Pulmonary Involvement in Patients with Positive Myositis Antibodies in Rheumatology: A Retrospective Monocentric Analysis

Background: Pulmonary involvement is the most common prognosis-related organ involvement in idiopathic inflammatory myopathy (IIM). Owing to the large number of antibodies, the evidence for lung involvement and rare antibodies is limited. In everyday clinical practice, the interpretation of positive myositis antibodies represents a challenge. Methods: This study is a retrospective monocentric analysis. The data collection regarding positive myositis antibodies and possible pulmonary involvement was carried out from July 2019 to May 2022. Data analysis revealed positive results for one of the following antibodies: EJ, PL7, OJ, PL12, Mi-2α, TIF1γ, MDA5, SAE, NXP2, SRP, Ku, PM-Scl100 and PM-Scl75. In our analysis, patients with IIM, patients with inflammatory rheumatic disease other than IIM and patients without inflammatory rheumatic disease are described. The results of high-resolution computed tomography (HRCT), pulmonary function tests, echocardiographic examinations and their associated clinical findings are examined. Results: In the entire cohort, 209 patients with positive myositis antibodies were detected. In total, 22 (10.5%) patients had interstitial lung disease (ILD) patterns on HRCT. In the subgroup of patients with IIM, a significantly higher proportion of patients with lung involvement (n = 13, 35.1%) was found than in the group with other inflammatory rheumatic diseases (IRDs) (n = 6, 6.7%) or in the group without IRDs (n = 3, 3.7%). When the antibody groups were considered, the PL12-positive patients had the largest proportion of ILD (42%), followed by the MDA5-positive patients (40%). Conclusions: In patients with IIM, myositis antibodies are highly relevant for assessing the risk of lung involvement. In groups with other IRD or without IRD, antibody detection does not represent this high relevance for lung involvement. A differentiated assessment of the various MSAs or MAAs detected, as well as clinical parameters, allows for further important risk assessment for prognosis-relevant lung involvement. Full article

Fine particulate matter (PM2.5) exposure has been linked to increased lung damage due to compromised vascular barrier function, while 3-deoxysappanchalcone (3-DSC), a chalcone derived from Caesalpinia sappan, is known for its pharmacological benefits such as anti-cancer, anti-inflammatory, and antioxidant effects; however, its potential role in mitigating PM2.5-induced pulmonary damage remains unexplored. To confirm the inhibitory effects of 3-DSC on PM2.5-induced pulmonary injury, this research focused on evaluating how 3-DSC influences PM2.5-induced disruption of the barrier of the endothelial cells (ECs) in the lungs and the resulting pulmonary inflammation. Permeability, leukocyte migration, proinflammatory protein activation, reactive oxygen species (ROS) generation, and histology were assessed in PM2.5-treated ECs and mice. This study demonstrated that 3-DSC effectively neutralized the reactive oxygen species (ROS) generated by PM2.5 exposure in the lung endothelial cells, suppressing ROS-triggered p38 MAPK activation while enhancing Akt signaling pathways critical to preserving vascular barrier function. In animal models, 3-DSC administration markedly decreased vascular permeability, attenuated the influx of immune cells into the lung tissue, and lowered inflammatory mediators like cytokines in the airways of PM2.5-exposed mice. These data suggest that 3-DSC might exert protective effects on PM2.5-induced inflammatory lung injury and vascular hyperpermeability. Full article

Powdery mildew (PM), caused by Sphaerotheca phaseoli, severely threatens mungbean (Vigna radiata) productivity and quality, yet the molecular basis of resistance remains poorly defined. This study employed transcriptome profiling to compare defense responses in a resistant genotype, SUPER5, and a susceptible variety, CN84-1, following pathogen infection. A total of 1755 differentially expressed genes (DEGs) were identified, with SUPER5 exhibiting strong upregulation of genes encoding pathogenesis-related (PR) proteins, disease resistance proteins, and key transcription factors. Notably, genes involved in phenylpropanoid and flavonoid biosynthesis, pathways associated with antimicrobial compound and lignin production, were markedly induced in SUPER5. In contrast, CN84-1 showed limited activation of defense genes and downregulation of essential regulators such as MYB14. Gene ontology (GO) and Kyoto Encyclopedia of Genes and Genomes (KEGG) enrichment analyses highlighted the involvement of plant–pathogen interaction pathways, MAPK signaling, and reactive oxygen species (ROS) detoxification in the resistant response. Quantitative real-time PCR validated 11 candidate genes, including PAL3, PR2, GSO1, MLO12, and P21, which function in pathogen recognition, signaling, the biosynthesis of antimicrobial metabolites, the production of defense proteins, defense regulation, and the reinforcement of the cell wall. Co-expression network analysis revealed three major gene modules linked to flavonoid metabolism, chitinase activity, and responses to both abiotic and biotic stresses. These findings offer valuable molecular insights for breeding PM-resistant mungbean varieties. Full article

