Temperature and Precipitation Extremes Under SSP Emission Scenarios with GISS-E2.1 Model

Atmospheric warming results in increase in temperatures for the mean, the coldest, and the hottest day of the year, season, or month. Global warming leads to a large increase in the atmospheric water vapor content and to changes in the hydrological cycle, which include an intensification of precipitation extremes. Using the GISS-E2.1 climate model, we present the future changes in the coldest and hottest daily temperatures as well as in extreme precipitation indices (under four main Shared Socioeconomic Pathways (SSPs)). The increase in the wet-day precipitation ranges between 6% and 15% per 1 °C global surface temperature warming. Scaling of the 95th percentile versus the total precipitation showed that the sensitivity for the extreme precipitation to the warming is about 10 times stronger than that for the mean total precipitation. For six precipitation extreme indices (Total Precipitation, R95p, RX5day, R10mm, SDII, and CDD), the histograms of probability density functions become flatter, with reduced peaks and increased spread for the global mean compared to the historical period of 1850–2014. The mean values shift to the right end (toward larger precipitation and intensity). The higher the GHG emission of the SSP scenario, the more significant the increase in the index change. We found an intensification of precipitation over the globe but large uncertainties remained regionally and at different scales, especially for extremes. Over land, there is a strong increase in precipitation for the wettest day in all seasons over the mid and high latitudes of the Northern Hemisphere. There is an enlargement of the drying patterns in the subtropics including over large regions around Mediterranean, southern Africa, and western Eurasia. For the continental averages, the reduction in total precipitation was found for South America, Europe, Africa, and Australia, and there is an increase in total precipitation over North America, Asia, and the continental Russian Arctic. Over the continental Russian Arctic, there is an increase in all precipitation extremes and a consistent decrease in CDD for all SSP scenarios, with the maximum increase of more than 90% for R95p and R10 mm observed under SSP5–8.5. Next to adrenergic signalling, purinergic pathways mediated by extracellular adenine nucleotides have been described to shape thermogenic and metabolic functions in brown adipose tissue (BAT). Here we describe high expression of P2X5 that is activated by ATP in mature adipocytes of BAT and differentiated brown adipocytes in vitro. The levels of other P2X family members were much lower, or expression was restricted to tissue-resident macrophages or endothelial cells. Global and brown adipocyte-specific P2rx5 deficiency resulted in lower expression of the uncoupling protein 1 (UCP1). However, indirect calorimetry studies showed that P2X5 did not affect systemic energy expenditure. Of note, glucose tolerance was impaired under chow and obesogenic high-fat diet conditions, which can be explained by lower glucose disposal into BAT but not into other organs. In summary, these data indicate a modulatory role of P2X5 in systemic and BAT-specific glucose metabolism. Full article

Accelerating climate change poses significant risks to water security and ecological stability in arid regions due to the increasing frequency and intensity of extreme precipitation events. As a climate-sensitive area, the inland river basin (IRB) of Northwest China—a critical water source for local ecosystems and socioeconomic activities—remains insufficiently studied in terms of future extreme precipitation dynamics. This study evaluated the spatiotemporal evolution of extreme precipitation in the IRB under a new low radiative forcing scenario (SSP1-1.9) by employing four global climate models (GCMs: GFDL-ESM4, MRI-ESM2, MIROC6, and IPSL-CM6A-LR). Eight core extreme precipitation indices were analyzed to quantify changes during the near future (NF: 2021–2050) and far future (FF: 2071–2100) periods. Our research demonstrated that all four models were capable of capturing seasonal patterns and exhibited inherent uncertainty. The annual total precipitation (PRCPTOT) in mountainous regions showed minimal variation, while desert areas were projected to experience a 2-6-fold increase in precipitation in the NF and FF. The Precipitation Intensity Index (SDII) weakened by approximately −10% in mountainous areas but strengthened by around +10% in desert regions. Most mountainous areas showed an increase in the maximum consecutive dry days (CDD), whereas desert regions exhibited extended maximum consecutive wet