Dear Colleagues,
This Special Issue, “Recent Advances of Anti-Infective Activity Compounds”, compiles high-level contributions centered on the chemical innovation, molecular optimization, and mechanistic characterization of compounds with activity against a broad spectrum of infectious agents. Emphasis is placed on advances in synthetic methodologies, molecular design strategies, and structural refinement aimed at enhancing potency, selectivity, physicochemical balance, and translational relevance. This edition highlights progress in the development of diverse scaffolds, including non-classical chemotypes, heterocyclic frameworks, hybrid structures, bioisosteric derivatives, metal-based entities, and prodrug systems activated by pathogen-specific biochemical transformations. Contributions applying integrated medicinal chemistry approaches, such as structure-guided optimization, ligand-based modeling, conformational control, and advanced computational workflows, are particularly valued. Studies employing state-of-the-art analytical, biophysical, and in vitro/in vivo methodologies to elucidate interaction profiles and stability, permeability, ADME characteristics, and PK/PD behavior further enrich this collection. Altogether, this Special Issue reflects the current landscape of anti-infective compound development and delineates emerging molecular strategies to address global challenges associated with infectious diseases and drug resistance.
