Fluoride is a crucial inorganic anion found in drinking water, which may pose serious health hazards to human health if consumed in excess amounts. The quantification of fluoride in drinking water with high sensitivity, selectivity, and cross-sensitivity is critical. Given these factors, the present work proposes a spectroelectrochemical sensing platform for fluoride sensing using 5,10,15,20-tetraphenyl-21H,23H-porphine iron (III) chloride (FeTPP), and tetrabutylammonium perchlorate (TBAP) as the electrolyte. The proposed spectroelectrochemistry (SEC) is a hybrid platform that concurrently provides spectroscopic and electrochemical information about a system susceptible to oxidation and reduction. An ensemble–based multivariate prediction model was developed to simultaneously analyze electrochemical and spectroscopic data to predict fluoride concentration with enhanced reliability and precision. The prediction model provided promising results with a coefficient of determination of 0.9923 ± 0.0063 and a MSE of 0.369 ± 0.0596. These encouraging results demonstrate the promising performance of the proposed spectroelectrochemical platform in complex real-world applications. Full article

Obesity is a chronic epidemic caused by abnormal fat metabolism. As a key digestive enzyme, pancreatic lipase (PL) is an important target for regulating fat metabolism. The inhibitory potential of 5,10,15,20-Tetrakis (4-aminophenyl) porphyrin (TAPP), 5,10,15,20-Tetrakis (4-hydroxyphenyl) porphyrin (THPP), meso-Tetra (4-carboxyphenyl) porphine (TCPP), Cu (II) meso-Tetra (4-carboxyphenyl) porphine (Cu-TCPP) on PL was studied by enzymatic kinetics, multi-spectral, and molecular simulation technology. THPP, TCPP, TAPP, and Cu-TCPP all had good PL inhibitory activity (IC50 range: 97.49–248.70 μM) and were uncompetitive inhibitors. The order of inhibitory ability was: THPP > TCPP > TAPP > Cu-TCPP. The fluorescence quenching mechanism of THPP to PL was a mixed quenching dominated by static quenching, while TCPP, TAPP, and Cu-TCPP were static quenching. The binding of THPP, TCPP and TAPP to PL was mainly driven by hydrogen bonds and van der Waals forces, while Cu-TCPP was mainly driven by a hydrophobic interaction. Four porphyrin compounds changed the conformation of PL, affected the microenvironment of Tyr and Trp residues, and induced changes in the secondary structure of PL, thereby reducing the stability and catalytic activity of PL. Hydrogen bonds played an important role in the binding stability of THPP, TCPP, TAPP, and PL. Full article

The photodynamic inactivation (PDI) of bacteria is a promising alternative to antibiotics in boar semen extenders. It was recently established using the illumination of semen samples containing 2 µM of the photosensitizer 5,10,15,20-tetrakis(N-methyl-4-pyridyl)-21H,23H-porphine (TMPyP) with white LED light. High concentrations of TMPyP require strict sample handling in the dark to avoid uncontrolled photodynamic effects caused by ambient light. This study was designed to examine whether lower concentrations of PS could be utilized along with a narrow band blue LED light source, which aligns with TMPyP’s Soret band, thereby minimizing light-induced disruption. A dose-response study with blue LED light exposure of sperm revealed no light toxicity. Importantly, substituting the established white light PDI with blue light illumination and 0.5 µM TMPyP resulted in robust antimicrobial efficiency and sperm compatibility in long-term stored semen samples. This modification led to the confirmation of the hypothesis that a diminished TMPyP concentration in concert with blue LED light facilitates semen handling in normal laboratory light while avoiding unintended light effects. In conclusion, this study plays a pivotal role in augmenting the practicality of the innovative PDI technology by establishing a method that is less susceptible to unanticipated effects of ambient light during sample management. In this study, we present a novel approach using amperometric microsensors to detect quercetin in cosmetic formulations and track its metabolic behavior after topical application. This method offers a sensitive, real-time alternative to conventional techniques, enabling the detection of quercetin’s bioavailability, its transformation into active metabolites, and its potential therapeutic effects when applied to the skin. Quercetin (Q) is a bioactive flavonoid known for its potent antioxidant properties, naturally present in numerous plants, particularly those with applications in cosmetic formulations. In response to the growing interest in developing novel plant-based dermo-cosmetic solutions, this study investigates the electrochemical detection of quercetin, a ketone-type flavonoid, extracted from Gingko biloba essential oil. Three newly designed amperometric microsensors were developed to assess their efficacy in detecting quercetin in botanical samples. The sensor configurations utilized two forms of carbon material as a foundation: graphite (G) and carbon nanoparticles (CNs). These base materials were modified with paraffin oil, chitosan (CHIT), and cobalt(II) tetraphenylporphyrin (Co(II)TPP) to enhance sensitivity. Differential pulse voltammetry (DPV) served as the analytical method for this investigation. Among the sensors, the CHIT/G–CN microsensor exhibited the highest sensitivity, with a detection limit of 1.22 × 10−7 mol L−1, followed by the G–CN (5.64 × 10−8 mol L−1) and Co(II)TPP/G–CN (9.80 × 10−8 mol L−1) microsensors. The minimum detectable concentration was observed with the G–CN and CoP/G–CN microsensors, achieving a threshold as low as 0.0001 μmol L−1. Recovery rates and relative standard deviation (RSD) values averaged 97.4% ± 0.43, underscoring the sensors’ reliability for quercetin detection in botanical matrices. Full article

