Helichrysum arenarium (L.) Moench is a European native species typically associated with dry, sunny, sandy, and nutrient-poor habitats. In European herbal practice, preparations from H. arenarium inflorescences are traditionally used for the symptomatic relief of digestive complaints. Polyphenolic constituents, particularly flavonoid and chalcone derivatives, are among the most characteristic compounds detected in the inflorescences. Intensive harvesting and degradation of natural habitats have increased pressure on wild populations. Since H. arenarium is legally protected in many countries, biotechnological approaches for plant biomass production represent a promising alternative. In the present study, shoot and callus cultures were established, and detailed extract profiling was performed using ultra-high-performance liquid chromatography coupled with diode-array detection and electrospray ionization multistage mass spectrometry (UHPLC-DAD-ESI-MS3). Cytotoxic, antioxidant, anti-inflammatory, wound-healing, anti-collagenase, and anti-hyaluronidase activities were also evaluated. In vitro-derived materials were compared with inflorescences and shoots of the maternal plant. The analyzed materials showed pronounced tissue-dependent metabolic differentiation. The highest callus growth rate was observed for the Ha-C-1 line, which also accumulated the highest levels of chlorogenic acid (25.80 ± 3.09 mg/g dry extract) and isochlorogenic acid A (55.83 ± 13.78 mg/g dry extract). Apigenin was detected in the inflorescence extract, reaching 3.55 ± 1.42 mg/g dry extract. The extracts did not markedly affect HaCaT viability at 15.63–500 µg/mL, while the strongest bioactivities were observed for inflorescence and maternal plant extracts. Full article

Helichrysum italicum (Roth) G. Don (Asteraceae) is a valuable medicinal and aromatic plant native to a variety of habitats across the Mediterranean region. However, genetic studies of this morphologically diverse species have been limited by the scarcity of species-specific DNA markers. To address this limitation, we generated the first de novo transcriptome assembly comprising 24,806 transcripts from young shoots containing leaves and flowers, developed EST-SSR markers, and evaluated their utility in population genetic analysis. Seventy-eight primer pairs were designed, of which 23 showed successful amplification, polymorphism, and transferability to Helichrysum litoreum Guss. and Helichrysum arenarium (L.) Moench. A subset of 12 EST-SSRs was used to genotype 270 individuals from 12 natural populations of H. italicum in Corsica (France), along with one outgroup population from Croatia. The polymorphic information content ranged from 0.250 to 0.796, and Shannon’s information index ranged from 0.588 to 1.843, indicating the markers’ suitability for population genetic studies. Analysis of molecular variance revealed that 15% of the total genetic variation was attributable to differences among populations. Discriminant analysis of principal components and Bayesian clustering in STRUCTURE identified distinct population clusters corresponding to geographic locations. Notably, the southernmost coastal populations were clearly differentiated from the others. Full article

Synthetic anthelmintic drugs not only contribute to the development of pathogen resistance and environmental pollution but also to the development of pathogen resistance. Therefore, identifying the anthelmintic properties of widely used medicinal plants could be of great practical interest to veterinary and human medicine. In our experiment, we evaluated the in vitro survivability of the noninvasive and invasive (L1–2 and L3, respectively) larvae of Strongyloides papillosus and Haemonchus contortus subject to aqueous solutions of ethanolic tinctures of traditional medicinal plants (46 species). Most of the plant species we studied belonged to the families Asteraceae (Achillea millefolium, Arctium lappa, Artemisia absinthium, Bidens tripartita, Calendula officinalis, Cynara cardunculus var. scolymus, Echinacea purpurea, Helichrysum arenarium, Inula helenium, Matricaria chamomilla, Silybum marianum, Tanacetum vulgare, Taraxacum officinale, Tragopogon porrifolius), Rosaceae (Agrimonia eupatoria, Fragaria vesca, Sanguisorba officinalis), and Lamiaceae (Leonurus cardiaca, Mentha × piperita, Origanum vulgare, Salvia officinalis, Thymus vulgaris). Other plant families were represented by 1–3 species: Fabaceae (Glycyrrhiza glabra, Hedysarum alpinum, Trifolium pratense), Salicaceae (Populus nigra, P. tremula, Salix alba), Fagaceae (Quercus robur), Betulaceae (Betula pendula), Juglandaceae (Juglans regia), Rhamnaceae (Frangula alnus), Acoraceae (Acorus calamus), Apiaceae (Foeniculum vulgare), Caprifoliaceae (Valeriana officinalis), Cucurbitaceae (Cucurbita pepo), Equisetaceae (Equisetum arvense), Ericaceae (Vaccinium vitis-idaea), Gentianaceae (Centaurium erythraea), Hypericaceae (Hypericum perforatum), Malvaceae (Althaea