Annona amazonica R.E. Fries (synonyms Annona amazonica var. lancifolia R.E. Fries), popularly known in Brazil as “envireira”, is a tropical tree belonging to the Annonaceae family and is traditionally used as a food source. In this work, the in vitro and in vivo anti-liver cancer effects of essential oil (EO) from A. amazonica leaves were investigated for the first time. The chemical composition of the EO was evaluated via GC–MS and GC–FID. The alamar blue assay was used to evaluate the cytotoxicity of EOs against different cancerous and noncancerous cell lines. Cell cycle analyses, YO-PRO-1/PI staining, and rhodamine 123 staining were performed via flow cytometry in HepG2 cells treated with EO. The in vivo antitumor activity of EO was evaluated in NSG mice that were xenografted with HepG2 cells and treated with EO at a dose of 60 mg/kg. The major constituents (>5%) of the EO were (E)-caryophyllene (32.01%), 1,8-cineole (13.93%), α-copaene (7.77%), α-humulene (7.15%), and α-pinene (5.13%). EO increased apoptosis and proportionally decreased the number of viable HepG2 cells. The induction of DNA fragmentation and cell shrinkage together with a significant reduction in the ΔΨm in EO-treated HepG2 cells confirmed that EO can induce apoptosis. A significant 39.2% inhibition of tumor growth in vivo was detected in EO-treated animals. These data indicate the anti-liver cancer potential of EO from A. amazonica leaves. Full article

Background: Nemaline myopathy is a rare congenital neuromuscular disease associated with progressive weakness and frequent respiratory complications. In emergency situations, families often serve as the first and only responders. The aim of this study is to explore how parents in Spain care for children with nemaline myopathy during emergency situations, focusing on the clinical responses performed at home and the organizational challenges encountered when interacting with healthcare systems. Methods: A qualitative phenomenological study was conducted with 17 parents from 10 families belonging to the Asociación Yo Nemalínica. Semi-structured interviews were performed via video calls, transcribed verbatim, and analyzed using Giorgi’s descriptive method and ATLAS.ti software (version 24). Methodological rigor was ensured through triangulation, reflexivity, and member validation. Results: Four themes were identified. First, families were described as acting under extreme pressure and in isolation during acute home emergencies, often providing cardiopulmonary resuscitation and respiratory support without professional backup. Second, families managed ambiguous signs of deterioration using clinical judgment and home monitoring tools, often preventing fatal outcomes. Third, parents frequently assumed guiding roles in emergency departments due to a lack of clinician familiarity with the disease, leading to delays or errors. Finally, the transition to the Pediatric Intensive Care Unit was marked by emotional distress and rapid decision-making, with families often participating in critical choices about invasive procedures. These findings underscore the complex, multidisciplinary nature of caregiving. Conclusions: Parents play an active clinical role during emergencies and episodes of deterioration. Their lived experience should be formally integrated into emergency protocols and the continuity of care strategies to improve safety and outcomes. Full article

Impact of a 10-Week Strength Training Program on Physical Performance and Match External Load in Young Elite Female Soccer Players

Background: Soccer is a physically demanding sport characterized by frequent high-intensity efforts, which are particularly relevant in women’s competitions. Improving high-speed running and aerobic capacity has been linked to better on-field performance. Strength training has shown promise in enhancing these physical attributes, but its application in young female soccer players remains underexplored. This study aimed to investigate the effects of a 10-week in-season strength training program on physical performance and match running demands in young female soccer players. Methods: Thirty-two U18 Danish female professional soccer players from two comparable teams voluntarily participated in the study. Teams were allocated to either an experimental group, performing twice-weekly strength training (EG, n = 16) or a control group (CG, n = 16). Vertical jump performance and Yo-Yo IR2 performance as an estimation for maximal oxygen uptake (VO2max) were assessed both pre and post intervention. Additionally, players’ match external demands (i.e., total distance, distance covered at speeds above 23 km·h−1, and maximum velocity achieved) were monitored using Global Positioning System devices during four matches before and after the intervention. Results: Significant within-group differences were observed across all variables for the EG (p = 0.001; ES = 1.08 to 1.45, large), without differences in the CG (p > 0.01). Between-group analysis indicated significant differences favoring the EG in all variables (F = 27.40 to 47.17; p = 0.001). Conclusions: The application of a 10-week strength training program led to improvements in physical and match running performance among young female soccer players, underscoring the importance of incorporating strength training programs into female soccer periodization to enhance performance. Background: Sports performance indicators are mainly based on male athletes, highlighting the importance of portraying the female reality, particularly in rural contexts. This study aims to characterize sports performance indicators (body composition and physical fitness) of young Portuguese female athletes. Methods: A cross-sectional study was conducted with 124 girls (13.66 ± 1.93 years) participating in federated team sports in a rural region of Portugal. Body composition was assessed using bioelectrical impedance, and physical fitness was evaluated through vertical jump tests (countermovement jump and squat jump), sprint (20 m), agility (T-test), handgrip strength, and cardiovascular endurance (Yo-Yo IR1). Results: Volleyball players are taller; football and basketball players are heavier; football and volleyball players have more fat-free mass than handball players (p < 0.05). Body mass index and % body fat did not differ between sports (p > 0.05). Volleyball players performed better in the countermovement jump (F = 4.146, p = 0.008) and squat jump (F = 7.686, p < 0.001) when compared to basketball, football, and handball players. No differences were observed in the speed or cardiorespiratory endurance tests (p > 0.05). Conclusions: The results revealed that, despite some specific differences between sports, most physical fitness indicators did not differ significantly between sports after controlling for age, menarche, and training experience. These findings suggest that shared contextual limitations in rural regions may take precedence over sport-specific adaptations in the early stages of sports participation. Full article

