Skin aging is influenced by intrinsic and extrinsic factors, leading to structural changes in the skin. Current anti-aging cosmetic trends emphasize innovative natural ingredients, including plant extracts, thermal waters, and botanical hydrolats. This work aims to develop six natural anti-aging cosmetics (two serums, two day creams, and two night creams) with innovative and non-irritating profiles. The rational design was guided by market analysis and ingredient properties. Prototype formulations were created with two core mixtures: (1) natural mineral water from Termas de Unhais da Serra and Thymus × citriodorus hydrolat and (2) natural mineral water from Termas de Chaves and aqueous Vaccinium myrtillus (blueberry) extract. The products were evaluated for stability (4 °C and 40 °C, 4 weeks), safety (according to EC Regulation No. 1223/2009), including in vitro testing for skin irritation potential (epiderm model; OECD TG 439). The market study highlighted a gap in anti-aging products combining natural extracts and thermal waters. All prototypes showed desirable textures and remained stable under test conditions. No irritating effects were observed. The results support the development of effective anti-aging cosmetics rooted in natural resources. These innovative products can meet the market demand for natural and sustainable skincare solutions. Full article

We present the numerical proof of a new sensor concept, based on the Resistive AC-Coupled Silicon Detectors (RSDs) paradigm and standard CMOS process, which benefits from having a 100% fill factor and embedded front-end electronics. The compatibility between these two technologies has been investigated, and our encouraging results suggest that this target could be reliably achieved, enabling the possibility to considerably boost the performance of current silicon detectors intended for timing and 4D-tracking. Natural mineral waters (NMWs) and plant extracts have long been valued for their therapeutic properties and skin benefits. This study investigated, in vitro, the role of five Portuguese NMWs (A-E), combined with plant extracts from five species (Ficus carica L., Rubus idaeus L., Vaccinium myrtillus, Cistus ladanifer and Thymus x citriodorus) as bioactive ingredients. Antioxidant capacity was assessed using the 2,2-Diphenyl-1-picrylhydrazyl (DPPH) method. Cellular biocompatibility was evaluated in fibroblasts (L929) and macrophages (RAW 264.7). Skin-repairing and anti-senescence properties were evaluated in L929 cells through the scratch-wound method and β-galactosidase assay. Superoxide dismutase (SOD) was quantified using a commercial kit, and lipopolysaccharide-induced reactive oxygen species (ROS) were quantified using a fluorescent probe (H2DCFDA) in RAW 264.7. The results highlighted the beneficial impact of extracts combined with NMWs. An increase in antioxidant capacity of up to 90% was observed in mixtures comprising Ficus carica L., compared with NMWs alone. In contrast, mixtures with Cistus ladanifer showed promising anti-aging potential, with a 40% decrease in senescent cells and a 33% ROS reduction. Rubus idaeus L. extract produced an increase in cell migration capacity (up to 50%), depending on the NMW. This study highlights the potential synergism of natural ingredients with plant extracts for anti-aging. Full article

Congenital cystic adenomatoid malformation (CCAM) is a rare fetal lung anomaly characterized by benign multicystic masses that can lead to severe complications, such as pulmonary hypoplasia, fetal hydrops, and neonatal death. This literature review examines current knowledge on antenatal therapies for CCAM, focusing on pharmacological, procedural, and surgical interventions. Betamethasone, the first-line pharmacological treatment, has shown efficacy in reducing lesion size and resolving hydrops, particularly in microcystic CCAM. Procedural options, such as thoracoamniotic shunting, are effective for macrocystic lesions but carry risks including preterm labor and thoracic deformities. Open fetal surgery remains a last-resort intervention for severe cases, while emerging techniques, like percutaneous laser ablation and sclerotherapy, offer promising minimally invasive alternatives. A proposed treatment algorithm emphasizes individualized care based on lesion type, gestational age, and the presence of complications. The authors searched the US National Library of Medicine Database, Google Scholar, and PubMed Central to gather information on antenatal therapies for CCAM. This review emphasizes that, despite significant advancements, considerable challenges persist, underscoring the need for prospective studies to refine therapeutic protocols and assess long-term outcomes. The early immune kinetics after SARS-CoV-2 vaccination remain poorly understood, particularly among individuals with low-count monoclonal B-cell lymphocytosis (MBLlo). We investigated the cellular and humoral kinetics in the blood of 50 non-MBL