Lupin (Lupinus spp.) is increasingly incorporated into processed foods as a gluten-free ingredient and alternative protein source, but it is also a regulated allergen in the European Union due to cross-reactivity with other legumes, especially peanut. Reliable methods for detecting undeclared lupin traces in complex food matrices are therefore essential for consumer protection. In this study, a loop-mediated isothermal amplification (LAMP) assay was developed for rapid and sensitive detection of lupin DNA. Several nuclear and chloroplast regions were evaluated for primer design, and gene encoding the Lup a 1 allergen was selected as the optimal target. Amplification was monitored by real-time fluorescence, agarose gel electrophoresis, and visual colorimetry. The selected primer set achieved a detection limit of 25 pg of lupin DNA and consistently detected lupin in binary mixtures down to 10 mg/kg, with no cross-reactivity against closely related legumes or tree nuts. Application to processed foods confirmed detection in products declaring lupin and revealed potential undeclared presence in some commercial samples. Colorimetric detection provided reliable results comparable to real-time monitoring, enabling simple readouts without specialized equipment. Overall, the developed LAMP assay represents a rapid, specific, and sensitive alternative to PCR-based methods for allergen monitoring and food safety management. Full article

Mountainous regions with complex terrain often generate mixed pixels, reducing the accuracy of land use classification. This study evaluates the applicability and performance of eight commonly used Land Use Products (LUPs) with resolutions from 10 m to 1000 m in a typical mountainous area of the Wumeng Mountains, using manually vectorized high-resolution imagery as reference data. The spatial patterns of classification inconsistency and topographic influences were systematically analyzed. Results show the following: (1) All LUPs underestimate Cropland, Forest, Built-up areas, and Waters, while overestimating Grassland. Most products perform poorly, with an Overall Accuracy (OA) below 0.7. (2) Classification performance varies by land cover type and location. In complex terrains, high fragmentation causes mixed pixels, leading to inconsistency. (3) XGBoost and SHAP analyses indicate elevation and Terrain Surface Complexity as dominant factors. Accuracy increases with elevation and is higher on shady than on sunny slopes. Greater surface complexity reduces performance. (4) Spatial resolution alone does not determine performance—algorithm design, sample distribution, and data fusion are critical. All eight LUPs show poor applicability in mountainous regions. Future research should emphasize terrain-stratified sample balancing, terrain-aware algorithms, and strategies to mitigate mixed pixel effects. Full article

Guanling-Zhenfeng County, a microcosm of the ecologically fragile karst area in southwest China, experiences rapid population growth and urban expansion which intensifies land use transformation and ecological landscape fragmentation. Exploring the spatiotemporal characteristics of landscape fragmentation and its causes in Guanling-Zhenfeng County is of great significance in maintaining the stability of the ecosystem and ecological protection in karst areas. In this study, a comprehensive landscape fragmentation index (FI), geographic probe, multi-scale geographically weighted regression (MGWR), and PLUS model were used to quantitatively explore the spatiotemporal characteristic heterogeneity, causes, and future scenario projections of landscape fragmentation in Guanling-Zhenfeng County from 2000 to 2020. The results showed that: (1) the distribution of each landscape index was characterized by obvious spatial differentiation. Among them, the spatial distribution trends of patch density (PD) and largest patch index (LPI) were opposite and the distribution trends of Shannon diversity index (SHDI) and Shannon evenness index (SHEI) were similar. There were fewer heterogeneous patches in the study area from 2000 to 2020, and the landscape shape was more regular and less fragmented. (2) The overall landscape fragmentation in Guanling-Zhenfeng County from 2000 to 2020 was dominated by moderate