Microbiome analysis has revolutionized our understanding of various biological processes, spanning human health and epidemiology (including antimicrobial resistance and horizontal gene transfer), as well as environmental and agricultural studies. At the heart of microbiome analysis lies the characterization of microbial communities through the quantification of microbial taxa and their dynamics. In the study of bacterial abundances, it is becoming more relevant to consider their relationship, to embed these data in the framework of network theory, allowing characterization of features like node relevance, pathways, and community structure. In this study, we address the primary biases encountered in reconstructing networks through correlation measures, particularly in light of the compositional nature of the data, within-sample diversity, and the presence of a high number of unobserved species. These factors can lead to inaccurate correlation estimates. To tackle these challenges, we employ simulated data to demonstrate how many of these issues can be mitigated by applying typical transformations designed for compositional data. These transformations enable the use of straightforward measures like Pearson’s correlation to correctly identify positive and negative relationships among relative abundances, especially in high-dimensional data, without having any need for further corrections. However, some challenges persist, such as addressing data sparsity, as neglecting this aspect can result in an underestimation of negative correlations. Full article

Background and Clinical Significance: Neuroleptic malignant syndrome (NMS) is a life-threatening condition usually caused by the exposure to antipsychotics. This case report presents a catatonia syndrome that may have developed in the context of a moderate NMS. Case Presentation: An 18-year-old male patient presented with a treatment-resistant catatonia syndrome that debuted 2 weeks prior to the presentation (creatin kinase levels = 4908 U/lL maximum temperature = 38.9°C, white blood count = 13.20 × 109/L, Bush–Francis Catatonia Rating Scale = 30 points). Possible organic causes of catatonia were ruled out, according to the negative results obtained. The patient’s condition improved under benzodiazepine treatment and he was later discharged. After discharge, the catatonia was attributed to a possible NMS with moderate severity. The diagnosis was supported by NMS Diagnosis Criteria Score = 85 points and the presence of Levenson’s triad. Conclusions: This case highlights the concomitant manifestation of both catatonia and NMS in the same patient and the difficulty of establishing a correct diagnosis involving both entities. Head and neck squamous cell carcinomas (HNSCCs) represent a heterogeneous group of tumors with a complex molecular profile. Despite therapeutic advances, patient prognosis remains poor, emphasizing the need for more effective treatment strategies. Traditional chemotherapy, with cisplatin and 5-fluorouracil (5-FU), remains the gold standard but is limited by toxicity and tumor resistance. Immunotherapy, particularly immune checkpoint inhibitors targeting programmed cell death protein 1 (PD-1) and its ligand (PD-L1), has improved overall survival, especially in patients with high PD-L1 expression. In parallel, targeted therapies such as poly (ADP-ribose) polymerase 1 (PARP1) inhibitors—which impair DNA repair and increase replication stress—have shown promising activity in HNSCC. Cyclin-dependent kinase (CDK) inhibitors are also under investigation due to their potential to correct dysregulated cell cycle control, a hallmark of HNSCC. This review aims to summarize current and emerging pharmacotherapies for HNSCC, focusing on chemotherapy, immunotherapy, and PARP and CDK inhibitors. It also discusses the evolving role of targeted therapies in improving clinical outcomes. Future research directions include combination therapies, nanotechnology-based delivery systems to enhance treatment specificity, and the development of diagnostic tools such as PARP1-targeted imaging to better guide personalized treatment approaches. Objectives: This study aimed to evaluate the 6-month real-world outcomes of switching to faricimab in patients with aflibercept-resistant neovascular age-related macular degeneration (nAMD). Methods: A retrospective review was conducted on the eyes of 60 patients with aflibercept-resistant nAMD that were switched to faricimab. Best-corrected visual acuity (BCVA) and optical coherence tomography (OCT) parameters, including central subfield thickness (CST), subfoveal choroidal thickness (SFCT), and both the maximum height and width of pigment epithelial detachment (PED), at baseline and 1, 3, and 6 months after switching were evaluated. The type of PED and retinal fluid were also analyzed. Results: The results showed that BCVA remained stable at month 6 (p = 0.150), while CST significantly decreased (p = 0.020), and SFCT remained unchanged (p = 0.072). The maximum PED height significantly decreased (p = 0.030), while the maximum PED width did not change (p = 0.07). The mean injection interval significantly increased from 6.8 ± 2.4 weeks before switching to 11.2 ± 1.7 weeks after switching (p = 0.068). Furthermore, the dry