Annona neoinsignis H. Rainer (Annonaceae) is a tree native to the Amazon rainforest. Its fruits are also suitable for human consumption in their natural state or are processed to make desserts. In this work, we characterized the chemical composition of the essential oil (EO) from the leaves of A. neoinsignis and evaluated its anti-liver-cancer potential via in vitro and in vivo approaches. Chemical composition analysis revealed β-elemene, (E)-caryophyllene, germacrene D, and germacrene B as the main constituents. The EO had IC50 values ranging from 12.28 to 37.50 μg/mL for B16-F10 cells and MCF-7 cells, whereas an IC50 value of >50 μg/mL was found for noncancerous MRC-5 cells. DNA fragmentation, YO-PRO-1 staining, and loss of mitochondrial transmembrane potential were detected in EO-treated HepG2 cells, indicating the induction of apoptosis. Significant in vivo growth inhibition of 53.7% was observed in mice bearing HepG2 cell xenografts treated with EO at a dosage of 40 mg/kg. These data suggest that EO from A. neoinsignis leaves is a drug source for liver cancer. Full article

Background/Objectives: Inappropriate use of emergency department (ED) services is widely acknowledged to have a negative impact on health systems as a whole. A minor portion of Frequent User (FU) patients are often responsible for the disproportionate use of ED services. Methods: A retrospective population study was conducted on the attendances of pediatric EDs from 1 January 2022 to 31 December 2022 at the Roma 1 Local Health Authority in Rome, a territory served by more than 13 EDs. Nested logistic regression analysis was used to investigate patient characteristics in predicting inappropriate use of EDs. Results: In 2022, 35,691 pediatric ED attendances were recorded, with 24,824 patients distributed among 904 PCP/GPs in the six districts. A total of 71.8% of patients had only one attendance in 2022. A total of 3.5% of the patients were FUs, who were responsible for more than 10% of the attendances. However, most of the attendances were not appropriate. FUs, younger age (<2 yo), and more severe clinical presentations were more likely to be associated with appropriate ED attendance. PCPs/GPs and districts do not have a role in determining a higher number of ED attendances. The single patient’s characteristics have a greater influence on this phenomenon. Conclusions: Frequent use of the ED is not associated with inappropriate use among children, mainly due to the characteristics and needs of specific patients. However, parents’ education for a more rational use of health system resources and the provision of local solutions to children’s health needs may allow for a more appropriate use of health service resources. Candidiasis is a major fungal infection worldwide, with invasive forms linked to high morbidity and mortality. The emergence of azole resistance in Candida parapsilosis causing candidemia led us to examine the epidemiology and antifungal susceptibility of Candida species at the University Hospital of Liège between January 2017 and December 2023. A total of 916 isolates from blood or sterile body fluids, tissues, and abscesses were analyzed. Species identification was performed using MALDI-TOF MS and antifungal susceptibility testing via Sensititre YO10 AST was interpreted according to the CLSI guidelines. Candida albicans remained the predominant species (56%), followed by Nakaseomyces glabratus (19%), Candida parapsilosis (8%), and Candida tropicalis (7%). No significant shift toward non-albicans Candida species (NAC) was observed even during the COVID-19 pandemic, supporting the use of narrow-spectrum empirical therapy in selected patients. Fluconazole