Dietary advanced glycation end-products (dAGEs) contained in high-sugar/fat and ultra-processed foods of the “Western diet” (WD) pattern predispose to several diseases by altering protein function or increasing oxidative stress and inflammation via RAGE (receptor for advanced glycation end-products). Although elevated endogenous AGEs are associated with loss of muscle mass and functionality (i.e., muscle wasting; MW), the impact of dAGEs on MW has not been elucidated. Here, we show that the most common dAGEs or their precursor, methylglyoxal (MGO), induce C2C12 myotube atrophy as endogenous AGE-derived BSA. ROS production, mitochondrial dysfunction, mitophagy, ubiquitin–proteasome activation, and inhibition of myogenic potential are common atrophying mechanisms used by MGO and AGE-BSA. Although of different origins, ROS are mainly responsible for AGE-induced myotube atrophy. However, while AGE-BSA activates the RAGE-myogenin axis, reduces anabolic mTOR, and causes mitochondrial damage, MGO induces glycolytic stress and STAT3 activation without affecting RAGE expression. Among thirty selected natural compounds, Vaccinium macrocarpon (VM), Camellia sinensis, and chlorophyll showed a surprising ability in counteracting in vitro AGE formation. However, only the standardized VM, containing anti-glycative metabolites as revealed by UHPLC-HRMS analysis, abrogates AGE-induced myotube atrophy. Collectively, our data suggest that WD-linked dAGE consumption predisposes to MW, which might be restricted by VM food supplements. Full article

Large-fruited cranberry (Vaccinium macrocarpon Aiton) is a species known for its highly valued fruit and is typically propagated vegetatively through the rooting of stem cuttings. Studies on the rooting of stem cuttings of large-fruited cranberry have shown that the morphological traits of the root system are a key indicator of the effectiveness of this process. To support rooting, gel coatings based on polysaccharides and containing auxins, especially the indole-3-butyric acid (IBA) W4 variant, were developed and applied. These significantly influenced root length (increase of 44.6% compared to control W0), surface area (increase of 32.4% compared to W0), volume (increase of 26.7% compared to W0), and average thickness, which translated into better nutrient uptake and a higher degree of plant nourishment. The W4 coating, combining mineral components, polysaccharides, and IBA, reduced transpiration and maintained moisture, promoting effective rooting. The associated metabolic changes were confirmed by analyses of oxidative stress markers and chlorophyll fluorescence. The study demonstrated that enhanced root system development was closely linked with the increased accumulation of macro- and micronutrients in the aerial parts of the plants, directly contributing to improved growth and potential yield. These findings highlight that effective rooting—achieved through the targeted metabolic stabilisation of the rooting environment—is essential for the successful vegetative propagation of large-fruited cranberry. The N fertilization to reach high cranberry (Vaccinium macrocarpon) yields resulted in high proportions of soft berries. Our objective was to define the mineral nutrient balance of cranberry to reach a high yield of firm berries. The database comprised 393 observations on cv. ‘Stevens’. Berries were analyzed for total S, N, P, K, Ca, Mg, B, Cu, Zn, Mn, Fe, Al, and Si. Random Forest and XGBoost machine learning models were run to predict yield and firmness classes using raw concentrations, centered log ratios (clr) accounting for nutrient interactions, and weighted log ratios (wlr) that also considered the importance of each dual interaction. The wlr returned the most accurate models. The wlr standards elaborated from the high-yielding and nutritionally balanced subpopulation most often differed between the high-yield class and the high-firmness class. The wlr Cu level was significantly (p ≤ 0.01) too high to reach the high-yielding class in the nutritionally imbalanced subpopulation. There was excessive Al and shortage of Si and Mg to reach high berry firmness in the nutritionally imbalanced subpopulation (p ≤ 0.01), indicating the large influence of soil genesis on berry firmness. Despite statistical evidence, cranberry response to Al and Si corrective measures should be tested to elaborate site-specific recommendations based on soil and tissue tests. Full article