healthy donors (HD) vs. 16 MBLlo subjects after SARS-CoV-2 vaccination, who were subclassified according to their history of previous exposure to SARS-CoV-2 into SARS-CoV-2 naïve and previously infected subjects. Overall, we found decreased neutrophil and lymphocyte counts at day +4 following each dose in non-MBL HD, together with an earlier and higher increase in plasma cell (PC) counts and SARS-CoV-2-specific antibody levels after the first vaccine in previously infected non-MBL HD. MBLlo subjects showed a similar profile, except for lower B-cell and higher PC counts after vaccination, and a trend towards a higher (but delayed) antibody response. In summary, we found different cell-kinetic profiles following vaccination in SARS-CoV-2 naïve vs. previously infected non-MBL HD (earlier PC and antibody responses in the latter group); additionally, MBLlo subjects had significantly lower B-cell and higher PC counts after vaccination, and a delayed SARS-CoV-2-specific antibody response. Background/Objectives: Excessive or inadequate use of antimicrobial drugs may lead to the emergence of resistant strains. For this reason, it is important to monitor consumption indicators to assess drugs’ utilization over time. This study aimed to analyze the consumption of medically prescribed azole antifungal drugs in mainland Portugal from 2014 to 2023, focusing on those directed to genital infections: fluconazole, isoconazole, itraconazole, and sertaconazole. Methods: For each drug, the evaluated parameters were the total number of packages, number of packages per 1000 inhabitants, defined daily dose (DDD) per 1000 inhabitants per day, and total costs. For this purpose, we used data from community pharmacies’ sales, which are available through INFARMED (the Portuguese national authority on medicines and health products). Results: Several trends emerged from data analysis. The COVID-19 pandemic negatively affected the consumption of all azole antifungal drugs included in this study. However, after 2020, fluconazole and sertaconazole consumption has been increasing. In the specific case of fluconazole, there was an increase in expenditure, although the total number of packages suffered a decrease over the 10-year study period. Additionally, the defined daily dose (DDD) per 1000 inhabitants per day for fluconazole and itraconazole was lower compared to estimates from the last available survey (2009). Conclusions: Although our findings represent a lesser pressure on fungi, further monitoring is needed to better understand the evolution of fluconazole and itraconazole consumption over time, particularly due to the trends observed in this study. Background: Eggshell contamination threatens the viability of hatching eggs. This contamination can be caused by harmless, opportunistic, or pathogenic bacteria. Although necessary, the use of synthetic antibiotics to treat eggshells can present several significant problems: They can be toxic and damage the shell, and, most worryingly, they can lead to bacterial resistance. Faced with these challenges, the objective of this research was to create and test a sanitizing plan for hatching eggs using essential oils derived from Citrus aurantifolia (CAEO), Ocimum basilicum (OBEO), or Allium sativum (ASEO). Methods: Sanitizing solutions containing specific concentrations of these essential oils were prepared, and their antimicrobial properties and contributions to poultry safety and hatching parameters were investigated. Results: The bacterial load was reduced in eggshells sanitized with essential oils, and the degree of bacterial inhibition, along with their safety profile, may be directly related to optimal hatchability rates, lower incidences of contaminated dead embryos, and the hatching of healthy chicks. Conclusions: Together, these results reinforce the importance of essential oils in the development of effective and safe treatments for managing hatching eggs. Full article

Hepatocellular carcinoma (HCC) represents a global health concern, ranking as the sixth most common malignancy worldwide and the third leading cause of cancer-related mortality. Despite advances in research, the diagnosis and prognosis of such malignancy remain challenging. Alpha-fetoprotein, the current serum biomarker used in the management of HCC, has limited sensitivity and specificity, making early detection and effective management more difficult. Thus, new management approaches in diagnosis and prognosis are needed to improve the outcome and survival of HCC patients. SNHG1 is a long noncoding RNA mainly expressed in the cell and cytoplasm of cells and is consistently upregulated in tissues and cell lines of HCC, where it acts as an important regulator of various processes: modulation of p53 activity, sponging of microRNAs with consequent upregulation of their target mRNAs, regulation of fatty acid, iron and glucose metabolism, and interaction with immune cells. The deregulation of these processes results