Xylella fastidiosa (XF) is a Gram-negative bacterium responsible for severe plant diseases affecting a wide range of host plants, some of them important crops. Since 2017, only the pauca subspecies (ST80) have been identified in Ibiza. XF is naturally transmitted by xylem sap-feeding insects; among them, only Philaenus spumarius (PS) and Neophilaenus campestris Fallén (NC) have been reported as potential insect vectors for XF in Ibiza. This study aims to investigate the dissemination of XF and to propose effective control strategies. The crops and the surrounding vegetation were sampled for potential vectors. DNA from insects was extracted and amplified by three qPCR methods, allowing subspecies identification. The results confirmed the continuous presence of adults of PS and NC in Ibiza and Formentera from May to December with important populations. During the summer period, PS captures predominantly took place within the surrounding woody vegetation adjacent to the plots. The main host plant for PS was Pinus halepensis Miller in Ibiza and Juniperus phoenicea subsp. turbinata (Guss.) Nyman in Formentera. In Ibiza, off the total PS captures, 4.47% tested positive for XF. These results confirm that PS is the main vector of XF on these islands, both in terms of captures and the percentage of positive insects for XF. In Formentera, despite the presence of potential vectors and the proximity and contact with Ibiza, no XF-positive insects were found, confirming the absence of the bacterium on the island. Olive Quick Decline Syndrome (OQDS) associated with Xylella fastidiosa subsp. pauca is one of the most destructive diseases of olive trees in the Salento Peninsula (Southern Italy), particularly on the cultivars Cellina di Nardò and Ogliarola Salentina. This study proposes the NuovOlivo protocol as a management strategy to permit coexistence between X. fastidiosa subsp. pauca and olive drupes and extra-virgin oil production. Thirty-two private olive orchards affected by OQDS and cultivated following the standard agronomic techniques in use in the area were surveyed during the 2019–2023 olive-growing seasons. Tested cultivars included Cellina di Nardò, Ogliarola Salentina, Coratina, Ascolana Tenera, Nociara, Leccino, and Bella di Cerignola. At the beginning of the protocol application, the susceptible plants showed OQDS symptom severity of 40–80% and did not produce olives or oil, while the resistant(?)/tolerant cultivars exhibited a 2–8% leaf scorch and a drupe production less than 1–2 kg/plant. After the removal of dry branches in January–February, plants were sprayed two times per year (preferably in March and October) with NuovOlivo®, a mixture of aqueous botanical extracts esterified in the presence of sodium hydroxide with vegetable oils and activated at the time of use with sodium bicarbonate. In all the orchards, a slow-release fertilizer was distributed, and weeds were controlled by mowing or chopping. Upon eventual appearance, the dry twigs were removed. Treated olive trees produced new vegetation, rebuilt their foliage, reduced OQDS symptoms, and turned out cluster inflorescence and drupes. The drupes yield was 6.67–51.36 kg per plant, with an average of 13.19% in extra-virgin olive oil (free acidity 0.01–0.2%). Plants used as controls showed OQDS symptoms and were unproductive, and newly formed shoots were desiccated. The proposed protocol promotes, supports, and restores new vegetation, flowers, fruits, and oil production of the treated olive plants affected by OQDS without losing susceptible olive plants. The Apulian landscape and economy, based on olive presence and production, could be also safeguarded. Full article

The meadow spittlebug Philaenus spumarius (Hemiptera: Aphrophoridae) is distributed in several habitats worldwide and has been recently recognized as the main vector of Xylella fastidiosa subsp. pauca. This bacterium has been associated with olive quick decline syndrome (OQDS) in the Salento Peninsula (Italy) and is responsible for extensive desiccation and die-off of olive trees. Current OQDS management strategies include the control of P. spumarius populations, mainly through the removal of weed hosts and insecticide treatments. In addition to the mandatory phytosanitary measures, the demand for new strategies compatible with integrated pest management is increasing. In this study, laboratory biological assays were performed to assess the potential toxic effect of vegetal formulations against P. spumarius adults. Two formulations were tested at different concentrations: Form A, an emulsion of 10% hot pepper-infused oil (Capsicum annuum subspecies Cayenna in olive oil) and Arabic gum in an aqueous solution of extracts of Salvia guaranitica, and Form B, an aqueous solution of extracts of Taxus baccata. Both Form A and B showed to be toxic against P. spumarius compared to the water control. The mean percentage of spittlebug mortality obtained with Form A one hour after treatments was dose–dependent; the lethal dose values were 0.13% (LD25), 0.36% (LD50), and 0.85% (LD75). At the same time, no significant differences in mortality rate were observed between the 0.75% treatment and the treatments with deltamethrin (about 90%). The mean percentage of spittlebug mortality obtained with Form B ranged from 21% to 53% one hour after treatment, but these values were significantly lower than those obtained with deltamethrin. The effectiveness of Form A on the P. spumarius population was also evaluated in the field. The averages of captures in the three experimental blocks were 1.8/trap for treated and 7.7/trap for untreated plots, and the spittlebug populations significantly decreased after treatments. Based on these results and the literature data, we hypothesize that the effectiveness of Form A is the result of the synergistic effect of all its components. No symptoms of phytotoxicity were recorded on olive trees treated with Form A, and the number of P. spumarius specimens collected on these plants was much lower than on untreated plants. These results suggest the potential use of Form A in the protection of olive trees. This vegetal formulation can thus be considered as a valid alternative to chemical insecticides for the control of the main vector of X. fastidiosa and could be integrated into a sustainable management system for OQDS. This is the first worldwide report of X. fastidiosa (Xf) subsp. pauca on Castanea sativa and the first characterization of Xf infection on this species. Plants located in three sites (in a long-term affected area in Apulia) were monitored for symptoms and bacterial concentrations in spring and summer, while microscopic analyses were performed to evaluate the pathogen distribution in the xylem vessels. All chestnut plants appeared asymptomatic but the Xf subsp. pauca strain “De Donno” was present at low concentrations (1.14 × 104 and 1.56 × 103 cfu mL−1 in April and June) and with a low incidence (respectively, 38% and 30%). The FISH-CLSM (Fluorescent In Situ Hybridization—Confocal Laser Scanning Microscope) analysis showed evident Xf occlusions but in a low percentage (9.2 ± 3.4%); these data can explain the lack of symptoms on the canopy. Furthermore, through a bibliographic analysis it emerged that Philaenus spumarius and Neophilaenus campestris, two Xf vectors present in Europe, are suckling feeding insects on chestnut trees and could be involved in the bacterial transmission to this species. Asymptomatic Xf host species can play a considerable role in new outbreak emergence or in the expansion of existing ones. So, it is essential to identify them to plan more effective monitoring activities. Full article

The olive tree (Olea europaea subsp. europaea var. europaea) represents the cornerstone crop of Apulian agriculture, which is based on the production of oil and table olives. The high genetic variability of the Apulian olive germplasm is at risk of genetic erosion due to social, economic, and climatic changes. Furthermore, since 2013, the spread of the Gram-negative bacterium Xylella fastidiosa subsp. pauca responsible for the olive quick decline syndrome (OQDS) has been threatening olive biodiversity in Apulia, damaging the regional economy and landscape heritage. The aim of this study was to investigate the differential response to X. fastidiosa infection in a collection of 100 autochthonous Apulian olive genotypes, including minor varieties, F1 genotypes, and reference cultivars. They were genotyped using 10 SSR markers and grown for 5 years in an experimental field; then, they were inoculated with the bacterium. Symptom assessments and the quantification of bacterium using a qPCR assay and colony forming units (CFUs) were carried out three and five years after inoculation. The study allowed the identification of nine putatively resistant genotypes that represent a first panel of olive germplasm resources that are useful both for studying the mechanisms of response to the pathogen and as a reserve for replanting in infected areas. Xylella fastidiosa subsp. pauca sequence type 53 was found to be associated with olive trees showing extensive twig and branch dieback and plant death in the Gallipoli area of Salento and the common name of “olive quick decline syndrome” (OQDS) was given to the disease. Repeated interceptions in Europe and Italy of ornamental coffee plants originated from Central America, jointly in phylogenetic analyses of many strains of the pathogen, indicated this origin as the most probable source of its introduction in Salento. Drought events could have been conducive to the initial outbreaks of the disease. Subsequently, the pathogen largely spread over the territory according to a “stratified dispersal” model. The high susceptibility of the local cultivars Ogliarola salentina and Cellina di Nardò, a low soil content of zinc, copper, and manganese, improper pruning, and adverse climatic events could have further contributed to the spread of the pathogen. The polyphagous insect Philaenus spumarius L. is the main vector of the bacterium in the area. The adults were detected X. f. subsp. pauca-positive in early May, and their incidence was higher during spring and early autumn when they efficiently spread the bacterium among the olive trees. Many other host plant species can host the bacterium, and some of them can act as a “reservoir” for the disease spread. The aggressive fungus Neofusicoccum mediterraneum Crous, M.J. Wingf. And A.J.L. Philips, could also be involved in OQDS. A sustainable control strategy for reducing the incidence and severity of X. f. subsp. pauca in the olive groves of Salento that allows the trees to produce is presented and discussed. Resilient trees of Ogliarola salentina and Cellina di Nardò have been observed in the heavily infected areas of Salento. Full article

Xylella fastidiosa subsp. pauca is the causal agent of “olive quick decline syndrome” in Salento (Apulia, Italy). On April 2015, we started interdisciplinary studies to provide a sustainable control strategy for this pathogen that threatens the multi-millennial olive agroecosystem of Salento. Confocal laser scanning microscopy and fluorescence quantification showed that a zinc-copper-citric acid biocomplex—Dentamet®—reached the olive xylem tissue either after the spraying of the canopy or injection into the trunk, demonstrating its effective systemicity. The biocomplex showed in vitro bactericidal activity towards all X. fastidiosa subspecies. A mid-term evaluation of the control strategy performed in some olive groves of Salento indicated that this biocomplex significantly reduced both the symptoms and X. f. subsp. pauca cell concentration within the leaves of the local cultivars Ogliarola salentina and Cellina di Nardò. The treated trees started again to yield. A 1H-NMR metabolomic approach revealed, upon the treatments, a consistent increase in malic acid and γ-aminobutyrate for Ogliarola salentina and Cellina di Nardò trees, respectively. A novel endotherapy technique allowed injection of Dentamet® at low pressure directly into the vascular system of the tree and is currently under study for the promotion of resprouting in severely attacked trees. There are currently more than 700 ha of olive groves in Salento where this strategy is being applied to control X. f. subsp. pauca. These results collectively demonstrate an efficient, simple, low-cost, and environmentally sustainable strategy to control this pathogen in Salento. Full article

Xylella fastidiosa subsp. pauca, strain CoDiRO is the bacterium responsible for the onset of the disease known as the olive quick decline syndrome, which has been causing a phytosanitary and economic emergency in the Apulia region since 2013. To date, three insect species have been identified as pathogenic carriers of X. fastidiosa. With the advancement of the infection front, and the possibility of pathogenic insects being “hitchhiked” over long distances, the monitoring of the vectors of X. fastidiosa in the Italian regions bordering Apulia is an increasingly contingent issue for the rapid containment of the bacterium and the protection of the olive-growing heritage. Accordingly, the present research concerned the capture and recognition of the vector insects of X. fastidiosa in the upper Ionian coasts of Calabria (Italy) to evaluate the possible presence of the bacterium through molecular diagnostic techniques. The sampling allowed us to ascertain the presence of Philaenus spumarius and Neophilaenus campestris and their preferential distribution in olive groves and meadows, whereas all the 563 individuals tested negative for the pathogen. During recent years; Xylella fastidiosa subsp. pauca (Xfp) has spread in Salento causing relevant damage to the olive groves. Measures to contain the spreading of the pathogen include the monitoring of