officinalis), Plantaginaceae (Linaria vulgaris, Plantago major), Poaceae (Zea mays), Polygonaceae (Polygonum aviculare), and Ranunculaceae (Nigella sativa). We determined Artemisia absinthium, Inula helenium, Matricaria chamomilla, Salvia officinalis, and Populus nigra, whose aqueous solutions of alcohol tinctures demonstrated nematocidal properties. Rhizosphere microorganisms effectively exploit nutrient resources within the rhizosphere, while growth-promoting bacteria in this environment play a vital role in regulating soil fertility and enhancing plant health. In this study, we utilized a comprehensive approach that included the isolation, purification, and identification of dominant microorganisms, alongside high-throughput sequencing technology. This methodology was employed to analyze the primary microbial groups and their diversity within the rhizosphere soil of Helichrysum arenarium (L.) Moench in Altay, Xinjiang, China. By isolating bacterial strains from the rhizosphere soil using a dilution coating method, we successfully obtained 43 distinct strains. Subsequently, selective media were employed to screen for growth-promoting characteristics among these isolated strains derived from the rhizosphere soil of H. arenarium (L.) Moench. The results, obtained through high-throughput amplification sequencing, revealed diverse bacterial communities belonging to 35 phyla, 93 orders, 215 families, 324 genera, and 231 species associated with H. arenarium (L.) Moench, as well as fungal communities comprising 14 phyla, 47 orders, 96 families, 204 genera, and 571 species present in the rhizosphere soil. Among these identified communities, Actinobacteriota emerged as the predominant bacterial phylum while Ascomycetes and Mortieromycetes were recognized as the principal fungal phyla found in the rhizospheric soil of H. arenarium (L.) Moench. Analysis of culturable bacteria’s promotion activity within this rhizospheric environment indicated that three strains—S16, S31, and S29—exhibited the highest solubility index for inorganic phosphorus; additionally, the screened strains S7 and S10 demonstrated nitrogen-fixing capabilities. Furthermore, ten strains exhibiting excellent iron-bearing capacities were identified; notably, strain S16 displayed the highest D/d value indicating, its superior iron-bearing capacity. The growth-promoting bacteria were identified as Kocuria rosea, Priestia megaterium, Bacillus mobilis, Bacillus bataviensis, three variants of Bacillus mycoides, Bacillus paramobilis, Bacillus sonorensis, and Alcaligenes faecalis. This study provides a foundational understanding of how microorganisms in the rhizosphere of H. arenarium (L.) Moench influence soil nutrient release and offers valuable insights into enhancing yield and quality cultivation by isolating, screening, and identifying growth-promoting bacteria from rhizosphere soil. Background: Acne is a prevalent dermatological condition characterized by the blockage of hair follicles and sebaceous glands, leading to the formation of acne. The anaerobe pathogen Cutibacterium acnes (formerly known as Propionibacterium acnes) plays an essential role in the pathogenesis of acne, for which generally antimicrobial treatment is required. Acne is a substantial health concern, and continuing research is being conducted to discover novel and efficacious remedies. The antimicrobial activity of plants has been demonstrated in numerous studies, and they are still targeted organisms in drug development. Studies showing that plants are effective against acne pathogens have also been reported. Methods: The antimicrobial activity of the hydroethanolic extracts prepared from 30 plant species was determined against C. acnes standard strains (C. acnes Scholz and Kilian ATCC 11827 and ATCC 11828) and 30 clinical isolates in our preliminary screening. Since acne is an inflammatory skin disease, the anti-inflammatory effect of six active extracts against C. acnes was determined through the in vitro inhibition of collagenase, lipoxygenase (LOX), hyaluronidase and xanthine oxidase (XO) enzymes. Results: Cotinus coggygria Scop. leaf extract displayed the highest hyaluronidase and collagenase inhibition (79.75% and 52.52%, respectively), while the extract from the aerial parts of Helichrysum arenarium (L.) Moench demonstrated a potent XO inhibitory effect (82.51%). Therefore, these two extracts have been chosen for further studies, and LC/MS-MS was used to determine the phenolic profiles of these extracts. Conclusions: Subsequently, nanoemulgels were formulated with the active extracts to develop a prototype herbal anti-acne product, and characterization studies of the formulations were conducted. Background/Objectives: Cancer is the second leading cause of death globally. Medicinal plants have emerged as fundamental sources of bioactive compounds with anticancer potential, largely attributed to their diverse secondary metabolites. This study aimed to investigate the cytotoxic effects