The increasing resistance of microorganisms to currently used antimicrobials requires the urgent development of new effective treatments. Plant-based natural products can be an alternative solution. The aerial plant parts of the cranberry (Vaccinium macrocarpon) present a potential new source of antimicrobial secondary metabolites. Volatile essential oils were extracted from Stevens, Early Black, and Mullica Queen variety plants by steam distillation (SD) and the Clevenger method (CM), and their profiles were characterized by GC-MS. The extracts and two identified constituents, cinnamaldehyde and terpineol, were screened by the disc diffusion assay against Gram-positive B. cereus ATCC 11778 and S. aureus ATCC 25923 and Gram-negative bacteria E. coli ATCC 25922, P. aeruginosa ATCC 27853, and C. albicans ATCC 14053. Radical scavenging antioxidant activity was also determined using the DPPH assay. The CM extracts were rich in fatty acids, sesquiterpenes, and diterpenes, whereas the SD extracts contained more aldehydes, monoterpenes, and phenylpropanoids. All volatile extracts showed promising antioxidant activity; leaf extract activity was significantly higher than the vine (p < 0.05). The CM leaf and vine extracts exhibited antimicrobial activity against B. cereus, S. aureus, E. coli, and C. albicans compared to the SD, and the leaf extracts were more effective than the vine extracts. Individual constituents of leaf and vine extracts, cinnamaldehyde and α-terpineol, also showed antimicrobial activity against these organisms. The active constituents of the CM extracts are yet to be identified. A multivariate analysis revealed a particular pattern of inhibition of the tested organisms. Based on our results, cranberry volatile extracts have potential for future valorization as antibacterials, antifungals, and antioxidants. Full article

Alternative feed additives are being investigated due to the restriction of antibiotics use to decrease antimicrobial resistance (AMR) in food-producing animals. This study investigated the effects of dietary American cranberry (Vaccinium macrocarpon) and wild blueberry (V. angustifolium) pomaces on the cecal microbiota and resistome profiles as well as the short-chain fatty acid levels. Male broiler chickens Cobb500 were fed a basal diet with either 55 ppm bacitracin methylene disalicylate (BMD); 0.5% (CRP0.5) and 1% (CRP1) cranberry pomace; and 0.5% (LBP0.5) and 1% (LBP1) lowbush blueberry pomace with or without a multienzyme mixture (ENZ). The results showed that at 21 days of age, the total coliform counts decreased in the CRP0.5-fed birds compared to BMD (p < 0.05). The use of pomace significantly increased the abundance of Lactobacillus and Bacteroides regardless of ENZ, while CRP decreased the Proteobacteria phylum abundance. In-feed ENZ tended to increase the relative abundance of genes conferring aminoglycoside resistance. Treatment with CRP0.5 decreased the abundance of cepA genes encoding for macrolide (MACROLIDE) and lincomycin (InuD) resistance while increasing those for tetracycline (tetO and tetX) resistance. These results showed, for the first time, the potential of the studied enzymes in influencing berry pomace’s effects on antimicrobial resistance gene profiles in broilers. Full article

Background/Objectives: FimH adhesin, located at the tips of type 1 pili in Escherichia coli (E. coli), plays a crucial role in bacterial adhesion to the surface urothelial cells—a key step in the pathogenesis of urinary tract infections (UTIs). Given the rising concern over antimicrobial resistance (AMR), and considering that E. coli is one of the pathogens with the largest AMR burdens on a global scale, alternative strategies targeting bacterial adhesion are gaining increasing attention. Products that contain D-mannose and cranberry-derived phenolic compounds have shown promise in preventing E. coli colonization and infection. The aim of this study was to investigate the antiadhesive effects of cranberry-related phenolic compounds on FimH-mediated E. coli adhesion using a cellular hemagglutination inhibition assay, as well as to assess the synergistic effects of mannose and phenolic compounds on biofilm formation. Methods: A range of phenolic acids (benzoic, chlorogenic, hippuric, p-coumaric, ferulic and caffeic), resveratrol, (+)-catechin and procyanidin A, as well as a Vaccinium macrocarpon extract, were evaluated for their ability to inhibit FimH-mediated adhesion. A binocular microscope was used to observe agglutination, and we also evaluated the biofilm inhibition potential of the phenolic compounds in the presence of D-mannose. Results: Our results demonstrated that these compounds significantly reduced hemagglutination, with benzoic acid, chlorogenic acid, caffeic acid and resveratrol exhibiting strong inhibitory effects at concentrations as low as 0.25 mM. Furthermore, the addition of 1 mM solutions of these phenolic compounds to D-mannose resulted in a twofold reduction in the inhibition titer, suggesting synergistic interactions. In addition to their antiadhesive properties, the tested phenolic compounds contributed slightly to the inhibition of FimH-mediated biofilm formation, further supporting their potential roles in UTI prevention. Conclusions: These findings highlight the potential of cranberry-derived phenolics as natural antiadhesive agents against E. coli and warrant further investigation into their mechanisms of action and possible applications in infection control. Cranberry Oil: A Potent Natural Intimate Care Ingredient Displaying Antioxidant and Anti-Inflammatory Effects and Promoting Beneficial Vaginal Lactobacillus

