Amino acid composition and nitrogen quantification in grape must are of great importance given their usefulness for varietal characterization, influence on fermentation processes and identification of the aromatic potential of musts. The objective of this work was to determinate the amino acid and nitrogen compound profiles of 30 minority varieties of potential use in winemaking, all grown in the same ampelographic collection in Alcalá de Henares. The concentrations of 31 amino acids in must were identified and quantified using high-performance liquid chromatography (HPLC) during four seasons (2020 to 2023), and the average values of total free amino acids (TAN), yeast assimilable nitrogen (YAN) and aromatic precursor nitrogen (APN) were calculated for each variety. ‘Tazazonal’, a minority red grape variety, was found to exhibit high concentrations of yeast assimilable nitrogen (YAN), total amino nitrogen (TAN) and ammonia nitrogen (APN), comparable to those observed in ‘Tempranillo’ and ‘Garnacha Tinta’. These two cultivars are representative of traditional Spanish red grapevine varieties and are among the most widely cultivated in the country. In the case of white varieties, ‘Albillo del Pozo’, ‘Pintada’ and ‘Verdejo Serrano’ showed higher concentrations of these parameters than ‘Malvar’ and ‘Airén’, which are also widely grown in Spain. The results revealed distinct amino acid profiles for each variety, enabling their classification and supporting the identification of variants within the autochthonous germplasm. This approach aimed to highlight minority varieties of potential interest for future studies, given their relevance to both regional and national viticulture. Full article

Repeated expeditions across various regions of Georgia in the early 2000s led to the identification of 434 wild grapevine individuals (Vitis vinifera L. subsp. sylvestris (C.C. Gmel.) Hegi) across 127 different sites, with 45% of these sites containing only a single vine and only 7% more than 9 vines. A total of 70 accessions were propagated in a germplasm collection, 41 of them were descripted from the ampelographic point of view and 32 from the phenological one. The geographical and ecological analysis confirmed that wild grapevines primarily grow in humid environments with warm and fully humid climates, often near rivers. They favor deep, fertile, and evolved soils, mainly alluvial and cinnamonic types (80%), with a marginal presence on strongly eroded soils. Their main natural vegetations are forests and open woodlands, with some individuals in the Southeast found in steppes. The altitudinal range spans from 0 to 1200 m, with 80% of vines distributed between 400 and 900 m. The phenological analysis revealed significant differences among the accessions but no difference among populations, with only a slight variation in bud-break timing, indicating a high level of synchronicity overall. Flowering timing proved to be the most uniform stage, suggesting minimal environmental pressure on genetic adaptation. The mature leaf morphology exhibited significant polymorphism, though leaves were generally three- or five-lobed, weak-wrinkling, and -blistering, with a low density of hairs. Bunch and berry morphology were more uniform. Bunches were consistently very small, cylindrical, and never dense or winged. Berries were also very small, mostly globular, always blue-black in color, and non-aromatic. A striking feature was the frequency of red flesh coloration, which ranged from weak to strong, with uncolored flesh being rare. The Georgian population of wild grapevines was found to be fragmented, often consisting of scattered single individuals or small groups. Therefore, we believe it is urgent for Georgia to implement specific protection measures to preserve this vital genetic resource. Full article

Leaf chlorophyll content is a key indicator of plant physiological status in viticulture; therefore, regular evaluation to obtain data for nutrient supply and canopy management is of vital importance. The measurement of pigmentation is most frequently carried out with hand-held instruments, destructive off-site spectrophotometry, or remote sensing. Moreover, smartphone-based applications also ensure a promising way to collect colorimetric information that could correlate with pigmentation. In this study, four grapevine genotypes were investigated using smartphone-based RGB (Red, Green, Blue) and CIE-L*a*b* colorimetry and a portable chlorophyll meter. The objective of this study was to evaluate the correlation between leaf chlorophyll