Botanical gardens serve as vital centers for ex situ conservation, maintaining diverse plant species under controlled conditions. Terrestrial orchids, despite their wide diversity and distribution, often occur in small and declining populations, making their conservation increasingly urgent. This study aimed to examine the potential for establishing the first specialized orchidarium in Serbia, focusing on the native orchid species of the Fruška Gora region. A SWOT analysis, combined with site assessment data, was employed to identify key strengths, weaknesses, opportunities, and threats, informing the development of a functional zoning plan. The results indicate that such an orchidarium would offer a threefold benefit: strengthening ex situ conservation, advancing scientific research and environmental education, and promoting sustainable tourism. The proposed design consists of eight distinct zones, three of which reflect natural habitats of selected orchid species. The planned integration of a seed gene bank in the central zone, along with living plant collections and a nearby in vitro culture laboratory, establishes a comprehensive framework for the sustainable management of orchid genetic resources in the region, forming a foundation for future research and preservation. The Bignoniaceae family encompasses numerous species of ecological, medicinal, and cultural significance, yet its ethnobotanical value remains underexplored in many regions of Thailand. This study investigates the diversity, phenology, cultural relevance, and traditional uses of Bignoniaceae species in Maha Sarakham Province, Northeastern Thailand. Through semi-structured interviews with 260 local informants across 13 districts—alongside field observations and herbarium voucher collections—we documented 27 species across 21 genera. These integrated methods enabled the identification of key culturally significant species and provided insights into their traditional uses. Phenological data revealed clear seasonal patterns in flowering and fruiting, aligned with the regional climatic cycle. Quantitative ethnobotanical indices—including Species Use Value (SUV), Genera Use Value (GUV), Relative Frequency of Citation (RFC), Cultural Importance Index (CI), and Cultural Food Significance Index (CFSI)—were employed to evaluate species significance. Results indicate that species such as Dolichandrone serrulata, D. spathacea, and Oroxylum indicum hold high cultural and practical value, particularly in traditional medicine, spiritual practices, and local landscaping. These findings underscore the critical role of Bignoniaceae in sustaining biocultural diversity and emphasize the urgency of preserving traditional botanical knowledge amid environmental and socio-economic change. Moreover, the insights contribute to broader efforts in cultural heritage preservation and biodiversity conservation across tropical and subtropical regions. Full article

Bee pollen’s aroma combined with other floral components serve various purposes, including attracting pollinators and signaling the availability of food sources. The present study aimed to comparatively analyze the volatile profiles of unifloral pollen taxa. Bee pollen loads were collected using pollen traps and sorted based on their botanical origin, determined by color and pollen grain morphology. The separated pollen samples were analyzed using a Purge & Trap/GC-MS system, identifying the volatile profiles of pollen from 16 plant species. The analysis revealed distinguished differences in the total volatile organic compounds (VOCs) among the various pollen species. Notably, the pollen from Erica manipuliflora, Papaver rhoeas, and Sisymbrium irio contained the highest number of VOCs, with 54, 51, and 42 substances detected, respectively. Certain volatile compounds appeared to correlate with increased bee visitation. For instance, 4-methyl-5-nonanone was uniquely found in E. manipuliflora pollen, while isothiocyanate compounds were exclusively present in species of the Brassicaceae family. Therefore, given the significant impact of VOCs on honey bees’ preferences, it is essential to consider not only the nutritional value of bee pollen when evaluating its beekeeping value, but also its aroma profile. Pollen, the male gametophyte of flowering plants, is collected by honeybees as a primary source of protein and converted into bee pollen through the enzymatic activity of digestive secretions. The nutrients in bee pollen are available in amounts well beyond those of proteins, comprising macronutrients such as carbohydrates, lipids and dietary fiber, as well as micronutrients such as minerals, vitamins, organic acids, and phenolic compounds. This study aimed to determine the macro and trace mineral content of bee pollen from