Special Issue Information

Dear Colleagues,

 

Autoimmune diseases constitute a heterogeneous group of chronic conditions characterized by aberrant immune responses against self-antigens, leading to tissue damage and functional impairment. Despite advances in understanding their clinical manifestations, many autoimmune diseases remain poorly controlled with conventional therapies. Recent breakthroughs in molecular immunology have elucidated key pathogenic mechanisms, including genetically susceptible loci, epigenetic modifications, dysregulated cytokine networks, and the breakdown of immune tolerance. These discoveries have paved the way for novel therapeutic approaches aimed at selectively targeting immune pathways with improved efficacy and safety.

Biologic agents such as TNF-α, IL-6, and B-cell inhibitors have transformed the treatment landscape of conditions like rheumatoid arthritis, systemic lupus erythematosus, and multiple sclerosis. Moreover, the emergence of small-molecule inhibitors targeting intracellular signaling cascades (e.g., JAK-STAT pathway) offers additional options for patients with refractory disease. Advances in cell-based immunotherapies, microbiome modulation, and tolerance-inducing strategies further expand the horizon of precision medicine in autoimmunity. Integration of multi-omics technologies and artificial intelligence into clinical practice is expected to improve disease prediction, classification, and individualized treatment selection.

This Special Issue highlights the molecular underpinnings of autoimmunity and explores the latest therapeutic innovations, underscoring the shift from broad immunosuppression to personalized, mechanism-based interventions. Continued translational research and robust clinical trials are essential to realizing the full potential of these emerging therapies and to addressing unmet needs in autoimmune disease management.