macula rate was 43.3% at month 6. Conclusions: Switching to faricimab in aflibercept-resistant nAMD patients showed stable visual outcomes, significant anatomical improvements, and reduced treatment burden over 6 months in real-world clinical settings. Background/Objectives: Infectious endophthalmitis is a vision-threatening complication of intraocular surgery and intravitreal injections. Standard treatment involves intravitreal antibiotics; however, concerns regarding multidrug resistance and vancomycin-associated hemorrhagic occlusive retinal vasculitis (HORV) highlight the need for alternative antimicrobial strategies. This study aimed to evaluate the clinical efficacy and safety of a protocol combining intravitreal injection of 1.25% povidone-iodine (PI) with intraoperative irrigation using low concentrations of vancomycin and ceftazidime. Methods: We retrospectively analyzed 11 eyes from patients diagnosed with postoperative or injection-related endophthalmitis. Six of the eleven cases received an initial intravitreal injection of 1.25% PI, followed by pars plana vitrectomy with irrigation using balanced salt solution PLUS containing vancomycin (20 μg/mL) and ceftazidime (40 μg/mL). A second intravitreal PI injection was administered at the end of surgery in all cases. Additional PI injections were administered postoperatively based on clinical response. Clinical outcomes included best-corrected visual acuity (BCVA), microbial culture results, corneal endothelial cell density, and visual field testing. Results: All eyes achieved complete infection resolution without recurrence. The mean BCVA improved significantly from 2.18 logMAR at baseline to 0.296 logMAR at final follow-up (p < 0.001). No adverse events were observed on specular microscopy or visual field assessment. The protocol was well tolerated, and repeated PI injections showed no signs of ocular toxicity. Conclusions: This combination protocol provides a safe and effective treatment strategy for infectious endophthalmitis. It enables rapid and complete infection resolution while minimizing the risks associated with intravitreal antibiotics. These findings support further investigation of this protocol as a practical and globally accessible alternative to standard intravitreal antimicrobial therapy. Full article

The time-domain electromagnetic method has been widely applied in mineral exploration, oil, and gas fields in recent years. However, its response characteristics remain unclear, and there is an urgent need to study the response characteristics of the borehole-surface transient electromagnetic(BSTEM) field. This study starts from the time-domain electric field diffusion equation and discretizes the calculation area in space using tetrahedral meshes. The Galerkin method is used to derive the finite element equation of the electric field, and the vector interpolation basis function is used to approximate the electric field in any arbitrary tetrahedral mesh in the free space, thus achieving the three-dimensional forward simulation of the BSTEM field based on the finite element method. Following validation of the numerical simulation method, we further analyze the electromagnetic field response excited by vertical line sources.. Through comparison, it is concluded that measuring the radial electric field is the most intuitive and effective layout method for BSTEM, with a focus on the propagation characteristics of the electromagnetic field in both low-resistance and high-resistance anomalies at different positions. Numerical simulations reveal that BSTEM demonstrates superior resolution capability for low-resistivity anomalies, while showing limited detectability for high-resistivity anomalies Numerical simulation results of BSTEM with realistic orebody models, the correctness of this rule is further verified. This has important implications for our understanding of the propagation laws of BSTEM as well as for subsequent data processing and interpretation. Full article

Creating a comfortable microclimate in the premises of buildings is currently becoming one of the priorities in the field of architecture, construction and engineering systems. The increased attention from the scientific community to this topic is due not only to the desire to ensure healthy and favorable conditions for human life but also to the need for the rational use of energy resources. This area is becoming particularly relevant in the context of global challenges related to climate change, rising energy costs and increased environmental requirements. Practice shows that any technical solutions to ensure comfortable temperature, humidity and air exchange in rooms should be closely linked to the concept of energy efficiency. This allows one not only to reduce operating costs but also to significantly reduce greenhouse gas emissions, thereby contributing to sustainable development and environmental safety. In this connection, this study presents a parametric assessment of the influence of climatic and geometric factors on the aerodynamic characteristics of the air cavity, which affect the heat exchange process in the ventilated layer of curtain wall systems. The assessment was carried out using a combined analytical calculation method that provides averaged