Optimizing Bioactive Compound Recovery from Chestnut Shells Using Pressurized Liquid Extraction and the Box–Behnken Design

Chestnut (Castanea sativa Mill.) is an edible nut recognized for its nutritional attributes, particularly its elevated levels of carbohydrates (starch) and proteins. Chestnuts are popular for their health-promoting properties and hold significant environmental and economic importance in Europe. During this study, after the characterization of the fruit, attention was directed toward the valorization of chestnut shells, a predominant by-product of industrial chestnut processing that is typically discarded. Valuable bioactive compounds were extracted from the shells using Pressurized Liquid Extraction (PLE), a green, efficient, scalable method. Response surface methodology (RSM) was utilized to determine optimal extraction conditions, identified as 40% v/v ethanol as the solvent at a temperature of 160 °C for 25 min under a constant pressure of 1700 psi. High total polyphenol content (113.68 ± 7.84 mg GAE/g dry weight) and notable antioxidant activity—determined by FRAP (1320.28 ± 34.33 μmol AAE/g dw) and DPPH (708.65 ± 24.8 μmol AAE/g dw) assays—were recorded in the optimized extracts. Ultrahigh-performance liquid chromatography coupled with a hybrid trap ion mobility-quadrupole time-of-flight mass spectrometer (UHPLC-TIMS-QTOF-MS) was applied to further characterize the compound profile, enabling the identification of phenolic and antioxidant compounds. These findings highlight the possibility of using chestnut shell residues as a long-term resource to make valuable products for the food, medicine, cosmetics, and animal feed industries. This study contributes to the advancement of waste valorization strategies and circular bioeconomy approaches. Full article

Asian chestnut gall wasp (ACGW) (Dryocosmus kuriphilus Yasumatsu), native to China, is an invasive pest that causes significant economic losses in Castanea species. While some cultivars show full resistance by inhibiting insect development in buds, the underlying defense mechanisms remain unclear. In this study, the accumulation and distribution of hydrogen peroxide (H2O2) were investigated in dormant buds of chestnut cultivars that are resistant and susceptible to D. kuriphilus by using the 3,3′-diaminobenzidine (DAB) staining method. Buds were examined under a stereomicroscope during key stages of pest development, including oviposition, transition from egg to larva, gall induction, and gall development. Baseline levels of H2O2 were detected in all buds; however, these levels varied among cultivars, with resistant cultivars exhibiting lower basal levels. The degree of H2O2 accumulation was found to vary depending on plant–insect interaction, physiological processes, and cultivar-specific traits. Histochemical staining revealed that brown spots indicative of H2O2 accumulation were concentrated in the vascular bundles of leaf primordia and in the apical regions. In resistant hybrid cultivars, the defense response was activated at an earlier stage, while in resistant Castanea sativa Mill. cultivars, the response was delayed but more robust. Although consistently high levels of H2O2 were observed throughout the pest interaction in susceptible cultivars, gall development was not inhibited. During the onset of physiological bud break, increased H2O2 accumulation was observed across all cultivars. This increase was associated with endodormancy in susceptible cultivars and with both defense mechanisms and endodormancy processes in resistant cultivars. These findings highlight the significant role of H2O2 in plant defense responses, while also supporting its function as a multifunctional signaling molecule involved in gall development and the regulation of physiological processes. Wildfires are integral to many forest ecosystems, yet their ecological effects are often influenced by historical land use and management. In this study, we assess the short-term impacts of fire and management on Castanea sativa Mill. stands in the fayal-brezal zone of northern Tenerife (Canary Islands), where traditional agroforestry systems have been widely abandoned. We established 12 transects across four stands: managed-burned, managed-unburned, oldfield-burned, and oldfield-unburned. We analyzed forest structure, understory species richness and composition, and soil nutrient content one year after a large wildfire. Forest structure has primarily been determined by management history, with oldfield plots showing greater tree density, basal area, and basal sprouting. Fire has had a limited effect on tree mortality, affecting ~10% of individuals on average. Understory species richness was significantly higher in managed plots, particularly those affected by fire, suggesting a positive interaction between disturbance and management. Species composition differed significantly among treatments, with Indicator Species Analysis identifying distinct taxa associated with each condition. Fire in oldfield plots led to increased compositional similarity with managed stands, indicating fire’s potential homogenizing effect. Principal Component Analysis of soil nutrients did not reveal clear treatment-related patterns, which was probably due to microenvironmental variability and the short post-fire interval. Overall, our results highlight the dominant role of land-use legacy in structuring these forests, with fire acting as a secondary but influential driver, revealing significant changes in species composition as well as in species richness. These findings have direct relevance for conservation and restoration strategies as well as for maintenance in these stands of Castanea sativa. They should also encourage managers of these protected areas, where land abandonment and fire are increasingly shaping forest dynamics. Full article

