Despite the increasing recognition of the role of urban orchard ecosystems in sustainable urban development, the mechanistic understanding of how tree species soil biochemical heterogeneity drives microbial community assembly, the spatial patterns governing microbe-environment interactions, and their collective contributions to ecosystem multifunctionality remain poorly characterized. This study investigated how Prunus species and soil depth affect microbial biodiversity and metabolomic signatures in an urban orchard in Cluj-Napoca, Romania. Soil samples were collected from five fruit tree species (apricot, peach, plum, cherry, and sour cherry) across three depths (0–10, 10–20, and 20–30 cm), resulting in 225 samples. The microbial community structure was analyzed through phospholipid fatty acid (PLFA) profiling, whereas the soil metabolome was analyzed by mass spectrometry techniques, including gas chromatography–mass spectrometry (GC–MS/MS) and MALDI time-of-flight (TOF/TOF) MS, which identified 489 compounds across 18 chemical classes. The results revealed significant tree species-specific effects on soil microbial biodiversity, with bacterial biomarkers dominating and total microbial biomass varying among species. The soils related to apricot trees presented the highest microbial activity, particularly in the surface layers. Metabolomic analysis revealed 247 distinct KEGG-annotated metabolites, with sour cherry exhibiting unique organic acid profiles and cherry showing distinctive quinone accumulation. Depth stratification influenced both microbial communities and metabolite composition, reflecting oxygen gradients and substrate availability. These findings provide mechanistic insights into urban orchard soil biogeochemistry, suggesting that strategic species selection can harness tree species-soil microbe interactions to optimize urban soil ecosystem services and enhance urban biodiversity conservation. Sperm membrane lipids play a crucial role in male fertility, influencing sperm motility, viability, and functional competence. This study comprehensively characterizes the phospholipid and sphingolipid composition in highly motile human spermatozoa obtained through the swim-up method, a widely used technique in assisted reproductive technology (ART). Using two-dimensional thin-layer chromatography and phosphorus analysis, we identified choline glycerophospholipids (CGP, 45%), ethanolamine glycerophospholipids (EGP, 26%), and sphingomyelin (SM, 17%) as predominant phospholipids, with minor components including cardiolipin, lysophospholipids, phosphatidylinositol, phosphatidylserine, phosphatidic acid, and neutral lipids. Gas chromatography analysis of glycerophospholipids (GPL) revealed a high long chain (C20–C22) polyunsaturated fatty acids (PUFA) content (46.3%), particularly docosahexaenoic acid (DHA, 22:6n-3), which was more abundant in CGP (46%) than EGP (26%). Sphingolipid analysis indicated that ceramide (Cer) and SM shared similar fatty acid profiles due to their metabolic relationship, with very-long-chain (VLC) PUFA (≥C26) being more prevalent in SM (10%) than in Cer (6%). Additionally, argentation chromatography identified highly unsaturated VLCPUFA species in Cer, including 28:3n-6, 28:4n-6, and 30:4n-6, which had not been previously quantified in motile human spermatozoa. Given the essential function of sphingolipid metabolism in spermatogenesis, capacitation, and acrosomal exocytosis, our findings suggest that the balance of VLCPUFA-containing SM and Cer could play a role in sperm performance and fertilization potential. This study provides novel insights into the lipid signature of human sperm and highlights the relevance of membrane lipid remodeling for male fertility and ART outcomes. Full article

This study investigated the effect of rice bran derived from broken rice feeding on the lipid classes and fatty acid composition of oil extracted from crickets (Acheta domesticus). During the final seven days prior to cricket oil extraction, crickets were fed either rice bran or control feed. The lipid classes in both the cricket oil and feed materials (rice bran or control feed) were identified using thin-layer chromatography, while the proportion of neutral lipids, phospholipids, and glycolipids was quantified by solid-phase extraction. Fatty acid composition was determined using GC-FID. Cricket oil from the rice bran-fed group exhibited a significantly higher ratio of glycolipids, which are abundant in rice bran, compared to the control group. Although both oleic and linoleic acid levels were significantly higher than in the control feed, only the oleic acid proportion was significantly increased in the cricket oil from the rice bran-fed group. Additionally, the ω-6/ω-3 ratio in each lipid fraction was significantly higher in the rice bran-fed group compared to the control group. These results suggest that dietary supplementation with rice bran derived from broken rice significantly alters the lipid profile of crickets and may represent a valuable strategy for enhancing the nutritional quality of