Background/Objectives: Porcine circovirus type 2 (PCV2) infection causes porcine circovirus disease (PCVD), a global immunosuppressive disease in pigs. Its clinical manifestations include post-weaning multisystemic wasting syndrome (PMWS) and porcine dermatitis and nephropathy syndrome (PDNS), which cause significant economic losses to the swine industry. The Cap protein, which is the major protective antigen of PCV2, can self-assemble to form virus-like particles (VLPs) in the insect baculovirus expression system. Few studies have compared the expression of Cap proteins in different baculovirus expression systems. Methods: In this study, we compared two commonly commercialized baculovirus construction systems with the Cap protein expression in various insect cells. Results: The results demonstrate that the flashBAC system expressed the Cap protein at higher levels than the Bac-to-Bac system. Notably, when expressing four copies of the Cap protein, the flashBAC system achieved the highest protein yield in High Five cells, where it reached 432 μg/mL at 5 days post-infection (dpi) with 27 °C cultivation. Animal experiments confirmed that the purified Cap protein effectively induced specific antibody production in mice and swine. Conclusions: This study provides critical data for optimizing the production of the PCV2 Cap protein, which is of great significance for reducing the production cost of PCV2 vaccines and improving the industrial production efficiency. Weaning stress damages the intestines and disrupts the intestinal barrier in piglets, which significantly impacts the pig farming industry’s economy. We aimed to examine the effects of ShenQiGan extract (CAG) on intestinal barrier function and explore the underlying molecular mechanisms in stress-challenged weaned piglets. The experimental design involved 80 weaned piglets aged 28 days (with an average body weight of 7.78 ± 0.074 kg) that were randomly allocated into four groups: Control, LCAG (0.1% CAG), MCAG (0.5% CAG), and HCAG (1.0% CAG). After a 28-day trial period, the growth performance and incidence of diarrhea in piglets were evaluated. CAG increased the average daily gain of weaned piglets, reduced the feed-to-gain ratio, and decreased the incidence of diarrhea. It significantly lowered serum inflammatory cytokine levels while elevating immunoglobulin levels. The supplement notably enhanced concentrations of acetic acid, propionic acid, butyric acid, and isobutyric acid. Furthermore, CAG demonstrated intestinal morphology restoration and upregulation of tight junction proteins and MUC2 protein expression in jejunum. At the mRNA level, it significantly upregulated the expression of Occludin, Claudin1, and MUC2 genes. CAG improves growth performance and mitigates diarrhea in weaned piglets by enhancing intestinal barrier integrity, modulating systemic inflammatory responses, elevating immunoglobulin levels, and promoting short-chain fatty acids (SCFAs) production in the cecum. As a versatile microorganism, yeast generates a variety of bioactive compounds through fermentation, such as amino acids, vitamins, enzymes, and growth factors, which collectively contribute to improved growth and overall health in pigs. Iron-rich bauxite residue (red mud) is a hazardous alkaline solid waste produced during the production of alumina from high-iron bauxite, which poses severe environmental challenges due to its massive stockpiling and limited utilization. In this study, metallic iron was recovered from high-iron red mud using the smelting reduction process. Thermodynamic analysis results show that an increase in temperature and sodium oxide content, along with an appropriate mass ratio of Al2O3 to SiO2 (A/S) and mass ratio of CaO to SiO2 (C/S), contribute to the enhancement of the liquid phase mass fraction of the slag. During the smelting reduction process of high-iron red mud, iron recoveries for low-alkali high-iron red mud and high-alkali high-iron red mud under optimal conditions were 98.14% and 98.36%, respectively. The metal obtained through reduction meets the industrial standard for steel-making pig iron, which is also confirmed in the pilot-scale experiment. The smelting reduction process of high-iron red mud can be divided into two stages, where the reaction is predominantly governed by interfacial chemical reaction and diffusion control, respectively. The apparent activation energy of high-alkali high-iron red mud is lower than that observed for low-alkali high-iron red mud. The reduced slag can be used as a roadside stone material or cement clinker. This proposed method represents a sustainable process for the comprehensive utilization of high-iron red mud, which also promotes the minimization of red mud. Full article