different botanical and geographical origins, and to assess their bioaccessibility through simulated in vitro digestion, their dietary contribution, and potential health risks. Seven bee pollen samples were investigated, three with a monofloral origin of above 80%, from Nigella spp., Helianthus annuus and Castanea sativa, and four with a multifloral origin. Mineral composition revealed potassium as the most abundant element, while iron, manganese, and copper were found at trace levels. Castanea sativa pollen had the highest overall mineral content, whereas Nigella spp. showed the lowest values for calcium, magnesium, and copper. The bioaccessibility of bee pollen was highest during the gastric phase for most minerals except copper, where most of the samples peaked in the intestinal phase. Overall, mineral bioaccessibility after simulated digestion followed the order K > Mg > Cu > Ca > Mn > Fe > Zn. While for manganese, the consumption of bee pollen showed the highest contribution to recommended dietary intake (16% for women and 12% for men), calcium had the lowest, with less than 1% of the RDA at a consumption level of 40 g/day. Health risk assessment confirmed that consuming 40 g/day of bee pollen poses no risk because the target hazard quotient and hazard index are below the risk threshold of 1.0. Full article

Muehlenbeckia tuggeranong is an endangered subshrub with an estimated seven individuals remaining in its native habitat, and twelve held in an ex situ living collection in the Australian National Botanic Gardens, Canberra. We conducted a genetic analysis on all known individuals of the species both in situ and ex situ to inform the conservation management of one of the rarest plants in Australia, certainly the rarest in the Australian Capital Territory. We found recent seedlings did not result from hybridisation with M. axillaris but resulted from sexual reproduction within the ex situ collection, leading to greater genetic diversity ex situ than in situ. However, low genetic diversity across the species indicates a high risk of extinction. Through simulations we identified the optimal breeding pairs to minimise further genetic diversity loss and increase the number of available genotypes for future reintroduction. Our work highlights the need to incorporate genetically informed breeding programs into living collections management of endangered plant species, particular those with unique life history traits. Although the usage of genetic data for the conservation management of threatened species is growing rapidly, the assessment of the trajectory and persistence of ex situ and in situ populations suffers from a lack of monitoring of genetic indicators. To fill this gap, we conducted a comparative analysis of the genetic structure ex situ and its source population of Cochlearia polonica to improve conservation outcomes for this species. C. polonica, as a range-restricted endemic species in Poland, exists in the wild in a single reintroduced population, which was the source for establishing an ex situ population in the botanical garden, and both populations have been well studied in abundance time series until the extinction of artificial populations due to genetic constraints. We collected AFLP data from individuals sampled after the peak recovery phase following the founder effect in the ex situ population, as well as from the source population, to assess the genetic consequences of long-term cultivation. The genetic profile of both populations suggested their strong differentiation. The genetic variation in the ex situ population expressed by Shannon’s Information Index and expected heterozygosity (He) was approximately half that of the source population. While ex situ population growth was high, the gene pool was homogenised, and genetic diversity waned, leading to its extinction across merely 5–6 generations. Thus, the only reintroduced population of C. polonica left in the wild is more threatened with extinction than previously thought, and conservation action should be taken immediately. Full article

Impatiens reidii Hook.f., a presumed extinct and Kumaon endemic species only known from its type collection in 1886, is rediscovered from near type locality after a gap of 138 years. The identity of the historically ambiguous name Impatiens tingens Edgew. is reinvestigated based on studies of literature, type specimens, and recollection of live material from the type locality that confirmed the name I. tingens is conspecific with I. scullyi Hook.f. and thus latter name is synonymised. Impatiens inayatii Hook.f., another little-known species described by Hooker as endemic to Kali valley, Kumaon currently treated as a synonym of Impatiens bicornuta Wall., is recollected from near the type locality after 124 years. Our recollection confirms its identity as a totally distinct species from section Urticifoliae S.Akiyama and H.Ohba, thus reinstating name I. inayatii. Augmented description for all three species is provided with information on their botanical history, species etymology, key identification characteristics, distribution, phenology, ecology, and first photographic documentation of I. reidii and I. inayatii. Also, lectotypes are designated for the names I. reidii, I. tingens, I. laxiflora var. stracheyi Hook.f. and I. micranthemum Edgew. Full article