The consumption of organic foods is on the rise, as health-conscious consumers increasingly perceive them as superior both in nutritional value and for overall well-being. However, data on the nutritional and sensory properties of organic chestnuts remain scarce. This research aimed to evaluate and compare the nutritional and sensory characteristics of organic and conventional chestnuts. Results indicate that organic chestnuts exhibit a distinct sensory profile and achieve significantly higher overall scores in sensory analysis compared to conventional chestnuts. Specifically, organic chestnuts displayed stronger aromas of chestnut (9 vs. 8), hazelnut (5.87 vs. 5), almond (5 vs. 4), butter (3.96 vs. 3), and floral notes (5.95 vs. 4.96). Notably, organic chestnuts were strongly characterized by a caramel aroma, which was completely absent in conventional chestnuts (1.95 vs. 0), and probably due to the exclusive presence of decanal and 1-pentanol observed among the volatile compounds. Furthermore, organic chestnuts demonstrated a higher nutritional value, particularly in terms of antioxidant content. The total phenolic content (TPC) was significantly greater in organic chestnuts (6.54 mg GAE/g) compared to conventional samples (5.18 mg GAE/g). The relationships between attributes and consumers’ perceived liking revealed a strong association between liking and the attributes of caramel, floral, hazelnut, almond, and chestnut. These attributes are specific to organic chestnuts. As a result, both consumers and the trained panel prefer organic chestnut samples over conventional ones. Promoting the consumption of organic chestnuts by enhancing knowledge and awareness of their characteristics can encourage their use, contributing to key health and environmental sustainability goals. Chestnut flour, obtained through drying and milling of Castanea sativa fruits, has evolved from a subsistence food into a sought-after niche product, appreciated for its naturally gluten-free profile, high starch content, and richness in micronutrients. Over the past decade, its demand has steadily increased due to consumer perception of the health benefits associated with chestnut consumption. As the market for chestnut flour expanded from small-scale to large-scale production, alternative methods to the traditional process were developed. Its distinctive aroma and flavor are strongly influenced by processing methods, which are the focus of this study. Two drying approaches were compared: a traditional smoke-based method (drying house named metato) characterized by a wood-drying method and a controlled laboratory process using a forced-air dryer that maintained a constant temperature of 40 °C. The impact of these methods on the physico-chemical composition, volatile organic compounds (VOCs), and sensory properties of the flour was evaluated using chemical, instrumental, and sensory analyses. The traditional method enhanced the flour’s aromatic complexity and typicity through the application of smoke, which has been demonstrated to generate volatile organic compounds (VOCs), such as guaiacol, furfural, and o-creosol, that are associated with the smoked aroma. Nevertheless, if not properly managed, it can lead to undesirable sensory notes due to excessive smoke exposure. In contrast, the laboratory-controlled process ensured better preservation of bioactive compounds—such as polyphenols (351 mg GAE/100 g dm) and ascorbic acid (322 mg/kg dm)—while retaining the aroma notes associated with fresh chestnuts. Optimizing processing methods may support the valorization of chestnut flour as a high-quality ingredient in the modern gluten-free and functional food market. Full article

Ink disease, primarily caused by the pathogen Phytophthora xcambivora, significantly threatens the health and productivity of sweet chestnut (Castanea sativa Mill.) orchards, highlighting the need for accurate detection methods. This study investigates the efficacy of machine learning (ML) classifiers combined with high-resolution multispectral imagery acquired via unmanned aerial vehicles (UAVs) to assess chestnut tree health at a site in Tuscany, Italy. Three machine learning algorithms—support vector machines (SVMs), Gaussian Naive Bayes (GNB), and logistic regression (Log)—were evaluated against eight vegetation indices (VIs), including NDVI, GnDVI, and RdNDVI, to classify chestnut tree crowns as either symptomatic or asymptomatic. High-resolution multispectral images were processed to derive vegetation indices that effectively captured subtle spectral variations indicative of disease presence. Ground-truthing involved visual tree health assessments performed by expert forest pathologists, subsequently validated through leaf area index (LAI) measurements. Correlation analysis confirmed significant associations between LAI and most VIs, supporting LAI as a robust physiological metric for validating visual health assessments. GnDVI and RdNDVI combined with SVM and GNB classifiers achieved the highest classification accuracy (95.2%), demonstrating their superior sensitivity in discriminating symptomatic from asymptomatic