fragmentation, with the smallest percentage of extreme fragmentation, and heavy fragmentation was mainly distributed in the north-central part of the study area. (3) Natural and social factors jointly affect the landscape fragmentation in Guanling-Zhenfeng County, and there is a significant interactive enhancement effect among the factors, with population density being the most important influence factor. In addition, the effects of the factors on landscape fragmentation showed significant spatial non-stationarity. (4) The characteristics of landscape fragmentation changes in Guanling-Zhenfeng County under different scenarios varied significantly, with the largest percentage of increase in heavy landscape fragmentation under the business-as-usual scenario (BAU), the next under the land use planning scenario (LUP), and the smallest under the ecological protection scenario (ESP). Pterocarpus anglonesis DC is an indigenous medicinal plant belonging to the Pterocarpus genus of the Fabaceae family. It is used to treat stomach problems, headaches, mouth ulcers, malaria, blackwater fever, gonorrhea, ringworm, diarrhea, heavy menstruation, and breast milk stimulation. Column chromatography of the stem bark extracts resulted in the isolation of eight compounds, which included friedelan-3-one (1), 3α-hydroxyfriedel-2-one (2), 3-hydroxyfriedel-3-en-2-one (3), lup-20(29)-en-3-ol (4), Stigmasta-5-22-dien-3-ol (5), 4-O-methylangolensis (6), (3β)-3-acetoxyolean-12-en-28-oic acid (7), and tetradecyl (E)-ferulate (8). The structures were established based on NMR, IR, and MS spectroscopic analyses. Triple-negative breast cancer (HCC70), hormone receptor-positive breast cancer (MCF-7), and non-cancerous mammary epithelial cell lines (MCF-12A) were used to test the compounds’ cytotoxicity. Overall, the compounds showed either no toxicity or very low toxicity to all three cell lines tested, except for the moderate toxicity displayed by lupeol (4) towards the non-cancerous MCF-12A cells, with an IC50 value of 36.60 μM. Compound (3β)-3-acetoxyolean-12-en-28-oic acid (7) was more toxic towards hormone-responsive (MCF-7) breast cancer cells than either triple-negative breast cancer (HCC70) or non-cancerous breast epithelial (MCF-12A) cells (IC50 values of 83.06 vs. 146.80 and 143.00 μM, respectively). Current antineoplastic agents present multiple disadvantages, driving an ongoing search for new and better compounds. Four lupane-type triterpenes, 3α,24-dihydroxylup-20(29)-en-28-oic acid (1), 3α,23-dihydroxy-30-oxo-lup-20(29)-en-28-oic acid (2), 3α,23-O-isopropylidenyl-3α,23-dihydroxylup-20(29)-en-28-oic acid (3), and 3α,23-dihydroxylup-20(29)-en-28-oic acid (4), previously isolated from Phoradendron wattii, were evaluated on two cell lines of chronic (K562) and acute (HL60) myeloid leukemia. Compounds 1, 2, and 4 decreased cell viability and inhibit proliferation, mainly in K562, and exhibited an apoptotic effect from 24 h of treatment. Of particular interest is compound 2, which caused arrest in active phases (G2/M) of the cell cycle, as shown by in silico study of the CDK1/Cyclin B/Csk2 complex by molecular docking. This compound [3α,23-dihydroxy-30-oxo-lup-20(29)-en-28-oic acid] s a promising candidate for incorporation into cancer treatments and deserves further study. Full article

As the key tool for land use management in China, land use planning (LUP) is characterized by the advantage of being state-led, quota-oriented, and easily monitored. Nevertheless, increasing research has doubted its effectiveness, particularly in controlling urban expansion, while few studies have empirically confirmed it. To this end, we construct a planning constraint index (PCI), supported by land use quota, for evaluating the pressure from LUP on urban expansion, and then analyze the spatial equilibrium of PCI. We also explore the spatial effects of PCI on urban expansion via the geographically weighted regression model (GWR) by collecting the spatial data of 280 prefecture-level cities in China from 2009 to 2016. The results show that: (1) most cities present a shortage of urban land quotas, and spatial disequilibrium exist among regions; (2) PCI has significant negative effects on urban expansion for all of the 280 cities, and spatial disparity of these effects occurs as well; (3) PCI has a stronger influence in southwest China where the economy is relatively backward. This study aims to provide a reference for land use planners and policymakers to help improve the effectiveness of LUP. Full article

