Impact of N-PEP-12 Supplementation on Attentional Performance and Mental Wellbeing in Healthy Adults with Subjective Cognitive Complaints: A Randomized, Placebo-Controlled Trial

Background: Subjective cognitive complaints (SCCs) are common in middle-aged and older adults and may reflect early cognitive changes, alongside alterations in stress, mood, sleep, and quality of life. N-PEP-12 is a peptide-based nutritional supplement with potential neuroprotective effects, but evidence of its benefits in healthy adults with SCCs remains limited. Objective: To evaluate the effects of N-PEP-12 supplementation on attention, cognitive function, and mental wellbeing in healthy middle-aged and older adults with SCCs. Methods: In this prospective, randomized, double-blind, placebo-controlled trial, 276 participants aged 50–75 years with SCCs and no clinically significant cognitive impairment were randomized to placebo, N-PEP-12 45 mg, or N-PEP-12 90 mg. Assessments were performed at baseline and after 30, 90, and 180 days. Primary outcomes included Test of Attentional Performance measures. Secondary outcomes included WAIS-IV Digit Span Forward and Backward, perceived stress, mood, sleep quality, and EQ-5D-5L visual analog scale. Results: Across the three primary attention outcomes analyzed jointly in a multivariate repeated-measures model, there was a significant group-by-visit interaction (p = 0.003) and a significant effect of visit (p < 0.001), without a significant main effect of treatment arm (p = 0.133), indicating a time-dependent treatment effect. This result was obtained in a sensitivity analysis population in which missing values were imputed under assumptions least favorable to the active arms. In exploratory endpoint-specific comparisons at 90 days, both N-PEP-12 groups showed greater improvements than placebo in alertness, attention omissions and memory omissions, and Digit Span Forward and Backward scores also improved. In a subsequent uncontrolled extension phase, in which all participants received active treatment, participants initially assigned to placebo showed comparable improvements after switching to N-PEP-12 90 mg. These observations are exploratory. Adverse events were infrequent and similarly distributed across groups. Conclusions: N-PEP-12 supplementation was associated with improvements in attention, working memory, and mental wellbeing in healthy middle-aged and older adults with subjective cognitive complaints. These findings support further investigation of N-PEP-12 as a nutritional intervention for early subjective cognitive changes associated with aging. Full article

Mango fruits are popular for their flavor. Sugar, a key component of fruit nutrition and flavor, determines fruit quality via its composition and content. While many studies explore molecular regulatory pathways of sugar metabolism in various fruits, large-scale physiological screening of related enzymes via advanced mathematical methods is largely neglected, and the physiological mechanism of sugar accumulation in mango remains unclear. This study analyzed glucose, fructose, sucrose and starch contents, and the activities of 11 sugar metabolism-related enzymes (adenosine diphosphate glucose pyrophosphorylase (AGP); sucrose synthase (SS); sucrose phosphate synthase (SPS); protein kinase (PK); starch debranching enzyme (DBE); phosphoglucomutase (PGM); α-amylase; β-amylase; isoamylase (ISA); sucrose invertase (INV); acid invertase (AI)) from fruit growth to post-ripening stages of mango cultivar ‘Renong No.1’, determined the correlations among soluble sugar, starch and enzyme activities, and constructed mathematical models of inter-group relationships using R language. The results showed that glucose, fructose and starch accumulated during fruit growth, while sucrose accumulated during post-ripening. Fructose content was significantly positively correlated with AGP, α-amylase, AI, β-amylase and SPS activities; so was glucose content and AGP activity; sucrose content and AGP, AI, PK, α-amylase, ISA, INV and SPS activities; and sweetness value and AGP, α-amylase, AI, SPS, SS and β-amylase activities. AI, AGP and α-amylase were identified as the key enzymes influencing sweetness value. Ultimately, a new visualizing and digitizing statistical approach for roles of eleven key enzymes in sweet value and soluble sugar component content during mango development was shown. Full article