trees. Indices such as MCARI and SAVI showed limited discriminative power, underscoring the importance of selecting appropriate VIs that are tailored to specific disease symptoms. This study highlights the potential of integrating UAV-derived multispectral imagery and machine learning techniques, validated by LAI, as an effective approach for the detection of ink disease, enabling precision forestry practices and informed orchard management strategies. Monitoring vegetation phenology is crucial for understanding how plants respond to climate change and how the latter affects the role of vegetated ecosystems in biosphere cycles. It has been reported that the growing season has been extended, leading to an increase in global terrestrial productivity, but not much attention has been given to how different climatic variables affect specific tree species’ phenology. This study focuses on the main phenological events (SOS, Start Of Season; EOS, End Of Season; and LOS, Length Of Season) of two deciduous species (Fagus sylvatica L. and Castanea sativa Mill.) and the effects of temperature and precipitation on them. The analysis concerns a 23-year period (2000–2022) of various sites across southern Europe. The dates for each phenological event are estimated based on NDVI timeseries from MODIS satellite sensor. Both species show an elongation of their growing season, with SOS occurring 2.09 and 1.63 days/decade earlier and EOS 2.97 and 3.03 days/decade later for Fagus sylvatica and Castanea sativa, respectively, with this trend appearing more intense at lower altitudes. Temperature seems to be the major driver for these changes for both species, with higher temperatures before each phenological event leading to earlier SOS and delayed EOS. The effects of precipitation are less homogenous, showing different trends between sites and species. Full article

The aim of our study was to compare the composition of polyphenolic compounds between the Croatian acacia (Robinia pseudoacacia L.) and chestnut (Castanea sativa Mill.) honey from several aspects: production season, hive position (on the edge and in the middle of a series of hives), part of the hive (small or normal extension), and honey extraction method (centrifuging or draining honey combs). Additionally, in acacia honey, we also monitored the influence of different storage temperatures (room temperature (RT) and 4 °C) on the content of phenolic compounds. To separate, identify and quantify individual flavonoids and phenolic acids from the honey, we used the HPLC method. The total polyphenols and antioxidant activity of the samples, their antimicrobial activity and their elemental content were also measured. The significant influence of the season, hive position, and extraction method on the total identified phenolic compounds, phenolic acids, flavonoids, total phenols and antioxidant activity was detected in almost all the acacia and chestnut honey samples. Chestnut honey from 2013 had more total phenolics (TPs) and antioxidant capacity (FRAP) than chestnut from 2014 and 2015. Honey collected from smaller extensions of hives had significantly higher TPs and FRAP compared to normal hive extensions. Centrifugation reduced the TPs and FRAP in most cases, but not always uniformly. Storage at RT led to the predominance of gallic, p-coumaric and benzoic acid in acacia honey, while storage at 4 °C maintained p-coumaric acid as the dominant phenolic acid. Flavonoids, particularly pinobanksin in acacia honey and hesperetin/pinobanksin in chestnut honey, were less affected by the storage conditions compared to phenolic acids. The non-centrifuged chestnut sample from 2015 showed the lowest MIC values against the most tested pathogenic bacteria. All the honey samples showed an extremely low concentration of heavy metals and relatively high concentrations of potassium and calcium. Full article

The sustainability of agriculture is seriously threatened by climate change. In Europe, chestnut ecosystems, which are growing mainly in Mediterranean climate, are facing during summertime increasing of heat and drought stresses. These induce fragilities on trees, leading to a reduction in productivity and predisposing them to pest and disease attacks. The plasticity of chestnut species under contrasting climate is known. Understanding the specific adaptation of cultivars to different climate features is now important to anticipating climate changes. Caucasian Region is considered the origin center of chestnut (Castanea sativa), which is characterized by climatic transition from the Mediterranean to the Euro-Siberian area. Mostly, areas of chestnut are concentrated in the countries around the Mediterranean Basin, thriving in regions with humid and Pré-Atlantic bioclimates. In Portugal, more than 95% of the chestnut area is located in the Center and North side of Portugal. This is an anisohydry species, characterized by good hydroplasticity: 90% reduction in A occurs when Ψwstem drops to −1.25 MPa, and a 50% reduction in A occurs at values of −1.7 MPa. The highest fatty acid contents in chestnut chloroplasts are a-linolenic acid (18:3), ranging between 40 and 50% of the total amount and being the unsaturated/saturated 2.27 for Longal. New strategies are being investigated in order to increase tolerance against those abiotic factors in chestnut species. They include the use of innovative irrigation techniques, which can increase production 22–37%. Fertilization with silicone (Si) has been investigated to promote the tolerance of plants against heat and drought stresses. Breeding programs, mostly (in Europe) against ink disease, have been performed since the middle of the XX century to create new genotypes (such the Portuguese ColUTAD®). ClimCast, a network of orchards, was created in Portugal with the aim of responding to the new challenges facing orchards in the context of climate change. Full article