the areas bordering the so-called “infected” zone and the tree eradication in case of positive detection. In order to provide a control strategy aimed to maintain the tree productivity in the infected areas, we further evaluated the in vitro and in planta mid-term effectiveness of a zinc-copper-citric acid biocomplex. The compound showed an in vitro bactericidal activity and inhibited the biofilm formation in representative strains of X. fastidiosa subspecies, including Xfp isolated in Apulia from olive trees. The field mid-term evaluation of the control strategy assessed by quantitative real-time PCR in 41 trees of two olive groves of the “infected” area revealed a low concentration of Xfp over the seasons upon the regular spraying of the biocomplex over 3 or 4 consecutive years. In particular, the bacterial concentration lowered in July and October with respect to March, after six consecutive treatments. The trend was not affected by the cultivar and it was similar either in the Xfp-sensitive cultivars Ogliarola salentina and Cellina di Nardò or in the Xfp-resistant Leccino. Moreover, the scoring of the number of wilted twigs over the seasons confirmed the trend. Recently, a new severe disease has been reported in the Salento area (Apulia region, southern Italy) in the multimillennial olive agro-ecosystem, given the common name “olive quick decline syndrome” (OQDS). Together with Xylella fastidiosa subsp. pauca, some pathogenic fungi such as Phaeoacremonium spp. have been found associated with the disease. The main predisposing factors to the disease seem to be local cultivar susceptibility, depletion of some micronutrients in the soil that could be related to some agronomical practices favoring the depletion of soil fertility, an incorrect pruning cycle, climatic changes that result in increased soil waterlogging, and frost and drought events. The possible synergistic action of microorganisms other than X. f. subsp. pauca cannot be excluded. The features characterizing the areas where OQDS first appeared and subsequently spread, described and discussed here, would point to a rather fragile environment where one or more adverse climatic and/or edaphic factors could have acted together. The intrinsic peculiarities and management of the Salento olive agro-ecosystem could also have played a fundamental role in enhancing the virulence of X. f. subsp. pauca once introduced from abroad. Xylella fastidiosa is a vector-borne bacterium that causes diseases in many plants of economic interest. The bacterium–vector initial interactions involve bacterial membrane-bound adhesins that mediate cell attachment to the foregut of insect vectors. We investigated the role of the afimbrial adhesin XadA2 in the binding and biofilm formation of X. fastidiosa subsp. pauca to vector surfaces in vitro, as well as its potential to disrupt pathogen transmission. We showed that XadA2 has binding affinity for polysaccharides on sharpshooter hindwings, used as a proxy for the interactions between X. fastidiosa and vectors. When in a medium without carbon sources, the bacterium used wing components, likely chitin, as a source of nutrients and formed a biofilm on the wing surface. There was a significant reduction in X. fastidiosa biofilm formation and cell aggregation on vector wings in competition assays with XadA2 or its specific antibody (anti-XadA2). Finally, pathogen acquisition and transmission to plant were significantly reduced when the vectors acquired X. fastidiosa from an artificial diet supplemented with anti-XadA2. These results show that XadA2 is important in mediating bacterial colonization in the insect and that it could be used as a target for blocking X. fastidiosa transmission. Full article

Xylella fastidiosa subsp. pauca is responsible for the “olive quick decline syndrome” (OQDS) in Salento (Apulia). The main epidemiological aspects of the syndrome are related to the pathogen spread and survival in the area, and to the biology of the insect vector. The assessment of the macro and microelements content (i.e., ionome) in soil and leaves could provide basic and useful information. Indeed, knowledge of host ionomic composition and the possibility of its modification could represent a potential tool for the management of diseases caused by X. fastidiosa. Therefore, soil and leaf ionomes of naturally infected, not infected, and zinc–copper–citric acid biocomplex treated trees of different areas of Apulia and the bordering Basilicata regions were compared. We observed that soil and leaf ionomic composition of olive farms growing in the pathogen-free areas north