days (CWD). Moderate rainfall (P1025) variations primarily ranged between −5% and +20%, with greater fluctuations in desert areas. Heavy rainfall (PG25) fluctuated between −40% and +40%, reflecting stark contrasts in extreme precipitation between arid basins and mountainous zones. The maximum 1-day precipitation (Rx1day) and maximum 5-day precipitation (Rx5day) both showed significant increases, which indicated heightened risks from extreme rainfall events in the future. Moreover, the IRB region experienced increased total precipitation, enhanced rainfall intensity, more frequent alternations between drought and precipitation, more frequent moderate-to-heavy rainfall days, and higher daily precipitation extremes in both the NF and FF periods. Objectives: The aim of this observational study was to evaluate whether the presence of periapical inflammatory cysts (PIC) is accompanied by a state of vitamin D (25OHD) 25(OH)D insufficiency or deficiency and biochemical variations in biomarkers of bone metabolism such as osteocalcin (OC), isoenzyme of bone alkaline phosphatase (BALP), and C-terminal telopeptide of type 1 collagen (CTX). Methods: A total of 56 patients (group P), 36 males and 20 females, of which 42 had one cyst (group P1) and 14 had multiple periapical cysts (group P2), alongside 56 healthy subjects (group H) were recruited. Rx-OPT and clinical evaluation were used to evaluate the presence of PIC. At the first visit, all subjects underwent venous sampling (group P and H) to measure bone biomarkers by the chemiluminescence method. The Mann–Whitney test was used to compare the different biomarkers in the H vs. P, H vs. P1, H vs. P2, and P1 vs. P2 groups. The Mann–Whitney test was used to compare biomarker levels between the study groups. ROC curves were used to search for the concentration of the different biomarkers in which the best sensitivity and specificity were found. Results: 25OHD and CTX showed a difference between H vs. P, H vs. P1, H vs. P2, and P1 vs. P1 groups (p < 0.05). The study of the ROC curves with a comparison between concentrations in the H vs. P group showed the best sensitivity and specificity for 25OHD at a concentration <19 ng/mL, highlighting a picture of 25OHD deficiency. Conclusions: The presence of apical cysts could be indicative of a vitamin D deficiency that should be appropriately treated. The findings suggest that vitamin D deficiency, given its role in bone metabolism and mineralisation, may contribute to a biological environment that favours the development or persistence of periapical cystic lesions. Rhodotorula mucilaginosa plays a key role in developing the taste of dry-cured ham, while the mechanism of Rhodotorula mucilaginosa proteases on myofibrillar protein (MP) hydrolysis and the evolution of taste substances has not been studied. The enzymatic characteristics, hydrolysis capacities for MPs, free amino acid contents, metabolite compositions, and taste attributes were investigated during the interactions of MPs and proteases. The proteases of R. mucilaginosa EIODSF019 (RE) and R. mucilaginosa XZY63-3 (RX) showed high hydrolytic activities at the conditions of pH 5.0~7.0 and 30~40 °C. Compared with RX, RE showed a lower Michaelis constant (Km) value and a better affinity for protein substrates. RE showed a higher capability to degrade myosin and actin compared with RX and P. kudriavzevii XS-5 proteases (PK). The microtopography of enzyme-treated MPs in RE presented a smoother surface and lower root mean square roughness than that in RX and PK. The total content of free amino acids significantly increased from 0.34 mg/100 mL of CK to 17.10 mg/100 mL of RE after 4 h of hydrolysis of MPs. Sixty-two metabolites were identified by LC-MS/MS, and γ-glutamyl peptides were the main components of MP hydrolysates. Sensory scores of umami, richness, and aftertaste showed the largest values in RE among these groups. Partial least squares discriminant analysis and correlation network demonstrated that γ-Glu-Lys, γ-Glu-Tyr, γ-Glu-Glu, γ-Glu-His, γ-Glu-Leu, γ-Glu-Cys, γ-Glu-Ala, and γ-Glu-Gln were positively correlated with the improvements of umami, richness, and aftertaste in RE. Background: Low birth weight in newborns is of multifactorial origin (fetal, maternal, placental, and environmental factors), and in one-third of cases, the cause is of unknown origin, with high infant morbidity and mortality. The main treatment for regaining weight and height in children with low birth weight is the application of growth hormones. However, their role as a protective factor to prevent an increase in body composition and the development of metabolic diseases is still poorly understood. Methodology: A case–control study