The aim of this study was to evaluate setting time, hardness, solubility, bioactivity and interaction with dentin of four calcium silicate-based sealers (CSBS). Three single-phase CSBS (AH Plus Bioceramic/AHB, CeraSeal/CSL, TotalFill BC/TFL), one powder/liquid CSBS (BioRoot RCS/BRT) and an epoxy control (AH Plus Jet/AHP) were investigated. Setting time was evaluated on glass (G1) and dentin (G2) surfaces, by adding 1%wt purified water to single-phase products. For hardness measurements, the Shore-D hardness test was used. Solubility was assessed according to the ISO 6876:2012 standard. For bioactivity screening, 1-week set specimens were immersed in SBF or water (30 days/37 °C) and examined by ATR–FTIR spectroscopy. Interaction with dentin was tested by ATR–FTIR before and after contact with the sealers. For setting time in G1, all CSBS failed to comply with the ISO standard, while in G2, most materials were set in the range of 6–8 h, except for CSL. The ranking of significant differences in hardness was AHP, BRT > CSL, AHB, TFL. Regarding solubility, AHB, BRT and AHP were found to comply with the ISO standard, whereas CSL and TFL failed. For bioactivity, characteristic peaks of calcium phosphates were found in all CSBS, with TFL being the most bioactive. A chemical interaction between CSBS and dentin was registered, with a strong reduction in collagen peaks and an increase in carbonates. The CSBS tested exhibited great variance in their behaviour regarding the properties assessed, although a strong deproteinating effect was registered on dentin for all. Full article

Objectives: This study evaluated and compared the sealing ability and elemental composition of a resin-based endodontic sealer (AH Plus) used with three root canal obturation techniques: single cone (SC), lateral compaction (LC), and warm vertical condensation (WVC). The investigation focused on microstructural characteristics, interfacial integrity, and elemental distribution within filled root canals. Material and Methods: Sixty extracted single-root teeth were instrumented using the ProTaper Gold system and randomly assigned to three groups (n = 20) according to the obturation technique. The AH Plus Jet sealer was applied in all cases. Following obturation, samples were subjected to radiographic investigation and analyzed using optical microscopy and scanning electron microscopy (SEM) coupled with energy-dispersive X-ray spectroscopy (EDX) to assess the sealing performance and chemical composition. Results: Radiographic and microscopic assessments indicated that the SC method showed strong gutta-percha adhesion to dentin with a thin cement layer, whereas WVC provided excellent adaptation and penetration of gutta-percha. The LC technique demonstrated good adhesion but displayed occasional structural irregularities. SC has the thicker adhesion layer with uneven distribution regarding coronal, median, and apical, regions ranging from 45 to 80 μm, while WVC ensures a thin and uniform sealing layer of about 35 μm in all regions. SEM and EDX analyses detailed the interfacial microstructure and confirmed the presence of carbon (C), oxygen (O), calcium (Ca), zinc (Zn), barium (Ba), and sulfur (S) across all groups. Conclusions: All three obturation techniques (SC, WVC, LC) achieved effective sealing when combined with the AH Plus sealer. The main difference between the methods consists of the sealer layer thickness and its even distribution regarding gutta-percha cones. Full article

This study aims to compare the osteogenic potential of premixed hydraulic calcium silicate-based sealers (HCSSs) with an epoxy resin-based sealer in human bone marrow-derived stem cells (hBMSCs). Three HCSSs (White Endoseal MTA, One-Fil, EndoSequence BC Sealer) were compared with AH Plus Jet, an epoxy resin-based sealer. Disk-shaped specimens were prepared using sterilized Teflon tubes and immersed in osteogenic medium to create eluates. hBMSCs were cultured in each eluate, and osteogenic potential was assessed by alkaline phosphatase (ALP) activity (n = 6), Alizarin Red-S (ARS) staining (n = 6), quantitative real-time polymerase chain reaction (qPCR) (n = 3), and Western blot analysis. Statistical analyses were conducted using SPSS (version 24.0), with significance set at p < 0.05. All experimental groups exhibited higher ALP activity than the control on day 4. ARS staining of HCSSs differed significantly from AH Plus Jet on day 14 (p < 0.05), while White Endoseal MTA exhibited the highest intensity. qPCR revealed that EndoSequence BC Sealer induced the highest SMAD1 expression on day 4, while One-Fil and EndoSequence BC Sealer significantly upregulated RUNX2 expression compared with AH Plus Jet (p < 0.05). Western blotting confirmed that EndoSequence BC Sealer induced the highest RUNX2 protein expression. Collectively, premixed HCSSs promoted superior mineralization and RUNX2 expression compared to conventional resin-based sealer in hBMSCs. Full article