concentration and RGB- or CIE-L*a*b*-based color indices. A further aim was to find an appropriate model for discriminating between the genotypes by leaf coloration. For these purposes, fully developed leaves of ‘Chardonnay’, ‘Sauvignon blanc’, and ‘Pinot noir’ clones 666 and 777 were investigated with the Color Grab smartphone application to obtain RGB and CIE-L*a*b* values. Using these color values, chroma, hue, and a further 31 color indices were calculated. Chlorophyll concentrations were determined using an Apogee MC100 device, and the values were correlated with color values and color indices. The results showed that the chlorophyll concentration and color indices significantly differed between the genotypes. According to the results, certain color indices show a different direction in their relationship with leaf pigmentation for different grapevine genotypes. The same index showed a positive correlation for the leaf chlorophyll concentration for one variety and a negative correlation for another, which raises the possibility that the relationship is genotype-specific and not uniform within species. In light of this result, further study of the species specificity of the commonly used vegetation indices is warranted. Support Vector Machine (SVM) analysis of the samples based on color properties showed a 71.63% classification accuracy, proving that coloration is an important ampelographic feature for the identification and assessment of true-to-typeness. The aim of this research work was to find historical varieties that existed in this area before phylloxera and to identify them on the basis of historical written sources and genetic analyses. At the beginning of the 19th century, around 300 grape varieties were cultivated in Styria. Between 2020 and 2022, old vineyards were monitored at 115 locations in Styria (between the Mura and Sava rivers) in Slovenia. The directly collected samples (340 grapevine accessions) were determined by molecular analysis with 24 SSR markers. A total of 66 different genotypes were detected. After comparison with the available databases, 29 historical varieties and 37 unknown historical genotypes were identified. Several parameters were calculated to evaluate the usefulness of the selected loci in this work, and a dendrogram representing the genetic similarities between the origins was created using the neighbor-joining method to investigate possible ancestry relationships in the sample set. The most common historical varieties were ‘Belina’ (‘Heunisch weiss’), ‘Vrbovec’ (‘Tantovina Eihenblaetrige’), ‘Ranfol’ (‘Ranfol beli’), and ‘Pelesovna’ (‘Vulpea’). Varieties from the current variety list were also frequently found, such as ‘Frankinja’ (‘Blaufraenkisch’) and ‘Žametovka’ (‘Kavčina črna’). In a few locations, one of the most important red varieties from the beginning of the 20th century was also found in this area (alongside ‘Frankinja’ and ‘Žametovka’), i.e., ‘Vranek’ (‘Zimmettraube balu’). At that time, this variety was planted in multi-variety vineyards and was preserved, but its importance in single-variety vineyards quickly declined due to female flower. In addition, genetic analyses have shown that 37 unknown historical genotypes have been found in this area. These genotypes need to be described ampelographically and technologically evaluated in the future. Most of the vegetative offsprings of these genotypes have already been transferred to the Meranova gene bank, where they can be accurately described ampelographically under the same pedoclimatic conditions. In this study, an analysis of the virome of 51 grapevines from the Don ampelographic collection named after Ya. I. Potapenko (Russia) was performed using high-throughput sequencing of total RNA. A total of 20 previously described grapevine viruses and 4 viroids were identified. The most detected were grapevine rupestris stem pitting-associated virus (98%), hop stunt viroid (98%), grapevine Pinot gris virus (96%), grapevine yellow speckle viroid 1 (94%), and grapevine fleck virus (GFkV, 80%). Among the economically significant viruses, the most present were grapevine leafroll-associated virus 3 (37%), grapevine virus A (24%), and grapevine leafroll-associated virus 1 (16%). For the first time in Russia, a grapevine-associated tymo-like virus (78%) was detected. After a bioinformatics analysis, 123 complete or nearly complete viral genomes and 64 complete viroid genomes were assembled. An analysis of the phylogenetic relationships with reported global isolates was performed. We discovered and characterized