Cranberry oil is known for nutritional benefits, and this work is aimed at studying its soothing properties and potential as an intimate care ingredient. The antioxidant, anti-inflammatory, and anti-irritation properties of cranberry oil were evaluated on epithelial cells and tissues, including the vaginal epithelium. The impact of the oil on vaginal microbiota was assessed in vitro. Cranberry oil reduced oxidative stress in keratinocytes (ROS −43%) and lowered inflammation by lessening the release of cytokines IL-8 (−33%) and TNF-α (−32%). Irritation induced by sodium dodecyl sulfate (SDS) in skin explants was lowered by 24%. Cranberry oil and fruit extract acted synergistically on inflammation, decreasing TNF-α release by 75% (vs. −34% and −16%, respectively). Cranberry oil reduced inflammation on EpiVaginal™ tissue, decreasing IL-6 by 36%. The minimum inhibitory concentration (MIC) of cranberry oil on the pathogenic vaginal microorganisms C. albicans and G. vaginalis was 0.5% and 0.1%, respectively. The oil promoted the growth of commensal L. jensenii (×79 at 0.1%) and favored a high proportion of lactic acid bacteria when co-cultured with C. albicans. Cranberry oil has antioxidant, anti-inflammatory, and soothing properties on skin. Anti-inflammatory activity was confirmed on vaginal epithelium, and initial in vitro evidence indicates that the oil can balance vaginal flora to prevent dysbiosis. Full article

Timing insecticide applications with insect emergence is critical for the management of cranberry pests like Sparganothis fruitworm (Sparganothis sulfureana, Lepidoptera: Tortricidae). The annual peak flight of S. sulfureana has previously been predicted using a degree-day model with a biofix date of 1 March; however, this biofix is not suitable for regions where winter and spring temperatures are warmer and flooding of cranberry beds is relied upon, which inhibits S. sulfureana development. In this study, we present two new degree-day models for predicting S. sulfureana peak flight based on six years of trapping data from New Jersey (USA): one with a biofix of 15 April, a date when drainage of cranberry beds occurs on average, and another using individual bed drainage dates. These models project peak flights at 525.5 and 521.0 degree-days using 15 April and water draw date as biofixes, respectively. These models can be used interchangeably, with both biofixes being suitable for regional grower guidance. Furthermore, differences in S. sulfureana peak flight were observed across four cranberry varieties; however, the effect of variety was influenced by year (significant variety-by-year interaction). This year-to-year variation in peak flight was strongly associated with spring (April–May) temperatures. Using these models, we project that with climate change, the peak flight of S. sulfureana in New Jersey cranberry beds may occur up to a week earlier by 2050. The use of a region-specific biofix and variety-specific models will help to better refine degree-day models for S. sulfureana, allowing for improved timing of management strategies against this pest. Anthocyanins are bioactive compounds responsible for various physiological processes in plants and provide characteristic colors to fruits and flowers. Their biosynthetic pathway is well understood; however, the enzymatic degradation mechanism is less explored. Anthocyanase (β-glucosidase (BGL)), peroxidase (POD), and polyphenol oxidase (PPO) are enzymes involved in degrading anthocyanins in plants such as petunias, eggplants, and Sicilian oranges. The aim of this work was to investigate the physicochemical interactions between these enzymes and the identified anthocyanins (via UPLC-MS/MS) in cranberry (Vaccinium macrocarpon) through molecular docking to identify the residues likely involved in anthocyanin degradation. Three-dimensional models were constructed using the AlphaFold2 server based on consensus sequences specific to each enzyme. The models with the highest confidence scores (pLDDT) were selected, with BGL, POD, and PPO achieving scores of 87.6, 94.8, and 84.1, respectively. These models were then refined using molecular dynamics for 100 ns. Additionally, UPLC-MS/MS analysis identified various flavonoids in cranberries, including cyanidin, delphinidin, procyanidin B2 and B4, petunidin, pelargonidin, peonidin, and malvidin, providing important experimental data to support the study. Molecular docking simulations revealed the most stable interactions between anthocyanase and the anthocyanins cyanidin 3-arabinoside and cyanidin 3-glucoside, with a favorable ΔG of interaction between −9.3 and −9.2 kcal/mol. This study contributes to proposing a degradation mechanism and seeking inhibitors to prevent fruit discoloration. Full article