cricket oil. Lipids are structurally diverse biomolecules that play essential roles in cellular function, energy storage, and signaling. The human lipidome, a dynamic and complex subset of the metabolome, is shaped by both endogenous factors, such as genetics, sex, age, and metabolic health, and exogenous influences like lifestyle, diet, and microbiota. Among these, diet stands out as one of the most modifiable and impactful determinants, influencing lipid composition across plasma, serum, and lipoprotein fractions. While traditional lipid profiling provides limited insight, lipidomics enables comprehensive characterization of lipid species, revealing mechanistic links between lipid metabolism and diseases such as cardiovascular disease (CVD), metabolic syndrome (MetS), and inflammatory disorders. This review explores: (1) the relationship between lipid profiles and CVD risk, (2) the internal and external modulators of the lipidome, and (3) current evidence on how specific dietary patterns, including Mediterranean, Nordic, low glycemic, and vegetarian diets, and individual nutrients such as omega-3 fatty acids (FAs), plant sterols, and mycoprotein, influence lipidomic profiles. Advances in lipidomics highlight that dietary fat quality, food matrix, and eating patterns can significantly modulate lipid species such as triacylglycerols (TAGs), ceramides (Cers), and phospholipids, with implications for cardiometabolic health. Notably, distinct responses are observed across plasma High-Density Lipoprotein (HDL) and Low-Density Lipoprotein (LDL) lipidomes, emphasizing the need for compartment-specific analyses. Understanding these diet-lipidome interactions offers promising avenues for precision nutrition and the development of lipid-based biomarkers for disease prevention and management. Full article

Phytoremediation is widely acknowledged as a viable method for soil remediation; however, its efficacy remains limited in soils co-polluted with petroleum hydrocarbons and heavy metals. To overcome this constraint, the present study explored an innovative approach utilizing ferrate-modified biochar (FeBC) to augment phytoremediation efficiency. Experimental findings revealed that ferrate treatment markedly modified the physicochemical characteristics of biochar, yielding thinner, smoother-surfaced structures with pronounced iron enrichment. At a 5% application rate alongside ryegrass cultivation, FeBC exhibited superior remediation performance, achieving 52.0% degradation of petroleum hydrocarbons (notably within the meso-aggregate fraction) and a 19.2% decline in zinc bioavailability via immobilization, thereby reducing zinc uptake in ryegrass tissues. Furthermore, FeBC amendment induced significant shifts in rhizosphere soil biochemistry and microbial ecology, characterized by diminished catalase activity but elevated urease and alkaline phosphatase activities. Phospholipid fatty acid profiling indicated a substantial rise in bacterial biomass (encompassing both Gram-positive and Gram-negative groups), particularly in meso- and micro-aggregates, whereas soil bacterial α-diversity declined markedly, accompanied by distinct compositional changes across aggregate size fractions. These results offer mechanistic insights into the synergistic interaction between FeBC and ryegrass in co-contaminated soil rehabilitation, the aggregate-dependent distribution of remediation effects, and microbial community adaptations to FeBC treatment. Collectively, this study advances the understanding of ferrate-modified biochar’s role in phytoremediation enhancement and clarifies its operational mechanisms in petroleum-zinc co-contaminated soil systems. Activation of TP53 signaling during ribosome biogenesis is an essential part of erythroid development, whereas the pathologic activation of TP53 in ribosomopathies such as Diamond-Blackfan anemia (DBA) and del (5q) myelodysplastic syndrome (MDS) prevents the normal expansion of erythroid precursors. TP53 can also be linked to the pathogenesis of DBA and MDS via ferroptosis, a form of iron-mediated cell death propagated by excess polyunsaturated fatty acid-containing oxidizable phospholipids and loss of lipid peroxide repair capacity. The primary objective of this work was to establish how overexpression and mutation of the TP53 gene influences lipid composition, erythroid differentiation, and ferroptosis sensitivity in K-562 cells, an in vitro model for studying erythropoiesis. Employing a reverse genetics approach, we generated four isogenic cell lines that either lacked functional TP53 expression, expressed wild-type (WT) TP53, or expressed one of the two most common TP53 mutation types, R175H or R282W. We then utilized non-targeted lipidomics to quantify and identify changes in specific lipid species that occur with induction of WT and mutant TP53 expression. We also analyzed differences in gene expression, ferroptosis sensitivity, and hemoglobinization by qPCR, CCK-8 cytotoxicity assay, and o-dianisidine staining, respectively. The abundance of 337 distinct lipid species was impacted by induction of WT TP53 expression compared to K-562 cells expressing a nonfunctional P53 protein. Yet only 17 lipid compounds were differentially impacted between cells expressing WT TP53 and either of the mutant TP53 genes tested. Similarly, while the TP53 null K-562 cells displayed modest sensitivity to ferroptosis, cells expressing both WT and mutant TP53 genes were remarkably resistant to ferroptosis. However, terminal differentiation and hemoglobinization were significantly impacted in R175H mutant TP53-expressing K-562 cells. Findings from this work provide novel insights into the role of TP53 in lipid metabolism and terminal erythropoiesis. Full article