Plastic Film Mulching Regulates Soil Respiration and Temperature Sensitivity in Maize Farming Across Diverse Hydrothermal Conditions

Soil respiration (Rt), consisting of heterotrophic (Rh) and autotrophic respiration (Ra), plays a vital role in terrestrial carbon cycling and is sensitive to soil temperature and moisture. In dryland agriculture, plastic film mulching (PM) is widely used to regulate soil hydrothermal conditions, but its effects on Rt components and their temperature sensitivity (Q10) across regions remain unclear. A two-year field study was conducted at two rain-fed maize sites: Anding (warmer, semi-arid) and Yuzhong (colder, drier). PM significantly increased Rt, Rh, and Ra, especially Ra, due to enhanced root biomass and improved microclimate. Yield increased by 33.6–165%. Peak respiration occurred earlier in Anding, aligned with maize growth and soil temperature. PM reduced Q10 of Rt and Ra in Anding, but only Ra in Yuzhong. Rh Q10 remained stable, indicating microbial respiration was less sensitive to temperature changes. Structural equation modeling revealed that Rt and Ra were mainly driven by soil temperature and root biomass, while Rh was more influenced by microbial biomass carbon (MBC) and dissolved organic carbon (DOC). Despite increased CO2 emissions, PM improved carbon emission efficiency (CEE), particularly in Yuzhong (+67%). The application of PM is recommended to enhance yield while optimizing carbon efficiency in dryland farming systems. Full article