of Helichrysum arenarium extracts from two distinct regions of Turkiye, Mersin, and Artvin, on cancerous (MDA-MB-231, RT4, T98G) and non-cancerous (ARPE-19, hGF) cell lines and to identify bioactive compounds responsible for these effects. Methods: H. arenarium plant extracts were prepared using ethanol and methanol as solvents, followed by lyophilization and dissolution in DMSO. The cytotoxic effects of the extracts were evaluated using Hoechst staining and MTS assays to assess cell viability. IC50 values and selectivity indices were calculated. Phytochemical composition was analyzed using Quadrupole Time-of-Flight mass spectrometry. Results: The ethanol extract from Mersin (HAE-M) demonstrated superior cytotoxicity, particularly against breast and bladder cancer cells, while showing minimal impact on non-cancerous cells. HAM-M, HAE-A, and HAM-A exhibited comparatively less potent effects. Phytochemical analysis of HAE-M identified 16 bioactive compounds, including Naringenin, Luteolin, and Quercitrin, known for their antioxidant and anticancer properties. Conclusions: These findings highlight the potential of H. arenarium extracts, particularly HAE-M, as a source of potent anticancer agents. This study is novel in its comprehensive analysis of different extraction methods and regional plant sources, combined with phytochemical profiling, to identify selective anticancer effects. Further investigations into the mechanisms of action of these extracts could contribute to the development of plant-derived anticancer therapies. Full article

Members of Asteraceae family have properties enabling their application for medical purposes. The major- and microelement content in 15 medicinal species of the Asteraceae family growing in the National Botanical Garden, Republic of Moldova was determined by instrumental neutron activation analysis. A total of 31 chemical elements, Na, Mg, Al, Cl, K, Ca, Sc, V, Cr, Mn, Fe, Co, Ni, Zn, As, Se, Br, Rb, Sr, Mo, Sb, Cs, Ba, La, Ce, Sm, Tb, Hf, Ta, Th and U, were identified in herbal samples. Potassium was found to be the most abundant major element; its content in plants ranged from 20,700 mg/kg in Artemisia lerchiana to 58,000 mg/kg in Matricaria recutita. The content of other major elements existed in the following ranges: Ca from 4700 to 14,200 mg/kg and Mg from 1710 to 3870 mg/kg. The maximum content of Mg, K and Ca in analyzed plants was higher compared to data presented in the literature. Among essential microelements the most abundant were Fe (83–910 mg/kg), Mn (23–150 mg/kg) and Zn (27–76 mg/kg). The daily intake of metal and the health risk index for selected elements (As, Ni, Sb, V, Mn, Cr, Co, U, Sr, Al, Fe, and Zn) were calculated. Health risk indexes were obtained for V and Co in Calendula officinalis, Tanacetum balsamita, Achillea clypeolata, Artemisia balchanorum, Artemisia lerchiana, Helichrysum arenarium, and Matricaria recutita. The principal component analysis showed three associations of elements, which can be defined as physiological, geological and anthropogenic sources of elements. Full article

Helichrysum italicum (Roth) G. Don is a Mediterranean medicinal plant with great potential in the cosmetics, culinary and pharmaceutical fields due to its unique bioactive compounds. Its recent introduction into agroecosystems has enhanced the exploitation of genetic diversity in natural populations, although limited molecular markers have made this challenging. In the present study, primers were designed for all 43 SSRs (72.1% mononucleotide, 21% dinucleotide and 6.9% trinucleotide repeats) identified in the chloroplast genome. Populations from Cape Kamenjak (Croatia) and Corsica (France) were analyzed with ten carefully selected cpSSR markers. From the initial set of 16 cpSSRs amplified in all samples, 6 cpSSR markers were removed due to low-length polymorphisms, size homoplasy and nucleotide polymorphisms that could not be detected with allele length. Of the 38 haplotypes detected, 32 were unique to their geographic origin. The highest number of private haplotypes was observed in the Cape Kamenjak population (seven out of nine detected). Based on clustering analyses, the Kamenjak population was the most similar to the Capo Pertusato (south Corsica) population, although only one sub-haplotype was shared. Other Corsican populations were more similar to each other. A cross-species transferability test with Helichrysum litoreum Guss. and Helichrysum arenarium (L.) Moench was successfully conducted and private alleles were identified. Full article