This study evaluated how varying calcium silicate (30–50%) and radiopacifier (45–65%) ratios affected the physicochemical properties of experimental sealers (P1–P3) compared to Bio-C Sealer, AH Plus Bioceramic, and AH Plus Jet. Properties such as flowability, solubility, and radiopacity were assessed per ISO 6876/2012, and setting time followed ASTM C266-2008. pH and volumetric changes were measured in acrylic teeth (n = 10) filled with each sealer, with pH evaluated at 3, 24, 72, and 168 h, and volume changes assessed via micro-CT at baseline and after 7 days. P1 showed setting time similar to AH Plus Bioceramic (p > 0.05). All calcium silicate-based sealers had greater solubility than AH Plus Jet (p < 0.05), with only P3 meeting ISO standards. P1 and AH Plus Jet exhibited higher radiopacity (p < 0.05). Bio-C Sealer and AH Plus Bioceramic had more volume change than P2 (p < 0.05). All experimental sealers showed stable volume and increasing alkalinity, peaking at 72h. Bio-C Sealer showed a consistent pH rise, except between 24 and 168 h (p > 0.05). Sealer properties were strongly influenced by the composition. P1 (30% calcium silicate/65% radiopacifier) had superior flowability and radiopacity, while P3 (50% calcium silicate/45% radiopacifier) was the only one to meet solubility standards. Full article

Background/Objectives: We aimed to assess the bond strength of AH Plus Jet (AH) and an epoxy resin-based root canal sealer prototype (K-0189) adhered to three different obturation points. Methods: A total of 120 single-rooted teeth were selected after radiographic analysis, and their root canals were instrumented with ProTaper Next files (PTN) up to size X5. The teeth were randomly assigned to two sealer groups (G) (G1: AH, G2: K-0189, each n = 60) and further divided into three subgroups: (A) ConformFit X5 points (PTN) cold obturation (CO), (B) ProTaper Universal F5 points (PTU) (CO), (C) GuttaCore X5 (GC) warm obturation (WO). After final irrigation (NaOCl 3%, EDTA 17%, NaOCl 3%) and drying, root canals were obturated and stored for 30 days at 37 °C in Simulated Body Fluid (SBF). The specimens were embedded in acrylate and sectioned horizontally; then, push-out bond strength (POBS) analysis was performed. Results: The median POBS values [MPa] were G1A: 2.03; G1B: 2.12; G1C: 3.2; G2A: 1.91; G2B: 2.56; and G2C: 3.36. WO showed significantly higher POBS (p < 0.001 *) than CO. The POBS was not significantly different between the two WO groups (p = 0.508). The POBS of G2B was significantly higher compared to all other CO groups. Conclusions: The epoxy resin sealer prototype demonstrated POBS values comparable to AH Plus when used with WO and PTU points, indicating significantly higher POBS values compared to all other CO points. Background/Objectives: Since biomineralization by calcium silicate-based sealers (CSBSs) was reported, retrieving canal filling materials may be challenging during endodontic retreatment due to their adhesion to dentin. This study aimed to evaluate the possibility of removing residual mineral deposits from two kinds of CSBSs compared to the AH Plus Jet (AHJ). Methods: Root canals of mandibular premolars were prepared, obturated with the sealer-based obturation method using a WOG medium gutta-percha cone and one of the following sealers: AHJ, AH Plus Bioceramic (AHB), and Ceraseal (CER) (n = 12/group). After 3 weeks, endodontic retreatment was conducted with the WOG files, followed by instrumentation with XP-endo Finisher (XPF). Micro-computed tomography scanning was obtained after canal filling, after retreatment with WOG, and after the use of XPF. The percentage of the removed filling volume was calculated. One-way ANOVA with Tukey’s test and a non-parametric test with Bonferroni’s correction were performed. Root canal dentin after retreatment was examined using a scanning electron microscope (SEM). Results: After supplementary instrumentation with XPF, the mean residual filling volumes for the AHJ, AHB, and CER groups were 1.35 mm3, 0.55 mm3, and 0.82 mm3, respectively. The AHJ group showed greater residual volume compared to the AHB group (p < 0.05). The AHB and CER groups demonstrated higher mean percentages of removed filling volume at 94.8%, and 92.5%, respectively, compared to 87.1% for the AHJ group (p < 0.05). More mineral deposits were observed in the CER group with SEM. Conclusions: AHB and CER are retrievable during endodontic retreatment, with CER preferable due to greater mineral deposits in dentinal tubules. Full article

