Seed germination is a critical phase in the plant lifecycle of Camellia oleifera (oil tea), directly influencing seedling establishment and crop reproduction. In this study, we examined transcriptomic and physiological changes across five defined germination stages (G0–G4), from radicle dormancy to cotyledon emergence. Using RNA sequencing (RNA-seq), we assembled 169,652 unigenes and identified differentially expressed genes (DEGs) at each stage compared to G0, increasing from 1708 in G1 to 10,250 in G4. Functional enrichment analysis revealed upregulation of genes associated with cell wall organization, glucan metabolism, and Photosystem II assembly. Key genes involved in cell wall remodeling, including cellulose synthase (CESA), phenylalanine ammonia-lyase (PAL), 4-coumarate-CoA ligase (4CL), caffeoyl-CoA O-methyltransferase (COMT), and peroxidase (POD) showed progressive activation during germination. A Kyoto Encyclopedia of Genes and Genomes (KEGG) pathway analysis revealed dynamic regulation of phenylpropanoid and flavonoid biosynthesis, photosynthesis, carbohydrate metabolism, and hormone signaling pathways. Transcription factors such as indole-3-acetic acid (IAA), ABA-responsive element binding factor (ABF), and basic helix–loop–helix (bHLH) were upregulated, suggesting hormone-mediated regulation of dormancy release and seedling development. Physiologically, cytokinin (CTK) and IAA levels peaked in G4, antioxidant enzyme activities were highest in G2, and starch content increased toward later stages. These findings provide new insights into the molecular mechanisms underlying seed germination in C. oleifera and identify candidate genes relevant to rootstock breeding and nursery propagation. As a unique woody oil crop in China, Camellia oleifera Abel. germplasm resources show significant genetic diversity in Ya’an City. This study measured 60 phenotypic traits (32 quantitative, 28 qualitative) of 302 accessions to analyze phenotypic variation, establish a classification system, and screen high-yield, high-oil germplasms. The phenotypic diversity index for fruit (H’ = 1.36–1.44) was significantly higher than for leaf (H’ = 1.31) and flower (H’ < 1), indicating genetic diversity concentrated in reproductive traits, suggesting potential genetic variability in these traits. Fruit quantitative traits (e.g., single fruit weight CV = 35.37%, fresh seed weight CV = 38.93%) showed high genetic dispersion. Principal component analysis confirmed the fruit factor and economic factor as main phenotypic differentiation drivers. Quantitative traits were classified morphologically, and correlation analysis integrated them into 13 key indicators classified using LSD and range methods. Finally, TOPSIS evaluation selected 10 excellent germplasms like TQ122 and TQ49, with fruit weight, fresh seed yield, and kernel oil content significantly exceeding the population average. This study provides data for C. oleifera DUS test guidelines and proposes a multi-trait breeding strategy, supporting high-yield variety selection and germplasm resource protection. Full article

Oil-tea (Camellia oleifera), a typical oilseed tree, produces high-quality edible vegetable oils that contain rich unsaturated fatty acids and diverse lipid-soluble active compounds such as squalene. Although squalene biosynthesis and its molecular regulation have been studied in several plants, the molecular mechanisms underlying squalene biosynthesis in oil-tea seeds remain uncertain. We investigated and determined squalene accumulation with seed development. We conducted comparative transcriptomic analyses using the RNA-seq technique at the early, fast biosynthesis, and late stages of squalene accumulation with oil-tea seed development and identified 13 squalene biosynthesis key enzyme genes (such as CoHMGR_4, CoAACT_2, CoFPS_1, and CoFPS_2) in developing oil-tea seeds. According to whether the expressions of key enzyme genes were associated with squalene accumulation we found that the precursor IPP of squalene biosynthesis obtained via the MVA pathway was dominant with oil-tea seed development. Based on the gene co-expression analyses, we identified multiple transcription factors potentially involved in regulating squalene biosynthesis such as CoMYC2, CoREM39, CobZIP5, CoERF and CoWRKY. Using yeast one-hybrid and dual-luciferase assay experiments we demonstrated that the transcription factor CoMYC2 could activate the expression of a key enzyme gene CoHMGR_4, suggesting that CoMYC2 might be a critical regulator during squalene biosynthesis in oil-tea seed development. This study gives not only insights into understanding the molecular basis of squalene biosynthesis in oil-tea developing seeds but also provides gene resources for developing genetically improved varieties with higher content of squalene in oil-tea. To address the low shelling rate and high seed breakage in existing oil tea fruit shelling devices, a novel laser scoring device was designed for fresh Camellia oleifera fruits. Experimental studies were conducted to optimize the key parameters of the custom-built laser scoring machine, aiming to improve scoring qualification rates. Through single-factor tests and response surface methodology, a regression model was developed to characterize the relationship between the scoring qualification rate and the following three variables: conveyor speed (12 mm/s), laser power (97 W), and defocusing distance (10 mm). The study revealed interactive effects among these parameters. After optimization and verification under ideal conditions, the device achieved a peak average qualification rate of 85.6%. Full article

