Perilla frutescens Britton var. acuta Kudo leaves are widely consumed in East Asia due to their culinary and medicinal properties, which are largely attributed to their high levels of bioactive metabolites such as rosmarinic acid. In this study, we investigated the variation in rosmarinic acid content and overall metabolite profile of P. frutescens leaves collected from six different provinces in Republic of Korea. Quantitative analysis of the rosmarinic acid content was performed using HPLC, revealing significant regional differences, with the highest concentration observed in the leaves collected in Gyeongsangbuk-do and the lowest concentration in those from Jeollanam-do. HRESIMS and 1H-NMR spectrometry were used to determine the chemical structure of the isolated rosmarinic acid. LC-Q-TOF/MS analysis identified ten major metabolites, including phenolic acids, flavonoids, and triterpenoids. Multivariate statistics (OPLS-DA) revealed distinct clustering of populations, indicating a strong relationship between metabolites and environmental parameters. The distribution of the metabolite fingerprints and rosmarinic acid contents in P. frutescens leaves were also found to differ according to the cultivation region, suggesting that secondary metabolite expression is influenced by environmental and geographic factors. This work shows that metabolome profiles can be used in quality control and the development of high-quality products derived from P. frutescens. Full article

The fatty acid composition of some seed oils from plants of emerging interest was studied. The benefits towards human health were evaluated by taking into account current recommendations regarding dietary intake of essential, polyunsaturated, and monounsaturated fatty acids and by discussing the pathologies for which such fatty acids exert protective action. Species studied were Hemp (Cannabis sativa), Flax (Linum usitatissimum), Milk Thistle (Silybum marianum), Perilla (Perilla frutescens), Borage (Borago officinalis), and Black Cumin (Nigella sativa). Seeds were subjected to cold milling in order to maintain their original nutritional characteristics. Chemical analyses were performed via the dual-detector gas chromatography technique by means of Flame Ionization Detection (FID) and mass spectrometry (MS) and by applying a modified version of the AOAC Official Method 991.39, thanks to which it was possible to obtain the fatty acid composition expressed as mg per gram of oil: such information is not always available in the literature for the species studied here. Comparison with the fatty acid international guidelines about the recommended intakes in g/day was made. This allowed us to evaluate whether such oils are suitable to be used as fatty acid food supplements to rebalance the Western diet, which is shown to be inadequate by numerous studies. Results show that seed oils from Cannabis sativa, Linum usitatissimum, and Perilla frutescens are suitable to be used as food supplements while seed oils from Silybum marianum, Borago officinalis, and Nigella sativa are not. It is important to note that any possible benefits from other parts of the plant (leaves, stems, flowers, and roots) are not studied or questioned by the present research, which focuses solely on fatty acids in the oil extracted from the seeds. Perilla seed oil (PSO) possesses various physiological functions, such as lowering blood lipids and preventing cancer; however, its poor water solubility, dispersibility, and oxidative stability severely limit its application scope. Epigallocatechin gallate (EGCG) is a natural antioxidant abundant in tea leaves. In this study, PSO–casein–EGCG microemulsions were prepared, and their stability and lipid-lowering effects were evaluated. The results showed that the PSO microemulsion had a particle size of 361.23 ± 14.85 nm, a zeta potential of −20.77 ± 0.68 mV, a polydispersity index (PDI) of 0.17 ± 0.07, and an encapsulation efficiency of 94.3%. PSO microemulsions remained stable at room temperature for 5 days without droplet aggregation. The stability of the microemulsions was good when the NaCl concentration was between 0.1 and 1 mM and the pH was between 5 and 9. PSO microemulsions enhanced the oxidative stability of PSO. Additionally, PSO microemulsions significantly reduced triglyceride levels in Caenorhabditis elegans (77.50%, p < 0.005). Finally, it was found that the average lipid droplet size of ZXW618 mutant nematodes decreased by 41.23% after PSO microemulsion treatment. Therefore, PSO microemulsions may reduce fat accumulation in C. elegans by decreasing lipid droplet size. This provides new insights for advancing the application of PSO in the food processing industry. Full article