was conducted in a cohort of patients consulted at the CES Pediatric Endocrinology Clinic, Medellín, Colombia, between 2008 and 2018. We evaluated sociodemographic and clinical variables. Additionally, the identification of differential patterns of genomic methylation between cases (treated with growth hormone) and controls (without growth hormone treatment) was performed. The groups were compared using Fisher’s exact test for qualitative variables and Student’s t-test for the difference in means in independent samples. The correlation was evaluated with the Pearson coefficient. Results: Regarding clinical manifestations, body mass index (BMI) was higher in children who did not receive growth hormone treatment, higher doses of growth hormone treatment helped reduce body mass index (R: −0.21, and p = 0.067), and the use of growth hormone was related to a decrease in triglyceride blood concentrations (p = 0.06); these results tended towards significance. Regarding genome-wide methylation patterns, the following genes were found to be hypermethylated: MDGA1, HOXA5, LINC01168, ZFYVE19, ASAH1, MYH15, DNAJC17, PAMR1, MROCKI, CNDP2, CBY2, ZADH2, HOOK2, C9orf129, NXPH2, OSCP1, ZMIZ2, RUNX1, PTPRS, TEX26, EIF2A4K, MYO1F, C2orf69, and ZSCAN1. Meanwhile, the following genes were found hypomethylated: C10orf71-AS1, ZDHHC13, RPL17, EMC4, RPRD2, OBSCN-AS1, ZNF714, MUC4, SUGT1P4, TRIM38, C3, SPON1, NGF-AS1, CCSER2, P2RX2, LOC284379, GGTA1, NLRP5, OR51A4, HLA-H, and TTLL8. Conclusions: Using growth hormone as a treatment in SGA newborns helps regain weight and height. Additionally, it could be a protective factor against the increase in adolescent body composition. The emergence of the AI era driven by Large Language Models (LLMs) and the next-generation high-definition multimedia interface for immersive technologies (AR/VR/metaverse) have created an unprecedented demand for high-bandwidth interconnects. While optical communication systems provide a broad bandwidth, their relatively low power efficiency continues to limit their deployment in new applications. This work addresses the power efficiency challenges in CMOS optical transceiver design, leveraging the inherent cost and integration advantages of CMOS technology. After outlining the design principles for low-power optical transmitter (Tx) and receiver (Rx) design, we present a comprehensive design of a low-power optical transceiver chipset implemented in 28 nm CMOS. The Tx features a high-impedance asymmetric current-steering output stage with a stacked architecture that facilitates unipolar power supply operation for the efficient anode driving of a common-cathode VCSEL array and achieved a power efficiency of 1.59 pJ/bit. The Rx incorporates a tail-current-controlled Cherry–Hooper-based variable gain amplifier (VGA), which achieved a transimpedance gain that ranged from 68.4 to 78.5 dBΩ and a power efficiency of 1.06 pJ/bit. The Rx–Tx back-to-back measurements confirmed successful data transmission at 4 × 20 Gbps, which demonstrated an overall power efficiency of 2.65 pJ/bit. Full article

Background: This study aimed to develop a model for understanding stress-induced sleep disturbances and to explore the potential interactions between sleep disturbances and mood disturbances. Methods: The chronic unpredictable mild stress (CUMS) group was established using the CUMS method, while the CUMS+Noise group was subjected to an additional 8-h exposure to noise in conjunction with the CUMS protocol. Each group was tested for anxiety and depressive-like behavior using the open-field, elevated plus maze, tail suspension, and forced swimming tests in male C57BL/6J mice. Subsequently, we assessed sleep status using sleep recordings and a standardized scoring system alongside the pentobarbital sodium-induced sleep test. Results: The mice in both model groups exhibited anxiety-like behavior. Sleep disturbances observed in the CUMS+Noise group were characterized by disruptions in sleep duration and circadian rhythm. This observation was supported by a marked reduction in multiple sleep time intervals and single sleep duration, as well as a significant increase in sleep duration at the final time interval of ZT23-24. To further investigate the potential mechanisms of interaction, we conducted an analysis of hub genes present in the hippocampal sequencing data utilizing weighted gene co-expression network analysis (WGCNA). Pearson correlation analysis revealed a significant association between the hub genes Alb, P2rx1, and Npsr1 and key phenotypic traits. However, PCR experiments indicated that only Alb showed a significant difference, which aligns with the sequencing results. Conclusions: Albumin is a crucial transporter protein for thyroid hormones and plays a vital role in their metabolism. The interaction between sleep disorders and anxiety-like behavior may be closely linked to the dysfunctional transportation of thyroid hormones by albumin. Full article

