Dear Colleagues,
Recent advances in cancer therapeutics have shifted focus toward understanding the interplay of epigenetics, me-tabolism, and the tumor microenvironment (TME), recognizing their crucial roles in tumor initiation, progression, and resistance to therapies. Metabolic changes are closely linked to the TME, which influences epigenetic modifica-tions that regulate gene expression and, consequently, contribute to cancer cell plasticity. Exploiting epigenetic al-terations through the development of epigenetic modulators offers the possibility to reverse malignant phenotypes. Simultaneously, metabolic reprogramming offers novel targets for disrupting tumor growth. Furthermore, modu-lating the TME, including immune infiltration and stromal interactions, provides opportunities to overcome re-sistance and improve responses to chemotherapy and immunotherapy. Integrating these approaches holds the po-tential for designing novel, personalized, and more effective therapeutic regimens. This Issue aims to explore inno-vative strategies targeting these interconnected pathways in order to enhance treatment efficacy and therapeutic outcome.
