This study aimed to evaluate the impact of intercropping Brassica oleracea. with three perennial grasses (Poa pratensis L., Lolium perenne L., and Festuca rubra L.) under varying levels of iron (Fe) availability (Fe0, Fe1 and Fe5) in nutrient solutions. The research focused on biomass accumulation, photosynthetic efficiency, root development, nutrient uptake, and oxidative stress response. In the absence of Fe, Brassica sp. exhibited chlorosis, reduced biomass, and increased ferric chelate reductase (FCR) enzyme activity as an adaptive response. Brassica plants intercropped with Poa sp. maintained higher chlorophyll (Chl) levels and photosystem II efficiency (Fv/Fm values), mitigating Fe deficiency effects. Catalase activity and polyphenol production varied with intercropping species, indicating differential stress response mechanisms. Intercropping improved Zn, Mn, and P accumulation, with Poa sp. facilitating greater Zn and Mn uptake. Intercropping Brassica sp. with specific grass species offers potential agronomic benefits by improving Fe use efficiency, mitigating stress, and enhancing nutrient uptake. Future research should focus on optimizing intercropping combinations for sustainable agricultural practices. Full article

In plants, ferric-chelate reductase (FRO) plays a critical role in mediating extracellular iron (Fe) reduction, a process essential for cellular Fe homeostasis and abiotic stress tolerance. However, the biological functions and regulatory mechanisms of FRO proteins in fruit crops remain poorly characterized. Here, six VvFRO genes were identified in the table grape cultivar ‘Yanhong’. Transcriptional analysis revealed that root expression of these genes was mainly induced under Fe deficiency, Fe depletion, NaCl stress, and PEG-induced drought stress, respectively, but remained unchanged by low temperature (4 °C) or heat treatment (45 °C). Among them, VvFRO3 exhibited the highest constitutive expression, predominantly in leaves, and was significantly up-regulated under Fe deficiency, Fe depletion, or NaCl treatment. Functional complementation assays demonstrated that heterologous overexpression of VvFRO3 in the Arabidopsis thaliana fro2 knockout mutant rescued its growth retardation phenotype, particularly under Fe-deficient conditions. This study advances our understanding of Fe uptake, transport, and homeostasis mechanisms in perennial fruit crops. Full article

The PYL protein family are crucial sensors of the core signals of abscisic acid (ABA) and significantly influence the plant’s response to ABA-mediated abiotic stresses as well as its growth and development. However, research on the role of the MhPYL4 gene in iron (Fe) deficiency in apple trees is limited. Studies have shown that the MhPYL4 gene, when exposed to Fe-deficiency stress, exhibits more rapid transcriptional upregulation than other genes’ quickly elevated transcription. However, the precise mechanism by which it alleviates this stress remains unclear. The MhPYL4 gene (ID:103432868), isolated from Malus halliana, was analyzed to elucidate its function. Arabidopsis plants engineered to overexpress the MhPYL4 gene exhibited increased leaf chlorosis and slower growth in response to Fe stress compared to the unmodified controls. The transgenic plants also exhibited elevated levels of superoxide dismutase (SOD), peroxidase (POD), and catalase (CAT) activities, as well as ferric chelate reductase (FCR) activities. Levels of malondialdehyde (MDA), hydrogen peroxide (H2O2), and superoxide anion (O2) were increased. In addition, these transgenic plants had lower concentrations of proline (Pro) and Fe2+, which indicated that their stress tolerance was reduced. Similarly, the overexpression of MhPYL4 in apple calli resulted in inhibited growth and increased susceptibility under Fe stress conditions. Physiological evaluations indicated that the overexpression of MhPYL4 in Arabidopsis reduced its Fe stress tolerance by inhibiting chlorophyll synthesis. In apple calli, it altered pH levels, antioxidant enzyme activity, and Fe-reducing capabilities under the same stress conditions. In summary, the elevated expression of the MhPYL4 gene reduced the tolerance of both Arabidopsis and apple calli to Fe stress, suggesting that MhPYL4 acts as a negative regulator in response to Fe deficiency. Under iron (Fe)-limited conditions, plants have developed strategies for acquiring this essential micronutrient. Several Fe sources have been studied as potential fertilizers, with Fe synthetic chelates being the most used to prevent and correct Fe chlorosis in crops. The determination of the activity of the Fe chelate reductase (FCR) enzyme has long been described in the literature to understand the efficiency of Strategy I plants in acquiring Fe from fertilizers under deficient conditions. Other experiments have focused on the translocation of Fe to the plant to define the effectiveness of Fe fertilizers. Yet, both assays are relevant in knowing the capacity of a novel Fe source and other compounds alleviating Fe chlorosis in Strategy I plants. This work reviews the methodologies