In this work, we present the green synthesis of complex AE derived from β-hidroxymethylidene indanones by ultrasound, which allowed for the obtaining of compounds in a shorter time and with good yields. These organotin complexes were then doped with cobalt porphine and incorporated into a poly(3,4-ethylenedioxythiophene):poly(4-styrenesulfonate) matrix to manufacture composite semiconductor films. The semiconductor films were characterized through atomic force microscopy, examining their topography, Knoop hardness (around 17 HK), and tensile strength, which varied from 5 × 10−4 to 7 × 10−2 Pa. The optical behavior was evaluated, revealing that the changes in these characteristics are related to the type of organotin complex present in the composite film: the transmittance ranged from 77% to 86%, while the reflectance varied from 13% to 17%. The band gap, calculated using the Kubelka–Munk function F(KM), was approximately 3.7 ± 0.19 eV for all the semiconductor films. Finally, we assessed the electrical behavior of the composite films through current–voltage (I–V) measurements under different lighting conditions. The I–V curves demonstrated that they share a saturation current density of 3.46 mA/mm2. However, they differ in their conduction rates within the ohmic regimen. These composite films’ optical and electrical properties suggest their potential use in developing electronic devices like organic diodes. Full article

This study presents the development of zirconium polycarboxylate gel systems as substrates for advanced fluorescence sensing devices. Zirconium-based metal–organic gels (MOGs) offer a promising alternative due to the robustness of the Zr–O bond, which provides enhanced chemical stability. In this work, zirconium polycarboxylate gels were synthesized using green solvents in a rapid room temperature method. Fluorescein, naphthalene-2,6-dicarboxylic acid, and 4,4′,4″,4‴-(porphine-5,10,15,20-tetrayl)tetrakisbenzoic acid were incorporated as fluorophores to give the gel luminescent properties, enabling it to be used as a sensor. These fluorophores produce specific changes in the perceived color and intensity of the fluorescence emission upon interaction with different analytes in a solution, allowing a qualitative identification of different solvents and compounds. However, the fragile structure of neat gels hinders reproducible quantitative analysis of fluorescence emission. Therefore, to increase their mechanical stability during manipulation, a composite material was developed by combining the MOGs with quartz microcrystals, which proved to be a more reliable fluorescent system. The results show that the material can identify univocally different solvents and analytes in aqueous solutions by the quantitative analysis of the emission intensities. This work presents an innovative approach to create advanced fluorescence sensors with improved mechanical properties and stability using zirconium polycarboxylate gels and multiple fluorophores. Background: Glioblastoma multiforme (GBM) is the most common highly aggressive, primary malignant brain tumor in adults. Current experimental strategies include photodynamic therapy (PDT) and new drug delivery technologies such as nanoparticles, which could play a key role in the treatment, diagnosis, and imaging of brain tumors. Objectives: The purpose of this study was to test the efficacy of PDT using AGuIX-TPP, a polysiloxane-based nanoparticle (AGuIX) that contains TPP (5,10,15,20-tetraphenyl-21H,23H-porphine), in biological models of glioblastoma multiforme and to investigate the vascular mechanisms of action at multiple complexity levels. Methods: PDT effects were studied in monolayer and spheroid cell culture, as well as tumors in chicken chorioallantoic membranes (CAMs) and in mice were studied. Results: Treatment was effective in both endothelial ECRF and glioma U87 cells, as well as in the inhibition of growth of the glioma spheroids. PDT using AGuIX-TPP inhibited U87 tumors growing in CAM and destroyed their vascularization. The U87 tumors were also grown in nude mice. Their vascular network, as well as oxygen partial pressure, were assessed using ultrasound and EPR oximetry. The treatment damaged tumor vessels and slightly decreased oxygen levels. Conclusions: PDT with AGuIX-TPP was effective against glioma cells, spheroids, and tumors; however, in mice, its efficacy appeared to be strongly related to the presence of blood vessels in the tumor before the treatment. In the past few years, there has been an increasing interest in mycosporines—UV-absorbing molecules—bringing important insights into their intrinsic properties as natural sunscreens. Herein, mycosporine-serinol and gadusol (enolate form)/gadusolate were exposed to UV radiation via a solar simulator and the photostability was assessed in pure water and different natural matrices like river, estuary and ocean water. In general, this study revealed that the photodegradation of gadusolate and mycosporine-serinol was higher in natural matrices than in pure water due to the generation of singlet oxygen on UV irradiation. In pure water, in terms of photostability, both gadusolate and mycosporine-serinol were found to offer good protection and high performance in terms of photodegradation quantum yield ((0.8 ± 0.2) × 10−4 and (1.1 ± 0.6) × 10−4, respectively). Nonetheless, the photostability of mycosporine-serinol was found to be superior to that of gadusolate in natural water, namely, ocean, estuary and river. The present work highlights how mycosporine-serinol and gadusolate resist photodegradation, and supports their role as effective and stable UV-B sunscreens. Full article

