Dear Colleagues,
Mitochondria are no longer viewed as mere “powerhouses of the cell,” but as dynamic signaling organelles that integrate metabolic, immunologic, and stress responses to regulate cellular homeostasis and fate. This Special Issue aims to provide a holistic view of recent advances redefining mitochondrial biology beyond ATP generation. Mitochondria coordinate diverse processes such as ROS signaling, apoptosis, innate immune activation, and epigenetic regulation through metabolite flux and post-translational modifications, and their dysfunction drives cancer, neurodegeneration, metabolic disease, and aging. This collection will feature original research and reviews exploring mitochondrial dynamics and quality control; mitonuclear communication; mitochondrial nucleic acids in immune signaling; and organelle crosstalk with the ER, lysosome, and nucleus. It will also highlight innovations in mitochondrial imaging, multi-omics, and genome editing that reveal new regulatory mechanisms. By integrating insights from molecular biology, metabolism, and immunology, this Special Issue will position mitochondria as central hubs of cellular communication, emphasizing their pivotal role in linking metabolic states to immune and stress responses, and their potential as therapeutic targets in complex diseases.