that are used in FCR assays to evaluate novel Fe fertilizers, including the factors modulating the results obtained for FCR assay activity, such as the Fe substrate, the Fe level during the growing period and during the FCR assay, the pH, the choice of an in vivo or in vitro method, and the plant species. A discussion of the benefits of the concurrence of FCR and Fe uptake assays is then presented alongside a proposed methodology for assessing the effectiveness of Fe fertilizers, emphasizing the importance of understanding chemical and physiological plant interactions. This methodology unifies key factors that modify FCR activity and combines these with the use of the 57Fe tracer to enhance our comprehension of the efficacy of Fe-based fertilizers’ effectiveness in alleviating Fe chlorosis. This comprehensive approach not only contributes to the fundamental understanding of Fe-deficient Strategy I plants but also establishes a robust method for determining the efficiency of novel sources for correcting Fe deficiency in plants. Iron (Fe) deficiency is one of the most common micronutrient deficiencies limiting crop production globally, especially in arid regions due to decreased availability of Fe in alkaline soils. The ATP sulfurylase (ATPS) gene has been reported to participate in regulating various abiotic stresses. Transcriptome data and qRT-PCR analysis revealed that the ATP sulfurylase gene MhATPS1 was notably induced by Fe-deficiency stress. Consequently, MhATPS1 (103410737) was isolated from Malus halliana, and transgenic tobacco and transgenic apple calli were successfully obtained by genetic transformation. Compared with the wild type (WT), transgenic MhATPS1 lines (transgenic tobacco and transgenic apple calli) displayed stronger resistance to Fe-deficiency treatment. To be specific, transgenic plants exhibited better growth, accumulated more Fe2+ content, had higher ferric chelate reductase (FCR) activity, and a greater active oxygen scavenging capacity. Furthermore, transgenic MhATPS1 lines up-regulated the expression of Fe uptake genes under Fe-deficit stress. Additionally, MhATPS1 transgenic lines secreted more H+ content compared to the WT. In summary, these findings indicate that the MhATPS1 gene may play a positive role in Fe-deficiency stress in both tobacco and apple calli. Full article

The ferric chelate reductase (FRO) family plays a vital role in metal ion homeostasis in a variety of locations in the plants. However, little is known about this family in peanut (Arachis hypogaea). This study aimed to identify FRO genes from the genomes of peanut and the two diploid progenitors (A. duranensis and A. ipaensis) and to analyze their gene/protein structures and evolution. In addition, transcriptional responses of AhFRO genes to Fe deficiency and/or Cu exposure were investigated in two peanut cultivars with different Fe deficiency tolerance (Silihong and Fenghua 1). A total of nine, four, and three FRO genes were identified in peanut, A. duranensis, and A. ipaensis, respectively, which were divided into three groups. Most AhFRO genes underwent WGD/segmental duplication, leading to the expansion of the AhFRO gene family. In general, clustered members share similar gene/protein structures. However, significant divergences occurred in AhFRO2 genes. Three out of five AhFRO2 genes were lowly expressed in all tissues under normal conditions, which may be beneficial for avoiding gene loss. Transcription analysis revealed that AhFRO2 and AhFRO7 genes might be involved in the reduction of Fe/Cu in plasma membranes and plastids, respectively. AhFRO8 genes appear to confer Fe reduction in the mitochondria. Moreover, Fe deficiency induced an increase of Cu accumulation in peanut plants in which AhFRO2.2/2.4/2.5 and FRO7.1/7.2 might be involved. Our findings provided new clues for further understanding the roles of AhFRO genes in the Fe/Cu interaction in peanut. Full article

Iron is an essential micronutrient for citrus, playing an important role in photosynthesis and yield. The aim of this paper was to evaluate the tolerance to Fe deficiency of five citrus rootstocks: sour orange (S), Carrizo citrange (C), Citrus macrophylla (M), Troyer citrange (T), and Volkamer lemon (V). Plants were grown for 5 weeks in nutrient solution that contained the following Fe concentrations (in µM): 0, 5, 10, 15, and 20. At the end of the experiment, biomass (dry weight—DW), leaf area, total leaf chlorophyll (CHL), and the activity of root chelate reductase (FCR) were recorded. Additionally, the mineral composition of roots (R) and shoots (S) was evaluated. Principal component analysis was used to study the relationships between all parameters and, subsequently, the relations between rootstocks. In the first component, N-S, P-S, Ca-S, Cu-S, Zn-S, Mn-S, Zn-R, and Mn-R concentrations were related to leaf CHL and FCR. Increases in leaf CHL, Mg-R, and DW (shoots and roots) were inversely related to Cu-R, which was shown in the second component. The values obtained were consistent for V10, C15, and C20, but in contrast for S0 and S5. In conclusion, micronutrient homeostasis in roots and shoots of all rootstocks were affected by Fe stress conditions. The Fe/Cu ratio was significantly related to CHL, which may be used to assist rootstock performance. Full article

