The development of implantable neural interfaces is essential for enabling bidirectional communication between the nervous system and prosthetic devices, yet their evaluation still relies primarily on in vivo models which are costly, variable, and ethically constrained. Here, we report a bio-inspired artificial nerve simulator engineered as a reproducible ex vivo platform for pre-implantation testing of plug-type electrodes. The simulator is fabricated from a conductive hydrogel composite based on reduced graphene oxide (rGO), polyaniline (PANI), agarose, sucrose, and sodium chloride, with embedded conductive channels that replicate the fascicular organization and conductivity of peripheral nerves. The resulting construct exhibits impedance values of ~2.4–2.9 kΩ between electrode needles at 1 kHz, closely matching in vivo measurements (~2 kΩ) obtained in Sus scrofa domesticus nerve tissue. Its structural and electrical fidelity enables systematic evaluation of electrode–nerve contact properties, signal transmission, and insertion behavior under controlled conditions, while reducing reliance on animal experiments. This bio-inspired simulator offers a scalable and physiologically relevant testbed that bridges materials engineering and translational neuroprosthetics, accelerating the development of next-generation implantable neural interfaces. Xishu Gardens, an exemplary narrative of classical Chinese gardens, faces challenges in preserving its commemorative spatial structures while accommodating modern visitors’ needs. While trajectory analysis is critical, existing studies struggle to interpret multi-dimensional perception-preference data owing to spatiotemporal mismatches in multi-source datasets. This study adopted an improved Ward–K-medoids hybrid clustering algorithm to analyze 885 trajectory samples and 34,384 synchronized data points capturing emotional valence, cognitive evaluations, and dwell time behaviors via panoramic digital twins across three heritage sites (Du Fu Thatched Cottage, San Su Shrine, and Wangjiang Tower Park). Our key findings include the following: (1) Axial bimodal patterns: Type I high-frequency looping paths (27.6–68.9% recurrence) drive deep exploration, in contrast to Type II linear routes (≤0.5% recurrence), which enable intensive node coverage. (2) Layout-perception dynamics: single-axis layouts maximize behavioral engagement (DFTC), free-form designs achieve optimal emotional-cognitive integration (WTP), and multi-axis systems amplify emotional-cognitive fluctuations (SSS). (3) Spatial preference hierarchy: entrance and waterfront zones demonstrate dwell times 20% longer than site averages. Accordingly, the proposed model synchronizes Type II peak-hour throughput with Type I off-peak experiential depth using dynamic path allocation algorithms. This study underscores the strong spatial guidance mechanisms of Xishu Gardens, supporting tourism management and heritage conservation. Full article