The objective of the present study was to characterize the chestnut (Castanea sativa Mill.) tannin product, Farmatan Plus® (Tanin Sevnica d.d., Sevnica, Slovenia), and to subsequently examine its effects on the blood metabolic parameters of fattening lambs, particularly in relation to their health status. Thirty lambs were randomly divided into three treatment groups: a control group without added tannin and two groups that received 9.46 g of the tannin product/kg of the diet dry matter (DM) and 18.87 g of the tannin product/kg of the diet DM. Metabolic parameters such as contents of total protein, globulin, urea, and liver enzymes (AST and GGT) were measured over a trial period of 60 days to evaluate the effects of tannin supplementation. This study represents the first in-depth characterization of Farmatan Plus®, demonstrating its richness in bioactive compounds such as vescalin and castalagin. The results showed no significant adverse effects on lamb health, with all parameters remaining within normal physiological ranges (p > 0.05). These results support the safe inclusion of chestnut tannins in the diet of lambs and underline their potential as a functional feed additive that can positively influence the health and growth performance of ruminants. Since its introduction into Europe in the first half of the 20th century, Cryphonectria parasitica has been gradually spreading across the natural range of the sweet chestnut (Castanea sativa Mill.), infecting the trees and causing lethal bark cankers. Serendipitously, a hyperparasitic Cryphonectria hypovirus 1 (CHV1), which attenuates C. parasitica virulence in combination with more tolerant European chestnut species, was able to ward off the worst effect of the disease. In North America, unfortunately, the native Castanea dentata is now functionally extinct since it occurs only as root sprouts in eastern deciduous forests where it was once dominant. In our work, we investigated changes in C. parasitica populations over time and the regional variability in chestnut populations’ tolerance toward the blight disease. While vegetative compatibility (vc) type diversity and prevalence of hypovirulence remained similar as in previous studies, in the Buje population, unlike in previous studies, we were unable to find any hypovirulent fungal strains. The most common vegetative compatibility types (vc types) were EU-1, EU-2 and EU-12. However, several rare EU-types were found, including one previously unreported: EU-46. By inoculating several C. parasitica strains on tree stems from several chestnut populations, we observed that the induced lesion size was affected by the type of inoculum (CHV1-free or CHV1-infected), genotype-related individual chestnut stem and chestnut stem population of origin-related variability. The largest lesions were induced by CHV1-free fungal isolate DOB-G: 20.13 cm2 (95% C.I. 18.10–22.15) and the smallest by CHV1-infected L14/EP713: 2.49 cm2 (95% C.I. 1.59–3.39). Surprisingly, the size of the lesions induced by other CHV1-infected strains fell somewhere in between these extremes. The size of induced lesions was dependent on the population of origin as well and ranged from 11.60 cm2 (95% C.I. 9.87–13.33) for stems from the Moslavačka gora population to 17.75 cm2 (95% C.I. 15.63–19.87) for stems from Ozalj. The chestnut tree (Castanea sativa Mill.) is gaining importance in the mountainous regions of southern Europe due to the high value of its fruits. It is essential to establish effective cultivation protocols, considering that this species is still relatively understudied. In this study, we present the outcomes of the initial establishment of a chestnut orchard conducted through a nutrient omission trial for four years. The treatments included a fertilization plan with nitrogen, phosphorus, potassium, and boron (NPKB), the control, and four other treatments corresponding to the omission of each nutrient (-NPKB, N-PKB, NP-KB, NPK-B). The -NPKB and NP-KB treatments showed significantly lower trunk circumferences and canopy volumes compared to the other treatments. The NPK-B treatment resulted in the lowest fruit production, with a total accumulated yield (2020–2022) of 0.56 kg tree–1, a value significantly lower than that of NPKB (1.12 kg tree–1) and N-PKB (1.19 kg tree–1). The assessment of nutrient concentrations in the leaves revealed plants with deficient levels of B and K in treatments that did not receive these nutrients. Conversely, N levels in the leaves in the -NPKB treatment fell within the sufficiency range (20 to 28 g kg–1). This suggests that the sufficiency range should be adjusted to higher values, given the treatment’s effect on tree growth. It was also observed that the -NPKB treatment led to lower soil organic matter compared to the other treatments, likely due to reduced herbaceous vegetation development under the canopy, leading to decreased organic substrate deposition in the soil. The main findings of this study are that N and K were crucial elements for the optimal growth of chestnut trees, while B played a significant role in fruit production. Full article