Objective: This study investigates the relationship between social media use and consumer purchase intentions or out-of-home food choices in the Polish gastronomic market, focusing specifically on variations across generations. Methodology: Data were collected between May and July 2024 through a diagnostic survey using the Computer-Assisted Web Interviewing (CAWI) technique (N = 1099). Respondents were recruited via a non-probability quota sampling approach based on strict demographic inclusion criteria. To test the research hypotheses regarding generational variations in market behaviour, a multivariate discriminant function analysis was performed using Statistica 13.1 PL. A preliminary pilot study (N = 30) confirmed the initial questionnaire readability and overall consistency (Cronbach’s alpha = 0.87), while subscales were treated independently during the main analysis. Results: Multivariate models were found to be highly significant, revealing clear differences between age groups. The youngest cohort (aged 18–35) relies heavily on Instagram and TikTok, showing distinct patterns regarding visual triggers such as “instagrammable” aesthetics, digital validation, and menu uniqueness alongside weight loss claims. Conversely, seniors (aged 61 and older) lean unexpectedly towards X. This older segment displays pragmatic, utility-driven motives, searching for detailed textual data about ingredients and the health-promoting properties of food. General product quality and calorie control were identified as universal factors that do not vary by generation. Conclusions and Managerial Implications: The digital transformation of the Polish restaurant industry does not follow a single path. Social media is closely linked to modern customer journeys, with physical dining spots frequently serving as spaces for socialisation. Consequently, restaurant operators should move away from mass communication and adopt a selective omnichannel strategy, where message formats shift from visual appeal to factual, nutrition-oriented text that is tailored to the digital literacy and dietary needs of each generation. Pseudocereals such as quinoa, amaranth, and buckwheat have attracted increasing attention as ingredients for extruded snack products because of their nutritional value, gluten-free status, and content of bioactive compounds. The quality of extruded products is governed by complex interactions among processing variables, including barrel temperature, screw speed, feed moisture content, and formulation characteristics. As a result, mathematical modelling has become an important tool for predicting product properties and identifying suitable processing conditions. This review summarizes modelling approaches applied to extruded food products with a focus on pseudocereal extrusion. Particular emphasis is placed on response surface methodology (RSM), artificial neural networks (ANNs), adaptive neuro-fuzzy inference systems (ANFIS), support vector regression (SVR), and hybrid optimisation strategies. Published studies indicate that RSM remains the most commonly used approach because of its simplicity and interpretability, while ANN-based models generally provide much higher predictive accuracy when strong nonlinear relationships are present. The widespread use of small experimental datasets and limited external validation remains a major challenge for the practical implementation of advanced machine-learning models. This review examines the strengths and limitations of current modelling approaches and discusses future opportunities for integrating predictive models with digital manufacturing frameworks. Background/Objectives: The oral microbiome plays a central role in maintaining oral health from early life, with childhood representing a critical period for its establishment and long-term stability. While many environmental factors influence this dynamic microbial ecosystem, diet is distinct in being both universal and highly modifiable. This systematic review aims to evaluate and synthesize current evidence regarding the impact and mechanisms of distinct dietary habits, food matrices, and nutritional components on the composition, diversity, and ecological resilience of the pediatric oral microbiome. Methods: A literature review aligned with PRISMA guidelines was conducted via digital searches on PubMed, ScienceDirect, and Cochrane databases (January 2015–December 2025). Search strategies combined MeSH terms and keywords targeting “Microbiota”, “Mouth”, “Child”, “Diet”, and “Oral health”. Results: From 1068 records identified, 16 relevant articles met the inclusion criteria. Dietary habits may influence taxonomic and functional profiles. Frequent consumption of sugar-sweetened beverages, sucrose-rich sodas, and sweet treats induces notable dysbiosis and enriches acidogenic/aciduric taxa. Conversely, protective food matrices, including probiotic-fortified dairy products, polyol-based sugar-free chewing gums (xylitol and maltitol), bovine milk, and bioactive-rich agents like green tea, actively suppress cariogenic pathways (specifically Streptococcus mutans) and support commensal, health-associated genera without disrupting overall microbial structures. Conclusions: Diet represents an important modifiable factor shaping the pediatric oral