The nutritional value of lipids depends not only on their fatty acid composition but also on their stereospecific positioning on the glycerol backbone. This study investigated the fatty acid composition and sn-2 positional distribution of triacylglycerols (TAG), as well as the composition of major phospholipids in golden pompano (Trachinotus ovatus) juveniles (initial weight: 10 g) fed five diets including graded levels of dietary n-3 highly unsaturated fatty acids (HUFA; 0.64–2.10%) for 56 days. With increasing dietary n-3 HUFA levels, the proportions of eicosapentaenoic acid (EPA), docosapentaenoic acid (DPA), docosahexaenoic acid (DHA), and total n-3 HUFA in muscle TAG, phosphatidylcholine (PC), and phosphatidylethanolamine (PE) significantly increased. Phospholipids, especially PC and PE, were preferentially enriched with n-3 HUFA, and the sn-2 positions of TAG showed a significantly increased deposition of DHA and reduced n-6/n-3 ratios. RNA-Seq analysis was performed on muscle tissues of T. ovatus subjected to different dietary n-3 HUFA levels to further investigate the molecular mechanisms of lipid compositional and structural changes. A total of 126,792 unigenes were obtained, of which 47.78% were successfully annotated. KEGG pathway enrichment analysis implicated the glycerophospholipid, glycerolipid, and sphingolipid metabolism pathways in lipid composition and distribution regulation, identifying gpat4, agpat3, agpat8, lpeat1, and lpgat1 as potential regulators. These findings offer insights into lipid remodeling in marine fish and support strategies to enhance aquaculture product quality. Full article

An attempt was made to understand the interactive consequences of subjecting a rhizospheric microbial community of xTriticocereale (Triticale) to higher CO2 levels and soil nitrogen addition in the short term in a tropical agro-ecosystem. Open-top chambers (OTCs) were used to grow the test crops for a single season under ambient CO2 (AC) and elevated CO2 (EC) along with two variable N dosages: recommended (N0: 0.053 g N/kg of soil) and high (N2: 0.107 g of N/kg of soil) levels. Variations in the composition of microbial communities and abundances were investigated using phospholipid fatty acid analysis (PLFA). A significantly (p < 0.001) increased microbial biomass content (MB) was observed under EC compared to AC, while the addition of N had a minor effect. A decreased fungi/bacteria (F/B) ratio (~38%) was observed with high N application in the CO2 enrichment treatment. Bacteria were more abundant, while fungal abundance decreased under N2 and EC. Gram (+ve) bacteria used these conditions to thrive under N2 and EC, while Gram (−ve) bacteria declined. No significant effects on actinomycetes were noticed in any of the treatments. However, eukaryotes acquired more benefits and flourished in response to EC. Varied responses were noted for the Shannon diversity index (H’) under EC. Overall, (i) bacteria (Gram-positive) and eukaryotes dominated under EC and high N addition, while fungi decreased, and (ii) EC and high levels of N addition did not affect actinomycetes. Short-term exposure under the given conditions was found to alter the rhizospheric microbial community. However, multiple season studies are needed to elucidate whether these short-term responses are transient or continuous. Full article

Hemp possesses significant healthcare value due to its rich composition of unsaturated fatty acids and a distinctive golden ratio of linoleic acid to α-linolenic acid. As a promising special-oil crop, it holds substantial potential for development and utilization. However, the regulatory mechanisms underlying its lipid metabolic pathways remain poorly understood. In this study, the independently bred hemp seed variety Longdama No. 9 was used to construct a regulatory network of the fatty-acid and lipid metabolisms through integrative transcriptomic and lipidomic analysis. Transcriptomic profiling revealed differentially expressed genes (DEGs) involved in lipid biosynthesis across various tissues. In leaves, DEGs associated with glycerolipid synthesis were generally upregulated compared to in roots and seeds. In seeds, DEGs involved in fatty-acid synthesis and triacylglycerol (TAG) assembly were predominantly upregulated. Meanwhile, root tissues showed a higher abundance of upregulated DEGs related to phospholipid biosynthesis. Lipidomic analysis further highlighted