thermophysical parameters, such as mean air velocity (Vs), average internal surface temperature (tin.sav), and convective heat transfer coefficient (αs) within the air cavity. This study resulted in empirical average values, demonstrating that the air velocity within the cavity significantly depends on atmospheric pressure and façade height difference. For instance, a 10-fold increase in façade height leads to a 4.4-fold increase in air velocity. Furthermore, a three-fold variation in local resistance coefficients results in up to a two-fold change in airflow velocity. The cavity thickness, depending on atmospheric pressure, was also found to affect airflow velocity by up to 25%. Similar patterns were observed under ambient temperatures of +20 °C, +30 °C, and +40 °C. The analysis confirmed that airflow velocity is directly affected by cavity height, while the impact of solar radiation is negligible. However, based on the outcomes of the analytical model, it was concluded that the method does not adequately account for the effects of solar radiation and vertical temperature gradients on airflow within ventilated façades. This highlights the need for further full-scale experimental investigations under hot climate conditions in South Kazakhstan. The findings are expected to be applicable internationally to regions with comparable climatic characteristics. Ultimately, a correct understanding of thermophysical processes in such structures will support the advancement of trends such as Lightweight Design, Functionally Graded Design, and Value Engineering in the development of curtain wall systems, through the optimized selection of façade configurations, accounting for temperature loads under specific climatic and design conditions. Background/Objectives: The rapid emergence of antibiotic-resistant Escherichia coli in food and clinical environments necessitates new, clean-label antimicrobials. This study assessed eight Greek native essential oils—oregano, thyme, dittany, rosemary, peppermint, lavender, cistus and helichrysum—for activity against six genetically and phenotypically diverse E. coli strains (reference, pNorm, mecA, mcr-1, blaOXA and O157:H7). We aimed to identify oils with broad-spectrum efficacy and clarify the chemical constituents responsible. Methods: Disk-diffusion assays measured inhibition zones at dilutions from 50% to 1.56% (v/v). MIC and MBC values were determined by broth microdilution. GC–MS profiling identified dominant components, and Spearman rank-order correlations (ρ) linked composition to activity. Shapiro–Wilk tests (W = 0.706–0.913, p ≤ 0.002) indicated non-normal data, so strain comparisons used Kruskal–Wallis one-way ANOVA with Dunn’s post hoc and Bonferroni correction. Results: Oregano, thyme and dittany oils—rich in carvacrol and thymol—exhibited the strongest activity, with MIC/MBC ≤ 0.0625% (v/v) against all strains and inhibition zones > 25 mm at 50%. No strain-specific differences were detected (H = 0.30–3.85; p = 0.998–0.571; padj = 1.000). Spearman correlations confirmed that carvacrol and thymol content strongly predicted efficacy (ρ = 0.527–0.881, p < 0.001). Oils dominated by non-phenolic terpenes (rosemary, peppermint, lavender, cistus, helichrysum) showed minimal or no activity. Conclusions: Phenolic-rich EOs maintain potent, strain-independent antimicrobial effects—including against multidrug-resistant and O157:H7 strains—via a multi-target mode that overcomes classical resistance. Their low-dose efficacy and GRAS status support their use as clean-label food preservatives or adjuncts to antibiotics or bacteriophages to combat antimicrobial resistance. Full article

Background: The most common female endocrinopathy is polycystic ovary syndrome (PCOS), affecting 10–20% of women of reproductive age. It is associated with a wide range of hormonal and biochemical abnormalities and long-term metabolic and cardiovascular risks. It is characterized by infertility due to chronic anovulation, hyperandrogenism, polycystic ovarian morphology, and is often associated with insulin resistance (IR) and obesity. Hyperinsulinemia further increases androgen production and reduces sex hormone-binding globulin (SHBG) levels, thereby aggravating symptoms. In addition, vitamin D deficiency is often present in PCOS patients, and increasing evidence suggests that it may also be associated with insulin resistance and hyperandrogenism. Objective: This study aimed to evaluate the relationships between insulin resistance, vitamin D deficiency, body mass index (BMI), and androgen levels in women with PCOS. Method: A cross-sectional study was conducted in which data from 195 women diagnosed with PCOS and not yet receiving therapy at a gynecologic endocrinology unit of a university-based tertiary clinical center, between 2019 and 2024, were analyzed. The parameters recorded were age, body mass index (BMI), 25(OH) vitamin D levels, androgen hormone levels (testosterone, androstenedione), glucose-insulin responses during a 3-point oral glucose tolerance test (OGTT). Statistical analyses, including linear regression, Pearson, and Spearman correlation tests were used to assess associations between variables. Results: The mean age of the patients was 24.8 years (18–42), and the mean BMI was 