Background/Objectives: The porcine respiratory disease complex (PRDC) is a multifaceted, polymicrobial syndrome resulting from a combination of environmental stressors, primary infections (e.g., PRRSV) and secondary infectious agents (viruses and bacteria). PRDC causes severe lung pathology, leading to reduced performance, increased mortality rates, and higher production costs in the global pig industry. Our goal was to conduct a comprehensive study correlating both the anti-PRRSV immune response and 21 secondary infectious agents with PRDC severity. Methods: To this end, PRRSV-negative weaners were vaccinated with a PRRSV-2 MLV and put into a farm with a history of PRDC. Subsequently, anti-PRRSV cellular and antibody responses were monitored pre-vaccination, at 28 days post vaccination (dpv) and during PRDC outbreak (49 dpv). NanoString was used to quantify 21 pathogens within the bronchoalveolar lavage (BAL) at the time of necropsy (51 dpv). PRRSV-2 was present in 53 out of 55 pigs, and the other five pathogens (PCMV, PPIV, B. bronchiseptica, G. parasuis, and M. hyorhinis) were detected in BAL samples. Results: Although the uncontrolled settings of field trials complicated data interpretation, multivariate correlation analyses highlighted valuable lessons: (i) high weaning weight predicted animal resilience to disease and high weight gains correlated with the control of the PRRSV-2 field strain; (ii) most pigs cleared MLV strain within 7 weeks, and the field PRRSV-2 strain was the most prevalent lung pathogen during PRDC; (iii) all pigs developed a systemic PRRSV IgG antibody response which correlated with IgG and IgA levels in BAL; (iv) the induction of anti-field strain-neutralizing antibodies by MLV PRRSV-2 vaccination was both late and limited; (v) cellular immune responses were variable but included strong systemic IFN-γ production against the PRRSV-2 field strain; (vi) the most detected lung pathogens correlated with PRRSV-2 viremia or lung loads; (vii) within the six detected pathogens, two viruses, PRRSV-2 and PCMV, significantly correlated with the severity of the clinical outcome. Conclusions: While a simple and conclusive answer to the multifaceted nature of PRDC remains elusive, the key lessons derived from this unique study provide a valuable framework for future research on porcine respiratory diseases. The article presents issues related to the melting of cast iron with a limited or zero share of pig iron in the charge. The results of melts conducted in electric induction furnaces are presented. The elimination of pig iron and its replacement with steel or return scrap is highly significant in the context of sustainable production and product life cycle assessment (LCA). The paper presents the results of research carried out during melts conducted under both laboratory and industrial conditions. The chemical composition of the cast iron, its physicochemical properties obtained from the analysis of the cooling curve and its derivative, as well as the structure, were analyzed. It was found that cast iron produced using high-quality steel scrap contains fewer sulfur and phosphorus impurities. However, it was also observed that such cast iron exhibits reduced nucleation ability, which can be improved by applying an inoculation process. Full article

Determining the technological quality of fresh meat pieces is essential in the meat industry to ensure the production of high-quality products. For this purpose, nuclear magnetic resonance (NMR) is a non-destructive and non-invasive technique that appears as an alternative to traditional methodologies. The objective of this work is to determine the potential of magnetic resonance imaging (MRI) and time-domain (TD-NMR) relaxometry for determining the physicochemical characterization of fresh hams with different industrial destinations (both fresh and cured products, such as dry-cured ham). For this study, the biceps femoris, semimembranosus, and semitendinosus muscles of 20 fresh hind legs from white pigs, classified into four categories according to their fat content, were analyzed. The semitendinosus muscle was selected as a model, and positive and negative correlations were obtained between different physicochemical parameters and the longitudinal (T1) and transverse (T2) relaxation times obtained by MRI and TD-NMR. Regression models using T1 and T2 were also developed to predict the muscle water-holding capacity (WHC) and drip loss, using high, medium, and low magnetic field NMR (R2 > 0.80). Therefore, MRI and TD-NMR could be considered as highly suitable and accurate non-destructive techniques for the WHC determination in the meat industry. Full article