Chestnut (Castanea sativa Mill.) forests in the Mediterranean region are facing increasing abandonment due to a combination of factors, ranging from climate change to socioeconomic issues. The recovery of chestnut ecosystems and their preservation and valorization are key to ensuring the supply of the wide spectrum of ecosystem services they provide and to preventing detrimental environmental shifts. The study’s objective was to provide evidence on the effects of different management options on the ecophysiology of chestnut forests, with diverse pruning intensities (low, medium, and high intensity versus no pruning) tested in an abandoned chestnut stand in central Italy with the aim of recovering and rehabilitating it for fruit production. Innovative Internet of Things (IoT) ‘Tree Talker’ devices were installed on single trees to continuously monitor and measure ecophysiological (i.e., water transport, net primary productivity, foliage development) and microclimatic parameters. Results show a reduction in water use in trees subjected to medium- and high-intensity pruning treatments, along with a decrease in the carbon sequestration function. However, interestingly, the results highlight that trees regain their usual sap flow and carbon sink activity at the end of the first post-pruning growing season and fully realign during the following year, as also confirmed by the NDVI values. As such, this paper demonstrates the efficacy of recovering and managing abandoned chestnut forests, and the initial setback in carbon sequestration resulting from pruning is rapidly remedied with the advantage of reviving trees for fruit production. Additionally, the reduced water demand induced by pruning could represent a promising adaptation strategy to climate change, bolstering the resilience of chestnut trees to prolonged and intensified drought periods, which are projected to increase under future climate scenarios, particularly in the Mediterranean region. The chestnut tree (Castanea sativa Mill.) is a widespread plant in Europe, rich in high-value compounds, which can be divided mainly into monomeric polyphenols and tannins. These compounds exhibit various biological activities, such as antioxidant, as well as anticarcinogenic and antimicrobial properties. Chestnut wood (CW) extracts were prepared using different extraction techniques, process conditions, solvents, and their mixtures. This work aimed to test various extraction techniques and determine the optimal solvent for isolating enriched fractions of vescalagin, castalagin, vescalin, and castalin from CW residues. Supercritical CO2 extraction with a more polar cosolvent was applied at different pressures, which influenced solvent density. According to the results, the proportions of the components strongly depended on the solvent system used for the extraction. In addition, HPLC-DAD was used for semiqualitative purposes to detect vescalagin, castalagin, vescalin, and castalin. The developed valorization protocol allows efficient fractionation and recovery of the polyphenolic components of CW through a sustainable approach that also evaluates pre-industrial scaling-up. Castanea sativa Mill. (C. sativa) processing and pruning generate several by-products, including leaves, burs, and shells (inner and outer teguments), which are considered an important source of high-value phytochemicals. Ellagitannins from C. sativa leaf extracts have been described to impair H. pylori viability and inflammation in gastric cells. Furthermore, chestnut shells showed an important anti-inflammatory effect in gastric epithelial cells. Dietary polyphenols, including tannins, have been reported to interfere with targets of inflammation, including the nuclear factor κB (NF-κB). A promising role as a further therapeutical target for gut disorders has been recently proposed for the regulatory subunit of hypoxia-inducible factor (HIF-1α), as a potential stabilizer of intestinal barrier integrity. Therefore, the main objective of this work is the chemical characterization of several chestnut by-products (bud, spiny bur, wood, pericarp and episperm), together with the exploitation of their anti-inflammatory properties in intestinal cells, scavenging capacity, and stability following gastrointestinal digestion. The chemical characterization confirmed the presence of bioactive polyphenols in the extracts, including ellagitannins. In CaCo-2 cells stimulated by an IL-1β-IFN-γ cocktail, nearly all chestnut by-products (50 µg/mL) inhibited the release of proinflammatory mediators (CXCL-10, IL-8, MCP-1, ICAM), along with the NF-κB-driven transcription, and induced the HRE-driven transcription. The stability of the most promising extracts, identified through PCA and cluster analysis, was addressed by in vitro gastrointestinal digestion. Despite the significant reduction in total polyphenol index of chestnut bud and wood after gastric and intestinal digestion, the activity of these extracts on both scavenging and anti-inflammatory parameters remained promising. These data contribute to exploit the potential of chestnut by-products as sources of dietary polyphenols with anti-inflammatory properties at the intestinal level. Moreover, this study could represent an important step to encourage the recycling and valorization of chestnut by-products, promoting the circular economy and reducing the environmental impact related to the management of agriculture waste.