Emissions of carbon dioxide (CO2), nitrous oxide (N2O), and methane (CH4), particularly those from organic soils, need to be reduced in the context of climate change mitigation (CCM). Here, we estimated the greenhouse gas (GHG) fluxes from nutrient-poor organic soils in cranberry (Vaccinium macrocarpon) and highbush blueberry (Vaccinium corymbosum) plantations established on former peat extraction fields compared to active peat extraction fields and pristine raised bogs in Latvia. A two-year study (2016–2018) was conducted using the manual closed chamber method. In berry plantations and active peat extraction fields, annual net CO2 fluxes contributed the most to total GHG emissions, accounting for over 67%, and temperature had the most significant impact on CO2 fluxes. Conversely, annual CH4 fluxes were the primary contributor to total net GHG emissions in the pristine raised bog, which simultaneously acted as a slight CO2 sink. N2O fluxes were relatively low among all studied land use types. This study provided quantitative insights into the variation in GHG fluxes and the environmental variables influencing them, and the obtained data are valuable to estimate the impact of the establishment of berry plantations on former peat extraction fields on CCM in the hemiboreal region of Europe. A pesticide exposure baseline for honey bees was compiled for two New England cropping systems, the native North American plant species consisting of lowbush blueberry (Vaccinium angustifolium Aiton) and cranberry (Vaccinium macrocarpon Aiton). More unique pesticide compounds were applied in blueberry than cranberry, but the numbers of pesticides discovered in trapped honey bee pollen were similar between the two crop systems. Not all pesticides found in pollen were the result of the applications reported by growers of either crop. When comparing residues, number of pesticides detected, total concentration, and risk quotient varied between the two crops. Also, blueberry was dominated by fungicides and miticides (varroacides) and cranberry was dominated by insecticides and herbicides. When comparing reported grower applications that were matched with detection in residues, the proportion of pesticide numbers, concentrations, and risk quotients varied by crop system and pesticide class. In most cases, pesticide residue concentrations were of low risk (low risk quotient) to honey bees in these crops. Ozonation and UV irradiation are promising sustainable methods for extending the shelf life of stored fruits. The aim of this research was to evaluate the effectiveness of the constructed system, enabling simultaneous ozonation and UV-C irradiation of cranberry fruits in extending their shelf life. The effectiveness of this solution was determined by analyzing the properties of fruits subjected to the processes. The impact of these processes on the shelf life of fruits was assessed during 42 days of storage at a temperature of 8 °C and 80% humidity. It was demonstrated that applying the ozonation process to fruits, as facilitated by the developed solution’s throughput, led to a reduction in microbiological load (fermentation bacteria count reduction by 3.4 log cfu−1), resulting in an extension of their shelf life and a reduction in storage losses (8.98% by mass). The implementation of the developed solution, by reducing water loss and limiting fruit losses (approximately 5% less compared to the control group) during storage, contributes to tangible benefits for the producers and distributors of these fruits. The proposed modification has a positive environmental effect by reducing waste and makes the cultivation of cranberries more sustainable and environmentally friendly. Full article