Micronutrient iron (Fe) deficiency poses a widespread agricultural challenge with global implications. Fe deficiency affects plant growth and immune function, leading to reduced yields and contributing to the global “hidden hunger.” While conventional Fe-based fertilizers are available, their efficacy is limited under certain conditions. Most recently, nanofertilizers have been shown as promising alternatives to conventional fertilizers. In this study, three nanohematite/nanoferrihydrite preparations (NHs) with different coatings were applied through the roots and shoots to Fe-deficient cucumber plants. To enhance Fe mobilization to leaves during foliar treatment, the plants were pre-treated with various acids (citric acid, ascorbic acid, and glycine) at a concentration of 0.5 mM. Multiple physiological parameters were examined, revealing that both root and foliar treatments resulted in improved chlorophyll content, biomass, photosynthetic parameters, and reduced ferric chelate reductase activity. The plants also significantly accumulated Fe in their developing leaves and its distribution after NHs treatment, detected by X-ray fluorescence mapping, implied long-distance mobilization in their veins. These findings suggest that the applied NHs effectively mitigated Fe deficiency in cucumber plants through both modes of application, highlighting their potential as nanofertilizers on a larger scale. Iron deficiency is a worldwide nutritional problem affecting crop production. In Tunisia, this mineral disorder hampers the growth and yield of the major crops due to the abundance of calcareous soils that limit iron availability. The common bean is one of these crops suffering from lime-induced iron chlorosis. The exploration of the variability of common bean responses to iron deficiency allows us to screen tolerant cultivars and identify useful traits and indicators for further screening programs. To this end, two common bean cultivars (coco blanc, CB, and coco nain, CN) were cultivated hydroponically in standard nutrient solution (control) or nutrient solution deprived of iron (stressed). Analyses were reported on plant growth, photosynthetic pigments, photosynthesis, iron distribution, H-ATPase, and Fe-chelate reductase (Fe-CR) activities; important indicators were calculated; and convenient correlations were established. Current results demonstrated that iron deficiency stimulated specific symptoms of iron chlorosis on young leaves that were more precocious and severe in CB than CN. Spad index and chlorophyll pigments measurement confirmed these morphological changes and cultivar differences. Net photosynthesis (Pn) showed the same scheme of variation, with a significant decrease in Pn while respecting the previous cultivar’s variability. Plant growth is no exception to this general trend. The biomass decrease was two times higher in CB than CN. Otherwise, this mineral disorder significantly decreased Fe concentration in all plant organs. However, CN accumulated 40% more Fe than CB, resulting from its higher Fe Fe-CR and H-ATPase activities. Our results also demonstrated the close dependence of these metabolic functions on Fe availability in shoots and the strict relationship between Fe-CR and H-ATPase, photosynthesis, and chlorophyll content. Otherwise, CN demonstrated higher efficiency of Fe’s use (FeUE) for the key metabolic functions (photosynthesis, chlorophyll biosynthesis, and plant growth). The relative tolerance of CN as compared to CB was explained by its ability to establish a functional system less vulnerable to iron deficiency that operates effectively under problematic conditions. This system involves metabolic functions in shoots (photosynthesis, chlorophyll biosynthesis, Fe repartition, etc.) and others in roots (H-ATPase, Fe-CR), which are strictly interdependent. Evidence That PbrSAUR72 Contributes to Iron Deficiency Tolerance in Pears by Facilitating Iron Absorption

Iron is an essential trace element for plants; however, low bioactive Fe in soil continuously places plants in an Fe-deficient environment, triggering oxidative damage. To cope with this, plants make a series of alterations to increase Fe acquisition; however, this regulatory network needs further investigation. In this study, we found notably decreased indoleacetic acid (IAA) content in chlorotic pear (Pyrus bretschneideri Rehd.) leaves caused by Fe deficiency. Furthermore, IAA treatment slightly induced regreening by increasing chlorophyll synthesis and Fe2+ accumulation. At that point, we identified PbrSAUR72 as a key negative effector output of auxin signaling and established its close relationship to Fe deficiency. Furthermore, the transient PbrSAUR72 overexpression could form regreening spots with increased IAA and Fe2+ content in chlorotic pear leaves, whereas its transient silencing does the opposite in normal pear leaves. In addition, cytoplasm-localized