the genomes of five novel grapevine viruses: bipartite dsRNA grapevine alphapartitivirus (genus Alphapartitivirus, family Partitiviridae), bipartite (+) ssRNA grapevine secovirus (genus Fabavirus, family Secoviridae) and three (+) ssRNA grapevine umbra-like viruses 2, -3, -4 (which phylogenetically occupy an intermediate position between representatives of the genus Umbravirus and umbravirus-like associated RNAs). In this study, we analyzed the virome of 73 grape samples from two Dagestan ampelographic collections in Russia using high-throughput sequencing of total RNAs. Fourteen viruses and four viroids were identified, with one to eleven of them detected in each plant. For the first time in Russia, we identified grapevine leafroll-associated virus 7 and grapevine Kizil Sapak virus. A total of 206 genomes of viruses and viroids were obtained, and their phylogenetic analysis was carried out. The de novo assembly and tblastx analysis allowed us to obtain contigs of a novel (+) ssRNA genome of a plant virus from the genus Umbravirus, which was tentatively named grapevine umbra-like virus (GULV), as well as contigs of a novel dsDNA pararetrovirus from the genus Caulimovirus, which was tentatively named grapevine pararetrovirus (GPRV). Complete genomes of these viruses were obtained and used for Sequence Demarcation Tool (SDT) analysis and phylogeny studies. GULV and GPRV were detected in 16 and 33 germplasm samples from the Dagestan collections, respectively. Full article

A rapid and simple HPLC-DAD analytical method was developed and optimized for the determination of anthocynanins in three red Greek winegrape varieties (Kotsifali, Limnio, and Vradiano). The critical parameters, such as the acidifying solvent and the extraction temperature, which affect the extraction of anthocyanins from the grapes, were studied to find the optimum values. The developed methodology was validated in terms of selectivity, linearity, accuracy, and precision and presented satisfactory results. The limits of quantification (LOQs) ranged between 0.20 mg/kg to 0.60 mg/kg, and the limits of detection (LODs) ranged between 0.06 mg/kg and 0.12 mg/kg. The RSD% of the within-day and between-day assays were lower than 6.2% and 8.5%, respectively, showing adequate precision. The accuracy ranged between 91.6 and 119% for within-day assay and between 89.9 and 123% for between-day assay. Sixteen samples from the main regions of each variety as well as from the official ampelographic collections of Greece were collected during the 2020 growing season and were further analyzed by HPLC-DAD. Notable differences in the anthocyanin content were detected among the cultivars using hierarchical cluster analysis (HCA). Full article

Grapevine germplasm collections are unique repositories of grape cultivars; therefore, it is necessary to minimize their infection with pathogens, including viruses, and develop various programs to maintain them in a virus-free state. In our study, we examined the virome of the largest Russian grapevine germplasm collection, the Anapa Ampelographic Collection, using high-throughput sequencing of total RNAs. As a result of bioinformatics analysis and validation of its results by reverse transcription PCR (RT-PCR) and quantitative RT-PCR (RT-qPCR), we identified 20 viruses and 3 viroids in 47 libraries. All samples were infected with 2 to 12 viruses and viroids, including those that cause economically significant diseases: leafroll, fleck, and rugose wood complex. For the first time in Russia, we detected Grapevine virus B (GVB), Grapevine virus F (GVF), Grapevine asteroid mosaic-associated virus (GAMaV), Grapevine Red Globe virus (GRGV), Grapevine satellite virus (GV-Sat), Grapevine virga-like virus (GVLV), Grapevine-associated jivivirus 1 (GaJV-1) and Vitis cryptic virus (VCV). A new putative representative of the genus Umbravirus with the provisional name Grapevine umbra-like virus (GULV) was also identified in Russian grape samples. Full article