The flower buds of Magnolia biondii Pamp. (MBP), one of the botanical sources of Xinyi (Flos Magnoliae), are widely used in traditional medicine; however, their potential role in melanoma treatment remains unexplored. In this study, the phytochemical composition, antioxidant activity, and anti-melanoma mechanisms of MBP extracts were systematically investigated. Phytochemical profiling using UHPLC-Q-Exactive Orbitrap MS identified 26 bioactive compounds. The ethanol extract exhibited high total flavonoid and polyphenol contents, correlating with enhanced antioxidant capacity as demonstrated by DPPH and ABTS assays. Network pharmacology analysis highlighted the JAK/STAT signaling pathway, identifying STAT3 and STAT1 as core targets. Western blot analysis confirmed MBP significantly inhibited the phosphorylation of JAK1 and STAT1 in melanoma cells. Connectivity Map (CMap) and network analyses further pinpointed naringenin as a primary active constituent. In vitro assays demonstrated that MBP and naringenin inhibited the proliferation and migration of A375 and B16F10 melanoma cells, while exhibiting relatively low cytotoxicity toward normal keratinocytes. Molecular docking and dynamics simulations revealed strong and stable binding interactions between naringenin and JAK1/STAT1 proteins. These findings collectively support MBP and naringenin as promising candidates for melanoma treatment, providing mechanistic evidence for their targeted activity and laying a foundation for future research and clinical applications. Butterflies have nectar-feeding preferences based on various floral characteristics, including flower shape, size, color, fragrance, and nectar composition, which in turn affect their survival, reproduction, and roles in pollination. The National Botanical Garden (NBG) in Lalitpur, Nepal, holds a variety of flowering plants and butterfly populations, providing a suitable study site to test the hypotheses on floral preferences of butterflies. This study assessed the floral preferences of the butterfly community in the NBG based on flower color, the origin of flowering plants (native and alien), and the type of plants (herbs and shrubs). It also tested the association between butterfly proboscis lengths and corolla tube lengths of flowers. Data were collected from 10 blocks (each 5 × 5 m2) through direct observation during the spring and autumn seasons, from March to October 2022. A total of 24 species of butterflies were recorded during the study period, with the chocolate pansy (Junonia iphita) being the most abundant. The relative abundance of pink flowers was higher in the NBG, but the butterflies’ visitation frequency was significantly higher on yellow flowers (p < 0.05) than on other colors. The visitation frequencies of butterflies significantly varied with the flowers’ origin and types. Butterflies visited flowers of alien origin more frequently than native ones (p < 0.05) and those of herbs over shrubs (p < 0.05). Flowers from alien plants, such as Calluna vulgaris and Viola tricolor, were among the most frequently visited. The proboscis length of butterflies showed a significantly strong positive correlation with the corolla tube length of flowers (τ = 0.74, p < 0.001). These results can inform conservation practices and garden management strategies aimed at supporting butterfly diversity through the intentional selection of floral resources. Full article

The chemical composition of Aloe vera leaves was investigated by using liquid chromatography–high-resolution tandem mass spectrometry (LC-HRMS/MS). Five A. vera samples were collected across diverse geographical origins and cultivation practices: PO (Botanical Garden of Portici, Italy), CAN (Gran Canaria, Canary Islands), CA, E, and MM (Marine Reserve of Torre Guaceto, Brindisi, Italy). Analysis of hydroalcoholic organic extracts revealed 77 organic compounds, including ubiquitous primary metabolites (i.e., sugars, amino acids, and fatty acids) and natural products (i.e., phenols and aromatics, terpenes, and anthraquinones). Principal component analysis (PCA) on the raw dataset indicated a clear separation of samples depending on their geographical origins. PO samples showed high amounts of citric acid, the