The Kindlin family of scaffold proteins plays key roles in integrin-mediated processes. Kindlin-1 and -2, encoded by the FERMT1 and FERMT2 genes, respectively, are expressed in the epidermis. Kindlin-1 plays protective roles against the development of cutaneous squamous cell carcinomas (cSCCs) in epidermal keratinocytes. However, the role of Kindlin-2 in transformed epidermal keratinocytes has remained virtually unexplored. In this study, we used siRNA approaches to generate Kindlin-2-depleted cells in three isogenic transformed keratinocyte lines. PM1, MET1, and MET4 cells model, respectively, a precancerous lesion, a primary cSCC, and a metastatic lesion of the latter. MET1 cells express both Kindlin-1 and -2. However, Kindlin-1 was not detectable in PM1 and MET4 cells. FERMT2 silencing in PM1 and MET4, but not in MET1 cells, reduced proliferation and the ability to adhere to culture surfaces and spreading. Furthermore, Kindlin-2-depleted PM1 and MET4, but not MET1 cells, exhibited decreased numbers of focal adhesions, as well as an altered F-actin and microtubule cytoskeletal organization. Significantly, FERMT2 silencing reduced the directional migration in all three cell types. These findings are consistent with the concept that, in the absence of other Kindlin orthologues, Kindlin-2 plays a prominent role in the modulation of the proliferation, spreading, focal adhesion assembly, and motility of transformed keratinocytes, as exemplified by PM1 and MET4 cells. Nephrogenic diabetes insipidus (NDI) is a rare hereditary disorder characterized by renal resistance to arginine vasopressin (AVP), resulting in the kidneys’ inability to concentrate urine. Approximately 90% of NDI cases follow an X-linked inheritance pattern and are associated with pathogenic variants in the AVPR2 gene, which encodes the vasopressin receptor type 2. The remaining 10% are attributed to mutations in the AQP2 gene, which encodes aquaporin-2, and may follow either autosomal dominant or recessive inheritance patterns. We present the case of a male infant, younger than nine months of age, who was clinically diagnosed with NDI at six months. The patient presented recurrent episodes of polydipsia, polyuria, dehydration, hypernatremia, and persistently low urine osmolality. Despite adjustments in pharmacologic treatment and strict monitoring of urinary output, the clinical response remained suboptimal. Given the lack of improvement and the radiological finding of an absent posterior pituitary (neurohypophysis), the possibility of coexistent central diabetes insipidus (CDI) was raised, prompting a therapeutic trial with desmopressin. Nevertheless, in the absence of clinical improvement, desmopressin was discontinued. The patient’s management was continued with hydrochlorothiazide, ibuprofen, and a high-calorie diet restricted in sodium and protein, resulting in progressive clinical stabilization. Whole-exome sequencing identified a novel homozygous missense variant in the AQP2 gene (c.398T > A; p.Val133Glu), classified as likely pathogenic according to the American College of Medical Genetics and Genomics (ACMG) criteria: PM2 (absent from population databases), PP2 (missense variant in a gene with a low rate of benign missense variation), and PP3 (multiple lines of computational evidence supporting a deleterious effect)]. NDI is typically diagnosed during early infancy due to the early onset of symptoms and the potential for severe complications if left untreated. In this case, although initial clinical suspicion included concomitant CDI, the timely initiation of supportive management and the subsequent incorporation of molecular diagnostics facilitated a definitive diagnosis. The identification of a previously unreported homozygous variant in AQP2 contributed to diagnostic confirmation and therapeutic decision-making. The diagnosis and comprehensive management of NDI within the context of polyuria-polydipsia syndrome necessitates a multidisciplinary approach, integrating clinical evaluation with advanced molecular diagnostics. The novel AQP2 c.398T > A (p.Val133Glu) variant described herein was associated with early and severe clinical manifestations, underscoring the importance of genetic testing in atypical or treatment-refractory presentations of diabetes insipidus. Effects of Cognitive Behavioral Therapy-Based Educational Intervention Addressing Fine Particulate Matter Exposure on the Mental Health of Elementary School Children

Objectives: This study assessed the effectiveness of a cognitive behavioral therapy (CBT)-based fine dust education program, grounded in the Health Belief Model (HBM), on elementary students’ fine dust knowledge, related behaviors, and mental health (depression, anxiety, stress, sleep quality). Methods: From September to November 2024, 95 students (grades 4–6) living near a coal-fired power plant in midwestern South Korea were assigned to either an intervention group (n = 44) or a control group (n = 51). The intervention group completed a three-session CBT-based education program; the control group received stress management education. Assessments were conducted at weeks 1, 2, 4, and 8 using standardized mental health and behavior scales (PHQ: Patient Health Questionnaire, GAD: Generalized Anxiety Disorder Assessment, PSS: Perceived Stress Scale, ISI: Insomnia Severity Index). Results: A chi-square test was conducted to compare pre- and post-test changes in knowledge and behavior related to PM2.5. The intervention group showed significant improvements in seven fine dust-related knowledge and behavior items (e.g., PM2.5 awareness rose from 33.3% to 75.0%; p < 0.05). The control group showed limited gains. Regarding mental health, based on a mixed-design ANCOVA, anxiety scores significantly declined over time in the intervention group, with group and interaction effects also significant (p < 0.05). Depression scores showed time effects, but group and interaction effects were not significant. No significant changes were observed for stress, sleep, or group × PM2.5 interactions. Conclusions: The CBT-based education program effectively enhanced fine dust knowledge, health behaviors, and reduced anxiety among students. It presents a promising, evidence-based strategy to promote environmental and mental health in school-aged children. Full article

Effect of Repetitive Peripheral Magnetic Stimulation in Patients with Neck Myofascial Pain: A Randomized Sham-Controlled Crossover Trial