The expansion of the livestock industry has led to an increase in biogas slurry discharge, which contains high levels of nitrogen (N) and phosphorous (P). Struvite precipitation is an effective method for the recovery of N and P from biogas slurry, and the recovered N and P can be applied as a slow-release fertilizer in agricultural production. To form an industrial chain for struvite recovery and application in agriculture, we investigated the factors affecting struvite recovery from biogas slurry generated on a pig farm and evaluated its efficacy as a fertilizer. The N and P recovery efficiency was higher when magnesium chloride (MgCl2) was used as a magnesium (Mg) source compared with magnesium oxide (MgO), and the optimal reaction conditions were pH 10, a reaction time of 20 min, a stirring rate of 200 rpm, and a Mg/P/N ratio of 1.2:1.0:1.0, which achieved N and P recovery rates of 81.83% and 99.67%, respectively. To further investigate the commercial utility of using struvite recovered from biogas slurry as a fertilizer, the growth and content of nutrients in two common vegetables in China were measured. The vegetable quality-related parameters of bock choy (Brassica chinensis) improved as the proportion of struvite in the fertilizer increased. Fresh weight, soluble sugar, and soluble protein increased by 194.47%, 46.13%, and 82.42%, respectively. The quality-related parameters of water celery (Oenanthe javanica (Blume) DC.) increased with an increasing proportion of struvite (27.90 mg·g−1 soluble sugar and 42.20 mg·g−1 soluble protein). The application of struvite precipitated from biogas slurry in plant cultivation shows great potential and lays a solid foundation for the resourceful recovery and utilization of biogas slurry. Background: African swine fever (ASF) is a crucial socioeconomic setback to South Korea’s swine industry. This study aimed to determine seropositivity for ASF virus (ASFV) in pigs that appeared to be infected on farms with reported ASF outbreaks. Methods: A total of 2232 sera from ASF outbreaks (2019–2024) in South Korea were collected. Two enzyme-linked immunosorbent assay (ELISA) kits were used to detect ASFV antibodies, and an immunoperoxidase test (IPT) was used as a confirmatory test following the method recommended by the World Organisation for Animal Health in the Manual of Diagnostic Tests and Vaccines for Terrestrial Animals. Also, spatial clustering was identified using the Density-Based Spatial Clustering of Applications with Noise (DBSCAN) model to understand ASF hotspots in the wild boar population and assess the spatial relationship between the hotspots and ASF antibody-positive domestic pig farms. Results: Antibodies were first detected in Hwacheon in 2020, but by 2024, only 1.43% of pigs had detectable antibodies against ASFV. Although this percentage is still low, the number of antibody-positive pigs is gradually increasing. Additionally, 32 positive samples were found from nine pig farms with outbreaks, and these samples were confirmed positive in both the two ELISA tests and the IPT. The highest seropositivity was recorded at the finishing stage of pig production. When compared to the confirmatory IPT, both blocking and competition ELISA demonstrated high diagnostic sensitivities. The statistical association between ASF antibody-positive farms and wild boars were analyzed using Fisher’s exact test, yielding a significant p-value of 0.007. This indicates a strong correlation, as eight out of nine ASF-seropositive farms were located within hotspots that were significantly associated. Conclusions: Our findings provide valuable insights into ASFV antibody detection in South Korea and demonstrate a statistical association between farms housing pigs with ASFV antibodies and hotspots of ASFV-infected wild boars. Confirmatory tests, such as the IPT, are needed. These insights will contribute to the improvement of surveillance and biosecurity measures for swine farms. Full article

Pig slurry (PS) and wine vinasse (WV) pose environmental risks if not properly managed. Their composition makes them suitable for anaerobic co-digestion (AcoD), enhancing biomethane production and improving organic matter degradation efficiency. This research applies an innovative Design of Experiments (DoE) approach—specifically the Box–Behnken design (BBD)—to systematically optimize the AcoD process, surpassing traditional single-factor methods by efficiently evaluating the interactions. Variables such as temperature (35 °C, 52.5 °C, 70 °C), substrate ratio (25PS:75WV, 50PS:50WV, 75PS:25WV) and pH (7, 7.5, 8) were tested using a Box–Behnken design which studied the correlations between the experimental data and the model. In fact, the results showed that temperature, ratio, and their interaction significantly influenced biomethane production, being the pH the factor with the least influence on the response. Optimal conditions—pH of 8, temperature of 35 °C and a 50:50 substrate ratio—achieved a biomethane yield of 487.94 CH4/gVS (Volatile Solids). These results demonstrate the effectiveness of the DoE methodology in maximizing biomethane production and represent a significant advancement in valorizing wastes from pig farms and wineries, promoting a circular and sustainable economy. Background/Objectives: Foot-and-mouth disease virus (FMDV) poses a continuous threat to livestock health and