The phytochemical diversity and potential health benefits of V. oxycoccos and V. macrocarpon fruits call for further scientific inquiry. Our study aimed to determine the phytochemical composition of extracts from these fruits and assess their antioxidant, antibacterial, and anticancer properties in vitro. It was found that the ethanolic extracts of V. oxycoccos and V. macrocarpon fruits, which contained more lipophilic compounds, had 2–14 times lower antioxidant activity compared to the dry aqueous extracts of cranberry fruit, which contained more hydrophilic compounds. All tested cranberry fruit extracts (OE, OW, ME, and MW) significantly inhibited the growth of bacterial strains S. aureus, S. epidermidis, E. coli, and K. pneumoniae in vitro compared to the control. Cytotoxic activity against the human prostate carcinoma PPC-1 cell line, human renal carcinoma cell line (CaKi-1), and human foreskin fibroblasts (HF) was determined using an MTT assay. Furthermore, the effect of the cranberry fruit extract samples on cell migration activity, cancer spheroid growth, and viability was examined. The ethanolic extract from V. macrocarpon fruits (ME) showed higher selectivity in inhibiting the viability of prostate and renal cancer cell lines compared to fibroblasts. It also effectively hindered the migration of these cancer cell lines. Additionally, the V. macrocarpon fruit extract (ME) demonstrated potent cytotoxicity against PPC-1 and CaKi-1 spheroids, significantly reducing the size of PPC-1 spheroids compared to the control. These findings suggest that cranberry fruit extracts, particularly the ethanolic extract from V. macrocarpon fruits, have promising potential as natural remedies for bacterial infections and cancer therapy. The American cranberry is a perennial North American fruit plant that is grown successfully on commercial plantations in Poland. The purpose of this study was to recognize filamentous fungi that colonize roots, leaves, and fruits without visible disease symptoms. Pure fungal cultures were isolated from disinfected plant fragments in agar media and identified by sequencing common taxonomic DNA markers such as the ITS region, the TEF-1α, or RPB2 genes. Of the 141 isolates studied, 59% were identified as closely related to soil saprotrophs. They were classified primarily as showing the greatest similarity to type strains of Trichoderma amoenum, Trichoderma dorothopsis, Paraphaeosphaeria sporulosa, and Penicillium murcianum. Additionally, isolates that are most similar to strains of Penicillium crustosum, Aspergillus flavus, and Aspergillus versicolor that produced mycotoxins were detected. The fungi identified as closest to Alternaria geophila, Alternaria senecionicola, Paraphoma radicina, Pestalotiopsis unicolor, Pestalotiopsis scoparia, and Neopestalotiopsis spp., whose hosts are plants other than American cranberry, represented 33.81% of the isolates tested. Only 7.2% of the isolates corresponded to the species of Physalospora vaccinia, Diaporthe vaccinii, and Diaporthe eres, known cranberry pathogens. The results of this study can be used to identify latent plant infection and potential disease risks. Full article

This study investigated the in vitro antioxidant and biological properties of ethanol extracts obtained from the fruits of the highbush cranberry. The produced extracts exhibited a high content of polyphenols (1041.9 mg 100 g d.m.−1) and a high antioxidant activity (2271.2 mg TE g 100 d.m.−1 using the DPPH method, 1781.5 mg TE g 100 d.m.−1 using the ABTS method), as well as a substantial amount of vitamin C (418.2 mg 100 g d.m.−1). These extracts also demonstrated significant in vitro biological activity. Studies conducted on the Saccharomyces cerevisiae cellular model revealed the strong antioxidant effects of the extract, attributed to a significant reduction in the levels of reactive oxygen species (ROS) within the cells, confirming the utility of the extracts in mitigating oxidative stress. Moreover, inhibitory properties were demonstrated against factors activating metabolic processes characteristic of inflammatory conditions. It was observed that the cranberry extract inhibits the activity of cyclooxygenase-1 (COX-1) and cyclooxygenase-2 (COX-2) non-selectively. Additionally, the extract was found to be a highly active inhibitor of acetylcholinesterase (AChE), potentially suggesting the applicability of this extract in the prevention of neurodegenerative diseases, including Alzheimer’s disease.