microbiome, capable of either driving dysbiosis or reinforcing symbiosis. Cultivating a microbiome-informed dietary approach early in childhood supports a resilient microbial architecture, offering a non-invasive, public health framework for long-term oral and systemic disease prevention. Background: Obesity is a chronic, multifactorial disease that demands personalized and sustainable management approaches. Digital health technologies, such as artificial intelligence, wearable devices, mobile health apps, and virtual reality (VR), may support obesity care by providing enhanced behavioral monitoring, personalized feedback, and patient engagement. Objective: This critical narrative review discusses the current evidence on artificial intelligence, wearable technologies, and VR in the context of precision nutrition and obesity management and their possible clinical applications and limitations. Method: A critical narrative review was conducted using peer-reviewed literature published between 2019 and 2026 and identified through PubMed and Google Scholar. Search terms included combinations of “precision nutrition,” “personalized nutrition,” “obesity,” “weight management,” “metabolic health,” “digital health,” “artificial intelligence,” “machine learning,” “mobile health,” “wearable devices,” and “omics” using Boolean operators. Evidence from randomized controlled trials, systematic reviews, meta-analyses, and key conceptual studies was critically synthesized due to substantial heterogeneity in interventions and outcomes. Result: Wearables and mobile applications can enable continuous self-monitoring of physical activity, dietary intake, sleep, and physiological measures. Artificial intelligence may improve dietary personalization, risk prediction, glycemic control, and adaptive feedback. VR offers an immersive way to tackle behavioral and cognitive mechanisms related to overeating such as cravings, food cue reactivity, and inhibitory control. However, the evidence is heterogeneous, with many studies limited by short follow-up periods, small samples, variable adherence, and insufficient clinical validation. Conclusions: Artificial intelligence, wearable technologies, and VR are promising tools for precision obesity management, but their long-term clinical effectiveness remains uncertain. Future research should prioritize adequately powered trials, longer follow-up, standardized outcomes, transparent algorithms, ethical data governance, and integration with multidisciplinary nutrition and obesity care. Objective: The long-term management of cardiovascular diseases (CVDs) requires comprehensive rehabilitation strategies aimed at optimizing functional recovery and promoting behavioral changes. These interventions may include digital technologies that emerged as promising approaches to facilitate lifestyle changes, such as dietary behavior and physical activity (PA). Aim: This review aimed to synthesize the current evidence for the potential impact of remotely delivered nutrition interventions with or without a combination of PA interventions on biochemical biomarkers, cardiovascular indexes, anthropometric parameters, levels of PA, exercise and functional capacity, adherence to the Mediterranean Diet (MD), diet quality and quality of life, rehospitalization, urgent visits, death and acute events in adults (≥18 years) with CVDs. Secondary outcomes were accessibility, safety, usability, and adherence. Methods: The PubMed, Web of Science, and Scopus databases were comprehensively searched up to 2026 for Randomized Controlled Trials (RCTs) published in English, following the Preferred Reporting Items for Systematic Reviews and Meta-Analysis (PRISMA) guidelines. The methodological quality was assessed using the revised Cochrane Risk of bias tool for RCTs (RoB2). Results: 11 RCTs met the inclusion criteria. Sample sizes ranged from 66 to 879 participants. The duration of the interventions varied from 4 weeks to 12 months. The interventions were delivered remotely using a range of digital health technologies, including web-based programs, mobile phones, wearable devices, smartphone applications, and mini-apps. Favorable changes were observed in diet quality and in adherence to MD and PA levels, with no intervention-related adverse events reported. The RoB2 assessment indicated that the majority of included studies (6 out of 11) were classified as having some concerns regarding risk of bias. Conclusions: Remotely combined nutrition and PA interventions are a feasible and effective approach for improving mainly patient-reported outcomes in adults with CVDs. These findings support integrating remote multimodal rehabilitation into routine cardiovascular care and highlight the potential of telehealth-based models to enhance patients’ access to rehabilitation services. Background/Objectives: Although efficacious in