Two prostanglandins (luprostiol, LUP, and dinoprost, DIN) and two ovulation-inducing agents (human Chorionic Gonadotropin, hCG, and deslorelin, DES) were evaluated for luteolysis and estrus induction, and for ovulation induction, respectively, in embryo donor jennies. Twenty-six fertile Andalusian jennies were used. In Experiment 1, jennies (n = 112 cycles) were randomly treated with either LUP or DIN after embryo flushing. In Experiment 2, donors (n = 84 cycles) were randomly treated with either hCG or DES to induce ovulation. No differences were found between prostaglandins for all variables studied (prostaglandin–ovulation interval (POI), interovulatory interval (IOI), embryo recovery rate (ERR), positive flushing rate (PFR) and embryo grade (EG)). The ovulation rate was similar for hCG and DES (60.9% vs. 78.7%). However, the interval to ovulation (ITO) was affected (62.61 ± 7.20 vs. 48.79 ± 2.69 h). None of the other variables studied (ERR, PFR and EG) were affected (p > 0.05), except for embryo quality (p = 0.009). In short, both prostaglandins evaluated are adequate to induce luteolysis and estrus. Both ovulation-inducing agents hastened ovulation, but DES seems to be more effective than hCG. Follicular diameter affected the interval from treatment to ovulation, and high uterine edema was related to low embryo quality. Full article

Analyzing the interaction between land use patterns (LUPs) and socioeconomic factors (SEFs) could provide a basis for regional land spatial planning and management decisions in the future. In this study, population, gross domestic product (GDP) and land use intensity were selected to explain the relationship between SEFs and LUPs. The study designed a new method of sample line acquisition for wavelet analysis, and identified the interaction grid scales of LUP changes with SEFs in 1991, 2005 and 2019 by using cross wavelet transform analysis (XWT). Wavelet transform coherent analysis (WTC) was used to reveal the interaction direction and impact strength between LUPs and SEFs. The results showed that: (1) There were two ranges of 2978–5008 m and 24,400–29,738 m in which the grid scales showing interaction between LUPs and SEFs (population, GDP and land use intensity) from 1991 to 2019 were overlapping. (2) The interaction direction between LUPs and SEFs from 1991 to 2019 was almost negative on all sample lines, while the interaction directions of the middle sample line of population and GDP from 1991 to 2019, the end sample line of GDP in 2019, and the start sample line of land use intensity in 1991 were positive. (3) Dry land, grassland and construction land were most affected by SEFs, followed by paddy fields, forest land and other land, and the least affected were water areas during 1991 to 2019. The impact of population and GDP on LUPs was reduced, while the impact of land use intensity on LUPs was increased from 1991 to 2019. Overall, population, GDP and land use intensity were the important SEFs in the changes of LUPs, and were important factors for social progress and economic development. Full article

The declaration of lupine supplements is mandatory to avoid lupine allergy for sensitive individuals. However, reliable detection methods against lupine allergen remain critical to prevent the unintended consumption of allergen contaminated food. In this study, we have immunized an alpaca with lupine protein extracts and retrieved nanobodies (Nbs). Nevertheless, the target antigen has been recognized as Lup an 1, which has been classified as β-conglutin, and confirmed to connect with lupine allergy. After selection of the best Nb-pair, a sandwich enzyme-linked immunosorbent assay (ELISA) has been developed providing a linear range of 0.036–4.4 μg/mL with detection limit of 1.15 ng/mL. This immunoassay was confirmed by detecting the samples with spiked allergen, and a recovery from 86.25% to 108.45% with coefficient of variation (CV) less than 4.0% has been determined. Generally, this study demonstrated the selection of Nbs against allergen with crude protein content to develop the immunoassay for lupine surveillance in foods. Full article