agricultural economies. Current vaccines require high biosafety standards and are costly to produce. While novel vaccine technologies have been explored, most fail to meet industrial scalability, cost-efficiency, or multiserotype flexibility required for effective FMD control. This study aimed to evaluate the feasibility of using a high-cell density transient gene expression (TGE) system in CHO cells for the production of FMDV virus-like particles (VLPs) as a recombinant vaccine platform. Methods: VLP expression was optimized by adjusting cDNA and polyethyleneimine (PEI) concentrations. Expression yields were compared at 24 and 48 h post-transfection to determine optimal harvest timing. We further tested the system’s capacity to express different serotypes and chimeric constructs, incorporating VP1 sequences from various FMDV strains. Immunogenicity was evaluated in swine using VLPs from the A2001 Argentina strain as a model. Results: Optimal VLP expression was achieved at 24 h post-transfection. Chimeric constructs incorporating heterologous VP1 regions were successfully expressed. Immunized pigs developed protective antibody titers as measured by a virus neutralization test (VNT, log10 titer 1.43) and liquid-phase blocking ELISA (LPBE, titer 2.20) at 28 days post-vaccination (dpv). Titers remained above protective thresholds up to 60 dpv with a single dose. A booster at 28 dpv further elevated titers to levels comparable to those induced by the inactivated vaccine. Conclusions: Our results demonstrate the feasibility of using CHO cell-based TGE for producing immunogenic FMDV VLPs. This platform shows promise for scalable, cost-effective, and biosafe development of recombinant FMD vaccines. Full article

In recent years, much attention has been paid to environmental protection, not only by reducing emissions of harmful gases from industry, but also by reducing the excretion of biogenic compounds or ammonia emissions from agriculture, including animal production. The aim of this study was to determine the effects of complete diets with reduced inclusion levels of crude protein and limiting essential amino acids, fed to pigs in two- and three-phase feeding systems, and the feeding system on crude protein digestibility, nitrogen retention and utilization, fecal and urinary pH, fecal and urinary nitrogen and ammonia levels, and nitrogen excretion. Digestibility-balance trials were performed on 24 growing–finishing pigs housed in individual metabolism crates, in three groups, in two- and three-phase feeding systems. The pigs were fed the following diets: C-control diet; L-low-protein diet where the levels of crude protein and essential amino acids (lysine, methionine + cystine, threonine, and tryptophan) were reduced by 15% relative to diet C; L+AA-low-protein diet supplemented with crystalline lysine, methionine, threonine, and tryptophan to the standard levels (as in diet C). Diets L fed to pigs in two- and three-phase feeding systems significantly decreased crude protein digestibility and nitrogen retention, particularly in the three-phase system. The supplementation of diets L+AA with crystalline essential amino acids improved crude protein digestibility and nitrogen retention and utilization, especially in the two-phase system. Reduced concentrations of crude protein and essential amino acids in diets L contributed to a significant increase in feces and urine acidity in both two- and three-phase feeding systems. The supplementation of diets L+AA with essential amino acids resulted in a significant increase in urinary pH and a non-significant increase in fecal pH. This experimental factor had no effect on fecal ammonia concentration in group L+AA. The values of pH and total fecal nitrogen were somewhat higher in the two-phase system than in the three-phase system. Fecal ammonia concentration was similar in both systems. The three-phase feeding system contributed to a decrease in urinary pH and total urinary nitrogen. The analyzed feeding systems had no significant effect on urinary ammonia concentration. It was estimated that a reduction in crude protein (by 20–25 g/kg) and essential amino acid levels in pig diets, relative to the standard levels, reduced nitrogen excretion by 18.7% and 15.6% in two- and three-phase feeding systems, respectively. The supplementation of low-protein diets (L) with lysine, methionine, threonine, and tryptophan induced a further reduction in nitrogen excretion. A comparison of the effects of feeding systems (two-phase system vs. three-phase system) on crude protein digestibility and nitrogen retention and utilization revealed that better results were obtained in the two-phase feeding system. The intensifying global demand for food presents significant challenges for sustainable pig production, particularly in the context of escalating input costs, environmental degradation, and resource scarcity. Life cycle assessment provides a comprehensive