Pesticide residue analysis of agricultural produce is vital because of associated health concerns, highlighting the need for effective non-destructive techniques. This study introduces a method that combines short-wavelength infrared hyperspectral imaging with spectral unmixing to detect chlorfenapyr and azoxystrobin residues on perilla leaves. Sixty-six leaves were treated with pesticides at concentrations between 0 and 0.06%. The study utilized multicurve resolution-alternating least squares (MCR-ALS), a spectral unmixing method, to identify and visualize the distribution of pesticide residues. This technique achieved lack-of-fit values of 1.03% and 1.78%, with an explained variance of 99% for both pesticides. Furthermore, a quantitative model was developed that integrates insights from MCR-ALS with Gaussian process regression to estimate chlorfenapyr residue concentrations, resulting in a root mean square error of double cross-validation (RMSEV) of 0.0012% and a double cross-validation coefficient of determination (R2v) of 0.99. Compared to other chemometric approaches, such as partial least squares regression and support vector regression, the proposed integrated method decreased RMSEV by 67.57% and improved R2v by 2.06%. The combination of hyperspectral imaging with spectral unmixing offers advancements in the real-time monitoring of agricultural product safety, supporting the delivery of high-quality fresh vegetables to consumers. Background/Objectives: Perilla frutescens var. acuta Kudo, a member of the Lamiaceae family, has been previously reported to reduce neuroinflammation and potentially decrease Aβ plaque accumulation in 5XFAD mice. In this study, we aimed to evaluate the anti-neuroinflammatory potential of a standardized 60% ethanol extract of Perilla leaves (PE), optimized for commercial application. Methods: The inflammatory response was assessed in LPS-stimulated BV2 microglial cells, and the cognitive improvement was evaluated in an AD animal model induced by intracerebroventricular injection of Aβ. Results: Using LPS-stimulated BV2 microglial cells and an Aβ-injected ICR mouse model of Alzheimer’s disease, we found that PE significantly suppressed the LPS-induced production of nitric oxide and pro-inflammatory mediators, including IL-6, TNF-α, NF-κB, iNOS, and COX-2, along with inhibition of JNK and p38 MAPK activation. Furthermore, PE upregulated CREB and BDNF expression. In vivo, PE administration alleviated Aβ-induced cognitive deficits, which were associated with reduced expression of JNK, NF-κB, iNOS, and COX and increased CREB/BDNF signaling in the hippocampus. Behavioral assessments—including passive avoidance, Morris water maze, novel object recognition, and Y-maze tests—confirmed the improvement in cognitive function. Conclusions: Collectively, these findings demonstrate that PE exerts significant anti-neuroinflammatory and neuroprotective effects, supporting its potential as a functional ingredient for cognitive enhancement. Inflammatory arthritis (IA) is a chronic condition marked by joint dysfunction and pain, posing significant challenges for effective drug delivery. This study separated Perilla frutescens leaf-derived exosome-like nanovesicles (PFE) to effectively penetrate the stratum corneum barrier. These nanovesicles and indomethacin (IND) were subsequently developed into a nanogel designed for topical drug delivery systems (PFE-IND-GEL). PFE exhibited a typical vesicular structure with a mean diameter of 98.4 ± 1.3 nm. The hydrodynamic size and zeta potential of PFE-IND-GEL were 129.6 ± 5.9 nm and −17.4 ± 1.9 mV, respectively. Mechanistic investigations in HaCaT keratinocytes showed that PFE significantly downregulated tight junction proteins (ZO-1 and Occludin, p < 0.01) via modulation of the IL-17 signaling pathway, as evidenced by transcriptomic analysis. In a sodium urea crystal-induced rat IA model, the topical application of PFE-IND-GEL significantly reduced joint swelling (p < 0.05) and serum levels of inflammatory cytokines (IL-6, IL-1α, TNF-α) compared to control groups. Histopathological analysis confirmed the marked attenuation of synovial inflammation and cartilage preservation in treated animals. These findings underscore the dual role of PFE as both a topical permeation enhancer and an anti-inflammatory agent, presenting a promising strategy for managing IA. Full article