Background: Celiac disease (CD) is an immune-mediated disease characterized by disruptions of the small intestine. Factors such as viral and bacterial infections can trigger CD. Recently, the reactivation of Human Endogenous Retroviruses (HERVs) has also been implicated, but little is known about their specific role in patients with celiac disease. Methods: The purpose of this study is to explore the humoral immune response mounted against epitopes derived from the envelope portion of three families of HERVs (HERV-K, HERV-H, and HERV-W) in CD patients. Reactivity against the HERV-K, HERV-H, and HERV-W env-su peptides was tested by indirect ELISAs in plasma of 40 patients with celiac disease and 41 age-matched healthy subjects (HCs). Results: HERV-K, HERV-H, and HERV-W env-su peptides triggered different antibody responses in CD patients compared to HCs, with a stronger reactivity (p = 0.0001). Conclusions: Present results show, for the first time, that epitopes of HERV-K, HERV-H, and HERV-W are more recognized in patients with CD. Taking into consideration their proinflammatory and autoimmune features, this might suggest that HERVs may contribute to the development of CD or its exacerbation in genetically predisposed subjects. Finally, to elucidate the interplay between gut inflammation and HERVs during the inflammatory process, further studies are required. Those investigations should focus on the expression levels of HERVs and their relationship with the immune response, specifically examining anti-transglutaminase 2 (TG2) antibody levels under both gluten-free and gluten-containing dietary conditions. Metabolic dysfunctions are among the best documented hallmarks of ageing. Cardiovascular disease, Alzheimer’s disease, cancer, type 2 diabetes mellitus, metabolic-dysfunction-associated steatosis liver disease, and fragility fractures are diseases of hyperinsulinaemia that reduce life and healthspan. We studied the effect of suppressing ketosis in 10 lean (BMI 20.5 kg/m2 ± 1.4), metabolically healthy, pre-menopausal women (age 32.3 ± 8.9 years) maintaining nutritional ketosis (NK) for an average of 3.9 years (± 2.3) who underwent three 21-day phases: nutritional ketosis (NK; P1), suppressed ketosis (SuK; P2), and returned to NK (P3). Ketosis suppression significantly increased insulin, 1.83-fold (p = 0.0006); glucose, 1.17-fold (p = 0.0088); homeostasis model assessment for insulin resistance (HOMA-IR), 2.13-fold (p = 0.0008); leptin, 3.35-fold (p = 0.0010); total osteocalcin, 1.63-fold (p = 0.0138); and uncarboxylated osteocalcin, 1.98-fold (p = 0.0417) and significantly decreased beta-hydroxybutyrate, 13.50-fold (p = 0.0012) and glucagon-like peptide-1 (GLP-1), 2.40-fold (p = 0.0209). Sustained NK showed no adverse health effects and may mitigate hyperinsulinemia. All biomarkers returned to basal P1 levels after removing the intervention for SuK, indicating that metabolic flexibility was maintained with long-term euketonaemia. Full article

Flavour changing neutral current (FCNC) processes are described by loop diagrams in the Standard Model (SM), while in 331 models, based on the gauge group SU(3)C×SU(3)L×U(1)X, they are dominated by tree-level exchanges of a new heavy neutral gauge boson Z. By exploiting this feature, observables related to FCNC decays of K, Bd and Bs mesons can be considered in several variants of 331 models. The variants are distinguished by the value of a parameter β that plays a key role in this framework. Imposing constraints on the ΔF=2 observables, we select possible ranges for the mass of the Z boson in correspondence to the values β=±k/3, with k=1,2. The results are used to determine the impact of 331 models on bs processes and on the correlations among them, in the light of new experimental data recently released. Full article

This study employed metal additive manufacturing technology to fabricate oscillating heat pipes using SUS316L as the material and conducted related printing parameter experiments and thermal performance tests. The initial experimentation involved testing the relative density and size error of the metal additive manufacturing process. Density measurement was performed using the Archimedes method, and further X-ray CT scanning was utilized to observe the internal structure and compactness. The outcomes indicate that suitable laser parameters yield favorable results in producing oscillating heat pipes, achieving good compactness and minimal dimensional error with proper parameter adjustments. Following relevant pre-processing and post-processing on the oscillating heat pipe, leakage experiments were conducted to ensure experimental accuracy. The oscillating heat pipe had dimensions of 120 mm in length and 51 mm in width, with five turns of 2 mm × 2 mm cross-sectional channels inside. Interval design was employed to address inter-channel thermal interaction commonly encountered in flat heat pipes for comparison. Methanol was selected as the working fluid to investigate the oscillating characteristics and thermal performance under different input powers (20 W, 30 W, 40 W, 60 W, 80 W). The results indicated that the inter-channel spacing can significantly decrease the lateral thermal interaction and enhance the oscillation effect during the operation of the oscillating heat pipe, resulting in improved thermal performance. The experiments demonstrated that at 20 W, the equivalent thermal conductivity of the heat pipes with and without inter-channel spacing was 2428 and 1743 (W/mK), respectively, and at 80 W, it was 2663 and 2511 (W/mK), respectively. These results indicate that reducing thermal interaction can significantly improve the oscillation effect, leading to higher equivalent thermal conductivity at low power. Full article