Lupin protein isolate was treated using the combination of enzymatic hydrolysis (Papain, Alcalase 2.4 L and Pepsin) and lactic acid fermentation (Lactobacillus sakei ssp. carnosus, Lactobacillus amylolyticus and Lactobacillus helveticus) to investigate the effect on functional properties, sensory profile and protein integrity. The results showed increased foaming activity (2466–3481%) and solubility at pH 4.0 (19.7–36.7%) of all fermented hydrolysates compared to the untreated lupin protein isolate with 1613% of foaming activity and a solubility of 7.3 (pH 4.0). Results of the SDS-PAGE and Bead-Assay showed that the combination of enzymatic hydrolysis and fermentation of LPI was effective in reducing L. angustifolius major allergen Lup an 1 to a residual level of <0.5%. The combination of enzymatic hydrolysis and fermentation enables the production of food ingredients with good functional properties in terms of protein solubility and foam formation, with a balanced aroma and taste profile. A new lupane caffeoyl ester, lup-20(29)-ene 3β-caffeate-30-al (7), and a new oleanane-type triterpene, 3β-hydroxyolean-13(18)-en-12-one (17), were isolated from the aerial parts of Dobera glabra (Forssk), along with ten known triterpenes, including seven lupane-type lupeol (1), 30-nor-lup-3β-ol-20-one (2), ∆1-lupenone (3), lup-20(29)-en-3β,30-diol (4), lupeol caffeate (5), 30-hydroxy lup-20(29)-ene 3β-caffeate (6), and betunaldehyde (8); three oleanane-type compounds were also identified, comprising δ-amyrone (15), δ-amyrin (16), and 11-oxo-β-amyrin (18); together with six sterols, comprising β-sitosterol (9), stigmasterol (10), 7α-hydroxy-β-sitosterol (11), 7α-hydroxy-stigmasterol (12), 7-keto-β-sitosterol (13), and 7-keto-stigmasterol (14). Their structures were elucidated using a variety of spectroscopic techniques, including 1D (1H, 13C, and DEPT-135 13C) and 2D (1H–1H COSY, 1H–13C HSQC, and 1H–13C HMBC) nuclear magnetic resonance (NMR) and accurate mass spectroscopy. Subsequently, the different plant extracts and some of the isolated compounds (1–9, 11 and 13) were investigated for their possible cytotoxic activity in comparison to cisplatin against a wide array of aggressive cancer cell lines, such as colorectal cancer (HCT-116), hepatocellular carcinoma (HepG-2), and prostate cancer (PC-3) cell lines. Compound 11 displayed broad cytotoxicity against all of the tested cell lines (IC50 ≅ 8 µg/mL in all cases), and a high safety margin against normal Vero cells (IC50 = 70 µg/mL), suggesting that 11 may be a highly selective and effective anticancer agent candidate. Notably, the evidence indicated that the mode of action of compound 11 could possibly consist of the inhibition of phosphodiesterase I (80.2% enzyme inhibition observed at 2 µM compound concentration). The increasing prevalence of lupin allergy as a consequence to the functional characteristics of a growing number of sweet lupin-derived foods consumption makes the imperious necessity to develop analytical tools for the detection of allergen proteins in foodstuffs. The current study developed a new highly specific, sensitive and accurate ELISA method to detect, identify and quantify the lupin main allergen β-conglutin (Lup an 1) protein in natural and processed food. The implementation of accurate standards made with recombinant conglutin β1, and an anti-Lup an 1 antibody made from a synthetic peptide commonly shared among β-conglutin isoforms from sweet lupin species was able to detect up to 8.1250 ± 0.1701 ng (0.0406 ± 0.0009 ppm) of Lup an 1. This identified even lupin traces present in food samples which might elicit allergic reactions in sensitized consumers, such as β-conglutin proteins detection and quantification in processed (roasted, fermented, boiled, cooked, pickled, toasted, pasteurized) food, while avoiding cross-reactivity (false positive) with other legumes as peanut, chickpea, lentils, faba bean, and cereals. This study demonstrated that this new ELISA method constitutes a highly sensitive and reliable molecular tool able to detect, identify and quantify Lup an 1. This contributes to a more efficient management of allergens by the food industry, the regulatory agencies and clinicians, thus helping to keep the health safety of the consumers. Full article