Background/Objectives: Neck pain caused by myofascial pain syndrome (MPS) is a highly prevalent musculoskeletal condition. Repetitive peripheral magnetic stimulation (rPMS) is a promising treatment option; however, its therapeutic effect and optimal treatment frequency remain unclear. This study aimed to investigate the therapeutic effect and duration of effect of rPMS in patients with MPS of the neck. Methods: In this randomized, sham-controlled, crossover trial, 27 patients with neck MPS and baseline visual analog scale (VAS) scores ≥ 40 were enrolled. The mean age was 43.8 ± 9.1 years, and 63% were female. Participants were randomly assigned to receive either an initial rPMS treatment (a 10 min session delivering 3900 pulses at 5–10 Hz) or sham stimulation. After 7 days, groups crossed over. Pain intensity (VAS), disability (Neck Disability Index; NDI), and analgesic use were recorded daily for seven consecutive days. A linear mixed-effects model was used for analysis. Results: At baseline, the VAS and NDI scores were 61.8 ± 10.5 and 26.0 ± 6.3, respectively. rPMS produced a significantly greater reduction in both VAS and NDI scores, with the greatest differences observed on Day 4: the differences were −24.1 points in VAS and −8.5 points in NDI compared to the sham group. There was no significant difference in analgesic use between the two groups. Conclusions: A single rPMS session provides short-term improvement in pain and disability in neck MPS. Based on the observed therapeutic window, more frequent sessions (e.g., twice weekly) may provide sustained benefit and should be explored in future studies. Full article

Background/Objectives: Anxiety before ophthalmic surgery under local anesthesia may hinder patient cooperation and surgical outcomes. Nurse-led auditory interventions offer a promising non-pharmacological approach to perioperative anxiety management. This study evaluated the effectiveness of superimposed binaural beats (SBBs)—classical music layered with frequency differentials—in reducing anxiety during pterygium surgery with conjunctival autografting. Methods: In this randomized controlled trial, 111 adult patients scheduled for elective pterygium excision with conjunctival autografting under local anesthesia were allocated to one of three groups: SBBs, plain music (PM), or silence (control). A trained perioperative nurse administered all auditory interventions. The patients’ anxiety was assessed using the State–Trait Anxiety Inventory—State (STAI-S), and physiological parameters (blood pressure, heart rate, respiratory rate, and oxygen saturation) were recorded before and after surgery. Results: The SBB group showed significantly greater reductions in their STAI-S scores (p < 0.001), systolic blood pressure (p = 0.011), heart rate (p = 0.003), and respiratory rate (p = 0.009) compared to the PM and control groups. No adverse events occurred. Conclusions: SBBs are a safe, nurse-delivered auditory intervention that significantly reduces perioperative anxiety and supports physiological stability. Their integration into routine nursing care for minor ophthalmic surgeries is both feasible and beneficial. Satellite communications are pivotal to global Internet access, connectivity, and the advancement of information warfare. Despite these importance, the open nature of satellite channels makes them vulnerable to eavesdropping, making the enhancement of interception resistance in satellite communications a critical issue in both academic and industrial circles. Within the realm of satellite communications, polarization modulation and quadrature techniques are essential for information transmission and interference suppression. To boost electromagnetic countermeasures in complex battlefield scenarios, this paper integrates multi-parameter weighted-type fractional Fourier transform (MP-WFRFT) with directional modulation (DM) algorithms, building upon polarization techniques. Initially, the operational mechanisms of the polarization-amplitude-phase modulation (PAPM), MP-WFRFT, and DM algorithms are elucidated. Secondly, it introduces a novel variable polarization-amplitude-phase modulation (VPAPM) scheme that integrates variable polarization with amplitude-phase modulation. Subsequently, leveraging the VPAPM modulation scheme, an exploration of the anti-interception capabilities of MP-WFRFT through parameter adjustment is presented. Rooted in an in-depth analysis of simulation data, the anti-scanning capabilities of MP-WFRFT are assessed in terms of scale vectors in the horizontal and vertical direction. Finally, exploiting the potential of the robust anti-scanning capabilities of MP-WFRFT and the directional property of antenna arrays in DM, the paper proposes a secure transmission technique employing directional variable polarization modulation with MP-WFRFT. The performance simulation analysis demonstrates that the integration of MP-WFRFT and DM significantly outperforms individual secure transmission methods, improving anti-interception performance by at least an order of magnitude at signal-to-noise ratios above 10 dB. Consequently, this approach exhibits considerable potential and engineering significance for its application within satellite communication systems. Full article

