameters. Only alanine aminotransferase enzyme activity was lower (p < 0.0001) in OC-treated groups compared to the CON group. In summary, incorporating OC at 75 g/kg and 150 g/kg levels into the diets of black goat kids had positive comparable effects on some parameters related to growth performance, carcass attributes, and meat quality. Importantly, utilizing olive cake led to cost savings in production and may serve as a viable alternative feed source in goat nutrition. Full article

Reproductive traits are the basic economic traits of goats and important indicators in goat breeding. In this study, Dazu black goats (DBGs; n = 150), an important Chinese local goat breed with excellent reproductive performance, were used to screen for important variation loci and genes of reproductive traits. Through genome-wide association studies (GWAS), 18 SNPs were found to be associated with kidding traits (average litter size, average litter size in the first three parity, and average litter size in the first six parity), and 10 SNPs were associated with udder traits (udder depth, teat diameter, teat length, and supernumerary teat). After gene annotation of the associated SNPs and in combination with relevant references, the candidate genes, namely ATP1A1, LRRC4C, SPCS2, XRRA1, CELF4, NTM, TMEM45B, ATE1, and FGFR2, were associated with udder traits, while the ENSCHIG00000017110, SLC9A8, GLRB, GRIA2, GASK1B, and ENSCHIG00000026285 genes were associated with litter size. These SNPs and candidate genes can provide useful biological information for improvement of the reproductive traits of goats. Full article

Retinol-binding protein 4 (RBP4), a member of the lipocalin family, is a specific carrier of retinol (vitamin A) in the blood. Numerous studies have shown that RBP4 plays an important role in mammalian embryonic development and that mutations in RBP4 can be used for the marker-assisted selection of animal reproductive traits. However, there are few studies on the regulation of reproduction and high-prolificacy traits by RBP4 in goats. In this study, the 5′ flanking sequence of RBP4 was amplified, and a G>C polymorphism in the promoter region -211 bp (g.36491960) was detected. An association analysis revealed that the respective first, second and third kidding number and mean kidding number of nanny goats with CC and GC genotypes (2.167 ± 0.085, 2.341 ± 0.104, 2.529 ± 0.107 and 2.189 ± 0.070 for CC and 2.052 ± 0.047, 2.206 ± 0.057, 2.341 ± 0.056 and 2.160 ± 0.039 for GC) were significantly higher (p < 0.05) than those with the GG genotype (1.893 ± 0.051, 2.027 ± 0.064, 2.107 ± 0.061 and 1.74 ± 0.05). The luciferase assay showed that luciferase activity was increased in C allele individuals compared with that in G allele individuals. A competitive electrophoretic mobility shift assay (EMSA) showed that individuals with the CC genotype had a stronger promoter region binding capacity than those with the GG genotype. In addition, transcription factor prediction software showed that the RBP4 g.36491960G>C mutation added a novel binding site for transcription factor DP-1 (TFDP1). RT–qPCR results showed that the expression of TFDP1 was significantly higher in the high-prolificacy group than in the low-prolificacy group, and the expression of RBP4 was higher in both the CC and GC genotypes than that in the GG genotype. TFDP1 overexpression significantly increased the expression of RBP4 mRNA (p < 0.05) and the expression of the cell proliferation factors cyclin-D1, cyclin-D2 and CDK4 (p < 0.05). The opposite trend was observed after interference with TFDP1. Both the EdU and CCK-8 results showed that TFDP1 expression could regulate the proliferation of goat ovarian granulosa cells. In summary, our results showed that RBP4 g.36491960G>C was significantly associated with fecundity traits in goats. The g.36491960G>C mutation enhanced the transcriptional activity of RBP4 and increased the expression of RBP4, thus improving the fertility of Yunshang black goats. Full article

