Our study investigated how different levels of antioxidants and contrasting proportions of native legumes in the diet affect lamb meat quality. Twenty-four male Texel lambs were randomly assigned to three groups: two groups on a natural pasture in southern Brazil (Pampa Biome), each at a different proportion of legumes: Low-legume (LL, 4.37%) and High-legume (HL, 14.01%); the other group was stall-fed (Control) to achieve the same growth rates as the grazing groups. Cold carcass yield from the Control lambs was higher than HL. The meat from pasture-fed animals had a higher deposition of muscle α-tocopherol and lower lipid oxidation (TBARS values) after 9 days of storage. LL lambs had higher subcutaneous fat thickness, which promoted better sensory quality of the meat, as assessed by a trained panel. Pasture-based diets enhanced odd- and branched-chain fatty acids (OBCFAs), trans vaccenic acid, and total conjugated linoleic acids (CLAs), while decreasing elaidic acid. Despite the lower ∆9-desaturase activity, the higher proportion of Desmodium incanum (condensed tannin-rich native legume) in the HL diet did not impact meat nutritional quality. Finishing lambs on the Pampa Biome grasslands is an option for improving the oxidative stability and beneficial fatty acid content of lamb meat, which improves product quality and human consumer health. Cow’s milk and dairy products are the primary sources of OBCFAs, which have beneficial health properties. The goal of this study was to identify the factors that influence the content of OBCFAs in cow’s milk and to indicate which OBCFAs can serve as biomarkers for fermentation processes. The content of OBCFAs in milk depends on the species of ruminants, with studies showing that this varies between 3.33% (in goat’s milk) and 5.02% (in buffalo’s milk). These differences also stem from the animals’ energy balance, lactation phases, forage-to-concentrate ratio, and the presence of bioactive compounds in feeds, as well as management practices and environmental conditions. The OBCFAs in milk fat mainly come from rumen bacteria, but can also be synthesized de novo in the mammary gland, making them potentially useful noninvasive indicators of rumen fermentation. The concentration of BCFA is lower in colostrum and transitional milk than in full lactation milk. The proportions of total OBCFAs are higher in first- and second-parity cows. The most effective predictors of the biohydrogenation of fatty acids in the rumen are likely C18:2 cis-9, trans-11, iso-C16:0, and iso-C13:0. OBCFAs have been identified as potential biomarkers for rumen function, because their synthesis depends on specific bacteria. Strong predictors of subclinical ruminal acidosis include iso-C14:0, iso-C13:0, and C15:0. The concentration of ∑ OBCFA >C16 in milk is associated with fat mobilization and serves as a significant marker of the energy balance in cows. Full article

The odd- and branched-chain fatty acids (OBCFA) accumulated in ruminant products are a class of beneficial fatty acids for human health. Since biotin and leucine are involved in OBCFA synthesis, this study aimed to evaluate their effect on OBCFA synthesis in vitro. There were four treatments: the control group that only provided the basal diet, or the basal diet supplemented with biotin (4 mg/kg dry matter, DM), leucine (4 g/kg DM), or a combination of biotin (4 mg/kg DM) and leucine (4 g/kg DM). The results showed that biotin promoted the degradation of DM (p < 0.10), while leucine significantly increased the concentration of branched-chain volatile fatty acids and valerate (p < 0.05). The concentrations of total odd-chain fatty acids, total iso, total anteiso, total branched-chain fatty acids, total OBCFA, and total fatty acids were significantly increased by the supplementation of biotin or leucine (p < 0.05). Biotin and leucine significantly stimulated the activities of acetyl-CoA carboxylase, fatty acid synthase, and malonyl-CoA, with a significant interaction effect (p < 0.05). In conclusion, the results of this study suggested that biotin and leucine can be used as effective nutrition strategies to promote OBCFA synthesis. Full article