anthraquinone aloe-emodin, and the alkaloids tropine and tropinone. CAN samples showed high content of malic, citramalic, citraconic, erucic, and 3-dehydroquinic acids. CAN and PO samples contained high amounts of jasmonic, quinic, and 4-methoxycinnamic acids along with aloesin, tyramine, coumarin, and saponarin. Among the Brindisi samples, MM contained high amounts of limonene and α-linolenic acid. CA, E, and MM samples presented high amounts of eritrose-4-phosphate, glucose-1-phosphate, and fructosyl valine along with ginsenoside, proline, and ascorbic acid. These findings indicate that geographical origins and cultivation practices affect to different extents the metabolite profile of A. vera plants. Full article

Phlox are ornamentals of great decorative value, grown throughout the world for their attractive flowers. Phlox cultivar collections at the Tsitsin Main Botanical Garden and the Botanical Garden of Lomonosov Moscow State University (both Moscow, Russia) were surveyed for virus diseases. Tobacco streak ilarvirus (TSV), beet ringspot nepovirus (BRSV), and BRSV satellite RNA (satRNA) were first detected in phlox when viromes of symptomatic Phlox paniculata plants were studied using high-throughput sequencing. The nearly complete genomes of three TSV and BRSV isolates and two BRSV satRNAs were assembled and characterized. TSV isolates shared 96.9–99.7% nucleotide sequence identity and were 82.2–89.1% identical to their closest relatives from broad bean, dahlia, and echinacea. BRSV isolates were distantly related to each other (83.7–89.3% identity) and were closest to those from oxalis and potato. BRSV satRNAs shared 90.6% identity and were 87.8–94.1% identical to satRNAs associated with tomato black ring virus L and S serotypes. Thus, TSV, BRSV, and BRSV satRNA were for the first time detected in a new natural host P. paniculata in Russia, adding to the list of known phlox viruses and expanding information on the host range, geographic distribution, and genetic diversity of these viruses. Background: In many African areas, herbal products still represent a significant source of healthcare. However, a major gender bias is evident in the literature, as most of the work is carried out by male researchers, collecting data from male traditional practitioners, and thus often neglecting women’s specific health issues. This warrants a detailed review of the current knowledge about the major medicinal plants historically and still used for women’s health. Objective: This study aims to compile and critically analyze published data on the use of traditional herbal remedies by African women in addressing specific health conditions, in order to evaluate the potential of traditional medicine as a viable alternative or complementary approach to modern healthcare for women globally. Methods: Data were retrieved from databases by combining the following relevant keywords: “abortion, adverse, Africa, attendant, birth, botanical, delivery, developing, drug, ethnomedicine, ethnopharmacology, folk, gynecological, healing, infertility, herb, indigenous, lactation, medicine, native, obstetric, phytomedicine, plant, pregnancy, remedy, side, sub-Saharan, traditional, treatment, women”. Results: More than 125 studies, carried out across 12 African nations, revealed that up to 80% of African women resort to herbal medicines. An estimated 200 medicinally important plant species are reported to be utilized by women in different African countries, including Benin, Cameroon, Côte d’Ivoire, Egypt, Ethiopia, Ghana, Kenya, Mali, Nigeria, South Africa, Tanzania, and Zimbabwe. These herbs have many applications, mostly focused on infertility, pregnancy, painful menstruation, breast feeding, breast cancer, and contraception. Interestingly, according to their occurrence of usage, the plants most commonly reported for these conditions that are important to women are ambivalent plants (i.e., used both as foods and medicines) that include Zingiber officinale Roscoe, Allium sativum L., Cucurbita pepo L., and Ricinus communis L. Conclusions: Even though most women, in most African countries, do use traditional medicine, the amount of work published remains quite limited and no data are available in many countries. This study explores the sustainability of Islamic decorative arts by examining the symbolic, cultural, and spiritual dimensions of botanical decorations in Makkah’s architectural heritage. Grounded in Carl Jung’s theory of the collective unconscious and Lamya Al-Faruqi’s philosophy of Tawhid, the research uncovers the profound psychological and spiritual meanings embedded in these motifs. Employing a qualitative methodology, the study integrates symbolic analysis, cultural interpretation, and historical documentation, supported by digital design