randomized trials, the real-world adoption of AI-driven dietary management applications remains uncertain across diverse dining contexts and populations. Methods: This field-based observational study was conducted over 18 days at three real-world dining sites in Shanghai, China, enrolling 181 participants stratified into three groups based on food service style and customer attribute. A cross-sectional survey was administered on day 9, followed by a 9-day prospective usage tracking period. Results: After adjusting for sex, Group 2 (staff cafeteria with fixed-portion dishes) had the highest adjusted mean usability score at 71.20 (p < 0.001). Group 3 (community canteen) had the highest mean scores for information quality (16.57, p = 0.03) and perceptions of intended use in nutrition (12.01, p = 0.08). However, Group 3 recorded zero active usage sessions despite favorable initial perceptions. Conclusions: Favorable user perceptions of this AI-driven dietary management tool did not automatically translate into adoption. Scenario-specific usability and digital divide constraints define the boundary of real-world efficacy; moreover, AI may amplify existing dietary self-management behaviors rather than creating them de novo. Artificial intelligence (AI) is increasingly being applied in healthcare, with growing relevance to clinical nutrition. This narrative review examines current and emerging uses of AI in nutrition care within the Nutrition Care Process framework, with attention to assessment, monitoring and evaluation, diagnosis, intervention, and clinical support tools. Current applications include AI-assisted dietary assessment using image recognition, wearable sensors, analysis of continuous glucose and other physiologic data for early risk detection, and support for malnutrition screening and diagnosis. AI is also being explored for identifying micronutrient deficiencies and complications of nutrient excess, as well as for screening and early intervention in eating disorders. In nutrition intervention, AI has potential to support personalized dietary planning, nutrition support in intensive care settings, behavioral interventions, and precision nutrition approaches such as digital twins. Additional applications include clinical decision support and documentation assistance. However, despite its usefulness, concerns about AI systems exist. Its performance depends on the quality of the data used to train it; it can introduce bias, and it can produce inaccurate or misleading outputs. In addition, overreliance on AI may also reduce clinician attentiveness and contribute to cognitive errors. For these reasons, AI should be regarded as a support tool rather than a replacement for human clinical care. Overall, AI offers substantial opportunities to improve the personalization, efficiency, and scalability of clinical nutrition practice, but its safe and effective implementation will require continued validation, careful oversight, and integration with clinical expertise. Background/Objectives: Patients with inflammatory bowel disease (IBD) frequently experience food-related symptoms and express a strong need for dietary guidance, yet evidence-based recommendations remain limited. This often leads to self-imposed dietary restrictions, potentially compromising nutritional intake and food-related quality of life (Fr-QoL). The primary aim of this study was to evaluate changes in Fr-QoL, following personalized feedback on an IBD-specific food frequency questionnaire and access to an online nutritional knowledge portal over a 6-month period. Secondary exploratory outcomes included changes in dietary intake and diet quality. Methods: In this nationwide pre–post study, 108 patients with IBD completed baseline and 6-month assessments. Dietary intake was measured using the validated GINQ-FFQ and Fr-QoL using a 29-item questionnaire. Nutrient intake was compared with EFSA reference values. Participants received automated personalized feedback and had access to educational tools and dietitian resources. Changes over time were analyzed using paired statistical tests. Results: A significant improvement in Fr-QoL was observed (mean difference 8.1 points, 95% CI 5.5–10.7; p < 0.001; Cohen’s d = 0.39). Baseline intake was characterized by high fat and low fiber consumption. After 6 months, total energy intake decreased (p < 0.001), along with reductions in protein, fat, carbohydrates, fiber, and several micronutrients, including iron, folate, vitamin B6, vitamin C, potassium, magnesium, and zinc. Diet quality did not improve, and adherence to EFSA guidelines remained suboptimal. The digital tools were positively evaluated. Conclusions: In this single-arm pre–post study, Fr-QoL scores were higher at follow-up than at baseline among participants who completed both assessments, whereas dietary intake and overall diet quality did not improve. The findings support the feasibility and acceptability of the developed digital tools for dietary assessment and nutrition education, although their effectiveness requires further evaluation in controlled studies. Additional strategies, including structured dietitian-led support combined with digital resources, may be required to optimize nutritional outcomes in patients with IBD. Full article