In this work, we interpret the newly observed η1(1855) resonance with exotic JPC=1+ quantum numbers in the I=0 sector, reported by the BESIII Collaboration, as a dynamically generated state from the interaction between the lightest pseudoscalar mesons and axial-vector mesons. The interaction is derived from the lowest order chiral Lagrangian from which the Weinberg–Tomozawa term is obtained, describing the transition amplitudes among the relevant channels, which are then unitarized using the Bethe–Salpeter equation, according to the chiral unitary approach. We evaluate the η1(1855) decays into the ηη and KK¯*π channels and find that the latter has a larger branching fraction. We also investigate its SU(3) partners, and according to our findings, the π1(1400) and π1(1600) structures may correspond to dynamically generated states, with the former one coupled mostly to the b1π component and the latter one coupled to the K1(1270)K¯ channel. In particular, our result for the ratio Γ(π1(1600)f1(1285)π)/Γ(π1(1600)ηπ) is consistent with the measured value, which supports our interpretation for the higher π1 state. We also report two poles with a mass about 1.7 GeV in the I=1/2 sector, which may be responsible for the K*(1680). We suggest searching for two additional η1 exotic mesons with masses around 1.4 and 1.7 GeV. In particular, the predicted η1(1700) is expected to have a width around 0.1 GeV and can decay easily into KK¯ππ. Full article

Parameter-free theoretical predictions based on a dual shell mechanism within the proxy-SU(3) symmetry of atomic nuclei, as well as covariant density functional theory calculations using the DDME2 functional indicate that shape coexistence (SC) based on the particle-hole excitation mechanism cannot occur everywhere on the nuclear chart but is restricted on islands lying within regions of 7–8, 17–20, 34–40, 59–70, 96–112, 146–168 protons or neutrons. Systematics of data for even-even nuclei possessing K=0 (beta) and K=2 (gamma) bands support the existence of these islands, on which shape coexistence appears whenever the K=0 bandhead 02+ and the first excited state of the ground state band 21+ lie close in energy, with nuclei characterized by 02+ lying below the 21+ found in the center of these islands. In addition, a simple theoretical mechanism leading to multiple-shape coexistence is briefly discussed. Full article

The aim of the current study was to identify the major genes and pathways involved in the process of hypertrophy and skeletal muscle maturation that is common for Bos taurus, Ovis aries, and Sus scrofa species. Gene expression profiles related to Bos taurus, Ovis aries, and Sus scrofa muscle, with accession numbers GSE44030, GSE23563, and GSE38518, respectively, were downloaded from the GEO database. Differentially expressed genes (DEGs) were screened out using the Limma package of R software. Genes with Fold Change > 2 and an adjusted p-value < 0.05 were identified as significantly different between two treatments in each species. Subsequently, gene ontology and pathway enrichment analyses were performed. Moreover, hub genes were detected by creating a protein–protein interaction network (PPI). The results of the analysis in Bos taurus showed that in the period of 280 dpc–3-months old, a total of 1839 genes showed a significant difference. In Ovis aries, however, during the period of 135dpc–2-months old, a total of 486 genes were significantly different. Additionally, in the 91 dpc–adult period, a total of 2949 genes were significantly different in Sus scrofa. The results of the KEGG pathway enrichment analysis and GO function annotation in each species separately revealed that in Bos taurus, DEGs were mainly enriched through skeletal muscle fiber development and skeletal muscle contraction, and the positive regulation of fibroblast proliferation, positive regulation of skeletal muscle fiber development, PPAR signaling pathway, and HIF-1 signaling pathway. In Ovis aries, DEGs were mainly enriched through regulating cell growth, skeletal muscle fiber development, the positive regulation of fibroblast proliferation, skeletal muscle cell differentiation, and the PI3K-Akt signaling, HIF-1 signaling, and Rap1 signaling pathways. In Sus scrofa, DEGs were mainly enriched through regulating striated muscle tissue development, the negative regulation of fibroblast proliferation and myoblast differentiation, and the HIF-1 signaling, AMPK signaling, and PI3K-Akt signaling pathways. Using a Venn diagram, 36 common DEGs were identified between Bos taurus, Ovis aries, and Sus scrofa. A biological pathways analysis of 36 common DEGs in Bos taurus, Ovis aries, and Sus scrofa allowed for the identification of common pathways/biological processes, such as myoblast differentiation, the regulation of muscle cell differentiation, and positive regulation of skeletal muscle fiber development, that orchestrated the development and maturation of skeletal muscle. As a result, hub genes were identified, including PPARGC1A, MYOD1, EPAS1, IGF2, CXCR4, and APOA1, in all examined species. This study provided a better understanding of the relationships between genes and their biological pathways in the skeletal muscle maturation process. Full article

