CareCoach seeks to enhance self-efficacy in family caregivers of people living with dementia and has been feasibility tested in a multicentre randomised controlled trial. The intervention offers two face-to-face sessions with a trained coach and access to an online platform with nine modules. This paper reports findings from an embedded qualitative process evaluation assessing implementation from the implementer’s (‘coach’s’) (n = 8) perspective using individual interviews and implementer group discussions. Qualitative data were transcribed verbatim, inductively coded and analysed using Normalisation Process Theory. Implementers demonstrated (1) ‘Coherence’ by seeking to understand how CareCoach compared to current practice, highlighting the importance of supporting coaches to differentiate and identify boundaries between their new ‘coach role’ and usual practice; (2) ‘Cognitive Participation’ by reviewing training and resources to understand their role own responsibilities and facilitate delivery of coaching sessions; group supervision and peer support were also emphasised; (3) ‘Collective Action’ through interactions with carers to deliver key behavioural aspects such as goal setting, problem solving, and providing feedback; and (4) ‘Reflexive Monitoring’ by appraising the intervention to gain useful insights that could facilitate refinement of CareCoach training and delivery. This study provides a theoretically informed understanding of the implementation of CareCoach for caregivers of people living with dementia and provides recommendations to enhance training for coaches, intervention delivery and carer engagement. Objective: To develop and validate a ResNet-50-based deep learning model for automatic detection of osteomyelitis (DFO) in plain radiographs of patients with diabetic foot ulcers (DFUs). Research Design and Methods: This retrospective study included 168 patients with type one or type two diabetes and clinical suspicion of DFO confirmed via a surgical bone biopsy. An experienced clinician and a pretrained ResNet-50 model independently interpreted the radiographs. The model was developed using Python-based frameworks with ChatGPT assistance for coding. The diagnostic performance was assessed against the histopathological findings, calculating sensitivity, specificity, the positive predictive value (PPV), the negative predictive value (NPV), and the likelihood ratios. Agreement between the AI model and the clinician was evaluated using Cohen’s kappa coefficient. Results: The AI model demonstrated high sensitivity (92.8%) and PPV (0.97), but low-level specificity (4.4%). The clinician showed 90.2% sensitivity and 37.8% specificity. The Cohen’s kappa coefficient between the AI model and the clinician was −0.105 (p = 0.117), indicating weak agreement. Both the methods tended to classify many cases as DFO-positive, with 81.5% agreement in the positive cases. Conclusions: This study demonstrates the potential of IA to support the radiographic diagnosis of DFO using a ResNet-50-based deep learning model. AI-assisted radiographic interpretation could enhance early DFO detection, particularly in high-prevalence settings. However, further validation is necessary to improve its specificity and assess its utility in primary care. Full article

Cold-water soft corals are a known habitat for juvenile basket stars (Gorgonocephalus sp.), but the role of this relationship in the earliest life stages of basket stars warrants further investigation. Here, basket stars and colonies of the soft coral Gersemia rubiformis were collected together from the Funk Island Deep Marine Refuge (NW Atlantic) and maintained in a laboratory setting for observation. During this time, two developing (<1 mm disc diameter) basket stars were discovered on coral colonies and could be seen retracting with the coral polyp into the colony. The basket stars were recorded unharmed once the polyps were expanded again and continued to retract within the colony over the period of observation. The results of this study show that developing basket stars can spend time inside the coral colony, which could be a form of protection. Full article