Understanding historical climatic extremes and variability is crucial for effective climate change adaptation, particularly for urban flood management in developing countries. This study investigates historical precipitation trends in the Kathmandu Valley, Nepal, focusing on precipitation frequency, intensity, and the influence of the El Niño–Southern Oscillation (ENSO), using extreme precipitation indices and the precipitation concentration index (PCI). The results reveal sharply fluctuating short-term precipitation from 1980 to 2022, with the exception of an increasing trend during spring (1.17 mm/year) and a decreasing trend in November and December. Trends in extreme precipitation indices are mixed: RX7day shows an increasing trend of 0.1 mm/year, with decadal analysis (1980–2001 and 2002–2022) indicating similar upward patterns. In contrast, RX1day, RX3day, RX5day, and R95pTOT exhibit inconsistent trends, while R99pTOT demonstrates a decreasing trend over the full period (1980–2022). Although the number of days with precipitation ≥ 35 mm has declined, the increasing trend in 7-day maximum precipitation, coupled with no significant change in total annual precipitation and highly variable short-term rainfall, points to a rising risk of unexpected extreme precipitation events. Precipitation patterns in the Kathmandu Valley remain highly irregular across seasons, except during summer. ENSO exhibits a negative correlation with annual precipitation, extreme precipitation indices, and the PCI but shows a positive correlation with the annual and summer PCI as well as 1-day maximum precipitation, emphasizing its significant influence on precipitation variability. These findings highlight the urgent need for targeted climate adaptation strategies and provide valuable insights for hydrologists, meteorologists, policymakers, and urban planners to enhance climate resilience and improve flood management in the Kathmandu Valley. Accurate simulation of extreme precipitation events is crucial for managing climate-vulnerable sectors in Southern Africa, as such events directly impact agriculture, water resources, and disaster preparedness. However, global climate models frequently struggle to capture these phenomena, which limits their practical applicability. This study investigates the effectiveness of three bias correction techniques—scaled distribution mapping (SDM), quantile distribution mapping (QDM), and QDM with a focus on precipitation above and below the 95th percentile (QDM95)—and the daily precipitation outputs from 11 Coupled Model Intercomparison Project Phase 6 (CMIP6) models. The Climate Hazards Group Infrared Precipitation with Stations (CHIRPS) dataset was served as a reference. The bias-corrected and native models were evaluated against three observational datasets—the CHIRPS, Multi-Source Weighted Ensemble Precipitation (MSWEP), and Global Precipitation Climatology Center (GPCC) datasets—for the period of 1982–2014, focusing on the December-January-February season. The ability of the models to generate eight extreme precipitation indices developed by the Expert Team on Climate Change Detection and Indices (ETCCDI) was evaluated. The results show that the native and bias-corrected models captured similar spatial patterns of extreme precipitation, but there were significant changes in the amount of extreme precipitation episodes. While bias correction generally improved the spatial representation of extreme precipitation, its effectiveness varied depending on the reference dataset used, particularly for the maximum one-day precipitation (Rx1day), consecutive wet days (CWD), consecutive dry days (CDD), extremely wet days (R95p), and simple daily intensity index (SDII). In contrast, the total rain days (RR1), heavy precipitation days (R10mm), and extremely heavy precipitation days (R20mm) showed consistent improvement across all observations. All three bias correction techniques enhanced the accuracy of the models across all extreme indices, as demonstrated by higher pattern correlation coefficients, improved Taylor skill scores (TSSs), reduced root mean square errors, and fewer biases. The ranking of models using the comprehensive rating index (CRI) indicates that no single model consistently outperformed the others across all bias-corrected techniques relative to the CHIRPS, GPCC, and MSWEP datasets. Among the three bias correction methods, SDM and QDM95 outperformed QDM for a variety of criteria. Among the bias-corrected strategies, the best-performing models were EC-Earth3-Veg, EC-Earth3, MRI-ESM2, and the multi-model ensemble (MME). These findings demonstrate the efficiency of bias correction in improving the modeling of precipitation extremes in Southern Africa, ultimately boosting climate impact assessments. Full article