The textile industry is one of the most complex sectors, in terms of the materials and chemical processes used from petroleum and the environmental degradation during its production and disposal. It is therefore a sector looking for new possibilities and for more sustainable materials and applications. One option is to use natural dyes, as they are considered biodegradable, do not pollute the environment, and have potential use for many sectors, including the fashion industry. In this study, Alanya silk was dyed by a natural dyeing method with crocus sativus, Helichrysum arenarium, and Glycyrrhiza glabra L., plants that grow in and around the Alanya region. Quercus aegilops L. grown in the region was preferred as mordant, a natural binder, and is one of the plants with the highest tannin content, and it was used with a more environmentally friendly and sustainable approach to increase the binding in natural dyeing instead of chemical mordants. The aim is to provide an environmental and scientific contribution to the dyeing producers in this region. According to the MAUT (Multi-Attribute Utility Theory) method, the best dyes in terms of fastness and color efficiency were determined as the dyes made with the Glycyrrhiza glabra L. plant. In the present study, vortex-assisted matrix solid-phase dispersion (VA-MSPD) extraction was used to isolate the major bioactive compounds from H. arenarium. To reduce the negative environmental impact of the conventionally used organic solvents, four different choline chloride-based natural deep eutectic solvents (NADES) were investigated as possible eluents. The most influential VA-MSPD extraction parameters: stationary phase (adsorbent), adsorbent/sample ratio, vortex time, and volume of extraction solvent were systematically optimized. Ultrasound-assisted extraction with 80% MeOH was used as the standard method for the comparison of results. The stability of the obtained extracts was studied over a period of 0 to 60 days at three different temperatures (−18 °C, 4 °C, and 25 °C). All extracts were evaluated both spectrophotometrically (determination of total phenolic content (TPC) and antioxidant activity by ABTS and FRAP assay) and chromatographically (HPLC-UV). NADES based on choline chloride and lactic acid (ChCl-LA) was selected as the most effective extractant, with a determined TPC value of its extract of 38.34 ± 0.09 mg GA/g DW (27% higher than the methanolic VA-MSPD extract) and high antioxidant activity. The content of individual phenolic compounds (chlorogenic acid, dicaffeoylquinic acid isomers, naringenin isomers, and chalcones) in the ChCl-LA extract, determined by HPLC-UV, was comparable to that of the conventionally obtained one. Moreover, the stabilization effect of ChCl-LA was confirmed for the studied compounds: chlorogenic acid, naringenin-4′-O-glucoside, tomoroside A, naringenin-5-O-glucoside, isosalipurposide, and naringenin. The optimum VA-MSPD conditions for the extraction of H. arenarium polyphenols were: florisil/sample ratio of 0.5/1, a vortex time of 2 min, and an elution volume of ChCl-LA of 10 mL. Full article

Helichrysum italicum (Roth) G. Don (HI) and Helichrysum arenarium (L.) Moench (HA) are rich in polyphenols and their infusions have beneficial effects for patients with metabolic syndrome. To investigate whether these effects are mediated by the gut microbiota, we analysed the effects of daily consumption of HI or HA infusion on the composition of gut microbiota, inflammatory status, and zonulin, a marker of gut barrier permeability. The study was a randomized, double-blind comparative trial. Thirty participants were randomly assigned to two groups and received either HA or HI tea filter bags, each containing 1 g of dried plant material, for daily consumption lasting 4 weeks. The results show that consumption of both infusions resulted in a reduction of some genera belonging to Firmicutes and in a slight but significant reduction in Shannon diversity index. Consumption of HI infusion significantly reduced serum levels of proinflammatory markers and zonulin alongside with the observed trend of Proteobacteria reduction. It can therefore be concluded that the HI and HA infusions could act as prebiotics and thus improve the intestinal environment. In addition, HI infusion has a positive impact on microbial dysbiosis and gut barrier dysfunction that occur in obesity and metabolic syndrome. Full article

Helichrysum arenarium (L.) Moench, belonging to the Asteraceae family, is known in traditional medicine for its diuretic, choleretic, and anti-inflammatory properties. This review focuses on the superiority of Helichrysum arenarium (sandy everlasting) over other known plants as a source of active pharmacological compounds used in complementary medicine to prevent digestive and hepatobiliary illnesses. Because the species exists both in spontaneous flora and in crops, this article highlights the development of a controlled culture of H. arenarium, following the reproducible quality of the plant as a biological material. The diversity of phytocompounds, especially well-characterized flavonoid extracts, and the differences between extraction procedures are discussed. Antiviral, antibacterial, and antifungal activities against human pathogens proved by different plant extracts and their mechanisms of action are analyzed. This study aims to contribute to the insufficient knowledge