This study focuses on multiple origins of green-back purple and dual-faced purple Perilla frutescens, employing field cultivation experiments combined with detection methods, such as HPLC, LC-MS, and GC-MS, to compare the differences in yield, quality, and metabolic products of the different colored P. frutescens. The results indicate that green-back purple P. frutescens significantly outperformed dual-faced purple P. frutescens in terms of leaf, stem, and seed yields, while the effective component contents in the leaves and seeds of dual-faced purple P. frutescens are higher than those of dual-faced green P. frutescens. An analysis of the anthocyanin components in P. frutescens leaves and the volatile components in P. frutescens seeds shows that the total anthocyanin content in dual-faced purple P. frutescens leaves is 34.63% higher than that in green-back purple P. frutescens, whereas the total volatile components in the seeds of green-back purple P. frutescens exceeds those in dual-faced P. frutescens by 12.99%. The Mantel test indicates a potential correlation mechanism between the anthocyanin components in P. frutescens leaves and the volatile components in P. frutescens seeds, which are significantly associated with the yield quality of both P. frutescens leaves and seeds. This study found that P. frutescens with blue–green leaves yields more than double-sided purple P. frutescens, although the quality of its leaves and seeds is inferior to that of double-sided purple P. frutescens. Furthermore, the anthocyanin components in P. frutescens leaves and the volatile components in P. frutescens seeds exhibit significant correlations with the yield and quality of both leaves and seeds, offering important insights for the production and application of P. frutescens. Perilla frutescens (Perilla) has great potential for utilization in food and medicine due to the abundance of secondary metabolites, especially phenolic compounds. However, the molecular mechanism underlying phenolic compound synthesis in perilla remains poorly understood. By targeted metabolome analysis, we found nine differentially accumulated metabolites among QS2, QS6, and QO7 leaves and nine unique metabolites which only exist in QS6. Furthermore, transcriptome analysis showed the differential expression genes (DEGs) were significantly enriched into phenylpropanoid- and flavonoid-related pathways. Moreover, the integrated transcriptome and metabolome analyses indicated 14 candidates highly correlated with phenolic compound biosynthesis. In addition, phenylpropanoid- and flavonoid-biosynthesis-related DEGs, including one PAL, one CYP73A, one 4CL, two CHI, two F3H, one FLS, three CHS, two CYP75B1, one ANS, and two DFR, were isolated. The results in this study provide useful information for the metabolic regulation of phenolic compounds and serve as essential gene resources for future breeding programs. Full article

This study investigated the accumulation of nitrate and nitrite, as well as the vitamin C content, in various leafy vegetables, including amaranth greens, katuk, morning glory, squash leaves, vine spinach, leaf lettuce, romaine lettuce, Vietnamese basil, Vietnamese perilla, komatsuna, leeks, and spinach, grown with either organic or synthetic fertilizers. The findings indicate that the type of fertilizer significantly influences nitrate accumulation and vitamin C content in these vegetables. Organic fertilizers were found to reduce nitrate levels and increase vitamin C content in amaranth greens, katuk, morning glory, squash leaves, vine spinach, leaf lettuce, Vietnamese basil, Vietnamese perilla, and spinach compared to the results for synthetic fertilizers. However, high nitrate concentrations in leaf lettuce, komatsuna, and spinach pose potential health risks. The study also identified elevated nitrate levels in vegetables that are not currently regulated. Furthermore, more than half of the samples contained nitrite, for which no maximum permissible level has been established. These findings underscore the importance of organic vegetable cultivation in enhancing both human health and environmental sustainability. Full article

Objectives: This study aimed to identify and develop a novel, safe, and effective transdermal penetration enhancer derived from the leaves of Perilla frutescens (L.) Britt, and to explore the underlying mechanisms of its penetration enhancement effects. Methods: To evaluate the safety profile of the penetration enhancer, both skin irritation tests and histopathological analyses were conducted. The transdermal enhancement capabilities of the penetration enhancer were assessed in vitro using five model drugs. Furthermore, to gain insights into the penetration enhancement mechanism of this novel penetration enhancer, a range of analytical methods were used, including a spectroscopic technique, differential scanning calorimetry, micro-optical techniques, and molecular