Though previous studies have demonstrated the protective benefits of sports participation against illicit drug use for a general population, how these findings apply to LGBTQ youth remains unknown. This study specifically looks at the relationship between sports participation and prescription drug misuse among sexual and gender minority youth. Using secondary data from the 2019 YRBS, we analyze associations among sports participation, sexual orientation, gender identity, and prescription drug misuse among a representative sample of U.S. high school students in Florida. Our results show that sexual and gender minority youth are at increased risk for prescription drug misuse compared to their heterosexual and cisgender peers. Moreover, sports participation is associated with higher rates of prescription drug misuse among all students, and the nuances of these trends are discussed with particular attention paid to sexual and gender minority youth. These results challenge conventional wisdom about sports participation. Without the addition of new demographic survey questions and LGBTQ youth participation in the YRBS, common myths about sports might have persisted. Our findings point to the meaningful presence of LGBTQ youth in sports, call for research and programming on LGBTQ athletes’ unique needs regarding substance misuse risk, and encourage LGBTQ-inclusive policies and practices within schools and sports programs in particular. Background/Objectives: In the last few decades, the dengue virus, a prevalent flavivirus, has demonstrated various epidemiological, economic, and health impacts around the world. Dengue virus serotype 2 (DENV2) plays a vital role in dengue-associated mortality. The RNA-dependent RNA polymerase (RdRp) of DENV2 is a charming druggable target owing to its crucial function in viral reproduction. In recent years, streptomycetes natural products (NPs) have attracted considerable attention as a potential source of antiviral drugs. Methods: Seeking prospective inhibitors that inhibit the DENV2 RdRp allosteric site, in silico mining of the Streptome database was executed. AutoDock4.2.6 software performance in predicting docking poses of the inspected inhibitors was initially conducted according to existing experimental data. Upon the assessed docking parameters, the Streptome database was virtually screened against DENV2 RdRp allosteric site. The streptomycetes NPs with docking scores less than the positive control (68T; calc. −35.6 kJ.mol−1) were advanced for molecular dynamics simulations (MDS), and their binding affinities were computed by employing the MM/GBSA approach. Results: SDB9818 and SDB4806 unveiled superior inhibitor activities against DENV2 RdRp upon MM/GBSA//300 ns MDS than 68T with ΔGbinding values of −246.4, −242.3, and −150.6 kJ.mol−1, respectively. A great consistency was found in both the energetic and structural analyses of the identified inhibitors within the DENV2 RdRp allosteric site. Furthermore, the physicochemical characteristics of the identified inhibitors demonstrated good oral bioavailability. Eventually, quantum mechanical computations were carried out to evaluate the chemical reactivity of the identified inhibitors. Conclusions: As determined by in silico computations, the identified streptomycetes NPs may act as DENV2 RdRp allosteric inhibitors and mandate further experimental assays. Full article

Background: Lysosomal-associated membrane protein 1 (LAMP1), typically localized to the lysosomal membrane, is increasingly implicated as a marker of cancer aggressiveness and metastasis when expressed on the cell surface. This study aimed to develop a LAMP1-targeted antibody-based PET tracer and assess its efficacy in mouse models of human breast and colon adenocarcinoma. Methods: To determine the source of LAMP1 expression, we utilized human single-cell RNA sequencing and spatial transcriptomics, complemented by in-house flow cytometry on xenografted mouse models. Tissue microarrays of multiple epithelial cancers and normal tissue were stained for LAMP-1, and staining was quantified. An anti-LAMP1 monoclonal antibody was conjugated with desferrioxamine (DFO) and labeled with zirconium-89 (89Zr). Human triple-negative breast cancer (MDA-MB-231) and colon cancer (Caco-2) cell lines were implanted in nude mice. PET/CT imaging was conducted at 24, 72, and 168 h post-intravenous injection of 89Zr-DFO-anti-LAMP1 and 89Zr-DFO-IgG (negative control), followed by organ-specific biodistribution analyses at the final imaging time point. Results: Integrated single-cell and spatial RNA sequencing demonstrated that LAMP1 expression was localized to myeloid-derived suppressor cells (MDSCs) and cancer-associated fibroblasts (CAFs) in addition to the cancer cells. Tissue microarray showed significantly higher staining for LAMP-1 in tumor tissue compared to normal tissue (3986 ± 2635 vs. 1299 ± 1291, p < 0.001). Additionally, xenograft models showed a significantly higher contribution of cancer cells than the immune cells to cell surface LAMP1 expression. In vivo, PET imaging with 89Zr-DFO-anti-LAMP1 PET/CT revealed detectable tumor uptake as early as 24 h post-injection. The 89Zr-DFO-anti-LAMP1 tracer demonstrated significantly higher uptake than the control 89Zr-DFO-IgG in both models across all time points (MDA-MB-231 SUVmax at 168 h: 12.9 ± 5.7 vs. 4.4 ± 2.4, p = 0.003; Caco-2 SUVmax at 168 h: 8.53 ± 3.03 vs. 3.38 ± 1.25, p < 0.01). Conclusions: Imaging of cell surface LAMP-1 in breast and colon adenocarcinoma is feasible by immuno-PET. LAMP-1 imaging can be expanded to adenocarcinomas of other origins, such as prostate and pancreas. Full article