Colorectal cancer (CRC) is the third most common cancer in the world, with an ongoing rising incidence. Despite secure advancements in CRC treatments, challenges such as side effects and therapy resistance remain to be addressed. Photodynamic therapy (PDT) emerges as a promising modality, clinically used in treating different diseases, including cancer. Among the main challenges with current photosensitizers (PS), hydrophobicity and low selective uptake by the tumor remain prominent. Thus, developing an optimal design for PS to improve their solubility and enhance their selective accumulation in cancer cells is crucial for enhancing the efficacy of PDT. Targeted photoactivation triggers the production of reactive oxygen species (ROS), which promote oxidative stress within cancer cells and ultimately lead to their death. Ruthenium (Ru)-based compounds, known for their selective toxicity towards cancer cells, hold potential as anticancer agents. In this study, we investigated the effect of two distinct arene-Ru assemblies, which lodge porphin PS in their inner cavity, and tested them as PDT agents on the HCT116 and HT-29 human CRC cell lines. The cellular internalization of the porphin-loaded assemblies was confirmed by fluorescence microscopy. Additionally, significant photocytotoxicity was observed in both cell lines after photoactivation of the porphin in the cage systems, inducing apoptosis through caspase activation and cell cycle progression disruptions. These findings suggest that arene-Ru assemblies lodging porphin PS are potent candidates for PDT of CRC. Full article

Photoactive artificial nanocatalysts that mimic natural photoenergy systems can yield clean and renewable energy. However, their poor photoabsorption capability and disfavored photogenic electron–hole recombination hinder their production. Herein, we designed two nanocatalysts with various microstructures by combining the tailored self-assembly of the meso-tetra(p-hydroxyphenyl) porphine photosensitizer with the growth of titanium dioxide (TiO2). The porphyrin photoabsorption antenna efficiently extended the absorption range of TiO2 in the visible region, while anatase TiO2 promoted the efficient electron–hole separation of porphyrin. The photo-induced electrons were transferred to the surface of the Pt co-catalyst for the generation of hydrogen via water splitting, and the hole was utilized for the decomposition of methyl orange dye. The hybrid structure showed greatly increased photocatalytic performance compared to the core@shell structure due to massive active sites and increased photo-generated electron output. This controlled assembly regulation provides a new approach for the fabrication of advanced, structure-dependent photocatalysts. Full article

Porphyrins were identified some years ago as a promising, easily accessible, and tunable class of organic photoredox catalysts, but a systematic study on the effect of the electronic nature and of the position of the substituents on both the ground-state and the excited-state redox potentials of these compounds is still lacking. We prepared a set of known functionalized porphyrin derivatives containing different substituents either in one of the meso positions or at a β-pyrrole carbon, and we determined their ground- and (singlet) excited-state redox potentials. We found that while the estimated singlet excited-state energies are essentially unaffected by the introduction of substituents, the redox potentials (both in the ground- and in the singlet excited-state) depend on the electron-withdrawing or electron-donating nature of the substituents. Thus, the presence of groups with electron-withdrawing resonance effects results in an enhancement of the reduction facility of the photocatalyst, both in the ground and in the excited state. We next prepared a second set of four previously unknown meso-substituted porphyrins, having a benzoyl group at different positions. The reduction facility of the porphyrin increases with the proximity of the substituent to the porphine core, reaching a maximum when the benzoyl substituent is introduced at a meso position. Metal–organic frameworks (MOFs) are frequently utilized as sensing materials. Unfortunately, the low conductivity of MOFs hinder their further application in electrochemical determination. To overcome this limitation, a novel modification strategy for MOFs was proposed, establishing an electrochemical determination method for cyanides in Baijiu. Co and Ni were synergistically used as the metal active centers, with meso−Tetra(4−carboxyphenyl)porphine (TCPP) and Ferrocenecarboxylic acid (Fc−COOH) serving as the main ligands, synthesizing Ni/Co−MOF−TCPP−Fc through a hydrothermal method. The prepared MOF exhibited improved conductivity and stable ratio signals, enabling rapid and sensitive determination of cyanides. The screen−printed carbon electrodes (SPCE) were suitable for in situ and real−time determination of cyanide by electrochemical sensors due to their portability, low cost, and ease of mass production. A logarithmic linear response in the range of 0.196~44 ng/mL was demonstrated by this method, and the limit of detection (LOD) was 0.052 ng/mL. Compared with other methods, the sensor was constructed by a one−step synthesis method, which greatly simplifies the analysis process, and the determination time required was only 4 min. During natural cyanide determinations, recommended readouts match well with GC−MS with less than 5.9% relative error. Moreover, this electrochemical sensor presented a promising method for assessing the safety of cyanides in Baijiu. Full article

