During the Pleistocene, the successive ice ages prompted the southward expansion of the “Mammoth Steppe” ecosystem, a prevalent habitat that supported species adapted to cold environments such as the mammoth, woolly rhinoceros, and reindeer. Previously, the earliest evidence for such cold-adapted species in the Iberian Peninsula dated back to Marine Isotope Stage 6 (MIS 6, ~191–123 ka). This paper reports the discovery of a reindeer (Rangifer) tooth from Unit GIII of the Galería site at the Atapuerca-Trinchera site complex, dated to MIS 8 (~300–243 ka). This find is significant as it represents not only the oldest evidence of glacial fauna in the Iberian Peninsula but also the southernmost occurrence of reindeer in Europe of this age. The presence of Rangifer at this latitude (42°21′ N) during MIS 8 suggests that the glacial conditions affected the Iberian fauna earlier and with greater intensity than previously understood. Over the subsequent climatic cycles, cold-adapted species spread further south, reaching Madrid (40°20′) during the penultimate glacial period and the province of Granada (37°01′) during the last glacial maximum. The coexistence of human fossils and lithic artefacts within Units GII and GIII at Galería indicates that early humans also inhabited these glacial environments at Atapuerca. This study elaborates on the morphological and archaeological significance of the reindeer fossil, emphasizing its role in understanding the biogeographical patterns of glacial fauna and the adaptability of Middle Pleistocene human populations. The Eastern Baltic area stands out as a unique location due to the finds of Europe’s youngest dated mammoth remains (12.6–11.2 ka cal BP). Our study explores the drastic climate and landcover changes during the extinction of these gigantic herbivores at the Pleistocene/Holocene boundary. We used macrofossil analysis to determine the major contemporary terrestrial plant genera present in the area and used corresponding pollen taxa for REVEALS model-based landcover reconstructions. Our results indicate that these last mammoths utilised the open landcover of the Eastern Baltic, which developed as the continental ice sheet retreated during the termination of the last glaciation. Due to climate warming during the initial stages of the Holocene interglacial, the Eastern Baltic became speedily populated by birch and pine forests. The abrupt disappearance of typical forb-dominated tundra indicators, such as Dryas octopetala, and the fast increase in tree birch marked a shift from an open, tundra-like landscape to a forested one, making the environment inhospitable for mammoths even in northernmost Estonia by the beginning of the Holocene. A comparison between the isotopic values of nitrogen (δ15N) and carbon (δ13C) obtained from mammoths’ molars from 14.3 and 11.3 to 43.5 and 39.1 ka cal BP showed that mammoths experienced a decline in the nutritional value of their diet, resulting in their demise in the Eastern Baltic. Paleoclimatic changes during the Pleistocene–Holocene transition is suggested as a main factor that led to species extinction, including the woolly mammoth (Mammuthus primigenius), Steller’s sea cow (Hydrodamalis gigas) and the Don-hare (Lepus tanaiticus). These species inhabited the territory of Eurasia during the Holocene, but eventually went extinct. The Don-hare is an extinct species of the genus Lepus (Leporidae, Lagomorpha), which lived in the Late Pleistocene–Early Holocene in Eastern Europe and Northern Asia. For a long time, the Don-hare was considered a separate species, but at the same time, its species status was disputed, taking into account both morphological data and mitochondrial DNA. In this study, mitochondrial genomes of five Don-hares, whose remains were found on the territory of Northeastern Eurasia were reconstructed. Firstly, we confirm the phylogenetic proximity of the “young” specimens of Don-hare and mountain or white hare, and secondly, that samples older than 39 Kya form a completely distinct mitochondrial clade. Rapid and cost-effective retrieval of endogenous DNA from ancient specimens remains a limiting factor in palaeogenomic research. Many methods have been developed to increase ancient DNA yield, but modifications to existing protocols are often based on personal experience rather than systematic testing. Here, we present a new silica column-based extraction protocol, where optimizations were tested in controlled experiments. Using relatively well-preserved permafrost samples, we tested the efficiency of pretreatment of bone and tooth powder with a bleach wash and a predigestion step. We also tested the recovery efficiency of MinElute and QIAquick columns, as well as Vivaspin columns with two molecular weight cut-off values. Finally, we tested the effect of uracil-treatment with two different USER enzyme concentrations. We find that neither bleach wash combined with a predigestion step, nor predigestion by itself, significantly increased sequencing efficiency. Initial results, however, suggest that MinElute columns are more efficient for ancient DNA extractions than QIAquick columns, whereas different molecular weight cut-off values in centrifugal concentrator columns did not have an effect. Uracil treatments are effective at removing DNA damage even at concentrations of 0.15 U/µL (as compared to 0.3 U/µL) of ancient DNA extracts. Full article

The remains of “early” mammoths from a number of localities of the late Middle—early Late Pleistocene on the territory of the South of European Russia (the basin of the Don River, Rostov Region) are described. The description of the teeth and bones of a postcranial skeleton is given. Teeth characteristics (number of plates, lamellar frequency and enamel thickness) allow determining the finds as Mammuthus intermedius, described from the territory of France but known from other regions of Western Europe and Western Siberia as well. In Eastern Europe, this form was a typical representative of the Khazarian theriocomplex and existed during the MIS 5–7 interval. This mammoth taxon differs noticeably from the typical woolly mammoth M. primigenius, which appeared in continental Europe during MIS 4. Full article