PbrSAUR72 exhibits root expression preferences and displays high homology to AtSAUR40/72. This promotes salt tolerance in plants, indicating a putative role for PbrSAUR72 in abiotic stress responses. Indeed, transgenic plants of Solanum lycopersicum and Arabidopsis thaliana overexpressing PbrSAUR72 displayed less sensitivity to Fe deficiency, accompanied by substantially elevated expression of Fe-induced genes, such as FER/FIT, HA, and bHLH39/100. These result in higher ferric chelate reductase and root pH acidification activities, thereby hastening Fe absorption in transgenic plants under an Fe-deficient condition. Moreover, the ectopic overexpression of PbrSAUR72 inhibited reactive oxygen species production in response to Fe deficiency. These findings contribute to a new understanding of PbrSAURs and its involvement in Fe deficiency, providing new insights for the further study of the regulatory mechanisms underlying the Fe deficiency response. Iron is a key metal involved in several biological processes such as DNA replication and repair, cellular proliferation and cell cycle regulation. Excess volumes of labile iron are toxic and can lead to the production of ROS (reactive oxygen species) via Fenton chemistry. Due to this reactive nature, it can contribute to DNA damage and genomic instability. Therefore, excess iron in the labile iron pool is associated with cancer, which has made the labile iron pool a crucial target for anticancer therapy by targeting iron. This iron can be incorporated into essential enzymes such as ribonucleotide reductase (RnR). Over several decades of research, iron chelators function as more than just RnR inhibitors. Indeed, a plethora of iron chelator mechanisms can result in therapeutic properties that can target critical steps of cancer cells’ aberrant biological abilities such as proliferation, migration and metastasis. One such mechanism is the production of redox-active complexes that can produce toxic levels of ROS in cancer cells. Cancer cells are potentially more susceptible to ROS production or modulation of antioxidant levels. Understanding iron metabolism is vital in targeting cancer. For instance, Fe-S clusters have recently been shown to play crucial roles in cell signalling by ROS through their incorporation into essential DNA replication and repair enzymes. ROS can also degrade Fe-S clusters. Iron chelators that produce toxic levels of ROS, therefore, could also target Fe-S centres. Thus, the design of iron chelators is important, as this can determine if it will participate in redox cycling and produce ROS or if it is solely used to remove iron. This review focuses on alterations in cancer iron metabolism, iron’s role in genomic stability and how the design of chelators can use Fenton chemistry to their advantage to cause DNA damage in cancer cells and potentially inhibit Fe-S centres. Iron deficiency causes chlorosis and growth inhibition in Cinnamomum camphora, an important landscaping tree species. Siderophores produced by plant growth-promoting rhizobacteria have been widely reported to play an indispensable role in plant iron nutrition. However, little to date has been determined about how microbial siderophores promote plant iron absorption. In this study, multidisciplinary approaches, including physiological, biochemical and transcriptome methods, were used to investigate the role of deferoxamine (DFO) in regulating Fe availability in C. camphora seedlings. Our results showed that DFO supplementation significantly increased the Fe2+ content, SPAD value and ferric-chelate reductase (FCR) activity in plants, suggesting its beneficial effect under Fe deficiency. This DFO-driven amelioration of Fe deficiency was further supported by the improvement of photosynthesis. Intriguingly, DFO treatment activated the metabolic pathway of glutathione (GSH) synthesis, and exogenous spraying reduced glutathione and also alleviated chlorosis in C. camphora. In addition, the expression of some Fe acquisition and transport-related genes, including CcbHLH, CcFRO6, CcIRT2, CcNramp5, CcOPT3 and CcVIT4, was significantly upregulated by DFO treatment. Collectively, our data demonstrated an effective, economical and feasible organic iron-complexing agent for iron-deficient camphor trees and provided new insights into the mechanism by which siderophores promote iron absorption in plants. Iron (Fe) is the fourth most abundant element on earth and represents an essential nutrient for life. As a fundamental mineral element for cell growth and development, iron is available for uptake as ferric ions, which are usually oxidized into complex oxyhydroxide polymers, insoluble under aerobic conditions. In these conditions, the bioavailability of iron is dramatically reduced. As a result, microorganisms face problems of iron acquisition, especially under low concentrations of this element. However, some microbes have evolved mechanisms for obtaining ferric irons from the extracellular medium or environment by forming small molecules often regarded as siderophores. Siderophores are high affinity iron-binding molecules produced