docking simulations. Results: Perilla essential oil contained 93.70% perilla ketone (PEK), which exhibited a safety profile superior to that of azone. PEK significantly increased the cumulative skin permeation of all the model drugs (p < 0.05). PEK exhibited the most obvious impact on puerarin penetration, with quantitative enhancement ratios of 2.96 ± 0.07 and 3.39 ± 0.21 at concentrations of 3% and 5% (w/v), respectively. A strong correlation between the enhancement effect of PEK and the physicochemical properties of the drugs was observed. Mechanistic studies revealed that PEK facilitates drug distribution from the solution phase to the stratum corneum (SC). Conclusions: PEK, seldom discussed in former studies, was observed to show extensive penetration enhancement effects by inducing conformational changes in SC lipids and disrupting the tightly ordered bilayer arrangement of lipids. These findings highlight the potential of PEK as a promising and safe natural transdermal penetration enhancer. Full article

Background/Objectives: Perilla frutescens has historically been used to protect against inflammation and redox stress. This has been partly attributed to its high polyphenolic content; however, polyphenolic components in Perilla extract remain incompletely defined. This study aimed to characterise the polyphenolic composition in Perilla extract and evaluate its effect on the transcription factor nuclear factor erythroid 2-related factor 2 (Nrf2), regulating antioxidant defenses during inflammation and oxidative stress. Methods: Hot water extraction from Perilla leaves was followed by fractionation using four solvents of different polarity, namely methanol, butanol, ethyl acetate and ether. The polyphenolic composition of these fractions was analysed using RP-HPLC, and some of these compounds were quantified. The total phenolic, flavonoid, and ortho-diphenolic contents of each Perilla fraction were determined. The antioxidant activity was assessed using metal cation reduction and radical scavenging assays. A dual-luciferase assay using a human NQO1 ARE-luciferase reporter plasmid was employed to quantify Nrf2 activation by the Perilla fractions. Results: HPLC analysis identified 35 polyphenolic compounds, with the highest phenolic content present in the polar fractions and rosmarinic acid being the major constituent. Radical scavenging tests (DPPH and ABTS) confirmed the highest antioxidant capacity in the polar fractions. On cells in vitro, the methanol Perilla fraction displayed the strongest antioxidant activity, showing up to a 1.5-fold increase in human NQO1 ARE-luciferase reporter induction. Conclusions: This study has shown that Perilla extract contains a diversity of polyphenolic compounds contributing to its potent antioxidant effects, with methanol and butanol being the most efficient extraction solvents. While rosmarinic acid is expected to be the major contributor towards providing protection against inflammation and redox stress, further work is required on the synergystic effects between different polyphenols. Perilla frutescens (L.) Britt. is a traditional medicinal and culinary plant with a long history of cultivation and significant potential for broader utilization. The basic helix-loop-helix (bHLH) gene family is essential for regulating plant growth, development, stress responses, and secondary metabolism. However, the bHLH gene family in P. frutescens has not yet been characterized. In this study, a total of 205 bHLH genes were identified in P. frutescens through genome mining and analysis. Phylogenetic analysis classified these PfbHLH genes into 23 distinct subfamilies. Promoter analysis revealed an enrichment of cis-acting elements linked to plant hormone signaling and stress responses, suggesting their potential regulatory roles in development, growth, and stress adaptation. Expression profiling based on publicly available RNA-seq data demonstrated tissue-specific expression patterns of PfbHLH genes in roots, stems, and leaves. Four PfbHLH genes (PfbHLH66, PfbHLH45, PfbHLH13, and PfbHLH5) showed significant responses to methyl jasmonate (MeJA) induction. Yeast one-hybrid assays confirmed that these PfbHLH proteins could bind to the cis-acting G-box (CACGTG) element. This study offers new perspectives on the evolution, regulatory mechanisms, and functional roles of the bHLH gene family in P. frutescens. The findings deepen our understanding of the functional diversity within this gene family and establish a foundation for genetic enhancement and the biosynthesis of medicinal compounds in the species. Leafy vegetables, mainly lettuces, are currently the main crop cultivated in controlled