The paper presents the first results of Hg determination in the hair of prehistorical animals (woolly mammoth, steppe bison, and woolly rhino). Hair of prehistorical mammals can be used as an archive that preserves changes of environmental pollution at the paleoscale. The aim of our study was to assess the levels of Hg exposure of ancient animals and to understand whether Hg concentration in hair could be used as a proxy indicating changes of mercury levels in the environment following global climate changes. We assessed changes of Hg exposure recorded in hairs of seven specimens of mammoth fauna mammals that inhabited the Yakutia region in the period from 45 to 10 ka yr BP. Hg concentrations in hair varied from 0.017 to 0.177 µg/g; the lowest Hg concentration were determined in older specimens (45–33 kyr yr BP). The two highest concentrations belonged sample from the Last Glacial Maximum and the Karginian interstadial (57–24 kyr BP) periods. Our hypothesis is the increase of Hg concentrations in hair reflecting environmental Hg level might be forced by high dust load in cold periods and thawing permafrost in warm climatic periods. Long-term variations of Hg level recovered from Ice Age animals’ hair correlate with Hg profiles of concentration and deposition reconstructed from the Antarctica ice core. Full article

De-extinction projects for species such as the woolly mammoth and passenger pigeon have greatly stimulated public and scientific interest, producing a large body of literature and much debate. To date, there has been little consistency in descriptions of de-extinction technologies and purposes. In 2016, a special committee of the International Union for the Conservation of Nature (IUCN) published a set of guidelines for de-extinction practice, establishing the first detailed description of de-extinction; yet incoherencies in published literature persist. There are even several problems with the IUCN definition. Here I present a comprehensive definition of de-extinction practice and rationale that expounds and reconciles the biological and ecological inconsistencies in the IUCN definition. This new definition brings together the practices of reintroduction and ecological replacement with de-extinction efforts that employ breeding strategies to recover unique extinct phenotypes into a single “de-extinction” discipline. An accurate understanding of de-extinction and biotechnology segregates the restoration of certain species into a new classification of endangerment, removing them from the purview of de-extinction and into the arena of species’ recovery. I term these species as “evolutionarily torpid species”; a term to apply to species falsely considered extinct, which in fact persist in the form of cryopreserved tissues and cultured cells. For the first time in published literature, all currently active de-extinction breeding programs are reviewed and their progress presented. Lastly, I review and scrutinize various topics pertaining to de-extinction in light of the growing body of peer-reviewed literature published since de-extinction breeding programs gained public attention in 2013. The reconstruction of ancient metagenomes from archaeological material, and their implication in human health and evolution, is one of the most recent advances in paleomicrobiological studies. However, as for all ancient DNA (aDNA) studies, environmental and laboratory contamination need to be specifically addressed. Here we attempted to reconstruct the tissue-specific metagenomes of a 42,000-year-old, permafrost-preserved woolly mammoth calf through shotgun high-throughput sequencing. We analyzed the taxonomic composition of all tissue samples together with environmental and non-template experimental controls and compared them to metagenomes obtained from permafrost and elephant fecal samples. Preliminary results suggested the presence of tissue-specific metagenomic signals. We identified bacterial species that were present in only one experimental sample, absent from controls, and consistent with the nature of the samples. However, we failed to further authenticate any of these signals and conclude that, even when experimental samples are distinct from environmental and laboratory controls, this does not necessarily indicate endogenous presence of ancient host-associated microbiomic signals. Eight anatomically and taxonomically different finds are presented in this paper, and they belong to four taxa: woolly mammoth (Mammuthus primigenius), giant deer (Megaloceros giganteus), red deer (Cervus elaphus), and dog (Canis familiaris). All specimens represent allochthonous Late Pleistocene and Holocene animal remains, and all were dredged during the gravel exploitation at the Sekuline site near Molve (Podravina region, SW Pannonian basin, NE Croatia). Mammoth remains (bone and tusk fragments) were radiocarbon dated, and these are the first absolute dates on mammoths in Croatia. One upper last left deciduous premolar (dP4 sin.) also belongs to the same species. Ascribed to a dog is one well-preserved skull with a peculiar abscess scar on the maxillary bone as the result of an inflammatory process on the carnassial (P4) premolar. The Late Pleistocene cervid remains are giant deer, while the other cervid finds were determined to be red deer of the Holocene age. Morphometrical and taphonomical data are presented for each specimen. Such fossil and recent bone/tooth aggregates are characteristic of fluvial deposits and selective collecting. Although lacking stratigraphic provenance, these finds help to fulfil the gaps in palaeoenvironmental, palaeoecological, and palaeoclimate reconstructions of Podravina and its neighbouring areas. Tusks of very young mammoths and other proboscideans are extremely rare in the fossil record. This article presents all deciduous tusks and newly-developing permanent tusks of woolly mammoths which are known from Dutch localities and the North Sea. Four deciduous tusks are from The Netherlands and one is from Germany. Five specimens of newly-developing permanent tusks are also discussed. In this article, we present several characteristics and details of those tusks such as presence/absence of roots, and cementum and enamel layers that should facilitate an identification when new findings are made. We also describe a dental element, a digitelle, that is easily mistaken for a tusk of a juvenile woolly mammoth. Full article