in abnormal cell division, angiogenesis, and apoptosis, thus promoting various aspects of tumorigenesis, including proliferation, invasion, and migration of cells. Clinically, a higher expression of SNHG1 predicts poorer clinical outcomes by significantly correlating with bigger, less differentiated, and more aggressive tumors, more advanced disease stages, and lower overall survival in HCC patients. This article comprehensively summarizes the current understanding of the multifaceted roles of SNHG1 in the pathogenesis of HCC, while also highlighting its clinicopathological correlations, therefore concluding that it has potential as a biomarker in HCC diagnosis and prognosis. While prior research has clearly established links between maternal responses and children’s emotion regulation (ER), the implications of these links for children’s behaviors, especially at school (as reported by their teachers), remain much less explored. This study examined the mediating role of children’s ER in the relationship between maternal reactions to both negative and positive emotions of children and the subsequent behaviors of these children at school. Participants included 56 Portuguese school-aged children (31 boys and 25 girls, aged 6–10 years, mean age = 8.27, SD = 1.27), their mothers (aged 26–55 years, mean age = 38.33, SD = 6.68), and their teachers (n = 7 female teachers) in a multi-informant study. Mothers provided reports on their responses to their children’s emotions and their perceptions of the children’s ER and lability/negativity, while teachers assessed the children’s behavior in the classroom. The results indicated that punitive maternal reactions were associated with greater child lability/negativity, which in turn correlated with increased conduct problems and hyperactivity at school. Conversely, maternal encouragement of expression was linked to reduced lability/negativity, which was associated with fewer emotional symptoms at school. Additionally, maternal problem-focused reactions and guided/empowering responses were associated with reduced child lability/negativity, which in turn correlated with fewer conduct problems and less hyperactivity at school. These findings suggest that maternal responses to children’s emotions can significantly influence children’s behaviors in the classroom via mechanisms involving children’s ER. Full article

Human papillomavirus (HPV)-associated cervical cancer is the most common cancer among women worldwide. The treatment options are strongly related to increased infertility in women. Imiquimod (IQ) is an imidazoquinoline, which has proven antiviral effects against persistent HPV infection by activating immune cells via Toll-like receptors 7/8 when formulated in carriers, like nanogels, for topical use. An effective alternative to conventional therapies is the nanoparticle drug delivery system. We studied lipidic nanoparticles with IQ (Lipo IQ) and functionalized them with a DNA aptamer, AT11 (Lipo IQ AT11), to improve the selectivity for cervical cancer cells combined with the efficacy of essential oils. The formulations showed that the physicochemical properties are adequate for vaginal drug delivery and have antimicrobial activity at higher concentrations (with MIC50 starting from 0.625%). The final formulations exhibited cytotoxicity in cancer cells, enhanced by essential oils without affecting healthy cells, resulting in less than 10% cell viability in HeLa cells and over 60% in NHDF cells. Essential oils potentiate Lipo IQ’s effectiveness, while AT11 increases the selectivity for cervical cancer cells. As suggested by the results of the permeation assay, the formulations were internalized by the cancer cells. Overall, the obtained results suggested that the synergistic effect of the essential oils and the nanosystem potentiate the cytotoxic effect of Lipo IQ and that Lipo IQ AT11 promotes selectivity towards cancer cells. Background: A nationwide, prospective, multicenter, cohort study (the Disease-Related caloric-protein malnutrition EChOgraphy (DRECO) study) was designed to assess the usefulness of ultrasound of the rectus femoris for detecting sarcopenia in hospitalized patients at risk of malnutrition and to define cut-off values of ultrasound measures. Methods: Patients at risk of malnutrition according to the Malnutrition Universal Screening Tool (MUST) underwent handgrip dynamometry, bioelectrical impedance analysis (BIA), a Timed Up and Go (TUG) test, and rectus femoris ultrasound studies. European Working Group on Sarcopenia in Older People (EWGSOP2) criteria were used to define categories of sarcopenia (at risk, probable, confirmed, severe). Receiver operating characteristic (ROC) and area under the curve (AUC) analyses were used to determine the optimal diagnostic sensitivity, specificity, and predictive values of cut-off points of the ultrasound measures for the detection of risk of sarcopenia and probable, confirmed, and severe sarcopenia. Results: A total of 1000 subjects were included and 991 of them (58.9% men, mean age 58.5 years) were evaluated. Risk of sarcopenia was detected in 9.6% patients, probable sarcopenia in 14%, confirmed sarcopenia in 9.7%, and severe sarcopenia in 3.9%, with significant differences in the distribution of groups between men and women (p < 0.0001). The cross-sectional area (CSA) of the rectus femoris showed a significantly positive correlation with body cell mass of BIA and handgrip strength, and a significant negative correlation with TUG. Cut-off values were similar within each category of sarcopenia, ranging between 2.40 cm2 and 3.66 cm2 for CSA, 32.57 mm and 40.21 mm for the X-axis, and 7.85 mm and 10.4 mm for the Y-axis. In general, these cut-off values showed high sensitivities, particularly for the categories of confirmed and severe sarcopenia, with male patients also showing better sensitivities than women. Conclusions: Sarcopenia in hospitalized patients at risk of malnutrition was high. Cut-off values for the better sensitivities and specificities of ultrasound measures of the rectus femoris are established. The use of ultrasound of the rectus femoris could be used for the prediction of sarcopenia and be useful to integrate nutritional study into real clinical practice. Full article

The aetiology of acute appendicitis (AA), the most frequent abdominal surgical emergency, is still unclarified. Recent epidemiologic, clinical and laboratorial data point to an allergic component in the pathophysiology of AA. Mastocytes participate in the Th2 immune response, releasing inflammatory mediators from their granules upon stimulation by IgE-specific antigens. Among the well-known mediators are histamine, serotonin and tryptase, which are responsible for the clinical manifestations of allergies. We conducted a prospective single-centre study to measure histamine and serotonin (commercial ELISA kit) and tryptase (ImmunoCAP System) concentrations in appendicular lavage fluid (ALF) and serum. Consecutive patients presenting to the emergency department with a clinical diagnosis of AA were enrolled: 22 patients with phlegmonous AA and 24 with gangrenous AA The control group was composed of 14 patients referred for colectomy for colon malignancy. Appendectomy was performed during colectomy. Tryptase levels were strikingly different between histological groups, both in ALF and serum (p < 0.001); ALF levels were higher than serum levels. Tryptase concentrations in ALF were 109 times higher in phlegmonous AA (APA) (796.8 (194.1–980.5) pg/mL) and 114 times higher in gangrenous AA (AGA) (837.4 (272.6–1075.1) pg/mL) than in the control group (7.3 (4.5–10.3) pg/mL. For the diagnosis of AA, the discriminative power of serum tryptase concentration was good (AUC = 0.825), but discriminative power was weak (AUC = 0.559) for the differential diagnosis between APA and AGA. Mastocytes are involved in AA during clinical presentations of both phlegmonous and gangrenous appendicitis, and no significant differences in concentration were found. No differences were found in serum and ALF concentrations of histamine and serotonin between histological groups. Due to their short half-lives, these might have elapsed by the time the samples were collected. In future research, these determinations should be made immediately after appendectomy. Our findings confirm the hypersensitivity type I reaction as an event occurring in the pathogenesis of AA: tryptase levels in ALF and serum were higher among patients with AA when compared to the control group, which is in line with a Th2 immune response and supports the concept of the presence of an allergic reaction in the pathogenesis of acute appendicitis. Our results, if confirmed, may have clinical implications for the treatment of AA. Full article

We assessed the accuracy of a prototype radiation detector with a built in CMOS amplifier for use in dosimetry for high dose rate brachytherapy. The detectors were fabricated on two substrates of epitaxial high resistivity silicon. The radiation detection performance of prototypes has been tested by ion beam induced charge (IBIC) microscopy using a 5.5 MeV alpha particle microbeam. We also carried out the HDR Ir-192 radiation source tracking at different depths and angular dose dependence in a water equivalent phantom. The detectors show sensitivities spanning from (5.8 ± 0.021) × 10−8 to (3.6 ± 0.14) × 10−8 nC Gy−1 mCi−1 mm−2. The depth variation of the dose is within 5% with that calculated by TG-43. Higher discrepancies are recorded for 2 mm and 7 mm depths due to the scattering of secondary particles and the perturbation of the radiation field induced in the ceramic/golden package. Dwell positions and dwell time are reconstructed within ±1 mm and 20 ms, respectively. The prototype detectors provide an unprecedented sensitivity thanks to its monolithic amplification stage. Today, type 2 diabetes mellitus (T2DM) and skeletal muscle atrophy (SMA) have