The aim of the study was to compare odd and branched-chain fatty acids (OBCFA) of milk from sheep, goat, cow, buffalo, donkey, human, and formula milk. Ruminant, monogastric, and human milks have different concentrations of these fatty acids (FA). To highlight the differences on OBCFA, a total of 282 individual milk samples were analyzed by gas chromatography. The OBCFA were found higher in ruminant than non-ruminant milks (p < 0.05). Among ruminants, sheep milk had the highest OBCFA (4.5 g/100 g of total FAME), whereases the lowest values were found in formula milk (0.18 g/100 g of total FAME). Regarding individual linear odd-chain FA (linear-OCFA), C11:0 was found higher in donkey milk than others, while sheep and buffalo milks had the greatest concentration of C15:0. Among BCFA, the iso-BCFA were higher than anteiso-BCFA in all considered milks. The isoC17:0 showed the highest concentration in all milks except for donkey and buffalo, which showed higher concentration of isoC16:0 than others. In conclusion, ruminant milks are different in terms of these FA compared to human milk and its substitutes. However, the greatest differences were found with formula milk, suggesting that this product needs the implementation of these FA to be more similar to human milk composition. Full article

The fatty acid profile, vitamins A and E, cholesterol, antioxidant power colour and the phenols profile of Sarda sheep milk from 11 commercial sheep flocks managed under permanent grassland were investigated. In each farm, the structural and managerial data and milk samples were collected during four periods (sampling dates, SD): January, March, May, and July. Data from the milk composition (fat, protein, casein, lactose, and somatic cell count), 68 fatty acids, 7 phenols, 1 total gallocatechin equivalent, ferric reducing antioxidant power, vitamins A and E, cholesterol, degree of antioxidant protection, and the colour (b *, a * and L *) were analyzed by multivariate factorial analysis using a principal component analysis approach. A proc mixed model for repeated measurement to point out the studied factors affecting significant macro and micro milk composition was also used. Only the first five components were detailed in this paper, with approximately 70% of the explained variance detected. PC1 presented the highest positive loadings for milk lactose, de novo FA synthesis and the BH intermediate, whereas OBCFA had negative loadings values. The PC2, LCFA, UFA, MUFA, vitamins E, and DAP showed positive loadings values, while SFA had a negative value. The PC3 showed a high positive loading for total phenols and non-flavonoids. PC4 presented a high positive loading for the milk macro-composition and negative values for n-3 FAs. The PC5 is characterized by high positive loadings for the a * and L * colour parameters whereas negative loadings were detected for the milk flavonoids content. These preliminary results could help to establish future threshold values for the biomarkers in milk sourced from grazing dairy sheep in natural, permanent pasture-based diets. Full article

The purpose of this study was to investigate the effects of linseed supplementation on milk yield and quality, serum biochemistry and, in particular, to evaluate its possible effects on the production of odd- and branched-chain fatty acids (OBCFA) in the milk of Cilentana grazing goats. Twelve pregnant Cilentana dairy goats were divided into two groups (CTR, control, and LIN, linseed supplementation group). After kidding, the goats had free access to the pasture and both groups received a supplement of 400 g/head of concentrate, but the one administered to the LIN group was characterized by the addition of linseed (in a ratio of 20% as fed) to the ingredients. During the trial, milk samples were taken from April to August in order to evaluate milk production, composition, and fatty acid profile. In addition, blood samples were taken for evaluating the effects of linseed supplementation on goats’ health status. The health status of the goats was not influenced by the linseed supplementation, as confirmed by blood analyses. Concerning the effects on milk, the supplementation positively affected (p < 0.001) milk production and fat percentage and the fatty acid profile was markedly influenced by the lipid supplementation. In particular, milk from the LIN group was characterized by significantly lower concentrations of saturated fatty acids (FA; p < 0.001) and higher proportions of monounsaturated FA, polyunsaturated FA, and conjugated linoleic acids (CLAs) than milk from the CTR group (p < 0.001). In contrast, the OBCFA were negatively influenced by the linseed supplementation (p < 0.0001). Further studies are needed to test the effects of different fat sources and other nutrients on the diets. Full article

This review highlights the importance of odd and branched chain fatty acids (OBCFAs) and dietary factors that may affect the content of milk OBCFAs in dairy cows. Historically, OBCFAs in cow milk had little significance due to their low concentrations compared to other milk fatty acids (FAs). The primary source of OBCFAs is ruminal bacteria. In general, FAs and OBCFAs profile in milk is mainly affected by dietary FAs and FAs metabolism in the rumen. Additionally, lipid mobilization in the body and FAs metabolism in mammary glands affect the milk OBCFAs profile. In cows, supplementation with fat rich in linoleic acid and α-linolenic acid decrease milk OBCFAs content, whereas supplementation with marine algae or fish oil increase milk OBCFAs content. Feeding more forage rather than concentrate increases the yield of some OBCFAs in milk. A high grass silage rate in the diet may increase milk total OBCFAs. In contrast to saturated FAs, OBCFAs have beneficial effects on cardiovascular diseases and type II diabetes. Furthermore, OBCFAs may have anti-cancer properties and prevent Alzheimer’s disease and metabolic syndrome. Full article