Monitoring antibiotic retention in human body fluids after treatment and controlling heavy metal content in water are important requirements for a healthy society. Therefore, the approach proposed in this study is based on developing new optical sensors using porphyrin or its bifunctional hybrid materials made with AuNPs to accomplish the accurate detection of chloramphenicol and cobalt. To produce the new optical chloramphenicol sensors, 2,7,12,17-tetra-tert-butyl-5,10,15,20-tetraaza-21H,23H-porphine (TBAP) was used, both alone in an acid medium and as a hybrid material with AuNPs in a water–DMSO acidified environment. The same hybrid material in the unchanged water–DMSO medium was the sensing material used for Co2+ monitoring. The best results of the hybrid materials were explained by the synergistic effects between the TBAP azaporphyrin and AuNPs. Chloramphenicol was accurately detected in the range of concentrations between 3.58 × 10−6 M and 3.37 × 10−5 M, and the same hybrid material quantified Co2+ in the concentration range of 8.92 × 10−5 M–1.77 × 10−4 M. In addition, we proved that AuNPs can be used for the detection of azaporphyrin (from 2.66 × 10−5 M to 3.29 × 10−4 M), making them a useful tool to monitor porphyrin retention after cancer imaging procedures or in porphyria disease. In conclusion, we harnessed the multifunctionality of this azaporphyrin and of its newly obtained AuNP plasmonic hybrids to detect chloramphenicol and Co2+ quickly, simply, and with high precision. In this study, we assessed the electrical and optical behavior of semiconductor hybrid films fabricated from octaethyl-21H,23H-porphine copper (CuP), embedded in polymethylmethacrylate (PMMA), and polystyrene (PS). The hybrid films were characterized structurally and morphologically using infrared spectroscopy (IR), atomic force microscopy (AFM), scanning electron microscopy (SEM), and X-ray diffraction (XRD). Subsequently, the PMMA:CuP and PS:CuP hybrid films were evaluated optically by UV–vis spectroscopy, as well as electrically, with the four-point collinear method. Hybrid films present a homogeneous and low roughness morphology. In addition, the PS matrix allows the crystallization of the porphin, while PMMA promotes the amorphous structure in CuP. The polymeric matrix also affects the optical behavior of the films, since the smallest optical gap (2.16 eV) and onset gap (1.89 eV), and the highest transparency are obtained in the film with a PMMA matrix. Finally, the electrical behavior in hybrid films is also affected by the matrix: the largest amount of current carried is approximately 0.01 A for the PS:CuP film, and 0.0015 A for the PMMA:CuP film. Thanks to the above properties, hybrid films are promising candidates for use in optoelectronic devices. Full article

The organic dyes used in printing and dyeing wastewater have complex components, diverse structures and strong chemical stability, which make them not suitable for treatment and difficult to degrade in the environment. Porphyrins are macromolecules with 18 π electrons formed by four pyrrole molecules connected with a methylene bridge that has a stable structure. Porphyrin combines with iron to form an active intermediate with a structure similar to the cytochrome P450 enzyme, so they are widely used in the biomimetic field. In the current study, 5,10,15,20-tetra (4-carboxyphenyl) porphine ferric chloride (III) (Fe(III)TCPP) was used as a catalyst and iodosobenzene was used as an oxidant to explore the catalytic degradation of triphenylmethane dyes, such as rhodamine B (RhB) and malachite green (MG). The results of UV-Vis spectral analysis have shown that the conversion rate of the rhodamine B was over 90% when the amount of Fe(III)TCPP was 0.027 mM and the amount of iodosobenzene was eight equivalents. When the catalyst was 0.00681 mM and the amount of the oxidant was five equivalents, the conversion rate of the malachite green reached over 95%. This work provides a feasible method for the degradation of triphenylmethane dyes. Full article