Metagenomic approaches used for virus diagnostics allow for rapid and accurate detection of all viral pathogens in the plants. In order to investigate the occurrence of viruses and virus-like organisms infecting grapevine from the Ampelographic collection Kromberk in Slovenia, we used Ion Torrent small RNA sequencing (sRNA-seq) and the VirusDetect pipeline to analyze the sRNA-seq data. The used method revealed the presence of: Grapevine leafroll-associated virus 1 (GLRaV-1), Grapevine leafroll-associated virus 2 (GLRaV-2), Grapevine leafroll-associated virus 3 (GLRaV-3), Grapevine rupestris stem pitting-associated virus (GRSPaV), Grapevine fanleaf virus (GFLV) and its satellite RNA (satGFLV), Grapevine fleck virus (GFkV), Grapevine rupestris vein feathering virus (GRVFV), Grapevine Pinot gris virus (GPGV), Grapevine satellite virus (GV-Sat), Hop stunt viroid (HSVd), and Grapevine yellow speckle viroid 1 (GYSVd-1). Multiplex reverse transcription-polymerase chain reaction (mRT-PCR) was developed for validation of sRNA-seq predicted infections, including various combinations of viruses or viroids and satellite RNA. mRT-PCR could further be used for rapid and cost-effective routine molecular diagnosis, including widespread, emerging, and seemingly rare viruses, as well as viroids which testing is usually overlooked. (1) Background: Currently, some ampelographic methods are developing in order to identify grapevine varieties. For this purpose, morpho-colorimetric parameters in leaves have been analyzed by digital imagen analysis, but some environmental conditions may affect their determinations. (2) Methods: A research study was conducted to characterize leaf morpho-colorimetric parameters in five grapevine varieties growing under different plant water status and to discriminate them under these conditions. Leaves were collected in vines, and twelve leaf morpho-colorimetric and fractal dimension variables were assessed. (3) Results: Merlot presented the highest values of perimeter and area to perimeter ratio in leaves and higher leaf area than Chardonnay in both plant water conditions. Most of the leaf morpho-colorimetric variables allowed discriminating the grapevine varieties under the contrasted hydric conditions. Under non-water stress, Carmenère was not related to any measured parameters. Merlot was positively related to most of the leaf morphometric parameters, whereas Chardonnay presented the opposite behavior. RGB color system variables allowed discriminating the grapevine varieties under water stress conditions, and Sauvignon Blanc was not related to any measured parameter. Chardonnay and Pinot Noir were positively related to green color and negatively related to most of the leaf morphometric parameters, whereas Merlot showed the opposite behavior. (4) Conclusions: Leaf morpho-colorimetric and fractal dimension parameters were affected by plant water stress and more variables should be incorporated into the new ampelographic methods in order to characterize leaf morpho-colorimetric parameters of the different grapevine varieties more clearly. The protection of grapevine biodiversity and the safeguarding of genetic variability are certainly primary and topical objectives for wine research, especially in territories historically devoted to viticulture. To assess the autochthonous germplasm of three different districts of Southern Umbria (Central Italy), the plant material of 70 grapevines retrieved from reforested land plots or old vineyards was collected, and their genetic identity was investigated using 13 microsatellite markers (SSR). The results revealed the presence of 39 unique genotypes, divided into 24 already-known cultivars and 15 never-reported SSR profiles. Most of the grapevine accessions were then vegetatively propagated and cultivated in a vineyard collection both to be protected from extinction and to be evaluated at the ampelographic level. Overall, this work emphasizes the need for recovering the threatened genetic variability that characterizes minor neglected grapevine cultivars or biotypes of Southern Umbria germplasm, and the requirement to revalue and exploit the more valuable genetic resources to enhance the local agri-food economy. Morocco, with its Mediterranean climate and its various potentialities, conceals an important space for the extension of viticulture, in particular that of traditional grape varieties, which has undergone profound upheavals linked to economic, social and environmental constraints which have had a negative impact on genetic diversity. The present study aimed to evaluate the richness of the local phylogenetic heritage of the traditional vines in the North-West of Morocco. For this, we carried out a characterization of a collection of 1617 leaves, taken from 162 vine plants and belonging to 27 different traditional varieties. Thereafter, we used an ampelometric and ampelographic approach with the Super Ampelo software. The ANOVA test revealed the most discriminating parameters, which were the angles, the depth of the lateral sinuses in relation to the lengths of the ribs, and the relationships between all the