(1) Background: Continuous monitoring of tacrolimus (TAC), mycophenolic acid (MPA), and creatinine (Cre) after renal transplantation is vitally important. In this study, we developed a new method based on volumetric absorptive microsampling (VAMS) combined with Ultra Performance Liquid Chromatography–Tandem Mass Spectrometry (UPLC-MS/MS) to simultaneously quantify three analytes including TAC, MPA, and Cre in whole blood. (2) Methods: The VAMS-based UPLC-MS/MS assay used a shared extraction and a single injection to simultaneously quantify the included TAC, MPA, and Cre. Development and validation were carried out following the Food and Drug Administration and European Medicines Agency guidelines for the validation of bioanalytical methods. Moreover, clinical validation for the three analytes was performed in both dried blood spot (DBS) and VAMS. Furthermore, a willingness survey was conducted using the system usability scale (SUS) for renal transplant recipients. (3) Results: The assay was successfully validated for all analytes. No interference, carryover, or matrix effects were observed, and extraction recoveries and process efficiencies were >90.00%. Analysis was unaffected by hematocrit (0.20~0.60, L/L) and anticoagulants (EDTA-2K). Dried VAMS samples were stable for 7 days at ambient temperature and stable for at least 1 month at −20 °C. During clinical validation, the measured TAC, corrected MPA, and Cre concentrations of VAMS samples met the analytical standards (95.00%, 88.57%, and 92.50%). When more stringent clinical acceptance criteria were set, the results obtained by VAMS (90.00%, 71.43%, and 85.00%) better than DBS (77.50%, 62.86%, and 70.00%). Compared with DBS, the survey found that renal transplant recipients are more inclined to use VAMS. (4) Conclusions: A robust extraction and UPLC-MS/MS analysis method in VAMS tips was developed and fully validated for the simultaneous quantification of TAC, MPA, and Cre concentrations. This method provides analytical support for the one-sample remote monitoring of both immunosuppressive drug concentrations and renal function in allo-renal recipients. Based on the good consistency between this method and the routine detection of venous blood samples and higher patient satisfaction than DBS, we believe that VAMS sampling can be a better alternative to venous whole-blood sampling. Aldehyde dehydrogenase 2 (ALDH2) enzyme is required for alcohol detoxification. ALDH2 belongs to the aldehyde dehydrogenase family, the most important oxidative pathway of alcohol digestion. Two main liver isoforms of aldehyde dehydrogenase are cytosolic and mitochondrial. Approximately 50% of East Asians have ALDH2 deficiency (inactive mitochondrial isozyme), with lysine (K) for glutamate (E) substitution at position 487 (E487K). ALDH2 deficiency is also known as Alcohol Flushing Syndrome or Asian Glow. For people with an ALDH2 deficiency, their face turns red after drinking alcohol, and they are more susceptible to various diseases than ALDH2-normal people. This study performed a machine learning analysis of ALDH2 sequences of thirteen other species by comparing them with the human ALDH2 sequence. Based on the various quantitative metrics (physicochemical properties, secondary structure, Hurst exponent, Shannon entropy, and fractal dimension), these fourteen species were clustered into four clusters using the unsupervised machine learning (K-means clustering) algorithm. We also analyze these species using hierarchical clustering (agglomerative clustering) and draw the phylogenetic trees. The results show that Homo sapiens is more closely related to the Bos taurus and Sus scrofa species. Our experimental results suggest that the testing for discovering medicines may be done on these species before being tested in humans to alleviate the impacts of ALDH2 deficiency. Full article