Investigating the factors influencing rice grain yield (GY) is critical for optimizing nitrogen (N) management and enhancing resource use efficiency in rice cultivation. However, few studies have comprehensively investigated the factors affecting rice GY, considering an entire influence chain encompassing rice N uptake, growth indicators, and GY components. In this study, field experiment with six different N fertilizer rates (0, 60, 120, 180, 225, and 300 kg N ha−1, i.e., N0, N60, N120, N180, N225, and N300) was conducted in the Jianghan Plain in the Middle Reaches of the Yangtze River, China, to comprehensively elucidate the factors influencing rice GY from aspects of rice N uptake, growth indicators, and GY components and determine the optimal N fertilizer rate. The results showed that rice GY and N uptake initially increased and then either stabilized or declined with higher N fertilizer rate, while apparent N loss escalated with increased N fertilizer rate. The application of N fertilizer significantly promoted the increase in straw N uptake, which was significantly positively correlated with growth indicators (p < 0.05). Among all GY components, panicle number per hill was the most significant positive factor influencing rice GY, and it was significantly positively correlated with all rice growth indicators (p < 0.05). In addition, N180 was the optimal N fertilizer rate, ensuring more than 95% of maximum GY and reducing N loss by 74% and 39% compared to N300, respectively. Meanwhile, the average N balance for N180 remained below 60 kg N ha−1. In conclusion, optimizing the N fertilizer application in paddy fields can effectively maintain stable rice GY and minimize environmental pollution. Full article

Lidar technology is a key sensor for autonomous driving due to its precise environmental perception. However, adverse weather and atmospheric conditions involving fog, rain, snow, dust, and smog can impair lidar performance, leading to potential safety risks. This paper introduces a comprehensive methodology to simulate lidar systems under such conditions and validate the results against real-world experiments. Existing empirical models for the extinction and backscattering of laser beams are analyzed, and new models are proposed for dust storms and smog, derived using Mie theory. These models are implemented in the CARLA simulator and evaluated using Robot Operating System 2 (ROS 2). The simulation methodology introduced allowed the authors to set up test experiments replicating real-world conditions, to validate the models against real-world data available in the literature, and to predict the performance of the lidar system in all weather conditions. This approach enables the development of virtual test scenarios for corner cases representing rare weather conditions to improve robustness and safety in autonomous systems. Full article

Droughts are predicted to intensify with climate change, posing a serious threat to global crop production. Increasing drought tolerance in plants requires an understanding of the underlying mechanisms. This study measured the physiological, phytohormonal and transcriptomic responses to drought in two apple rootstocks to identify drought ‘biomarkers’ and investigated whether the application of acibenzolar-S-methyl (ASM) to the roots could enhance drought tolerance. Two potted-plant trials were conducted on dwarfing (M9) and semi-dwarfing (CG202) apple rootstocks. In both trials, the response patterns in the roots and leaves were compared between irrigated and non-irrigated plants over a 14-day period. In trial 2, ASM was applied 14 days before and immediately before withdrawing irrigation. Drought induced significant decreases in transpiration, photosynthesis and stomatal conductance in both trials. This was accompanied by the accumulation of abscisic acid (ABA) metabolites and the upregulation of ABA pathway transcripts (CYP707A1/A2 and NCED3), a decrease in 12-oxophytodienoic acid (cis-OPDA) and the downregulation of ABA receptor genes (PYL4). The responses to drought were greater in the roots than the leaves, broadly similar across both rootstocks, but differed in strength and timing between the rootstocks. The application of ASM to the roots did not significantly affect the responsiveness to drought in either rootstock. The identified phytohormonal and transcriptomic biomarkers require further validation across a broader range of genotypes. Full article

