Viticulture is increasingly seeking sustainable practices that enhance grape quality while reducing reliance on synthetic inputs. Among bio-based strategies, biochar has attracted growing interest for its potential to improve soil fertility and influence plant metabolism. However, its effects can vary depending on formulation and application methods. This study evaluated the effects of the use of solid (SB) and liquid biochar (LB) on the biochemical and nutritional composition in leaves, berry skins, and must of a grapevine (Vitis vinifera L., cv. Sangiovese) cultivated in a vineyard in Tuscany (Italy). SB was applied once to the soil at 2.5% (w/w), while LB was applied five times/season at 10% (v/v) via fertigation. Results revealed formulation-specific effects on grapevine physiology and fruit composition. SB maintained leaf chlorophyll concentrations, increased total soluble proteins (+65%), total polyphenols (+57%), and tannin content (+33%) in berry skins and must, and reduced Cu and Zn. Conversely, LB reduced leaf chlorophyll concentrations (−24%) and nutrient contents (P, Fe, Cu, and Zn), but increased total flavonoids (+13%), antioxidant capacity (+20%), and vitamin C (+18%) in berry skins, alongside higher fructose and reducing sugars in musts. The findings highlight biochar’s potential as a bio-based input in viticulture, emphasizing the importance of formulation and application strategy. SB appears suited to long-term soil improvement and enhanced phenolic richness, while LB may modulate sugar metabolism for targeted enological outcomes. Full article

Plants constantly face the environment that surrounds them and fight for survival against biotic and abiotic stress factors. To deal with harmful conditions, plants have developed a multilayer defence system, making them capable of recognising threats and promptly recovering from them. This phenomenon, which takes advantage of the “memory effect”, is referred to as bio-priming and represents a new frontier in terms of crop protection. Here, we investigated the “indirect” protective mechanisms of a new yeast extract formulate in Vitis vinifera cv. Sangiovese plants at both the biochemical and genic levels. The formulate was applied once a week for three consecutive weeks, and grapevine leaves were sampled from the first to the fifth day after treatment (dat) at every week of the experiment. Increased levels of jasmonic acid (every week at 2 dat; +70% as average) and abscisic acid (at 1 dat of the first week, more than 1.7-fold higher than the control) and the underproduction of salicylic acid (from 2 dat; −18%) confirmed that these signalling molecules/”specialised compounds” are actively involved in the early activation of defence pathways in treated vines. In addition, pr2 and chit1b, two genes involved in regulating hormonal crosstalk, were significantly up-regulated (both in the first and second week of the trial) and were also found to underlie upstream molecular activation. The results obtained by this investigation confirm the use of this new product to prime and protect grapevines from a wide range of fungal and fungal-like plant pathogens through the induction of defence responses. The intensification of highly specialized viticulture has led to a dramatic decrease of soil fertility that can be restored by increasing soil organic matter using organic fertilizers. The aim of the present experiment was to evaluate the effect of different organic amendments on vine vegetative growth and nutritional status, soil N availability and microbial biomass, as well as on yield and grape quality. The experiment was carried out in 2020 and 2021, on cv. Sangiovese (Vitis vinifera L.) vines grafted on 110 Richter (V. berlandieri × V. rupestris) planted in February 2019. Plants were fertilized yearly in spring with (1) mineral fertilization (MIN), (2) municipal organic waste compost (MOW), and (3) sewage sludge compost (SS). The application of SS increased nitrate availability in both years, while the supply of organic matter (no matter the source) enhanced soil microbial biomass content. Plant nutritional status was in the optimal range for all treatments, with an increase of N in SS and K in MOW. Fruit yield in 2020 was not influenced by treatments, while in 2021 it was enhanced by MIN and MOW, which also induced a higher berry quality. Plant vegetative growth was stimulated by the application of SS. In conclusion, from these preliminary results we observed a higher N availability as a consequence of SS supply that resulted in a higher plant biomass, but reduced yield and berry quality, supporting the theory that for vineyards, N should be carefully managed to reach an equilibrium between vegetative and reproductive activity. Grapevine “bois noir”, related to the presence of ‘Candidatus Phytoplasma solani’ (‘Ca. P. solani’), represents a serious threat in several vine-growing areas worldwide. In surveys conducted over two years, mild and/or moderate symptoms and lower pathogen titer were mainly associated with ‘Ca. P. solani’ strains harboring a secY gene sequence variant (secY52), whereas severe symptoms and higher titer were mainly observed in grapevines infected by phytoplasma strains carrying any one of another four variants. A comparison of amino acid sequences of the protein SecY of ‘Ca. P. solani’ strains revealed the presence of conservative and semi-conservative substitutions. The deduced three-dimensional (3D) protein analysis unveiled that one semi-conservative substitution identified in the sequence variant secY52 is responsible for a structural disordered region that probably confers a flexibility for binding to distinct molecular complexes. In fact, the other analyzed variants show an organized structure and the 3D in silico prediction allowed the identification of β-sheets. Thus, differences in symptom severity and pathogen concentration observed in grapevines infected by ‘Ca. P. solani’ strains carrying distinct secY gene sequence variants suggest a possible relationship between SecY protein structure and phytoplasma strain virulence. Full article