Background: Restrictive dieting is a common behavior among young adults and may be associated with increased vulnerability to disordered eating. University students represent a population potentially exposed to multiple influences affecting eating behaviors, including academic stress, social pressures, and appearance-related standards promoted through digital media. This study aimed to analyze the relationship between restrictive dietary practices, eating behaviors, and indicators of vulnerability to disordered eating among university students in Poland. Materials and Methods: A cross-sectional study was conducted among 420 university students aged 19–34 years. Data were collected using an anonymous online questionnaire consisting of sociodemographic questions, the original Restrictive Diet Motivation Questionnaire (RDMQ), and the validated Three-Factor Eating Questionnaire-13 (TFEQ-13). Motivational factors were categorized into aesthetic, social, health-related, eating control, and self-discipline, and personal beliefs regarding restrictive dieting. Statistical analyses included the Gamma correlation coefficient, chi-square test, and ordinal logistic regression, with statistical significance set at p < 0.05. Results: Restrictive dieting was reported by a substantial proportion of participants, with women reporting this behavior more frequently than men. The most prominent motivations were related to appearance, weight control, social influences, and personal beliefs regarding restrictive diets. Participants who followed restrictive diets demonstrated higher levels of cognitive restraint compared with those who did not engage in restrictive dieting. Strong associations were observed between cognitive restraint and aesthetic motivations, personal beliefs regarding restrictive dieting, social motivations, eating control and self-discipline, and health-related motivations. Multivariable analysis confirmed that aesthetic motivation, personal beliefs, previous restrictive dieting, and female sex were independent predictors of higher cognitive restraint. Conclusions: Restrictive dieting among university students is strongly associated with appearance-related motives, dieting-related beliefs, and increased cognitive restraint. In this study, eating disorder risk was inferred from behavioral indicators, particularly cognitive restraint, rather than from clinical diagnostic criteria. These findings highlight the need for preventive strategies promoting evidence-based nutrition knowledge, healthy eating behaviors, and a positive body image among young adults. Full article

Climate change is transforming heat stress (HS) from a seasonal challenge into a major constraint on the sustainability of dairy cattle production. Increasing temperatures, more frequent and prolonged heat waves, and reduced opportunities for nocturnal recovery adversely affect animal health, welfare, productivity, reproductive performance, and resource-use efficiency. This narrative review provides an integrated critical synthesis of current evidence on HS in dairy cattle within a sustainable development framework. It examines the biological mechanisms underlying disruption of homeostasis together with the consequences for milk production, product quality, health, welfare, reproduction, and developmental programming. The review further evaluates biomarkers of thermal adaptation, precision livestock farming, and environmental, nutritional, managerial, genetic, and technological mitigation strategies, considering their biological effectiveness, practical applicability, economic feasibility, and environmental trade-offs. Rather than addressing these components independently, the review integrates their interactions within an original conceptual framework and comparatively evaluates adaptation strategies to support practical decision-making. Current evidence indicates that sustainable climate adaptation requires integrated, context-specific approaches combining environmental management, nutrition, genetic improvement, precision monitoring, and digital technologies. Future progress will increasingly depend on advances in omics, artificial intelligence, and predictive livestock management to enhance climate resilience while improving productivity, animal welfare, and environmental sustainability. We conducted an online cross-sectional study to assess malnutrition risk, food and nutrition insecurity, and perceived barriers and