parameters. In this sense, the qualitative parameters (OIV Codes) confirmed the presence of morphological diversity within the grape varieties studied; the study of general averages made it possible to specify the varieties with the large values of distance/angles and of ratio, and we have shown the presence of a great intra-varietal diversity in addition to that which is inter-varietal. The analysis in principal components allowed the grouping of the grape varieties into five groups according to their expressions by the quantitative parameters, and it confirmed the hypothesis of the influence of the external environment in addition to the gene pool on the grape varieties. This leads us to provide more efforts to maintain inter- and intra-varietal variability and to fight against genetic erosion and the threat of environmental changes. Full article

Wine has been produced in Georgia since the 6th millennium BC. The processes of cultivar selection and breeding started with wild grapes Vitis vinifera L. ssp. sylvestris Gmel. and included multiple introgression events—from the wild to domestication. This article aims at improving the knowledge concerning the history of winemaking through a comparison of the Vitis vinifera subsp. sylvestris and subsp. sativa. Grapes of Vitis vinifera subsp. sylvestris were grown in an ampelographic collection and vintages 2017–2020 were analyzed. The obtained data were compared to a wider dataset available in literature concerning Vitis vinifera subsp. sativa, demonstrating the central role of grape morphology in the domestication process. This evidence suggests that the technological value of the cultivars played an important role in the selection process. In vintages 2017, 2018, and 2019, wines were produced with Vitis vinifera subsp. sylvestris grapes and compared with Cabernet Sauvignon and Saperavi vinifications. For all the vintages, the fermentations took shorter time for wild grape, despite the highest content of total phenols. Learning from the past, Vitis vinifera subsp. sylvestris might still be an interesting genetic resource for future breeding programs. Furthermore, the possible combination of wild and domesticated grapes can make possible the production of wines with long ageing, exalting their own characteristics. Full article

A prospecting work at the Axarquia region (Malaga, Spain) was carried out in order to identify local red grapevine cultivars preserved in ancient vineyards. A total of 11 accessions were collected in seven different plots from four municipalities and analyzed using 25 microsatellite loci for cultivar identification. The accessions analyzed were identified as eight different genotypes, seven of them corresponding to known cultivars as ‘Cabernet Sauvignon’, ‘Jaen Tinto’, ‘Molinera’, ‘Monastrell’, ‘Muscat of Alexandria’, ‘Parrel’, and ‘Romé’. In addition, one of them is referred to as the new genotype for ‘Cabriel’ cultivar. Additionally, an ampelographic characterization was carried out with 30 International Organisation of Vine and Wine (OIV) descriptors for two consecutive years for the eight accessions identified as local cultivars. This allowed the identification of a somatic variant of the ‘Muscat of Alexandria’ cultivar that affects the color of the berry and another of ‘Romé’ regarding bunch compactness. Full article

Climate change affects all characteristics of grapes, including the taste of fresh berries and wine. The purpose of this study was to reveal the dependence of fresh grapes taste scores (GS) and wine taste scores (WS) on the origin of the varieties and weather conditions of the harvest year in the northern zone of industrial viticulture in Russia. The material used in the study were taste scores of 232 grape varieties from the Don Ampelographic Collection (47°25′ N 40°03′ E) 1981–2019. The correlation analysis, ANOVA, and regression were used. It was shown that GS negatively correlated with productivity, and WS with the berry mass of variety. In recent decades, GS have increased in groups of varieties of various interspecies origin, and WS have not changed. Regression models revealed that the reason for GS improvement was in the growth of average temperature above 15 °C, while WS varied depending on the sum of precipitation for the period with temperatures above 20 °C, which did not have a reliable trend. Together with data on the growth of grape yield and sugar content, the results indicate that observed climate warming is favorable for grapevine cultivation in the northern zone of industrial viticulture in Russia.