Background: Pekin ducks are well-known meat-type ducks, whereas Shaoxing ducks are bred for their egg-laying abilities. Growth and development of poultry species is well studied; however, very little is known regarding differences in intestinal gene expression between Pekin and Shaoxing ducks. Methods: To investigate intestinal differences between Pekin and Shaoxing ducks, we conducted transcriptome analysis on ileal and cecal tissues from five 42-day-old ducks per breed, raised under identical housing and feeding conditions to minimize environmental influences. Results: The results showed that a total of 379 differentially expressed genes (DEGs) with p < 0.05 and |log2FoldChange| > 1 were identified in the ileum when Pekin ducks were compared to Shaoxing ducks, among which 158 were upregulated and 221 were downregulated. Compared to Shaoxing ducks, a total of 367 DEGs with p < 0.05 and |log2FoldChange| > 1 were identified in the ceca of Pekin ducks, among which 204 were upregulated and 163 were downregulated. Among these DEGs, nine genes were reported to be associated with growth and metabolism, namely, P2RX6, KCNJ6, CASQ2, EHHADH, ACSBG1, ELOVL4, AIF1L, VILL, and FABP1. Functional enrichment analyses using the Gene Ontology (GO) and Kyoto Encyclopedia of Genes and Genomes (KEGG) databases indicated that the DEGs were significantly involved in pathways such as calcium signaling, unsaturated fatty acid biosynthesis, fatty acid degradation, and tryptophan metabolism. Conclusions: In conclusion, our study identified transcriptome differences in the intestines of meat-type and laying-type ducks, offering insights into the genetic basis of their growth and metabolic differences. Future studies should validate key genes and explore environmental influences on gene expression. Full article

Crohn’s disease is an inflammatory bowel disease (IBD) that currently lacks satisfactory treatment options. Therefore, new targets for new drugs are urgently needed to combat this disease. In the present study, we investigated the transcriptomics-based mRNA expression of intestinal biopsies from patients with Crohn’s disease. We compared the mRNA expression profiles of the ileum and colon of patients with those of healthy individuals. A total of 72 genes in the ileum and 33 genes in the colon were differentially regulated. Among these, six genes were overexpressed in both tissues, including IL1B, TCL1A, HCAR3, IGHG1, S100AB, and OSM. We further focused on OSM/oncostatin M. To confirm the responsiveness of intestinal tissues from patients with Crohn’s disease to oncostatin M inhibition, we examined the expression of the oncostatin M using immunohistochemistry in patient biopsies as well as in kindlin-1−/− and kindlin-2−/− knockout mice, which exhibit an inflammatory bowel disease (IBD) phenotype, and found strong oncostatin M expression in all samples examined. Next, we conducted a drug-repurposing study using the supercomputer MOGON and bioinformatic methods. A total of 13 candidate compounds out of 1577 FDA-approved drugs were identified by PyRx-based virtual drug screening and AutoDock-based molecular docking. Their lowest binding energies (LBEs) ranged from −10.46 (±0.08) to −8.77 (±0.08) kcal/mol, and their predicted inhibition constants (pKi) ranged from 21.62 (±2.97) to 373.78 (±36.78) nM. Ecamsule has an interesting stereostructure with two C2-symmetric enantiomers (1S,4R-1′S,4′R and 1R,4S-1′R,4′S) (1a and 1b) and one meso diastereomer (1S,4R-1′R,4′S) (1c). These three stereoisomers showed strong, albeit differing, binding affinities in molecular docking. As examined by nuclear magnetic resonance and polarimetry, the 1S,4R-1′S,4′R isomer was the stereoisomer present in our commercially available preparations used for microscale thermophoresis. Ecamsule (1a) was chosen for in vitro validation using recombinant oncostatin M and microscale thermophoresis. Considerable dissociation constants were obtained for ecamsule after three repetitions with a Kd value of 11.36 ± 2.83 µM. Subsequently, we evaluated, by qRT-PCR, the efficacy of ecamsule (1a) as a potential drug that could prevent oncostatin M activation by inhibiting downstream inflammatory marker genes (IL6, TNFA, and CXCL11). In conclusion, we have identified oncostatin M as a promising new drug target for Crohn’s disease through transcriptomics and ecamsule as a potential