tissue-specific lipid distributions. The galactolipid monogalactosyldiacylglycerol (MGDG) was most abundant in the leaves. While phosphatidylglycerol (PG) had the highest molar percentage in the seeds, most other major phospholipids were predominantly found in the roots. The prevalence of the C36:6 molecular species in the MGDG and digalactosyldiacylglycerol (DGDG) indicates that hemp is a typical 18:3 plant. The combined transcriptomic and lipidomic analysis revealed that tissue-specific transcriptional regulation contributes to the unique lipid profile of hemp. These findings provide valuable insights into the regulation of lipid metabolism in hemp and identify key genes involved in oil biosynthesis, which can lay a theoretical foundation for the development and utilization of hemp as a special-oil crop. Background/Objectives: The synergistic effects of epigallocatechin gallate (EGCG) and taurine in modulating lipid metabolism abnormalities in rats were investigated, and along with their potential mechanisms. Methods/Result: Compared to intervention with EGCG/taurine alone, EGCG combined with taurine (1:3) not only reduced triglyceride (TG) generation in HepG2 cells (46.2%, 75.2%, respectively), but also significantly decreased the total cholesterol (TC) (33.3%, 41.8%), low-density lipoprotein cholesterol (LDL-C) (32.3%, 29.2%) in rats, while the high-density lipoprotein cholesterol (HDL-C) increased by 12.7% and 33.5%. In addition, the combination of EGCG and taurine not only inhibited lipogenic enzyme activity, but also enhanced the levels of lipid catabolic enzymes and antioxidant enzymes, and alleviated hepatic injury. Furthermore, it significantly modulated gut microbiota composition by altering the abundances of Bacteroidetes, Firmicutes, and Proteobacteria, improving intestinal flora balance. Metabolomic profiling showed that reducing N-linoleoyl proline, cortisol, and 3-isocholanolic acid, and increasing phospholipid metabolites are the main ways methods for normalizing lipid metabolism in rats. The combination also elevated short-chain fatty acid (SCFA) synthesis, preserving intestinal barrier integrity; it also promoted lipid catabolism and energy expenditure via activating Peroxisome proliferator- activated receptor alpha (PPARα) and suppressing hepatic fatty acid synthase (FAS)- mediated lipogenesis. Conclusion: These findings indicated that EGCG and taurine can synergistically regulate lipid metabolism abnormalities, which may offer a strategy for regulating lipid metabolism anomalies. The microbial community (as measured by phospholipid fatty acid analysis) and soil physicochemical properties (bulk density, organic matter, nitrogen, phosphorus, and potassium) were determined in soil samples collected from apple plantations of various ages (0-, 8-, 22-, 29-, and 36-year) in Gangdi Village, Xingtai, China. The soil samples were collected from depths of 0–20, 20–40, and 40–60 cm. The findings of the present study demonstrate that with increasing duration of apple cultivation, the soil bulk density and porosity decreased and increased, respectively. Initially, the content of soil nutrients such as organic matter, nitrogen, and phosphorus increased, eventually stabilizing, accompanied by a decline in pH. The soil microbial biomass significantly increased, accompanied by discernible alterations in the composition of the microbial community. Organic matter was found to be the primary factor influencing the structure and diversity of microbial communities. It is evident from forward analysis that the soil Gram-negative and actinomycete communities were predominantly influenced by soil pH, bulk density, and total phosphorus. In contrast, the Gram-positive and eukaryote communities were less affected by soil environmental factors. Notably, the soil bacterial community presented a greater degree of sensitivity to the duration of apple cultivation than did the fungal community. A marked vertical difference in the soil quality indicators was evident, with the increase in surface soil quality exceeding that of deeper soil depths. Full article

Soil microbial communities are crucial for ecosystem services, soil fertility, and the resilience of agroecosystems. This study investigated how long-term (31 years) agronomic practices—tillage, NPK fertilization, and cropping system—along with measured environmental variables influence the microbial biomass and its community composition in Chernozem soil under corn cultivation. The polyfactorial field experiment included three tillage treatments ((moldboard (MT), ripped (RT), strip (ST)), two fertilization regimes (NPK (N: 160; P: 26; K: 74 kg/ha), and unfertilized control) and two cropping systems (corn monoculture and corn–wheat biculture). The soil samples (0–30 cm) were collected in June and September 2023. Microbial biomass and community structure were quantified using phospholipid fatty acid (PLFA) analysis, which allowed the estimation of total microbial biomass and community composition (arbuscular mycorrhizal (AM) fungi, fungi, Gram-negative (GN) and Gram-positive (GP) bacteria, actinomycetes). Our results showed that microbial biomass increased from June to September, rising by 270% in unfertilized plots and by 135% in NPK-fertilized plots, due to higher soil moisture. Reduced tillage, especially ST, promoted significantly higher microbial biomass, with biomass reaching 290% and 182% of that in MT plots in June and September, respectively. MT had a higher ratio of bacteria-to-fungi compared to RT and ST, indicating a greater sensitivity of fungi to disturbance. NPK fertilization lowered soil pH by about one unit (to 4.1–4.8) and reduced microbial biomass—by 2% in June and 48% in September—compared to the control, with the particular suppression of AM fungi. The cropping system had a smaller overall effect on microbial biomass. Full article

Krill oil (KO) and fish oil (FO) are rich sources of long-chain polyunsaturated fatty acids, with eicosapentaenoic acid (EPA) and docosahexaenoic acid (DHA) bound to distinct molecular carriers (phospholipids vs. triglycerides). These oils have been the subject of considerable research interest over the past few years owing to their roles extensively studied for their antioxidant and anti-inflammatory properties relevant to disease prevention and therapy in certain diseases. This review aimed to provide a comparative summary of the antioxidant and anti-inflammatory activities of KO and FO, based on their bioactive components, and highlight the similarities and differences in their prospective mechanisms of action. Both oils exert antioxidant and anti-inflammatory activities, aligning with the review focus. The bioactivities of both oils stem from their distinct molecular compositions: KO delivers EPA/DHA via phospholipids, alongside astaxanthin, while FO provides EPA/DHA bound to triglycerides. In some cases, they exhibit similar outcomes, whereas in others, one may be more effective than the other. Further comparative studies examining dose-dependent effects, bioavailability kinetics, and tissue-specific molecular pathways are warranted. Full article

The Qinghai–Tibet Plateau, a region susceptible to global change, stores substantial amounts of soil organic carbon (SOC) in its alpine grassland. However, little is known about how SOC is regulated by soil microbial communities, which vary with elevation, mean annual temperature (MAT), and mean annual precipitation (MAP). This study integrates phospholipid fatty acid (PLFA) analysis to simultaneously resolve microbial biomass, community composition, and membrane lipid adaptations along an elevational gradient (2861–5090 m) on the Qinghai–Tibet Plateau. This study found that microbial PLFAs increased significantly with rising MAP, while the relationship with MAT was nonlinear. PLFAs of different microbial groups all had a positive effect on SOC storage. At higher altitudes (characterized by lower MAP and lower MAT), Gram-positive bacteria dominated bacterial communities, and fungi dominated the overall microbial community, highlighting microbial structural adaptations as key regulators of carbon storage. Saturated fatty acids with branches of soil microbial membrane dominated across sites, but their prevalence over unsaturated fatty acids decreased at high elevations. These findings establish a mechanistic link between climate-driven microbial community restructuring and SOC vulnerability on the QTP, providing a predictive framework for carbon–climate feedbacks in alpine systems under global warming. Background: Fatty acid status during the perinatal period is important for optimal offspring growth and development. Objectives: We aimed to test the association between maternal fatty acid (FA) intake during the third trimester of pregnancy and the FA composition of placental phospholipids, a marker of maternal fatty acid status. Methods: This cohort study was performed on 54 mothers participating in the PREOBE study. Maternal dietary intake was assessed with prospective 7-day food diaries at 34 weeks of gestation. Placenta samples were collected immediately after delivery and phospholipid FA was quantified with established methods. Data were analyzed with Pearson correlations and linear regression models, with adjustment for confounding factors. Results: Total energy intake was 2019 ± 527 kcal/d (mean ± SD) and total fat intake of the mothers was 87 ± 35 g/day. Myristic, stearic, oleic, and α-linolenic acid intakes were modestly correlated with placental percentages, with r-values ≤ 0.33. Only docosahexaenoic (DHA) acid intake (%-energy, %-fat, and g/d) showed r-values > 0.4 for the correlation with placenta phospholipids. Intake of other fatty acids, including arachidonic acid, was not associated with the placenta percentage. Linear regression models considering confounders showed only dietary DHA intake significant associations. Total fat intake did not interfere with the association of DHA intake with placental incorporation. Conclusions: DHA and arachidonic acid are enriched in the placenta, but only placental DHA content seems modifiable by maternal dietary DHA intake. Full article