The inverse association between the groups of odd-chain (OCFA) and branched-chain (BCFA) and the development of diseases in humans have generated interest in the scientific community. In experiment 1, the extent of the passage of odd- and branched-chain fatty acids (OBCFA) from milk fat to fresh cheese fat was studied in sheep and goats. Milk collected in two milk processing plants in west Sardinia (Italy) was sampled every 2 weeks during spring (March, April and May). In addition, a survey was carried out to evaluate the seasonal variation of the OBCFA concentrations in sheep and goats’ cheeses during all lactation period from January to June. Furthermore, to assess the main differences among the sheep and goat cheese, principal component analysis (PCA) was applied to cheese fatty acids (FA) profile. Concentrations of OBCFA in fresh cheese fat of both species were strongly related to the FA content in the unprocessed raw milk. The average contents of OBCFA were 4.12 and 4.13 mg/100 mg of FA in sheep milk and cheese, respectively, and 3.12 and 3.17 mg/100 mg of FA in goat milk and cheese, respectively. The OBCFA concentration did no differed between milk and cheese in any species. The content of OBCFA was significantly higher in sheep than goats’ dairy products. The OBCFA composition of the cheese was markedly affected by the period of sampling in both species: odd and branched FA concentrations increased from March to June. The seasonal changes of OBCFA in dairy products were likely connected to variations in the quality of the diet. The PCA confirmed the higher nutritional quality of sheep cheese for beneficial FA, including OBCFA compared to the goat one, and the importance of the period of sampling in the definition of the fatty acids profile. There is a growing interest of researchers in meat authentication in terms of geographical and dietary background of animals, and several analytical methods have been proposed for the purpose of investigating this. We hypothesized that the odd- and branched-chain fatty acid (OBCFA) profile in intramuscular fat (IMF) might suffice to distinguish lamb meat entering the food chain supply on the basis of the type of diet fed to lambs during the fattening period. A total of 30 individual OBCFA profiles, quantified by gas chromatography, of IMF of Manchego lambs were used. During the fattening period (42 days), the lambs were fed three diets differing in concentrate composition: (i) Control, concentrate typical of commercial fattening rations, rich in starch and based on cereals and soybean meal; (ii) Camelina, similar to Control but replacing 50% of the soybean meal with camelina meal; and (iii) Fibrous, concentrate rich in neutral detergent fiber (NDF), based on fibrous by-products and not including cereals nor soybean meal. The OBCFA were grouped into three classes (linear odd, iso and anteiso fatty acids) and were then submitted to a linear discriminant analysis, using the feeding treatments as grouping variable and the OBCFA class contents in IMF as quantitative variables. The results suggested that a high NDF to starch ratio of the concentrate, being the lowest for Control (CON) treatment and the highest for Fibrous (FIB) treatment, would be negatively related to the odd/anteiso ratio and positively related to the iso/(anteiso+odd) FA ratio in IMF. Determination of OBCFA profile in lamb meat would be useful to monitor the feeding regime (starch- or NDF-rich) of lambs entering the food chain supply. Full article