framework for quantifying environmental impacts and identifying production hotspots within pig production systems. Feed production and manure management are consistently identified as major contributors, emphasising the need for targeted interventions. Although soybean meal remains a key protein source, its association with deforestation and biodiversity loss is driving an interest in more sustainable alternatives. In temperate climates, faba beans offer a promising, locally sourced option, though their wider adoption is limited by amino acid imbalances and anti-nutritional factors. Grain preservation is another critical consideration, as post-harvest losses and fungal contamination compromise feed quality and animal health. Organic acid preservation has emerged as an energy-efficient, cost-effective alternative to industrial drying, improving storage stability and reducing fossil fuel dependence. Additional nutritional strategies, including dietary crude protein reduction, carbohydrate source modification, feed additive inclusion, and maternal nutritional interventions, can enhance nutrient utilisation, intestinal health, and herd resilience while mitigating environmental impact. This review explores practical feed-based strategies to support sustainable, resilient, and resource-efficient pig production and contribute to global food security. Full article

Aggressive behaviour in pigs poses significant challenges to animal welfare, production efficiency, and economic performance in the pork industry. This review explores the multifaceted causes of pig aggression, focusing on genetic, environmental, and physiological factors. Aggression in pigs is categorized into social, maternal, fear-induced, play, and redirected aggression, with early-life hierarchies and environmental stressors playing critical roles. Physiological markers, such as elevated cortisol and reduced serotonin levels, are closely linked to aggressive behaviour, while dietary interventions, including tryptophan supplementation, have shown promise in mitigating aggression. Environmental factors, such as overcrowding, noise, and heat stress, exacerbate aggressive tendencies, whereas enrichment strategies, like music and improved housing conditions, can reduce stress and aggression. Genome-wide analyses have pinpointed specific polymorphisms in neurotransmitter genes (DRD2, SLC6A4, MAOA) and stress-response loci (RYR1) as significant predictors of porcine aggression. Advances in genomic technologies, including genome-wide association studies (GWASs) and transcriptomic analyses, have further elucidated the genetic and epigenetic underpinnings of aggressive behaviour. Practical application in breeding programmes remains challenging due to aggression polygenic nature and industry hesitancy toward genomic approaches. Future research should focus on integrating genetic markers into breeding programmes, developing multitrait selection indices, and exploring epigenetic modifications to improve animal welfare and production efficiency. By addressing these challenges, the pork industry can enhance both the well-being of pigs and the sustainability of production systems. Full article

The improvement of carcass traits is a key focus in pig genetic breeding programs. To identify quantitative trait loci (QTLs) and genes linked to key carcass traits, we conducted a genome-wide association study (GWAS) using whole-genome sequencing data from 1118 commercial pigs (Duroc sires and Yorkshire/Landrace F1 dams). This study focused on six phenotypes: iodine value, belly firmness, belly side fat, total side thickness (belly SThK), belly subcutaneous fat (Subq), and belly seam. Phenotypes were measured using image analysis, DEXA, and fatty acid profiling, and genotyping was performed using low-pass sequencing (SkimSeq). After quality control, 18,911,793 single nucleotide polymorphisms (SNPs) were retained for further analysis. A GWAS was conducted using a linear mixed model implemented in GCTA. Key findings include a significant QTL on SSC15 (110.83–112.23 Mb), which is associated with the iodine value, containing genes such as COX15, CHUK, SCD, and HIF1AN, which have known roles in fatty acid metabolism. Additionally, PNKD, VIL1, and PRKAG3 (120.74–121.88 Mb on SSC15) were linked to belly firmness, influencing muscle structure and fat composition. Three QTLs for belly side fat were identified on SSC1, SSC2, and SSC3, highlighting genes like SLC22A18, PHLDA2, and OSBPL5, which regulate fat deposition and lipid metabolism. The results provide novel molecular markers that can be incorporated into selective breeding programs to improve pork quality, fat distribution, and meat composition. These findings enhance our understanding of the genetic mechanisms underlying carcass belly traits while offering tools to improve pork quality, optimize fat composition, and align with consumer preferences in the meat production industry.