susceptibility was high in C. albicans (98.8%), whereas N. glabratus and C. tropicalis showed high resistance rates with 10.1% and 16.9%, respectively. C. parapsilosis showed stable fluconazole susceptibility across the study period. Echinocandins demonstrated excellent activity (95.6–100%), and amphotericin B was effective against nearly all isolates. This seven-year surveillance at the University Hospital of Liège confirms that while C. albicans remains the predominant and highly susceptible species, rising azole resistance in non-albicans Candida—particularly N. glabratus and C. tropicalis—highlights the critical need for ongoing local epidemiological monitoring to guide effective and targeted antifungal therapy. Background/Objectives: Non-saccular aneurysms are a rare subtype of intracranial aneurysms with complex morphologies. Although treatment strategies for ruptured non-saccular and saccular aneurysms differ significantly, large-scale comparisons of the outcomes between the two types remain limited. We aimed to compare the clinical characteristics, procedure-related complications, and functional outcomes between patients with subarachnoid hemorrhage (SAH) caused by non-saccular or saccular aneurysms. Methods: We retrospectively analyzed 1176 consecutive patients with aneurysmal SAH from a population-based stroke registry in Kochi Prefecture, Japan. Aneurysms were classified as saccular or non-saccular based on the morphology, and clinical variables, radiological features, and treatment modalities were compared. Additionally, 840 patients who underwent intervention for their aneurysms within 3 days of onset were further investigated to evaluate the impact of the non-saccular aneurysm shape on poor functional outcomes, defined as a modified Rankin Scale score ≥ 3 at discharge. Results: Non-saccular aneurysms were more common in younger patients and located in the posterior circulation. Procedure-related ischemic complications were more likely to occur in non-saccular aneurysms than in saccular aneurysms (odds ratio [OR]: 2.57, 95% confidence interval [CI]: 1.56–4.97, p < 0.001). In a multivariable logistic regression analysis, a non-saccular morphology was an independent risk factor of poor outcomes (OR: 2.92, 95% CI: 1.34–6.32, p = 0.007) after adjustment for potential confounders. Interaction and subgroup analyses revealed that the negative effects of non-saccular aneurysms on functional outcomes were more prominent in younger patients aged ≤ 60 years. Conclusions: Non-saccular aneurysms are independently associated with ischemic complications and poor outcomes after SAH, particularly in younger patients. Full article

Na-layered Li-containing Mn-based cathodes (NaxLiyMn1-yO2, NLMOs) with additional Na+ storage ability resulting from the anionic redox reaction (ARR) hold great promise for sodium-ion batteries (NIBs). However, practical applications of NLMOs encounter challenges, such as migration of transition metal Mn, loss of lattice oxygen, and voltage decay during cycling. Here, we show that Cu plays an important role in enhancing the ARR via the reductive coupling mechanism (RCM). Results shows that a Cu2+/Fe3+ modified NLMO sample delivers a Na+ storage capacity as high as 174 mA h g−1 at 0.2C, higher than that of a Zn2+/Fe3+ modified NLMO sample (130 mA h g−1) and NLMO (154 mA h g−1). Both in situ and ex situ characterization results indicate that the obvious improvement in the electrochemical performance of the Cu2+/Fe3+ modified NLMO is due to the additional overlaps between the Cu 3d and O 2p orbitals, which is beneficial for the RCM. As a result, the ARR is enhanced so as to increase the Na+ storage capacity. Full article