Northern Thailand frequently suffers from severe PM2.5 air pollution, especially during the dry season, due to agricultural burning, local emissions, and transboundary haze. Understanding how pollution moves across regions and identifying source–receptor relationships are critical for effective air quality management. This study investigated the spatial and temporal dynamics of PM2.5 in northern Thailand. Specifically, it explored how pollution at one monitoring station influenced concentrations at others and revealed the seasonal structure of PM2.5 transmission using network-based analysis. We developed a Python-based framework to analyze daily PM2.5 data from 2022 to 2023, selecting nine representative stations across eight provinces based on spatial clustering and shape-based criteria. Delaunay triangulation was used to define spatial connections among stations, capturing the region’s irregular geography. Cross-correlation and Granger causality were applied to identify time-lagged relationships between stations for each season. Trophic coherence analysis was used to evaluate the hierarchical structure and seasonal stability of the resulting networks. The analysis revealed seasonal patterns of PM2.5 transmission, with certain stations, particularly in Chiang Mai and Lampang, consistently acting as source nodes. Provinces such as Phayao and Phrae were frequently identified as receptors, especially during the winter and rainy seasons. Trophic coherence varied by season, with the winter network showing the highest coherence, indicating a more hierarchical but less stable structure. The rainy season exhibited the lowest coherence, reflecting greater structural stability. PM2.5 spreads through structured, seasonal pathways in northern Thailand. Network patterns vary significantly across seasons, highlighting the need for adaptive air quality strategies. This framework can help identify influential monitoring stations for early warning and support more targeted, season-specific air quality management strategies in northern Thailand. Full article

Life Cycle Assessment of a Digital Tool for Reducing Environmental Burdens in the European Milk Supply Chain

Food loss and waste from the European Union’s dairy supply chain, particularly in the management of fresh milk, imposes significant environmental burdens. This study demonstrates that implementing Radio Frequency Identification (RFID)-enabled digital decision-support tools can substantially reduce these impacts across the region. A cradle-to-grave life cycle assessment (LCA) was used to quantify both the additional environmental burdens from RFID (tag production, usage, and disposal) and the avoided burdens due to reduced milk losses in the farm, processing, and distribution stages. Within the EU’s fresh milk supply chain, the implementation of digital tools could result in annual net reductions of up to 80,000 tonnes of CO2-equivalent greenhouse gas emissions, 81,083 tonnes of PM2.5-equivalent particulate matter, 84,326 tonnes of land use–related carbon deficit, and 80,000 cubic meters of freshwater-equivalent consumption. Spatial analysis indicates that regions with historically high spoilage rates, particularly in Southern and Eastern Europe, see the greatest benefits from RFID enabled digital-decision support tools. These environmental savings are most pronounced during the peak months of milk production. Overall, the study demonstrates that despite the environmental footprint of RFID systems, their integration into the EU’S dairy supply chain enhances transparency, reduces waste, and improves resource efficiency—supporting their strategic value. Variants in ELAC2, a gene encoding the mitochondrial RNase Z enzyme essential for mitochondrial tRNA processing, have been associated with severe pediatric-onset mitochondrial dysfunction, primarily presenting with developmental delay, hypertrophic cardiomyopathy (HCM), and lactic-acidosis. We hereby report the case of a 25-year-old young woman presenting with dilated cardiomyopathy (DCM) and peripheral sensorimotor polyneuropathy, harboring a homozygous variant in ELAC2. The same variant has been reported only once so far in a case of severe infantile-onset form of HCM and mitochondrial respiratory chain dysfunction, with in vitro data showing a moderate reduction in the RNase Z activity and supporting the current classification as C4 according to the American College of Medical Genetics (ACMG) criteria (PS3, PM2, PM3, PP4). Our extensive clinical, imaging, histological, and genetic investigations support a causal link between the identified variant and the patient’s phenotype, despite the fact that the latter might be considered atypical according to the current state of knowledge. A detailed review of the existing literature on ELAC2-related disease is also provided, highlighting the molecular mechanisms underlying tRNA maturation, mitochondrial dysfunction, and the variable phenotypic expression. Our case further expands the clinical spectrum of ELAC2-related cardiomyopathies to include a relatively late onset in young adulthood and underscores the importance of comprehensive genetic testing in unexplained cardiomyopathies with multisystem involvement. Study on the Adaptability of FBG Sensors Encapsulated in CNT-Modified Gel Material for Asphalt Pavement