tools, to assess the relevance of these decorations in contemporary urban contexts. Findings reveal that botanical motifs, such as palm trees and pinecones, reflect universal archetypes of resilience and growth while symbolising divine unity through abstraction and harmony. The research highlights their integral role in architectural structures and their potential in cultural tourism and educational initiatives. However, challenges such as urbanisation necessitate urgent documentation and innovative preservation strategies. This study offers valuable insights into sustaining Makkah’s architectural identity by bridging psychological and philosophical perspectives. Its recommendations align with Saudi Vision 2030 and global sustainability goals, advocating for the revival and integration of these motifs into modern urban design to ensure the continued appreciation and recognition of Makkan architectural heritage. Background/Objectives: Parkinson’s disease (PD) is a common neurodegenerative disease characterized by motor symptoms caused by the loss of dopaminergic neurons. While the pathophysiology of PD is still not fully understood, it is recognized that oxidative stress plays a major role in its progression. Previous studies have shown that the aerial parts of Artemisia iwayomogi Kitamura (AIK) possess medicinal properties, including antioxidant activity. This study aimed to investigate whether AIK can alleviate neuronal loss and motor symptoms in a PD model and to explore its therapeutic mechanisms. Methods: For the in vitro study, PC12 cells were treated with AIK and 1-methyl-4-phenylpyridinium (MPP+). For the in vivo study, C57BL/6J mice were orally administered AIK for 12 days; they received intraperitoneal injections of 1-methyl-4-phenyl-1,2,3,6-tetrahydropyridine (MPTP) for 5 consecutive days, starting on the 8th day of AIK administration. Results: AIK treatment to PC12 cells in the presence of MPP+ enhanced the phosphorylation of the protein kinase B/glycogen synthase kinase-3β signaling pathway, which is a crucial regulator of nuclear factor erythroid 2-related factor 2 (Nrf2) translocation. Additionally, AIK treatment increased cell survival and induced an antioxidant response involving heme oxygenase-1, via increasing the level of Nrf2 in the nucleus. In an MPTP-induced mouse model of PD, AIK administration activated Nrf2 in dopaminergic neurons and prevented the loss of dopaminergic neurons in the brain, which in turn alleviated motor dysfunction. Conclusions: Collectively, these findings suggest that AIK is a potential botanical candidate for PD treatment by protecting dopaminergic neurons through antioxidant activity. Full article

Biofilms, structured microbial consortia embedded in self-produced extracellular matrices, pose significant challenges across the medical, industrial, and environmental sectors due to their resistance to antimicrobial therapies and ability to evade the immune system. Their resilience is driven by multifaceted mechanisms, including matrix-mediated drug sequestration, metabolic dormancy, and quorum sensing (QS)-regulated virulence, which collectively sustain persistent infections and contribute to the amplification of antimicrobial resistance (AMR). This review critically examines the potential of plant-derived essential oils (EOs) as innovative agents for biofilm control. EOs exhibit broad-spectrum antibiofilm activity through multi-target mechanisms, including disrupting initial microbial adhesion, degrading extracellular polymeric substances (EPSs), suppressing QS pathways, and compromising membrane integrity. Their ability to act synergistically with conventional antimicrobials at sub-inhibitory concentrations enhances therapeutic efficacy while reducing the selection pressure for resistance. Despite their potential, EO applications face technical challenges, such as compositional variability due to botanical sources, formulation stability issues, and difficulties in standardization for large-scale production. Clinical translation is further complicated by biofilm stage- and strain-dependent efficacy, insufficient in vivo validation of therapeutic outcomes, and potential cytotoxicity at higher doses. These limitations underscore the need for optimized delivery systems, such as nanoencapsulation, to enhance bioavailability and mitigate adverse effects. Future strategies should include combinatorial approaches with antibiotics or EPS-degrading enzymes, advanced formulation technologies, and standardized protocols to bridge laboratory findings to clinical practice. By addressing these challenges, EOs hold transformative potential to mitigate biofilm-associated AMR, offering sustainable, multi-target alternatives for infection management and biofilm prevention in diverse contexts.