by a repertoire of proteins found in the cytoplasm of cyanobacteria, bacteria, fungi, and plants. Common groups of siderophores include hydroxamates, catecholates, carboxylates, and hydroximates. The hydroxamate siderophores are commonly synthesized by fungi. L-ornithine is a biosynthetic precursor of siderophores, which is synthesized from multimodular large enzyme complexes through non-ribosomal peptide synthetases (NRPSs), while siderophore-Fe chelators cell wall mannoproteins (FIT1, FIT2, and FIT3) help the retention of siderophores. S. cerevisiae, for example, can express these proteins in two genetically separate systems (reductive and nonreductive) in the plasma membrane. These proteins can convert Fe (III) into Fe (II) by a ferrous-specific metalloreductase enzyme complex and flavin reductases (FREs). However, regulation of the siderophore through Fur Box protein on the DNA promoter region and its activation or repression depend primarily on the Fe availability in the external medium. Siderophores are essential due to their wide range of applications in biotechnology, medicine, bioremediation of heavy metal polluted environments, biocontrol of plant pathogens, and plant growth enhancement. Iron (Fe) deficiency is a first-order agronomic problem that causes a significant decrease in crop yield and quality. Paradoxically, Fe is very abundant in most soils, mainly in its oxidized form, but is poorly soluble and with low availability for plants. In order to alleviate this situation, plants develop different morphological and physiological Fe-deficiency responses, mainly in their roots, to facilitate Fe mobilization and acquisition. Even so, Fe fertilizers, mainly Fe chelates, are widely used in modern agriculture, causing environmental problems and increasing the costs of production, due to the high prices of these products. One of the most sustainable and promising alternatives to the use of agrochemicals is the better management of the rhizosphere and the beneficial microbial communities presented there. The main objective of this research has been to evaluate the ability of several yeast species, such as Debaryomyces hansenii, Saccharomyces cerevisiae and Hansenula polymorpha, to induce Fe-deficiency responses in cucumber plants. To date, there are no studies on the roles played by yeasts on the Fe nutrition of plants. Experiments were carried out with cucumber plants grown in a hydroponic growth system. The effects of the three yeast species on some of the most important Fe-deficiency responses developed by dicot (Strategy I) plants, such as enhanced ferric reductase activity and Fe2+ transport, acidification of the rhizosphere, and proliferation of subapical root hairs, were evaluated. The results obtained show the inductive character of the three yeast species, mainly of Debaryomyces hansenii and Hansenula polymorpha, on the Fe-deficiency responses evaluated in this study. This opens a promising line of study on the use of these microorganisms as Fe biofertilizers in a more sustainable and environmentally friendly agriculture. Full article

Short Term Elevated CO2 Interacts with Iron Deficiency, Further Repressing Growth, Photosynthesis and Mineral Accumulation in Soybean (Glycine max L.) and Common Bean (Phaseolus vulgaris L.)

Elevated CO2 (eCO2) has been reported to cause mineral losses in several important food crops such as soybean (Glycine max L.) and common bean (Phaseolus vulgaris L.). In addition, more than 30% of the world’s arable land is calcareous, leading to iron (Fe) deficiency chlorosis and lower Fe levels in plant tissues. We hypothesize that there will be combinatorial effects of eCO2 and Fe deficiency on the mineral dynamics of these crops at a morphological, biochemical and physiological level. To test this hypothesis, plants were grown hydroponically under Fe sufficiency (20 μM Fe-EDDHA) or deficiency (0 μM Fe-EDDHA) at ambient CO2 (aCO2, 400 ppm) or eCO2 (800 ppm). Plants of both species exposed to eCO2 and Fe deficiency showed the lowest biomass accumulation and the lowest root: shoot ratio. Soybean at eCO2 had significantly higher chlorophyll levels (81%, p < 0.0001) and common bean had significantly higher photosynthetic rates (60%, p < 0.05) but only under Fe sufficiency. In addition, eCO2 increased ferric chelate reductase acivity (FCR) in Fe-sufficient soybean by 4-fold (p < 0.1) and in Fe-deficient common bean plants by 10-fold (p < 0.0001). In common bean, an interactive effect of both environmental factors was observed, resulting in the lowest root Fe levels. The lowering of Fe accumulation in both crops under eCO2 may be linked to the low root citrate accumulation in these plants when grown with unrestricted Fe supply. No changes were observed for malate in soybean, but in common bean, shoot levels were significantly lower under Fe deficiency (77%, p < 0.05) and Fe sufficiency (98%, p < 0.001). These results suggest that the mechanisms involved in reduced Fe accumulation caused by eCO2 and Fe deficiency may not be independent, and an interaction of these factors may lead to further reduced Fe levels.