To prolong the service life of asphalt pavement and reduce its maintenance cost, a fiber Bragg grating (FBG) sensor encapsulated in carboxylated carbon nanotube (CNT-COOH)-modified gel material suitable for strain monitoring of asphalt pavement was developed. Through tensile and bending tests, the effects of carboxylated carbon nanotubes on the mechanical properties of gel materials under different dosages were evaluated and the optimal dosage of carbon nanotubes was determined. Infrared spectrometer and scanning electron microscopy were used to compare and analyze the infrared spectra and microstructure of carbon nanotubes before and after carboxyl functionalization and modified gel materials. The results show that the incorporation of CNTs-COOH increased the tensile strength, elongation at break, and tensile modulus of the gel material by 36.2%, 47%, and 17.2%, respectively, and increased the flexural strength, flexural modulus, and flexural strain by 89.7%, 7.5%, and 63.8%, respectively. Through infrared spectrum analysis, it was determined that carboxyl (COOH) and hydroxyl (OH) were successfully introduced on the surface of carbon nanotubes. By analyzing the microstructure, it can be seen that the carboxyl functionalization of CNTs improved the agglomeration of carbon nanotubes. The tensile section of the modified gel material is rougher than that of the pure epoxy resin, showing obvious plastic deformation, and the toughness is improved. According to the data from the calibration experiment, the strain and temperature sensitivity coefficients of the packaged sensor are 1.9864 pm/μm and 0.0383 nm/°C, respectively, which are 1.63 times and 3.61 times higher than those of the bare fiber grating. The results of an applicability study show that the internal structure strain of asphalt rutting specimen changed linearly with the external static load, and the fitting sensitivity is 0.0286 με/N. Combined with ANSYS finite element analysis, it is verified that the simulation analysis results are close to the measured data, which verifies the effectiveness and monitoring accuracy of the sensor. The dynamic load test results reflect the internal strain change trend of asphalt mixture under external rutting load, confirming that the encapsulated FBG sensor is suitable for the long-term monitoring of asphalt pavement strain. Daily Reference Evapotranspiration Derived from Hourly Timestep Using Different Forms of Penman–Monteith Model in Arid Climates

In arid and semi-arid climates, where water scarcity is a persistent challenge, accurately estimating reference evapotranspiration (ET) becomes essential for sustainable water management and agricultural planning. The objectives of this study are to compare hourly ET among P–M ASCE, P–M FAO, and P–M KSA mathematical models. In addition to the accuracy assessment of daily ET derived from hourly timestep calculations for the P–M ASCE, P–M FAO, and P–M KSA. To achieve these goals, a total of 525,600-min data points from the Riyadh region, KSA, were used to compute the reference ET at multiple temporal resolutions: hourly, daily, hourly averaged over 24 h, and daily as the sum of 24 h values, across all selected Penman–Monteith (P–M) models. For hourly investigation, the comparison between reference ET computed as average hourly values and as daily/24 h values revealed statistically and practically significant differences. The Wilcoxon test confirmed a statistically significant difference (p < 0.0001) with R2 of 94.75% for ASCE, 94.87% for KSA at hplt = 50 cm, 92.41% for FAO, and 92.44% for KSA at hplt = 12 cm. For daily investigation, comparing the sum of 24 h ET computations to daily ET measurements revealed an underestimation of daily ET values. The Wilcoxon test confirmed a statistically significant difference (p < 0.0001), with R2 exceeding 90% for all studied reference ET models. This comprehensive approach enabled a rigorous evaluation of reference ET dynamics under hyper-arid climatic conditions, which are characteristic of central Saudi Arabia. The findings contribute to the growing body of literature emphasizing the importance of high-frequency meteorological data for improving ET estimation accuracy in arid and semi-arid regions.