The purpose of this study was to evaluate the effects of fucoidan supplementation on serum biochemical parameters, small intestinal barrier function, and cecal microbiota of weaned goat kids. A total of 60 2-month-old weaned castrated male goat kids (Chuanzhong black goat) were used in this 30-day experiment. The goat kids were randomly divided into four groups: a control group (CON) fed the basal diet, and three other groups supplemented with 0.1%, 0.3%, and 0.5% fucoidan in the basal diet (denoted as F1, F2, and F3 groups, respectively). The results indicated that dietary fucoidan supplementation decreased (p < 0.05) the activity of lactate dehydrogenase (LDH) and the content of glucose (GLU) as measured on day 15. As measured on day 30, dietary fucoidan increased (p < 0.05) the content of total protein (TP) and decreased the activity of aspartate aminotransferase (AST), and supplementation with 0.3% and 0.5% fucoidan decreased (p < 0.05) the activity of LDH. Dietary fucoidan decreased (p < 0.05) the content of D-lactic acid (D-LA) and the activity of diamine oxidase (DAO). Dietary fucoidan increased (p < 0.05) the activity of catalase (CAT) in the duodenum. Dietary 0.3% and 0.5% fucoidan enhanced (p < 0.05) the activity of glutathione peroxidase (GSH-Px) in the ileum, the activity of total superoxide dismutase (T-SOD) in the jejunum and ileum, and the activity of CAT in the ileum. Dietary 0.3% and 0.5% fucoidan reduced the contents of malondialdehyde (MDA) in the duodenum, jejunum, and ileum and the content of hydrogen peroxide (H2O2) in the duodenum. Dietary fucoidan increased (p < 0.05) the content of secretory immunoglobulin A (sIgA) in the duodenum. Supplementation of 0.3% and 0.5% fucoidan upregulated (p < 0.05) the gene expression of ZO-1 and claudin-1 in the duodenum, jejunum, and ileum, and dietary supplementation of 0.3% and 0.5% fucoidan upregulated (p < 0.05) the gene expression of occludin in the jejunum and ileum. The 16S rRNA high-throughput sequencing results showed that at the phylum level, dietary fucoidan increased (p < 0.05) the abundance of Bacteroidetes while decreasing (p < 0.05) the abundance of Firmicutes. At the genus level, dietary 0.3% and 0.5% fucoidan increased (p < 0.05) the abundances of Unspecified_Ruminococcaceae, Unspecified_Bacteroidale, Unspecified_Clostridiales, and Akkermansia. In conclusion, dietary fucoidan supplementation had positive effects on intestinal permeability, antioxidant capacity, immunity function, tight junctions, and the cecal microflora balance in weaned goat kids. As a vital metabolic and immune organ in animals, the liver plays an important role in protein synthesis, detoxification, metabolism, and immune defense. The primary research purpose of this study was to reveal the effect of breast-feeding, weaning transition, and weaning on the gene expression profile in the goat kid liver and to elucidate the transcriptome-level signatures associated with liver metabolic adaptation. Therefore, transcriptome sequencing was performed on liver tissues, which was collected at 1 day (D1), 2 weeks (W2), 4 weeks (W4), 8 weeks (W8), and 12 weeks (W12) after birth in Laiwu black goats at five different time-points, with five goats at each time point. From 25 libraries, a total of 37497 mRNAs were found to be expressed in goat kid livers, and 1271 genes were differentially expressed between at least two of the five time points. Gene ontology (GO) and Kyoto Encyclopedia of Genes and Genomes (KEGG) pathway analyses revealed that these genes were annotated as an extracellular region fraction, exhibiting monooxygenase activity, positive regulation of T cell activation, mitotic spindle mid-region assembly, cytokinesis, cytoskeleton-dependent cytokinesis, regulation of cytokinesis, regulation of lymphocyte proliferation, and so on. In addition, it mainly deals with metabolism, endocrine, cell proliferation and apoptosis, and immune processes. Finally, a gene regulatory network was constructed, and a total of 14 key genes were screened, which were mainly enriched for cell growth and development, endocrine, immune, and signal transduction-related pathways. Our results provide new information on the molecular mechanisms and pathways involved in liver development, metabolism, and immunity of goats. Full article

The purpose of this study was to evaluate the effects of fucoidan dietary supplementation on growth performance, organs’ relative weight, serum anti-oxidation markers, immune function indices and intestinal morphology in weaned kids. A total of 60 2-month-old weaned castrated male kids (Chuanzhong black goat) were used for this 30-day experiment and randomly allocated to four groups. The control group (CON) fed a basal diet, while the other three groups were provided with the same diet further supplemented with fucoidan at 0.1%, 0.3% or 0.5%, namely, F1, F2 and F3 groups, respectively. The results indicated that dietary fucoidan supplementation significantly increased (p < 0.05) the activity of catalase (CAT) when compared to the CON group on day 15. Moreover, the addition of fucoidan at 0.3% and 0.5% significantly increased (p < 0.05) the activities of glutathione peroxidase (GSH-Px) and total superoxide dismutase (T-SOD). On day 30, dietary fucoidan supplementation significantly reduced (p < 0.05) the feed conversion rate (FCR), contents of tumor necrosis-α (TNF-α), interleukin-1β (IL-1β) and interleukin-6 (IL-6), while it significantly increased (p < 0.05) the activity of total superoxide dismutase (T-SOD), the content of immunoglobulin G (IgG) and the villus height (VH) of the duodenum. Moreover, dietary 0.3% and 0.5% fucoidan supplementation significantly increased (p < 0.05) the villus height (VH) of the jejunum and ileum and significantly reduced (p < 0.05) the crypt depth (CD) of ileum. In conclusion, dietary fucoidan had positive effects on growth performance, serum anti-oxidation, immune function and intestinal morphology of weaned kids. Full article

Predicting the Digestive Tract Development and Growth Performance of Goat Kids Using Sigmoidal Models

The transition from monogastric to rumination stage is crucial in ruminants’ growth to avoid stressors—weaning and neonatal mortalities. Poor growth of the digestive tract could adversely affect the performance of the animal. Modeling informative growth curves is of great importance for a better understanding of the effective development pattern, in order to optimize feeding management system, and to achieve more production efficiency. However, little is known about the digestive tract growth curves. For this reason, one big goat farm of Laiwu black breed was chosen as a basis of this study. Forty-eight kids belonging to eight-time points (1, 7, 14, 28, 42, 56, 70, and 84 d; 6 kids for each) were selected and slaughtered. The body weight, body size indices, rumen pH, and stomach parts were determined and fitted to the polynomial and sigmoidal models. In terms of goodness of fit criteria, the Gompertz model was the best model for body weight, body oblique length, tube, and rumen weight. Moreover, the Logistic model was the best model for carcass weight, body height, and chest circumference. In addition, the Quadratic model showed the best fit for dressing percentage, omasum weight, abomasum weight, and rumen volume. Moreover, the cubic model best fitted the ruminal pH and reticulum percentage. The Weibull model was the best model for the reticulum weight and omasum percentage, while the MMF model was the best model describing the growth of chest depth, rumen percentage, and abomasum percentage. The model parameters, R squared, inflection points, area under curve varied among the different dependent variables. The Pearson correlation showed that the digestive tract development was more correlated with age than body weight, but the other variables were more correlated with body weight than age. The study demonstrated the use of empirical sigmoidal and polynomial models to predict growth rates of the digestive tract at relevant age efficiently. Full article