30.6 kg/m2 (17–51). Vitamin D deficiency was observed in 84.1% of patients, hyperandrogenism in 45.8%, and insulin resistance in 44.5%. A significant inverse correlation was found between BMI and vitamin D levels (r = −0.31, p =< 0.01) indicating that higher BMI is associated with lower vitamin D status. Similarly, BMI also showed a significant negative correlation with SHBG levels (r = –0.45, p < 0.01), suggesting that increasing body weight is linked to reduced SHBG concentrations. In addition, BMI was significantly positively correlated with 2 h insulin levels (r = 0.43, p =< 0.01) and with testosterone levels (r = 0.21, p = 0.01). These findings suggest that increased adiposity intensifies insulin resistance and is linked to both vitamin D deficiency and elevated androgen levels. Moreover, the combination of hyperinsulinemia and low vitamin D further disrupts hormonal balance by promoting ovarian androgen production and decreasing SHBG levels, thereby increasing the bioavailability of testosterone. A significant inverse correlation was found between vitamin D levels and 2 h insulin levels (r = −0.28, p =< 0.01), indicating that lower vitamin D status is associated with increased insulin resistance. Furthermore, 2 h insulin levels showed a significant positive correlation with testosterone levels (r = 0.32, p =< 0.01), suggesting that greater insulin resistance is linked to higher androgen production. Additionally, vitamin D levels were inversely correlated with testosterone (r = −0.18, p = 0.02), demonstrating that a lower vitamin D status may further contribute to the hyperandrogenic environment. Vitamin D levels also showed a significant positive correlation with SHBG concentrations (r = 0.29, p < 0.01), indicating that a higher vitamin D status may be associated with increased SHBG levels. In contrast, 2 h insulin levels were inversely correlated with SHBG (r = −0.43, p < 0.01), reflecting the suppressive effect of hyperinsulinemia on SHBG production. Conclusions: Insulin resistance, BMI, and vitamin D deficiency are closely related to each other and to the severity of PCOS, which is confirmed by the correlations with androgen levels. The revealed relationships draw attention to the special importance of vitamin D supplementation and the correction of carbohydrate metabolism in alleviating the symptoms of the disease and reducing long-term health risks. Salinity, as one of the critical environmental factors in marine ecosystems, has complex and wide-ranging biological effects. However, the effects of salinity fluctuation on antibiotic resistance genes (ARGs) and virulence factor genes (VFGs) in the marine environment are not well understood. In this study, metagenomic sequencing analysis was used to reveal the response of ARGs and VFGs, hosted by low and high nucleic acid-content bacteria (HNA and LNA bacteria), to salinity, as it decreased from 26‰ to 16‰. The results showed that a total of 27 ARG types and 13 VFG types in HNA and LNA bacteria were found. Salinity changes had significant effects on the ARGs’ and VFGs’ composition and their hosts’ composition. In the network topology relationship, the complexity of the network between the ARGs and their host as well as the VFGs and their host differed with the decrease in salinity. The abundance of most genera of HNA and LNA bacteria was significantly corrected with the abundance of ARGs and VFGs, respectively. Overall, this study demonstrates the effects of salinity on ARGs and VFGs hosted by HNA and LNA bacteria in the marine environment and suggests the importance of salinity in regulating HNA and LNA bacterial communities and functions. Federated learning promotes the development of cross-domain intelligent applications under the premise of protecting data privacy, but there are still problems of sensitive parameter information leakage of multi-party data temporal alignment and resource scheduling process, and traditional symmetric encryption schemes suffer from low efficiency and poor security. To this end, in this paper, based on the modified NTRU-type multi-key fully homomorphic encryption scheme, an asymmetric algorithm, a secure computation scheme of multi-party least common multiple and greatest common divisor without full set under the semi-honest model is proposed. Participants strictly follow the established process. Nevertheless, considering that malicious participants may engage in poisoning attacks such as tampering with or uploading incorrect data to disrupt the protocol process and cause incorrect results, a scheme against malicious spoofing is further proposed, which resists malicious spoofing behaviors and not all malicious attacks, to verify the correctness of input parameters or data through hash functions and zero-knowledge proof, ensuring it can run safely and stably. Experimental results show that our semi-honest model scheme improves the efficiency by 39.5% and 45.6% compared to similar schemes under different parameter conditions, and it is able to efficiently process small and medium-sized data in real time under high bandwidth; although there is an average time increase of 1.39 s, the anti-malicious spoofing scheme takes into account both security and efficiency, achieving the design expectations. Quantum memory