High temperatures and the anomalous distribution of rainfall during the growing season may have a negative impact on grapevine yield and berry composition. In recent years, many studies have focused on the application of agronomical techniques to reduce the negative impact of heat waves on secondary metabolites such as phenols. In particular, treatments with kaolin have shown positive effects on reducing canopy temperatures, enhancing the accumulation of anthocyanins. In regard to the above, three treatments were evaluated: untreated control (C), kaolin (CAO), and chabasite-rich zeolitites (ZEO) applications on cv. Sangiovese in order to verify the cooling effects on leaves and bunches, and the impact on gas exchange, yield parameters, berry composition, and on both chemical and sensory notes of wine. Minerals were sprayed twice around the veraison on the entire canopy at a 3% concentration. The results showed that the application of the minerals was able to reduce the berry temperatures in both years of the trial as compared to the untreated control (C), without affecting vine gas exchange, yield, and soluble solid accumulation. Furthermore, the cooling effect determined an increase in anthocyanin on both the grapes and the wine. At testing, CAO and ZEO wines stood out regarding greater color intensity and were preferred by the judges. Full article

Bois noir (BN), associated with ‘Candidatus Phytoplasma solani’ (CaPsol), is the most widespread disease of the grapevine yellows complex worldwide. In this work, BN epidemiology was investigated in a case study vineyard where an unusual CaPsol strain, previously detected only in other host plants, was found to be prevalent in grapevine. Experimental activities included: symptom observation; sampling of symptomatic vines, Auchenorrhyncha specimens, and weeds; molecular detection and typing of CaPsol strains; statistical analyses for determining possible relationships between CaPsol relative concentration, strain type, and symptom severity. Among insects, Reptalus quinquecostatus was the most abundant and was found to be highly infected by CaPsol, while Hyalesthes obsoletus, the main CaPsol vector, was not caught. Moreover, R. quinquecostatus harbored CaPsol strains carrying uniquely the stamp sequence variant St10, also identified as prevalent in vines and in the majority of weeds, and all the secY variants identified in the vineyard. Statistical analyses revealed that CaPsol strains carrying the St10 variant are not associated with severe symptoms, suggesting their possible moderate virulence. Based on such evidence, a new BN epidemiological pattern related to these CaPsol strains and involving grapevine, R. quinquecostatus, and/or weeds is proposed. Furthermore, the possible presence of other players (vectors and weeds) involved in CaPsol transmission to grapevines was highlighted. Full article

Bois noir is a disease associated with the presence of phytoplasma ‘Candidatus Phytoplasma solani’ belonging to the Stolbur group (subgroup 16SrXII-A), which has a heavy economic impact on grapevines. This study focused on the changes induced by phytoplasma in terms of the profile and amount of secondary metabolites synthesized in the phenylpropanoid pathway in leaves of Vitis vinifera L. red-berried cultivar Sangiovese. Metabolic alterations were assessed according to the disease progression through measurements of soluble sugars, chlorophyll, and phenolic compounds produced by plant hosts, in response to disease on symptomatic and asymptomatic Bois noir-positive plants. Significant differences were revealed in the amount of soluble sugars, chlorophyll, and accumulation/reduction of some compounds synthesized in the phenylpropanoid pathway of Bois noir-positive and negative grapevine leaves. Our results showed a marked increase in phenolic and flavonoid production and a parallel decrease in lignin content in Bois noir-positive compared to negative leaves. Interestingly, some parameters (chlorophyll a, soluble sugars, total phenolic or flavonoids content, proanthocyanidins, quercetin) differed between Bois noir-positive and negative leaves regardless of symptoms, indicating measurable biochemical changes in asymptomatic leaves. Our grapevine cultivar Sangiovese results highlighted an extensive modulation of the phenylpropanoid biosynthetic pathway as a defense mechanism activated by the host plant in response to Bois noir disease. Water availability is a major limiting factor in plant productivity and plays a key role in plant species distribution over a given area. New technologies, such as terahertz quantum cascade lasers (THz-QCLs) have proven to be non-invasive, effective, and accurate tools for measuring and monitoring leaf water content. This study explores the feasibility of using an advanced THz-QCL device for measuring the absolute leaf water content in Corylus avellana L., Laurus nobilis L., Ostrya carpinifolia Scop., Quercus ilex L., Quercus suber L., and Vitis vinifera L. (cv. Sangiovese). A recently proposed, simple spectroscopic technique was used, consisting in determining the transmission of the THz light beam through the leaf combined with a photographic measurement of the leaf area. A significant correlation was found between the product of the leaf optical depth (τ) and the leaf surface area (LA) with the leaf water mass (Mw) for all the studied species (Pearson’s r test, p ≤ 0.05). In all cases, the best fit regression line, in the graphs of τLA as a function of Mw, displayed R2 values always greater than 0.85. The method proposed can be combined with water stress indices of plants in order to gain a better understanding of the leaf water management processes or to indirectly monitor the kinetics of leaf invasion by pathogenic bacteria, possibly leading to the development of specific models to study and fight them.