become increasingly common occurrences. Whether the onset of T2DM increases the risk of SMA or vice versa has long been under investigation. Both conditions are associated with negative changes in skeletal muscle health, which can, in turn, lead to impaired physical function, a lowered quality of life, and an increased risk of mortality. Poor nutrition can exacerbate both T2DM and SMA. T2DM and SMA are linked by a vicious cycle of events that reinforce and worsen each other. Muscle insulin resistance appears to be the pathophysiological link between T2DM and SMA. To explore this association, our review (i) compiles evidence on the clinical association between T2DM and SMA, (ii) reviews mechanisms underlying biochemical changes in the muscles of people with or at risk of T2DM and SMA, and (iii) examines how nutritional therapy and increased physical activity as muscle-targeted treatments benefit this population. Based on the evidence, we conclude that effective treatment of patients with T2DM-SMA depends on the restoration and maintenance of muscle mass. We thus propose that regular intake of key functional nutrients, along with guidance for physical activity, can help maintain euglycemia and improve muscle status in all patients with T2DM and SMA. Hydrogen-Based Energy Systems: Current Technology Development Status, Opportunities and Challenges

The use of hydrogen as an energy carrier within the scope of the decarbonisation of the world’s energy production and utilisation is seen by many as an integral part of this endeavour. However, the discussion around hydrogen technologies often lacks some perspective on the currently available technologies, their Technology Readiness Level (TRL), scope of application, and important performance parameters, such as energy density or conversion efficiency. This makes it difficult for the policy makers and investors to evaluate the technologies that are most promising. The present study aims to provide help in this respect by assessing the available technologies in which hydrogen is used as an energy carrier, including its main challenges, needs and opportunities in a scenario in which fossil fuels still dominate global energy sources but in which renewables are expected to assume a progressively vital role in the future. The production of green hydrogen using water electrolysis technologies is described in detail. Various methods of hydrogen storage are referred, including underground storage, physical storage, and material-based storage. Hydrogen transportation technologies are examined, taking into account different storage methods, volume requirements, and transportation distances. Lastly, an assessment of well-known technologies for harnessing energy from hydrogen is undertaken, including gas turbines, reciprocating internal combustion engines, and fuel cells. It seems that the many of the technologies assessed have already achieved a satisfactory degree of development, such as several solutions for high-pressure hydrogen storage, while others still require some maturation, such as the still limited life and/or excessive cost of the various fuel cell technologies, or the suitable operation of gas turbines and reciprocating internal combustion engines operating with hydrogen. Costs below 200 USD/kWproduced, lives above 50 kh, and conversion efficiencies approaching 80% are being aimed at green hydrogen production or electricity production from hydrogen fuel cells. Nonetheless, notable advances have been achieved in these technologies in recent years. For instance, electrolysis with solid oxide cells may now sometimes reach up to 85% efficiency although with a life still in the range of 20 kh. Conversely, proton exchange membrane fuel cells (PEMFCs) working as electrolysers are able to sometimes achieve a life in the range of 80 kh with efficiencies up to 68%. Regarding electricity production from hydrogen, the maximum efficiencies are slightly lower (72% and 55%, respectively). The combination of the energy losses due to hydrogen production, compression, storage and electricity production yields overall efficiencies that could be as low as 25%, although smart applications, such as those that can use available process or waste heat, could substantially improve the overall energy efficiency figures. Despite the challenges, the foreseeable future seems to hold significant potential for hydrogen as a clean energy carrier, as the demand for hydrogen continues to grow, particularly in transportation, building heating, and power generation, new business prospects emerge. However, this should be done with careful regard to the fact that many of these technologies still need to increase their technological readiness level before they become viable options. For this, an emphasis needs to be put on research, innovation, and collaboration among industry, academia, and policymakers to unlock the full potential of hydrogen as an energy vector in the sustainable economy.