is essential for the prolonged storage and retrieval of quantum information. Nevertheless, no current studies have focused on the creation of effective quantum memory for continuous variables while accounting for the decoherence rate. This work presents an effective continuous-variable quantum key distribution method with parameter optimization utilizing the Elitist Elk Herd Random Immigrants Optimizer (2E-HRIO) technique. At the outset of transmission, the quantum device undergoes initialization and authentication via Compressed Hash-based Message Authentication Code with Encoded Post-Quantum Hash (CHMAC-EPQH). The settings are subsequently optimized from the authenticated device via 2E-HRIO, which mitigates the effects of decoherence by adaptively tuning system parameters. Subsequently, quantum bits are produced from the verified device, and pilot insertion is executed within the quantum bits. The pilot-inserted signal is thereafter subjected to pulse shaping using a Gaussian filter. The pulse-shaped signal undergoes modulation. Authenticated post-modulation, the prediction of link failure is conducted through an authenticated channel using Radial Density-Based Spatial Clustering of Applications with Noise. Subsequently, transmission occurs via a non-failure connection. The receiver performs channel equalization on the received signal with Recursive Regularized Least Mean Squares. Subsequently, a dataset for side-channel attack authentication is gathered and preprocessed, followed by feature extraction and classification using Adaptive Depthwise Separable Convolutional Neural Networks (ADS-CNNs), which enhances security against side-channel attacks. The quantum state is evaluated based on the signal received, and raw data are collected. Thereafter, a connection is established between the transmitter and receiver. Both the transmitter and receiver perform the scanning process. Thereafter, the calculation and correction of the error rate are performed based on the sifting results. Ultimately, privacy amplification and key authentication are performed using the repaired key via B-CHMAC-EPQH. The proposed system demonstrated improved resistance to decoherence and side-channel attacks, while achieving a reconciliation efficiency above 90% and increased key generation rate. Full article

Ferroptosis is a type of cell death executed by phospholipid peroxidation in an iron-dependent manner. Ferroptosis plays a central role in inhibiting tumor growth, enhancing the immune response, and is now considered a strategy to combat resistance to anticancer therapies. The oncosuppressor p53 is one of the major regulators of ferroptosis and can either promote or inhibit ferroptosis, depending on the context and/or extent of the damage. p53 governs the transcription of many genes that modulate cell susceptibility to ferroptosis, using this manner of death to fulfill its role as tumor suppressor. The diverse functions of p53 are related to non-coding RNAs (ncRNAs), especially microRNAs (miRNAs), and long non-coding RNAs (lncRNAs), since they can either regulate p53 or be regulated by p53. Therefore, an intricate metabolic network between ncRNAs and p53 ensures the correct response. In this review, we will discuss recent studies on the molecular interplay between p53-mediated ferroptosis and ncRNAs and how this contributes directly or indirectly to the outcome of ferroptosis. The Love That Kills: Phaedra’s Challenges to a Philosophy of Eros

Focusing on the legend of Phaedra and Hippolytus as developed in Euripides and Seneca and especially in Racine’s Phèdre and taking into account as well its further development in works by Camillo Boito, Luchino Visconti, and Yukio Mishima, I make the following arguments: (1) Contrary to many theologians and philosophers of love, a pathological form of love that issues in murder and suicide should not be regarded as unworthy of serious attention. Racine’s tragedy provides a catharsis for universal experiences of unrequited love and jealousy, a major human phenomenon. (2) Contrary to Paul Valéry, Phèdre’s love cannot be called merely animal, since the analytical insight she develops into her morbid passion carries tremendous moral force and lies at the origin of the European psychological novel, as launched by Madame de La Fayette a year later. (3) Contrary to François Mauriac, even if she is a heroine of desire or concupiscence rather than of “true love” (in contrast to the relatively innocent affections of Hippolyte and Aricie), the incredible beauty of her language resists such an easy categorization. (4) Study of concrete presentations of “love” in literature confirms that the meaning and use of this word is marked by an irreducible pluralism. Philosophical and theological analysis of love has to come to terms with this. (5) The role of a work of art, in crystallizing archetypical emotions and situations in a way that carries authority, is to provide the middle ground between the abstractions of philosophy on the one hand and the uncontrollable diversity of the empirical on the other. Even psychologies or sociologies of love, which claim to be close to the concrete data, need to be anchored in and corrected by the special concrete vision that only great literature can bring.