Background: Beetroot juice, the most commonly used route of dietary nitrate supplementation, is theorized to enhance physical performance. However, its effects on different aspects of physical performance, different populations, and optimal supplementation strategies remain controversial. The purpose of this study was to investigate the effects of beetroot juice (nitrate-rich) on physical performance, to compare its effects between professional athletes and non-athletes (healthy individuals), and to determine the optimal supplementation strategy. Methods: A systematic search was conducted on the Web of Science, Embase, PubMed, Cochrane Database, SPORTDiscus, Scopus, and CINAHL databases. Reviewers conducted study screening and selection, data extraction, and assessment of methodological quality using the AMSTAR 2 tool. Fifteen meta-analyses were included in this Umbrella Review. A narrative and quantitative synthesis was performed. Standardized mean differences (SMD) and 95% confidence intervals (CI) were reported. Results: (1) Beetroot juice significantly improved muscle strength (SMD = 0.08, p < 0.001), but the effect size was negligible. (2) Aerobic Endurance: Beetroot juice significantly improved VO2max (SMD = 0.16, p = 0.033) in healthy adults; however, the effect size was negligible. (3) Lactate Tolerance: beetroot juice significantly improved TTE (SMD = 0.25, p = 0.034) and YO-YOIR1 (SMD = 0.27, p = 0.049) performance in healthy adults, but the effect size was small. (4) Subgroup analyses revealed significant population differences: professional athletes showed significant muscular strength benefits (SMD = 0.27, p = 0.007), whereas non-athletes had more pronounced aerobic endurance improvements (SMD = 0.26, p < 0.001), but the effect size was small. (5) Nitrate supplementation timing and dose–response analysis revealed that nitrate supplementation, whether administered 2–3 h before exercise or over a prolonged period (≥3 days), produces a significant enhancement in physical performance (p < 0.01). Notably, acute improvement only has a small effect size (SMD = 0.20), while the impact of chronic supplementation is essentially negligible (SMD = 0.13). A dosage of 8.3–16.4 mmol NO3 (515–1017 mg) showed a significant improvement (SMD = 0.14, p = 0.029), although the effect size was negligible. Conclusions: Acute (2–3 h pre-exercise) and chronic (≥3 days) supplementation with beetroot juice to achieve nitrate levels of 8.3–16.4 mmol (515–1017 mg/d) are recommended to enhance physical performance. Beetroot juice shows population-specific effects: proper beetroot juice supplementation improves muscular strength in professional athletes and aerobic endurance in non-athletes. Moreover, appropriate supplementation of beetroot juice can improve the lactate tolerance in healthy adults. Full article

Background: Osteosarcoma (OS) is the most common primary malignant bone tumor. Doxorubicin (DOX) is extensively used in OS chemotherapy, yet improving patient outcomes remains challenging. This study investigated the effect of free DOX combined with inductive moderate hyperthermia (IMH) on Saos-2 human OS cells. Methods: Cell viability was assessed by trypan blue exclusion. Flow cytometry analyzed apoptosis, necrosis, and reactive oxygen species (ROS) in cells exposed to control (no treatment), IMH (42 MHz frequency, 500 μT magnetic field induction, 564 V/m electric field strength, 15 W output power, and 30 min duration) alone, DOX (0.06 μg/mL) alone, or DOX combined with IMH. The expression of p14ARF tumor suppressor and epidermal growth factor receptor (EGFR) was evaluated by immunocytochemistry. Spatial autocorrelation analysis quantified the heterogeneity of p14ARF and EGFR distributions in acquired images. Results: The half maximal inhibitory concentration (IC50) of DOX in Saos-2 cells had minimal variation between 48 h (0.060 ± 0.01 μg/mL) and 72 h (0.055 ± 0.003 μg/mL). DOX + IMH resulted in a 15% increase in early apoptosis and a 20% elevation in ROS levels compared with DOX alone. Immunocytochemical analysis revealed a 37% increase in p14ARF and a 32% reduction in EGFR expression following combined treatment in comparison to DOX alone. Image analysis showed that DOX + IMH treatment caused the highest Moran’s index values for p14ARF and EGFR, reflecting less heterogeneous spatial distributions (p < 0.05). Conclusions: IMH enhanced DOX-induced cytotoxicity in Saos-2 cells by initiating ROS-mediated apoptosis and reducing heterogeneity of cellular responses. Full article

XGBoost-Based Modeling of Electrocaloric Property: A Bayesian Optimization in BCZT Electroceramics