of the Salento Barletta-Andria-Trani BAT (Apulia) and Potenza PZ (Basilicata, Apulia bordering region) provinces is significantly different from that shown by the infected olive groves of the Salento areas (LE, BR, TA provinces). In particular, a higher content of zinc and copper both in soil and leaves was found in the studied northern areas in comparison to the southern areas. This finding could partly explain the absence of OQDS in those areas. In the infected Salento areas, the leaf ionomic profile resulted as being markedly different for the biocomplex treated compared to the untreated trees. A higher zinc content in leaves characterized treated with respect to untreated trees. On the other hand, among the not-infected trees, Xylella-resistant Leccino showed higher manganese content when compared with the higher pathogen sensitive Ogliarola salentina and Cellina di Nardò. According to these results, soil and olive leaf ionome could provide basic information for the epidemiologic study and possible control of X. f. subsp. pauca in Apulia. Full article

Xylella fastidiosa subsp. pauca is a xylem-limited bacterial phytopathogen currently found associated on many hectares with the “olive quick decline syndrome” in the Apulia region (Southern Italy), and the cultivars Ogliarola salentina and Cellina di Nardò result in being particularly sensitive to the disease. In order to find compounds showing the capability of reducing the population cell density of the pathogen within the leaves, we tested, in some olive orchards naturally-infected by the bacterium, a zinc-copper-citric acid biocomplex, namely Dentamet®, by spraying it to the crown, once per month, during spring and summer. The occurrence of the pathogen in the four olive orchards chosen for the trial was molecularly assessed. A 1H NMR metabolomic approach, in conjunction with a multivariate statistical analysis, was applied to investigate the metabolic pattern of both infected and treated adult olive cultivars, Ogliarola salentina and Cellina di Nardò trees, in two sampling periods, performed during the first year of the trial. For both cultivars and sampling periods, the orthogonal partial least squares discriminant analysis (OPLS-DA) gave good models of separation according to the treatment application. In both cultivars, some metabolites such as quinic acid, the aldehydic form of oleoeuropein, ligstroside and phenolic compounds, were consistently found as discriminative for the untreated olive trees in comparison with the Dentamet®-treated trees. Quinic acid, a precursor of lignin, was confirmed as a disease biomarker for the olive trees infected by X. fastidiosa subsp. pauca. When treated with Dentamet®, the two cultivars showed a distinct response. A consistent increase in malic acid was observed for the Ogliarola salentina trees, whereas in the Cellina di Nardò trees the treatments attenuate the metabolic response to the infection. To note that in Cellina di Nardò trees at the first sampling, an increase in γ-aminobutyric acid (GABA) was observed. This study highlights how the infection incited by X. fastidiosa subsp. pauca strongly modifies the overall metabolism of olive trees, and how a zinc-copper-citric acid biocomplex can induce an early re-programming of the metabolic pathways in the infected trees. The disease outbreak of Xylella fastidiosa subsp. pauca strain CoDiRO (Complesso del Disseccamento Rapido dell’Olivo) in Salento (Apulia, South Italy) associated with severe cases of olive quick decline syndrome may represent not just a new disease paradigm, but a challenge for policy formulation and science communication in plant pathology. Plant health management can be achieved by applying a technocratic model, in which objective science is thought to directly inform policy-making, or via decisionistic or inclusive models, in which scientific considerations drive risk assessment. Each could be applied to X. fastidiosa and CoDiRO strain management, thanks to consistent literature related to pathogen/host interactions, hosts, vectors, and diagnostic tools, reviewed here. However, consensus among stakeholders seems to be necessary in order to avoid plant health management failures or gridlocks, due to environmental, economic, and social implications in the X. fastidiosa threat. Here we discuss the role of consensus in building scientific opinion, reporting different approaches of governance after severe disease outbreaks in Europe. These case studies, and the available risk analysis for Xylella strains, should drive policy formulations towards more cooperative networks. Full article