Halifax Harbour, a major seaport in Nova Scotia that is approximately 100 km southeast of the Bay of Fundy, comprises a deep inner region called Bedford Basin, connected to the adjacent ocean by a shallow channel called The Narrows. A study of sea level and currents reveals the presence of episodic oscillations in The Narrows, with a period of approximately 2 h. The oscillation strength varies from day to day and, to some extent, through the seasons. The median amplitude of the associated sea level variation is 18% that of the de-tided signal, rising to 32% at the 95-th percentile. Values this large may be of concern for the transit of deep-draft vessels through shallow parts of the harbour and for the clearance of tall vessels under the two bridges that span The Narrows. Another concerning issue is the matter of oscillations being superimposed on storm surges. In addition to such direct effects of sea level variation, shear associated with the oscillations may increase the turbulent mixing in the region, affecting the overall state of this estuarine system. We explore the nature of the oscillations as a first step towards the improvement of prediction schemes for sea level and currents in the region. This involves an analysis of the oscillations in the context of seiche and Helmholtz resonance theories and the use of a 2D numerical model to handle realistic bathymetric conditions and other complications that the simpler theories cannot address. We conclude that the predictions of Helmholtz resonance theory are in reasonable agreement with both the observations and the predictions of the numerical model. Full article

In this study, a new fibre combination for an interlayer hybrid fibre-reinforced polymer laminate was investigated to achieve pseudo-ductile behaviour in tensile tests. The chosen high-strain fibre for this purpose was S-Glass, and the low-strain fibre was flax. These materials were chosen because of their relatively low environmental impact compared to carbon/carbon and carbon/glass hybrids. An analytical model was used to find an ideal combination of the two materials. With that model, the expected stress–strain relation could also be predicted analytically. The modelling was based on preliminary tensile tests of the two basic components investigated in this research: unidirectional laminates reinforced with either flax fibres or S-Glass fibres. Hybrid specimens were then designed, produced in a heat-assisted pressing process, and subjected to tensile tests. The strain measurement was performed using distributed fibre optic sensing. Ultimately, it was possible to obtain repeatable pseudo-ductile stress–strain behaviour with the chosen hybrid when the specimens were subjected to quasi-static uniaxial tension in the direction of the fibres. The intended damage-mode, consisting of a controlled delamination at the flax-fibre/glass-fibre interface after the flax fibres failed, followed by a load transfer to the glass fibre layers, was successfully achieved. The pseudo-ductile strain averaged 0.52% with a standard deviation of 0.09%, and the average load reserve after delamination was 145.5 MPa with a standard deviation of 48.5 MPa. The integrated fibre optic sensors allowed us to monitor and verify the damage process with increasing strain and load. Japanese sake, a traditional alcoholic beverage, contains several organic acids that may contribute to its sour taste. To identify these, a precolumn derivatization reagent, benzyl 5-(2-aminoethyl)-3-methyl-4-oxoimidazolidine-1-carboxylate (CIM-C2-NH2), developed for labeling carboxyl groups, was synthesized and applied to liquid chromatography–tandem mass spectrometry (LC-MS/MS) analysis of organic acids in six commercial sake samples. The majority primarily contained lactic acid (LA), and dicarboxylic acids, such as succinic acid (SA), malic acid (MA), and citramalic acid (CMA). The organic acid concentrations and compositions in the sake differed among brands. Notably, both l- and d-forms of LA were detected in all samples, while only d-CMA was present. To estimate the total acidic content, neutralization titration with sodium hydroxide was performed. In four of the six samples, titration results closely matched LC-MS/MS data, suggesting that l-LA, d-LA, SA, MA, and d-CMA were the primary contributors for the sour taste in these sakes. The discrepancy between titration and LC-MS/MS data for the other samples was attributed to the presence of other organic acids, which will be investigated in future studies. Full article