environment agriculture (CEA), including vertical farming and plant factories. There is a rising demand to expand this portfolio with a wider variety of underutilized edible plants containing various bioactive compounds and sensory properties seeking to enrich human diets. However, the optimal cultivation conditions for these underutilized plants significantly differ from those optimized for lettuce, basil, and other popular CEA crops. Therefore, this study aims to explore the impacts of light-emitting diode (LED) lighting intensity (photosynthetic photon flux density, PPFD) on green leaf Perilla frutescens cultivated in CEA. Plants were grown under four levels of LED lighting PPFDs from 150 to 300 µmol m−2s−1 for 4 weeks. Plant biomass productivity, soluble sugar contents, antioxidant properties (DPPH, ABTS free radical scavenging activities, FRAP antioxidant power), and total contents of phenolic compounds in leaves were evaluated at harvesting time. Further, harvested plant material was stored in the dark, at +6 °C, and the water content, water loss and transpiration rate, leaf sugar contents, and antioxidant properties were monitored 1, 3, and 5 days after harvesting. The summarized data suggest that higher cultivation lighting PPFD results in better harvest quality preservation during post-harvest storage. Full article

Plant breeding has evolved significantly over time with the development of transformation and genome editing techniques. These new strategies help to improve desirable traits in plants. Perilla is a native oil crop grown in Korea. The leaves contain many secondary metabolites related to whitening, aging, antioxidants, and immunity, including rosmarinic acid, vitamin E, luteolin, anthocyanins, and beta-carotene. They are used as healthy and functional food ingredients. It is an industrially valuable cosmetics crop. In addition, perilla seeds are rich in polyunsaturated fatty acids, such as α-linolenic acid and linoleic acid. They are known to be effective in improving neutral lipids in the blood, improving blood circulation, and preventing dementia and cardiovascular diseases, making them excellent crops whose value can be increased through improved traits. This research will also benefit perilla seeds, which can increase their stock through various methods, such as the increased production of functional substances and improved productivity. Recently, significant attention has been paid to trait improvement research involving gene-editing technology. Among these strategies, CRISPR/Cas9 is highly adaptable, enabling accurate and efficient genome editing, targeted mutagenesis, gene knockouts, and the regulation of gene transcription. CRISPR/Cas9-based genome editing has enormous potential for improving perilla; however, the regulation of genome editing is still at an early stage. Therefore, this review summarizes the enhancement of perilla traits using genome editing technology and outlines future directions. Full article

(1) Background: This study aimed to outline the antioxidant, antitumoral, and cytotoxic proprieties of various types of Perilla frutescens extracts obtained from the leaves of the species. (2) Methods: We determined total polyphenols, flavonoids and anthocyanins contents, as well as the in vitro antioxidant, antitumoral, and cytotoxic actions in three types of ethanolic extracts (E1, E2, E3) and in three types of acetone: ethanol extracts (A1, A2, A3) of Perilla frutescens according to standardized procedures. (3) Results: We found that Perilla frutescens ethanolic extracts had the highest total phenol and anthocyanins concentrations. The flavonoids concentration was not statistically different between the extracts. The iron chelating capacity, hydroxyl radical scavenging capacity, superoxide anion radical scavenging capacity, and lipoxygenase inhibition capacity showed a significant increase with higher concentrations of Perilla frutescens extracts, particularly the ethanolic extracts. Perillyl alcohol had greater cytotoxic capacity in the MG-63 cell line and E1 extract showed similar significant cytotoxic effects in the A431 cell line. (4) Conclusions: Both ethanolic and acetone–ethanol extracts from Perilla frutescens exhibited important antioxidant and antitumoral actions in vitro, which proportionally increased with concentration. The cytotoxic threshold determined in this study for various types of extracts could help determine the best dosage with the maximum antioxidant and antitumoral potential. Our results could serve as a basis for further studies that will investigate the cytotoxic effects of Perilla frutescens variants on various types of cancer cell lines. Full article
