Dear Colleagues,
Neurodegenerative diseases are characterized by progressive neuronal loss and/or dysfunction resulting from complex changes in nervous system dynamics. Recent reports have shifted the focus of research on this topic from isolated molecular events to integrated circuit-level mechanisms involving synaptic plasticity, network connectivity, and neuroinflammation. Disruptions in excitatory–inhibitory balance, mitochondrial dysfunction, oxidative stress, and trace element homeostasis (particularly zinc) are increasingly recognized as significant contributors to neurodegenerative pathology.
Emerging experimental and computational approaches, including in vivo imaging, optogenetics, and systems neuroscience, have provided new insights into how these factors reshape neuronal circuits in conditions such as Alzheimer's disease, Parkinson's disease, and multiple sclerosis. Understanding these mechanisms is crucial for identifying novel therapeutic targets that aim to restore network function rather than simply address neuronal loss.
This Special Issue aims to highlight recent advances in the circuit mechanisms underlying neurodegenerative diseases and to develop environmental perspectives for future interventions by bridging the gap between molecular findings and system-level dysfunction.