We aimed to reveal the dispersal and gene flow of the local wild boar (Sus scrofa) population and find their genetic boundary in Fukushima Prefecture. After the nuclear incident in 2011, the land was considered a difficult-to-return zone, and the increase in the number of wild boars was pronounced. To provide an effective management strategy for the wild boar population, we used multiplexed inter-simple sequence repeat genotyping by sequencing (MIG-seq) and clarified the genetic structure of wild boars. We obtained 328 single-nucleotide polymorphisms from 179 samples. STRUCTURE analysis showed that the most likely number of population cluster was K = 2. Molecular analysis of variance showed significant genetic differences between groups of wild boars inhabiting in the east and west across the Abukuma River. The migration rate from the eastern population to the western population is higher than in the reverse case based on BayesAss analysis. Our study indicates that both the Abukuma River and anthropogenic urbanization along the river may affect the migration of wild boars and the population in western was established mainly by the migration from other neighboring prefectures. We present a chiral K¯N interaction model that has been developed and optimized in order to account for the experimental data of inelastic K¯N reaction channels that open at higher energies. In particular, we study the effect of the higher partial waves, which originate directly from the chiral Lagrangian, as they could supersede the role of high-spin resonances employed in earlier phenomenological models to describe meson-baryon cross sections in the 2 GeV region. We present a detailed derivation of the partial wave amplitudes that emerge from the chiral SU(3) meson-baryon Lagrangian up to the d-waves and next-to-leading order in the chiral expansion. We implement a nonperturbative unitarization in coupled channels and optimize the model parameters to a large pool of experimental data in the relevant energy range where these new contributions are expected to be important. The obtained results are encouraging. They indicate the ability of the chiral higher partial waves to extend the description of the scattering data to higher energies and to account for structures in the reaction cross-sections that cannot be accommodated by theoretical models limited to the s-waves. In this work, a new flexible and biocompatible microfluidic pH sensor based on surface acoustic waves (SAWs) is presented. The device consists of polyethylene naphthalate (PEN) as a flexible substrate on which aluminum nitride (AlN) has been deposited as a piezoelectric material. The fabrication of suitable interdigitated transducers (IDTs) generates Lamb waves (L-SAW) with a center frequency ≈500 MHz traveling in the active region. A SU-8 microfluidics employing ZnO nanoparticles (NPs) functionalization as a pH-sensitive layer is fabricated between the IDTs, causing a shift in the L-SAW resonance frequency as a function of the change in pH values. The obtained sensitivity of ≈30 kHz/pH from pH 7 to pH 2 demonstrates the high potential of flexible SAW devices to be used in the measurement of pH in fluids and biosensing. Full article

We extend recent results regarding finding shortest unique substrings (SUSs) to obtain new time-space tradeoffs for this problem and the generalization of finding k-mismatch SUSs. Our new results include the first algorithm for finding a k-mismatch SUS in sublinear space, which we obtain by extending an algorithm by Senanayaka (2019) and combining it with a result on sketching by Gawrychowski and Starikovskaya (2019). We first describe how, given a text T of length n and m words of workspace, with high probability we can find an SUS of length L in O(n(L/m)logL) time using random access to T, or in O(n(L/m)log2(L)loglogσ) time using O((L/m)log2L) sequential passes over T. We then describe how, for constant k, with high probability, we can find a k-mismatch SUS in O(n1+ϵL/m) time using O(nϵL/m) sequential passes over T, again using only m words of workspace. Finally, we also describe a deterministic algorithm that takes O(nτlogσlogn) time to find an SUS using O(n/τ) words of workspace, where τ is a parameter. Full article