facilitators to dietary management among adults (18–64 years old) with a self-reported clinical diagnosis of inflammatory bowel disease (IBD) in outpatient and community settings. We assessed malnutrition risk using the Saskatchewan IBD-Nutrition Risk screening tool, food security risk using the two-item Hunger Vital Signs questionnaire, and nutrition security risk using a validated four-item tool. Open-ended questions elucidated barriers and facilitators to dietary management of IBD. Among 57 participants who self-reported a clinical diagnosis of IBD, 52.6% (n = 30/57) had ulcerative colitis, and 47.4% (n = 27/57) had Crohn’s disease. Nutrition insecurity was higher among those at high risk of malnutrition than among those at low-to-medium risk (p = 0.018), and there were no differences in food security status (p = 0.081). Using univariate logistic regression, the odds of being classified as having a high risk of malnutrition were associated with being male, following a special diet or nutrition plan, experiencing IBD-related flare-ups, and nutrition insecurity. Furthermore, healthcare professionals and social support, as well as digital resources/nutrition-related apps, were identified as facilitators to dietary management, while barriers included food access and expense, limited nutrition knowledge and resources, and fear of IBD-related symptom flare-ups. Although self-reported malnutrition risk and food and nutrition insecurity are common, larger prospective studies incorporating validated disease-activity measures and comprehensive nutritional assessment are needed to clarify these relationships and evaluate integrated dietetic and social-support interventions. Behavioural nudges modify choice architecture to steer food choices without removing alternatives or substantially changing economic incentives. This critical narrative review synthesises 62 sources from behavioural public policy, public health nutrition, food ethics, and sustainable food-system governance. It distinguishes healthy food nudges, sustainable food nudges, green nudges, and responsible food-system interventions, and compares availability, placement, defaults, labels, digital architecture, and mixed policy packages. The evidence shows that nudges influence immediate food selection, but effects are heterogeneous and weaker for sustained environmental outcomes. Price instruments address affordability, nudges address attention and convenience, and boosts strengthen decision competence; none can substitute for the others. Ethical acceptability requires a transparent, calibrated opt-out process, equity monitoring, disclosure of commercial motives, democratic accountability, and integration with structural policy. Public institutions and profit-driven digital platforms therefore require different governance safeguards. Nudges are most defensible as components of coordinated food-system policy rather than stand-alone solutions. Full article

Home enteral nutrition (HEN) is essential for patients with a functioning gastrointestinal tract who cannot meet nutritional needs orally, yet outpatient management remains complex, resource-intensive, and supported by a limited evidence base. Artificial intelligence (AI) may augment HEN care by extending monitoring, education, risk assessment, documentation support, and operational coordination into the home environment. Consistent with the narrative review format, this article uses a pragmatic, transparent synthesis of influential HEN-specific literature, relevant clinical nutrition evidence, and background knowledge from adjacent fields, including home healthcare, chronic disease management, oncology nutrition, telehealth, and software regulation. Direct evidence in established HEN populations remains scarce; therefore, most AI applications should be considered hypotheses or early implementation opportunities rather than proven standards of care. The strongest near-term opportunities are clinician-supervised patient education, symptom triage, adherence support, remote monitoring, and workflow automation. Predictive analytics, smart pumps, and precision enteral prescription tools are promising but require prospective HEN-specific validation, interoperability with electronic health records and home-infusion systems, reimbursement pathways, and governance safeguards. Key barriers include dataset bias, limited external validation, alert fatigue, privacy and regulatory concerns, unclear accountability, digital equity, cost uncertainty, and the risk of dehumanizing care. AI should be viewed as a complement to multidisciplinary HEN expertise. Priorities for the near future include HEN registries, standardized outcomes, prospective validation, pragmatic implementation trials, health-economic evaluation, and transparent oversight that preserves clinician accountability and patient-centered care.