Background/Objectives: Microorganisms are the leading cause of infections in the root canal system, contributing to the failure of endodontic treatments. This in vitro study aimed to compare the antimicrobial effects of four different endodontic sealers: Endomethasone N (Septodont, Saint Maur-des-Fossés, France), Sealapex (Kerr Corporation, Orange, CA, USA), AH Plus Jet (Dentsply DeTrey GmbH, Konstanz, Germany), and MTA Fillapex (Angelus, Londrina, Brazil). Methods: The sealers were tested against common oral pathogens, including Enterococcus faecalis, Staphylococcus aureus, Escherichia coli, Candida albicans, and Streptococcus mutans, using the agar diffusion method. Inhibition zones were measured at 24, 48, and 72 h to assess antimicrobial efficacy. Results: The results showed that Endomethasone was the most effective sealer against all tested microorganisms, demonstrating consistent inhibition across all time intervals. MTA Fillapex also exhibited a significant antimicrobial effect, particularly against Streptococcus mutans, with its efficacy increasing over time. AH Plus Jet displayed limited effectiveness, showing significant results only against Staphylococcus aureus. Conclusions: Overall, this study confirms the superior antimicrobial performance of Endomethasone, while the other materials, particularly MTA Fillapex and Sealapex, also showed notable effects in experimental conditions. The antimicrobial activity of all materials, except AH Plus Jet, increased over the 72-h period. Full article

Background: Using endodontic sealers with long-term antimicrobial properties can increase the success of endodontic treatment. This study aimed to assess the antimicrobial activity over time of two calcium silicate (CS)-based sealers, AH Plus Bioceramic and BioRoot RCS, and to compare them with an epoxy resin-based sealer, AH Plus Jet, against mature polymicrobial biofilms grown on human radicular dentin. Methods: The antimicrobial activity of the sealers was tested using a direct contact test after 1 and 6 weeks of contact with the biofilms. Cell viability was determined by the adenosine triphosphate (ATP) method and flow cytometry (FC). The results of the ATP test were analyzed using an ANOVA with Welch’s correction, followed by the Games–Howell test. The number of cells with damaged membranes obtained by FC in each period was compared by means of an ANOVA and Duncan’s test. For the comparison between times, a Student’s t-test was used. Results: Globally, after a week of contact, the epoxy resin-based sealer obtained the best results. However, at 6 weeks, the two CSs showed the highest antimicrobial efficacy, with a significant increase in this activity over time. Conclusions: Calcium silicate-based sealers exert long-term antimicrobial activity against endodontic biofilms. Full article