new drug candidate for Crohn’s disease through a drug-repurposing approach both in silico and in vitro. In this study, a single-pole double-throw (SPDT) switch for Sub-6 GHz application is designed. In particular, a shunt inductor is connected to the antenna port of the switch for ESD (electrostatic discharge) protection in RF (radio frequency) front end module. The shunt inductor not only serves as an ESD protection device, but also serves as a component of a parallel resonance circuit to suppress insertion loss of the switch. In addition, in order to secure the power handling capability, transistors turned off in the transmit (Tx) mode are implemented as quadruple-gate transistors. An SPDT switch is fabricated using GaAs pHEMT provided in the 500 nm GaAs BiFET process to verify the feasibility of the proposed switch structure. The operating frequency is set from 3 GHz to 5 GHz. The insertion loss and isolation measured in the Tx mode are lower than 0.35 dB and higher than 31.6 dB, respectively. The insertion loss and isolation measured in the Rx mode are lower than 0.32 dB and higher than 33.9 dB, respectively. The chip size including test pads is 0.890 × 0.875 mm2. Full article

Investigating the Spatiotemporal Variation in Extreme Precipitation Indices in Iran from 1990 to 2020

This study examines the spatiotemporal characteristics of extreme precipitation indices in Iran. It analyzes data from 38 synoptic stations across the country, covering the period from 1990 to 2020, focusing on the 11 most common extreme precipitation indices defined by the Expert Team on Climate Change Detection and Indices (ETCCDI). The analysis employs the Mann–Kendall (M–K) trend test. The findings indicate that the indices PRCPTOT (annual total precipitation), R20 mm (very heavy precipitation days), R10 mm (heavy precipitation days), R25 mm (number of wet days), Rx1 day (maximum 1-day precipitation), Rx5 day (maximum 5-day precipitation), SDII (simple daily intensity index), R95p (very wet day precipitation), R99p (extremely wet day precipitation), and CWDs (consecutive wet days) showed the highest values in the northern and western regions of the country, particularly at stations like Ramsar, Hamedan, Ilam, Kermanshah, and Yasouj. Conversely, the eastern and southeastern parts of the country showed the lowest values for these indices. The Consecutive Dry Day (CDD) index exhibited the highest values at Zabol station (228 days) and Abadan station (193 days) in the southern region of the country. Generally, precipitation extremes in the western, northwestern, and Caspian Sea coasts showed an increasing trend, while the eastern, southeastern, and central parts of the country demonstrated a decreasing trend. The trend test results indicate significant mutations in all precipitation indices, except for SDII, with mutation points primarily occurring during the decade from 2000 to 2010. The magnitude of mutation for each index post-mutation is generally greater than before. This study provides valuable information for decision-makers in agriculture, food security, water, and the environment. It also serves as a resource for natural disaster prevention and mitigation. Trends of Extreme Precipitation Events in Serbia Under the Global Warming

This paper examines extreme precipitation events (EXPEs) and their trends based on daily precipitation values observed at 14 stations in Serbia for the period 1961–2020. The following EXPEs were investigated: RR10mm (heavy precipitation days), RR20mm (very heavy precipitation days), Rx1day (highest 1-day precipitation amount), Rx3day (highest 3-day precipitation amount), Rx5day (highest 5-day precipitation amount), R95p (very wet days) and R99p (extremely wet days). A positive trend for all EXPEs was dominant in Serbia from 1961 to 2020. All annual Rx1day time series show a positive trend, which is significant at 12 out of 14 stations. The highest values of all EXPEs were observed in 2014, when the annual precipitation totals were the highest at almost all stations in Serbia. To examine the potential influence of global warming, the mean values of the EXPEs were calculated for two periods: 1961–1990 and 1991–2020. In the second period, higher values were determined for all EXPEs than in the first period. The large-scale variability modes, such as the North Atlantic Oscillation (NAO), the East Atlantic Oscillation (EA), and the East Atlantic–West Russia (EAWR) pattern, were correlated with the EXPEs. A negative correlation was found between the EXPEs and the NAO and the EAWR, and a positive correlation between the EXPEs and the EA pattern. For future research, the contribution of high-resolution data will be examined. Full article