Electrocaloric materials, which exhibit adiabatic temperature change under an applied electric field, are promising for solid-state cooling technologies. In this study, the electrocaloric response of lead-free BaxCa1−xZryTi1−yO3 (BCZT) ceramics was modeled to investigate the effects of composition, processing, and measurement conditions on performance. A high-accuracy XGBoost regression model (R2 = 0.99, MAE = 0.02 °C) was developed using a dataset of 2188 literature-derived data points to predict and design the electrocaloric response of BCZT ceramics. The feature space incorporated compositional ratios, processing parameters, measurement settings, and atomic-level Magpie descriptors, along with Curie temperature to account for phase-transition behavior. Feature importance analysis revealed that electric field, measurement temperature, and proximity to the Curie point are the most critical factors influencing ΔTEC. Bayesian optimization was applied to navigate the design space and identify performance maxima under unconstrained and realistic constraints, offering valuable insights into the nonlinear interactions governing electrocaloric performance. Under room temperature and moderate-field conditions (24 °C, 40 kV/cm), the optimized ΔTEC achieved a value of 1.03 °C for Ba0.85Ca0.15Zr0.40Ti0.60, to be processed at 1090 °C for 3 h during calcination, 1300 °C for 2 h during sintering. By integrating experimental insight with machine learning and optimization, this study offers a refined, interpretable framework for accelerating the design of high-performance electrocaloric ceramics while reducing the experimental workload. Background: The risk of recurrence of early-stage cervical cancer (CC) is associated with prognostic factors such as tumor size, lymphovascular space invasion (LVSI), and deep stromal invasion (DSI). However, the adjuvant pelvic radiotherapy (RT) following surgery to reduce the risk of recurrence in “intermediate risk” remains controversial. This study aims to evaluate the role of adjuvant RT in the recurrence and identify prognostic factors. Methods: A systematic search of PubMed, Embase, and Cochrane databases was performed to identify studies comparing adjuvant RT versus no adjuvant treatment in early-stage CC patients with intermediate-risk factors defined by GOG-92 criteria. Outcomes were recurrence, local recurrence, death, 5-year overall survival (5y-OS), and 5-year disease-free survival (5y-DFS). Tumor size ≥ 4 cm, LVSI, and DSI were also evaluated as prognostic factors for recurrence. Statistical analysis was performed using Review Manager 7.2.0. Heterogeneity was assessed with I2 statistics. Results: A total of 1504 patients from nine studies were included; only one study was a randomized controlled trial, while the others were retrospective cohorts. Adjuvant RT was used to treat 781 patients (52%). Median follow-up ranged from 48 to 120 months. Recurrence (OR 0.75; 95% CI 0.38–1.46; p = 0.39), local recurrence (OR 0.73; 95% CI 0.44–1.20; p = 0.22), death (OR 0.97; 95% CI 0.52–1.80; p = 0.91), 5y-OS (OR 1.22; 95% CI 0.36–4.18; p = 0.75), and 5y-DFS (OR 0.78; 95% CI 0.42–1.43 p = 0.42) revealed no statistically significant differences between adjuvant RT and observation groups. TS ≥ 4 cm was an independent prognostic risk factor for recurrence (HR 1.83; 95% CI 1.12–2.97; p = 0.02). Conclusions: Our findings suggest that adjuvant RT does not reduce recurrence risk in early-stage cervical cancer. Consider TS ≥ 4 cm as a significant prognostic factor for recurrence. Adjuvant RT in intermediate-risk patients should be considered with caution due the lack of significant improvement in recurrence until the CERVANTES and GOG-0263 trial results become available. This study investigated changes in microbial communities and functional components during the fermentation of traditional Korean grain vinegars collected from various regions as well as the correlations among these components. Microbial community analysis revealed that Lactobacillus acetotolerans was the dominant microorganism, while Acetobacter pasteurianus numbers gradually increased during fermentation, playing a key role in acetic acid production. L. acetotolerans, known to thrive in acidic environments, contributed to increasing the acidity of the vinegar and enhanced its preservative properties. The rise in the levels of organic acids, particularly acetic acid, was influenced by the activity of these microorganisms. Additionally, the production of free amino acids, such as alanine, was influenced by interactions between the fermentation medium and microbial communities, significantly contributing to the vinegar’s sweetness. Volatile flavor compounds exhibited considerable diversity due to changes in microbial communities driven by raw-material differences. In particular, five-grain vinegar (YO) tended to generate more complex and intense flavor compounds, with uniformly elevated levels of aldehydes, acids, and ketones. These findings suggest that raw-material selection and fermentation conditions significantly influence the flavor and functional properties of grain vinegars, providing valuable foundational data for improving vinegar production processes to enhance flavor and functionality. Accurate identification of surface cracks is of great significance for the site selection of transmission towers, as it directly affects the safety and stability of power grid construction. Traditional manual inspection methods are labor-intensive and inefficient, making them inadequate for large-scale and high-precision applications. Consequently, intelligent crack recognition technologies are receiving increasing attention and adoption. This study proposes a novel surface crack identification method that integrates multiple neural networks, aiming to overcome the limitations of traditional crack identification approaches, such as low accuracy and poor generalization. The proposed framework incorporates a convolutional neural network (CNN)-based data filtering module and a crack segmentation module combining UNet and YOLOv8 architectures. Together, these components form a robust and accurate end-to-end crack identification system, which is further applied to the evaluation of rock fragmentation. To verify the effectiveness and accuracy of the proposed method, experiments were conducted on a rock crack dataset and compared against several existing approaches. The results demonstrate that the proposed method achieves superior performance in crack detection accuracy. Moreover, tests on various scenario datasets also yielded promising results, indicating the strong generalization ability and adaptability of the method. Full article