The aim of this study was to evaluate the relationship of milk odd- and branched-chain fatty acids (OBCFA) with urinary purine derivates and estimated ruminal microbial crude protein (MCP) synthesis. Forty-four lactating Holstein cows were used in a 4 × 4 Latin square design with 21-day periods comprised of a 13-day adaptation phase to diet followed by an 8-day sampling phase. Differences in estimated MCP yield and milk OBCFA composition were found by feeding total mixed rations containing forage (maize silage, MS; red clover silage, RCS) and concentrates (0.75:0.25) with targeted proportions of RCS to MS of 0.15:0.60, 0.30:0.45, 0.45:0.30, and 0.60:0.15 on a dry matter basis. The MCP was estimated from the total urinary purine derivate (PD) excretion (MCPPD) and intakes of metabolizable energy (MCPME) or digestible organic matter (MCPdOM). The Pearson correlations of individual OBCFA with urinary parameters (uric acid, allantoin, PD and nitrogen) were generally weak (r = −0.37 to 0.55). Yields of individual OBCFA correlated positively with MCPME and MCPdOM (r = 0.21 to 0.55). The prediction of urinary PD concentration was moderate (R2 = 0.64) when including the proportion of iso-C17:0. The prediction of total PD excretion was low (R2 = 0.21) with yields of iso-C15:0, anteiso-C17:0, and iso-C16:0. The prediction of MCPPD was high (R2 = 0.99) when including the iso-C16:0 and cis-9 C17:1 concentrations, while those of MCPME and MCPdOM were low (R2 = 0.37 and 0.36, respectively) when including yields of iso-C15:0, cis-9 C17:1, and iso-C18:0. The correlations and regression analyses demonstrate that the estimated MCP synthesis and urinary PD excretion can be only moderately predicted by yields and concentrations of individual or total OBCFA in cow’s milk. However, milk OBCFA can still be seen as a promising, non-invasive method for predicting rumen function and microbial protein supply in dairy cows because MCP flow was not directly measured in this study but instead indirectly estimated probably comprising considerable deviations of the assumed values from the true ones. The purpose of this research was to evaluate whether relationships exist between odd- and branched-chain fatty acids (OBCFAs) originating from milk fat and the corresponding data of ruminal fermentation parameters, microbial populations, and base contents that were used to mark microbial protein in rumen. Nine lactating Holstein dairy cows with similar body weights and parity were selected in this study, and the samples of rumen and milk were collected at the early, middle, and late stages, respectively. The rumen and milk samples were collected over three consecutive days from each cow, and the ruminal and milk OBCFA profiles, ruminal fermentation parameters, bacterial populations, and base contents were measured. The results showed that the concentrations of OBCFAs, with the exception of C11:0 and C15:0, were significantly different between milk and rumen (p < 0.05). The concentrations of anteiso-fatty acids in milk were higher than those in rumen, and the contents of linear odd-chain fatty acids were higher than those of branched-chain fatty acids in both milk and rumen. Significant relationships that existed between the concentrations of C11:0, iso-C15:0, anteiso-C15:0, C15:0, and anteiso-C17:0 in rumen and milk (p < 0.05). The total OBCFA content in milk was positively related to the acetate molar proportion but negatively correlated with isoacid contents (p < 0.05). The populations of Ruminococcus albus, R. flavefacients, and Eubacterium ruminantium were significantly related to milk C13:0 contents (p < 0.05). The adenine/N ratio was negatively related to milk OBCFA content (p < 0.05) but positively associated with the iso-C15:0/iso-C17:0 ratio (p < 0.05). Milk OBCFAs were significantly correlated with ruminal fermentation parameters, ruminal bacterial populations, and base contents. Milk OBCFAs had the potential to predict microbial nitrogen flow, and the prediction equations for ruminal microbial nitrogen flow were established for OBCFAs in dairy milk. Full article

According to the knowledge that the composition in fatty acids of milk is related to the production system, we determined the fatty acid composition of goat milk yielded in three different Italian farms. Two low-input system farms; one organic (LI-O) and one conventional (LI-C), and one high-input system conventional farm (HI-C) were involved in the study. Significant differences were detected among the different groups considering the fatty acid pattern of milk. Fatty acids (FA) strictly related to the rearing system, such as odd and branched chain fatty acids (OBCFA), linoleic acid (LA, 18:2 n6), alpha-linolenic acid (ALA, 18:3 n3), elaidic acid (EA, 18:1 n9), total n6 and total n3 FA, were identified as the most significant factors in the characterization of samples coming from low- or high-input systems. OBCFA amounts were found to be higher (p < 0.05) in the LI-O milk (4.7%), followed by the LI-C milk (4.5%) and then by the HI-C milk (3.4%). The same trend was observed for Σn3 FAs, mainly represented by ALA (0.72%–0.81% in LI-O systems and 0.41% in HI-system), and the opposite for Σn6 FAs, principally represented by LA (2.0%–2.6% in LI-systems and 3.1% in HI-system). A significant (p < 0.01) discrimination among samples clusters coming from the different systems was allowed by the principal component analysis (PCA). Full article