This study aimed to examine how heating affects the physical properties of a newly developed premixed calcium silicate-containing sealer (AH Plus Bioceramic Sealer; AHB), in comparison with EndoSequence BC Sealer (ES), AH Plus Jet (AH), and Pulp Canal Sealer. The setting time, flow, and film thickness were tested with or without heating at 100 °C for 30 or 60 s, in accordance with ISO6876:2012 standards. Ultrastructural and elemental analyses were performed with scanning electron microscopy (SEM) and energy-dispersive X-ray spectroscopy (EDS). Data were analyzed using one-way analysis of variance (ANOVA) with a Tukey post hoc test (α = 0.05). All sealers exhibited significantly shorter setting time and flow after heating at 100 °C for 30 and 60 s (p < 0.05). After heating, AHB showed a significantly higher film thickness compared to that of the other materials (p < 0.05). None of the tested properties of heat-applied AHB and ES met ISO standards, except the setting time in ES. The SEM/EDS results for AHB and ES were not affected by heating. The detected changes in physical properties can negatively impact the performance of premixed calcium silicate-containing sealers, particularly AHB, when warm vertical compaction is employed. Background: Once the chemo-mechanical preparation of root canals is finished, achieving a complete seal of the root canal system becomes crucial in determining the long-term success of endodontic treatment. The important goals of root canal obturation are to minimize leakage and achieve an adequate seal. Thus, a material that possesses satisfactory mechanical characteristics, is biocompatible, and has the ability to penetrate the dentine tubules adequately is needed. Aim: This study aimed to compare the penetrability and bond strength between two calcium silicate-based sealers and an epoxy resin-based sealer, as well as examine the relationship between penetrability and bond strength for the different sealers. Method and materials: Thirty-nine recently extracted single-rooted human premolar teeth were instrumented and divided evenly into three groups (n = 13), according to the sealer used for obturation: AH Plus Jet, EndoSequence, and AH Plus Bioceramic Sealer. Three teeth (30 slices) were randomly selected out of each for analysis using confocal laser scanning microscopy to assess penetrability. The remaining ten teeth (90 slices) in each group were subject to push-out tests using a universal testing machine. All teeth were sectioned into nine transverse slices of 0.9 mm thickness for their respective tests (apical, middle, coronal). Results: AH Plus Jet exhibited significantly lower penetrability and significantly higher bond strength compared to EndoSequence BC sealer (p = 0.002) and AH Plus Bioceramic Sealer (p = 0.006). There was no significant difference between EndoSequence BC sealer and AH Plus Bioceramic Sealer in terms of either penetrability or bond strength. No correlation was found between penetrability and bond strength. Conclusions: Within the limitation of this study and regardless of the location in the canal, the bioceramic based root canal sealers appeared to perform better than the epoxy resin-based sealer in terms of dentinal penetration rate. Further studies are required to compare other biomechanical properties of bioceramic sealers including setting characteristics and bacterial leakage. Full article

Aim: The aim of this study was to assess the impact of the final irrigation protocol on the push-out bond strength of calcium silicate-based sealers when compared to an epoxy resin-based sealer. Materials and Methods: Eighty-four single-rooted mandibular human premolars were shaped using the R25® instrument (Reciproc, VDW, Munich, Germany) and then divided into three subgroups of 28 roots each depending on the final irrigation protocol: EDTA (ethylene diamine tetra acetic acid) and NaOCl activation, Dual Rinse® HEDP (1-hydroxyethane 1,1-diphosphonate) activation or sodium hypochlorite (NaOCl) activation. Then, each subgroup was divided into two groups (14 each) according to the sealer used (AH Plus Jet® or Total Fill BC Sealer®) for single-cone obturation. Dislodgement resistance using a universal testing machine, samples’ push-out bond strength and failure mode under magnification were determined. Results: EDTA/Total Fill BC Sealer® showed significantly greater values of push-out bond strength compared with HEDP/Total Fill BC Sealer® and NaOCl/AH Plus Jet®, with no statistical difference with EDTA/AH Plus Jet®, HEDP/AH Plus Jet® and NaOCl/Total Fill BC Sealer®, whereas HEDP/Total Fill BC Sealer® showed significantly lower values of push-out bond strength. The apical third demonstrated higher means of push-out bond strength compared with middle and apical thirds. The most common failure mode was cohesive but showed no statistical difference compared to other types. Conclusions: Irrigation solution and final irrigation protocol affect the adhesion of calcium silicate-based sealers. Full article