Chemical investigation of Cordia millenii, Baker resulted in the isolation of a new depsidone, cordidepsine (1), along with twelve known compounds including cyclooctasulfur (2), lup-20(29)-en-3-triacontanoate (3), 1-(26-hydroxyhexacosanoyl)glycerol (4), glyceryl-1-hexacosanoate (5) betulinic acid (6), lupenone (7), β-amyrone (8), lupeol (9), β-amyrin (10), allantoin (11), 2′-(4-hydroxyphenyl)ethylpropanoate (12) and stigmasterol glycoside (13). Hemi-synthetic reactions were carried out on two isolated compounds (5 and 6) to afford two new derivatives, that is, cordicerol A (14) and cordicerol B (15), respectively. The chemical structures of all the compounds were established based on analysis and interpretation of spectroscopic data such as electron ionization mass spectrometry (EI–MS), high resolution electrospray ionization mass spectrometry (HR-ESI–MS), fast atom bombardment mass spectrometry (FAB–MS), one dimension and two dimension nuclear magnetic resonance (1D and 2D-NMR) spectral data as well as X-ray crystallography (XRC). Lupeol ester derivatives [Lup-20(29)-en-3-triacontanoate (3)], monoglycerol derivatives [1-(26-hydroxyhexacosanoyl)glycerol (4) and glyceryl-1 hexacosanoate (5)] were isolated for the first time from Cordia genus while sulfur allotrope [cyclooctasulfur (2)] was isolated for the first time from plant origin. Biological assays cordidepsine (1) exhibited significant anti-HIV integrase activity with IC50 = 4.65 μM; EtOAc extract of stem barks, EtOAc fraction of roots and leaves were not toxic against 3T3 cells. As an important contributor to pollutant emissions to the atmosphere, land use can degrade environmental quality. In order to assess the impact of land-use planning on the atmosphere, we propose a methodology combining the land-use-based emission inventories of airborne pollutants and the long-term air pollution multi-source dispersion (LAPMD) model in this study. Through a case study of the eastern Chinese city of Lianyungang, we conclude that (1) land-use-based emission inventorying is a more economical way to assess the overall pollutant emissions compared with the industry-based method, and the LAPMD model can map the spatial variability of airborne pollutant concentrations that directly reflects how the implementation of the land-use planning (LUP) scheme impacts on the atmosphere; (2) the environmental friendliness of the LUP scheme can be assessed by an overlay analysis based on the pollution concentration maps and land-use planning maps; (3) decreases in the emissions of SO2 and PM10 within Lianyungang indicate the overall positive impact of land-use planning implementation, while increases in these emissions from certain land-use types (i.e., urban residential and transportation lands) suggest the aggravation of airborne pollutants from these land parcels; and (4) the city center, where most urban population resides, and areas around key plots would be affected by high pollution concentrations. Our methodology is applicable to study areas for which meteorological data are accessible, and is, therefore, useful for decision making if land-use planning schemes specify the objects of airborne pollutant concentration. The phytochemical study on the leaves of Acanthopanax gracilistylus (Araliaceae) resulted in the discovery of a new lupane-triterpene compound, acangraciligenin S (1), and a new lupane-triterpene glycoside, acangraciliside S (2), as well as two known ones, 3α,11α-dihydroxy-lup-20(29)-en-23,28-dioic acid (3) and acankoreoside C (4). Their chemical structures were elucidated by mass, 1D- and 2D-nuclear magnetic resonance (NMR) spectroscopy. The chemical structures of the new compounds 1 and 2 were determined to be 1β,3α-dihydroxy-lup-20(29)-en-23, 28-dioic acid and 1β,3α-dihydroxy-lup-20(29)-en-23,28-dioic acid 28-O-[α-l-rhamnopyranosyl-(1→4)-β-d-glucopyranosyl-(1→6)-β-d-glucopyranosyl] ester, respectively. The anti-neuroinflammatory activity of the selective compounds, 1 and 3, were evaluated with lipopolysaccharide (LPS)-induced BV2 microglia. The tested compounds showed moderate inhibitory effect of nitric oxide (NO) production. Full article