Additive manufacturing has recently become popular and more cost-effective for building construction. This study presents a prospective life cycle assessment (LCA) of 3D-printed foamed geopolymer composites (3D-FOAM materials) incorporating construction and demolition waste. The materials were developed using fly ash, slag, sand, and a foaming agent, with recycled clay brick waste (CBW) and autoclaved aerated concrete waste (AACW) added as alternative raw materials. The material formulations were evaluated for their compressive strength and thermal conductivity to define two functional units that reflect structural and thermal performance. A prospective life cycle assessment (LCA) was conducted under laboratory-scale conditions using the ReCiPe 2016 method. Results show that adding CBW and AACW reduces environmental impacts across several categories, including global warming potential and ecotoxicity, without compromising material performance. Compared to conventional wall systems, the 3D-FOAM materials offer a viable low-impact alternative when assessed on a functional basis. These findings highlight the potential of integrating recycled materials into additive manufacturing to support circular economy goals in the construction sector. Full article

Diabetic foot osteomyelitis (DFO) is a severe complication of diabetes mellitus and a leading cause of non-traumatic lower extremity amputation. Treatment remains clinically challenging with high recurrence rates despite standard antibiotic therapy and surgical debridement. This narrative review synthesizes current evidence on novel operative and nonoperative therapies for DFO, focusing on emerging biomaterials, local antibiotic delivery systems, innovative surgical techniques, and adjunctive topical agents. Studies examining bioabsorbable and nonabsorbable antibiotic carriers, such as calcium sulfate beads, collagen sponges, and bioactive glass, demonstrate promising infection resolution rates and a potential to reduce the surgical burden, though most are limited by small cohorts and observational designs. Similarly, alternative surgical approaches (i.e., cancelloplasty, conservative bone excision, and tibial cortex distraction) have shown early success in limb preservation. Nonoperative strategies, including adjunct antimicrobials, antimicrobial peptides, and topical oxygen, offer additional options, particularly for patients unfit for surgery. While initial outcomes are encouraging, the supporting evidence is heterogeneous and primarily limited to case series and small, noncomparative trials. Overall, these novel therapies show potential as adjuncts to established DFO management, but further prospective research is indicated to define their long-term efficacy, safety, and role in clinical practice. Limit Reference Points and Equilibrium Stock Dynamics in the Presence of Recruitment Depensation

Depensation (or an Allee effect) has recently been detected in stock–recruitment relationships (SRRs) in four Atlantic herring stocks and one Atlantic cod stock using a Bayesian statistical approach. In the present study, we define the Allee effect threshold (BAET) for these five stocks and propose BAET as a candidate limit reference point (LRP). We compare BAET to traditional LRPs based on proportions of equilibrium unfished biomass (B0) and biomass at maximum sustainable yield (BMSY) assuming a Beverton–Holt or Ricker SRR with and without depensation, and to the change point from a hockey stick SRR (BCP). The BAET for the case studies exceeded 0.2 B0 and 0.4 BMSY for three of the case study stocks and exceedances of 0.2 B0 were more common when the Ricker form of the SRR was assumed. The BAET estimates for all case studies were less than BCP. When there is depensation in the SRR, multiple equilibrium states can exist when fishing at a fixed fishing mortality rate (F) because the equilibrium recruits-per-spawner line at a given F can intersect the SRR more than once. The equilibrium biomass is determined by whether there is excess recruitment at the initial projected stock biomass. Estimates of equilibrium FMSY in the case studies were generally higher for SRRs that included the depensation parameter; however, the long-term F that would lead the stock to crash (Fcrash) in the depensation SRRs was often about half the Fcrash for SRRs without depensation. When warranted, this study recommends exploration of candidate LRPs from depensatory SRRs, especially if Allee effect thresholds exceed commonly used limits, and simulation testing of management strategies for robustness to depensatory effects.