regarding the effects of Helichrysum species and to reveal that their extracts can be a valuable source for new active pharmaceutical ingredients. This study compared the essential oils (EO) composition of Helichrysum arenarium (Bulgarian populations) with that of the cultivated species H. italicum. The EO composition of H. arenarium and H. italicum were analyzed via gas chromatography. In general, 75 components were identified in H. arenarium EO and 79 in H. italicum EO. The predominant constituents in H. arenarium EO were α-pinene (34.64–44.35%) and sabinene (10.63–11.1%), which affirmed the examined population as a new chemical type. Overall, the main EO constituents of H. italicum originating in France, Bosnia and Corsica were neryl acetate (4.04–14.87%) and β-himachalene (9.9–10.99%). However, the EOs profile of H. italicum introduced from the above three countries differed to some extent. D-limonene (5.23%), italicene, α-guaiene and neryl acetate (14.87%) predominated in the H. italicum introduced from France, while α-pinene (13.74%), δ-cadinene (5.51%), α-cadinene (3.3%), β-caryophyllene (3.65%) and α-calacorene (1.63%) predominated in plants introduced from Bosnia. The EOs of the plants introduced from France and Corsica had similar chemical composition and antimicrobiological activity. Full article

While harmful effects of blue light on skin cells have been recently reported, there are few studies regarding natural products that alleviate its negative effects. Therefore, we investigated ameliorating effects of yellow chaste weed (YCW) (Helichrysum arenarium) extract and its components, apigenin and galangin, on blue light-irradiated HaCaT cells. In this study, we found that YCW extract improved the reduced proliferation of HaCaT cells induced by blue light-irradiation and reduced blue light-induced production of reactive oxygen species (ROS) levels. We also found that apigenin and galangin, the main components of YCW extract, showed the same activities as YCW extract. In experiments examining molecular mechanisms of YCW extract and its components such as apigenin and galangin, they all reduced expression of transient receptor potential vanilloid member 1 (TRPV1), its phosphorylation, and calcium ion (Ca2+) influx induced by blue light irradiation. In addition, apigenin and galangin regulated phosphorylation of mitogen-activated protein kinases (MAPKs). They also reduced phosphorylation of mammalian sterile 20-like kinase-1/2 (MST-1/2), inducing phosphorylation of Akt (protein kinase B), one downstream molecule of MST-1/2. Moreover, apigenin and galangin promoted translocation of Forkhead box O3 (FoxO3a) from the nucleus to the cytosol by phosphorylating FoxO3a. Besides, apigenin and galangin interrupted blue light influences on expression of nuclear and secretory clusterin. Namely, they attenuated both upregulation of nuclear clusterin and downregulation of secretory clusterin induced by blue light irradiation. We also found that they downregulated apoptotic protein Bcl-2 associated X protein (Bax) and conversely upregulated anti-apoptotic protein B-cell lymphoma 2 (Bcl-2). Collectively, these findings indicate that YCW extract and its components, apigenin and galangin, antagonize the blue light-induced damage to the keratinocytes by regulating TRPV1/clusterin/FoxO3a and MAPK signaling. Helichrysum arenarium (L.) Moench (sandy everlasting) is the only species from genus Helichrysum Mill that grows spontaneously in Lithuania. The chemical composition of the essential oils (EOs) from inflorescences and leaves of H. arenarium wild plants was analysed by GC-MS. Palmitic (≤23.8%), myristic (≤14.9%) and lauric (6.1%) acids, n-nonanal (10.4%), and trans-β-caryophyllene (≤6.5%) were the major constituents in the EOs. For comparison, the main components in EO from flowers (commercial herb material) of H.italicum were γ-curcumene (21.5%), β-selinene (13.6%), α-selinene (8.1%), β-eudesmol (8.3%), and α-pinene (6.5%). Composition of H. arenarium methanolic extracts was investigated by HPLC-DAD-TOF. The main compounds were the following: luteolin-7-O-glucoside, naringenin and its glucoside, apigenin, chlorogenic acid, arenol, and arzanol. Antioxidant activity of EOs and extracts was tested by DPPH and ABTS●+ assays. Sandy everlasting extracts exhibited significantly higher radical scavenging activities (for leaves 11.18 to 19.13 and for inflorescences 1.96 to 6.13 mmol/L TROLOX equivalent) compared to those of all tested EOs (0.25 to 0.46 mmol/L TROLOX equivalent). Antioxidant activity, assayed electrochemically by cyclic and square wave voltammetry correlated with total polyphenolic content in extracts and radical scavenging properties of EOs and extracts. The toxic activity of EOs of both Helichrysum species was evaluated using a brine shrimp (Artemia salina) bioassay. H. italicum inflorescence EO was found to be toxic (LC50 = 15.99 µg/mL) as well as that of H. arenarium (LC50 ≤ 23.42 µg/mL) oils. Full article