Camellia oleifera is a unique woody edible oil tree species in China, and the ovule development affects the yield of seeds. This study selected three different types of C. oleifera clones and used LC-MS, RNA-seq, and other techniques to compare the endogenous hormone contents, gene expression levels, and metabolite changes between normal and aborted ovules. The results showed that high levels of ABA, JA, and SA may lead to the phenotype of ovule abortion. A total of 270 differential metabolites were identified in the metabolome, with L-methionine, citrulline, L-tryptophan, L-phenylalanine, and indolepyruvate being downregulated to varying degrees in the aborted ovules. Genes involved in plant hormone synthesis and response, such as GH3.1, IAA14, PIN1, AUX22, ARF1_2, BZR1_2, GA2ox, ERFC3, ABF2, and PYL8, responded to ovule development. This study elucidates the physiological, metabolic, and transcriptional responses to ovule abortion, providing a theoretical basis for understanding ovule development and yield regulation in C. oleifera. Tea oil is an important high-quality edible oil derived from woody plants. Camellia oleifera is the largest and most widely planted oil-producing plant in the Camellia genus in China, and its seeds are the most important source for obtaining tea oil. In current research, improving the yield and quality of tea oil is the main goal of oil tea genetic breeding. The aim of this study was to investigate the degree of genetic variation in an early crossing population of C. oleifera and identify single nucleotide polymorphisms (SNPs) and genes significantly associated with fruit traits, which can provide a basis for marker-assisted selection and gene editing for achieving trait improvement in the future. In this study, we selected a crossing population of approximately 40-year-old C. oleifera with a total of 330 samples. Then, ddRAD sequencing was used for SNP calling and population genetic analysis, and association analysis was performed on fruit traits measured repeatedly for two consecutive years. The research results indicate that over 8 million high-quality SNPs have been identified, but the vast majority of SNPs occur in intergenic regions. The nucleotide polymorphism of this population is at a low level, and Tajima’s D values are mostly greater than 0, indicating that the change in this population was not suitable for the model of central evolution. The population structure analysis shows that the population has seven theoretical sources of genetic material and can be divided into seven groups, and the clustering analysis results support the population structure analysis results. Association analysis identified significant SNPs associated with genes related to the seed number of a single fruit and seed kernel oil content. Our findings provide a basis for molecular breeding and future genetic improvement of cultivated oil tea. Full article

Soil physicochemical properties, root characteristics, and fruit economic traits were determined in Camellia oleifera plantations under spontaneous vegetation + mowing (W1), spontaneous vegetation + glyphosate (W2), and no weeding (CK) treatments. Compared with CK, W1 reduced soil bulk density and increased total nitrogen, total phosphorus, ammonium nitrogen, nitrate nitrogen, and effective potassium content. W2 treatment resulted in higher bulk density than W1 and lower water-holding capacity, total nitrogen, total phosphorus, total potassium, ammonium nitrogen, nitrate nitrogen, and available potassium of the soil. Generally, both W1 and W2 inhibited weed morphological traits while favoring the C. oleifera root system, with the W1 treatment resulting in the greatest increase. Fruit transverse diameter, longitudinal diameter, yield, and oil yield were higher in W1 than in CK and W2 treatments. Weed root systems and C. oleifera root systems ultimately affect oil production and yield by affecting bulk density, ammonium nitrogen, nitrate nitrogen, fruit transverse diameter, seed yield, and seed kernel oil content. In summary, W1 treatment improved the physicochemical properties, root growth, fruit growth, and soil quality in C. oleifera plantations. Full article

Camellia oleifera C. Abel is a woody oil crop with multiple purposes. This study aims to investigate the mutagenic effects of 60Co-γ radiation on C. oleifera seedlings and to screen anthracnose-resistant mutants. Two C. oleifera varieties were investigated: ‘Xianglin 1’ (XL1) and ‘Xianglin 210’ (XL210). Seeds were irradiated with 0 Gy, 30 Gy, 50 Gy, and 80 Gy of 60Co-γ, and after one year of planting, the mutagenic lines were studied, and disease-resistant mutants were screened. Results showed that as the radiation intensity was increased, the emergence percentage of both C. oleifera XL210 and XL1 was significantly decreased. Radiation significantly changed the SOD and POD activities in both varieties. Furthermore, 80 Gy irradiated lines showed reduced anthracnose resistance in both varieties. However, 50 Gy irradiated lines showed enhanced disease resistance in XL210 while reducing it in XL1. The 30 Gy irradiated lines did not affect the disease resistance of either variety. Colletotrichum gloeosporioides infection tests were conducted on 94 mutant C. oleifera seedlings, resulting in 8 highly resistant mutants (A3, A8, A10, A19, A21, A32, A35, B17) and 3 susceptible mutants (A4, B15, B27) in XL210 and XL1. Differences in SOD and POD activities led to variations in disease resistance among different mutants. Additionally, the expression levels of CoSOD1, CoPOD, CoIDD4, and CoWKRY78 were varied among the different mutants. This study delivers the screening of disease-resistant mutants in C. oleifera through mutagenic breeding, providing material for the development of new C. oleifera varieties and serving as a resource for further research in mutagenic breeding. In this study, we conducted tests on the seeds from four Taiwanese native Camellia species (C. japonica, C. furfuracea, C. laufoshanensis, and C. formosensis) and three commercialized species (C. oleifera, C. brevistyla, and C. sinensis) for comparison. We examined various aspects of these species, such as seed oil content, suitability for mechanical pressing, volatile components (edible flavor), and oil stability (suitability for cooking), to assess the feasibility of using these four native Taiwanese Camellia seeds as sources of edible oil. The results from solvent extraction tests and mechanical pressing experiments confirm that the seeds from C. furfuracea, C. japonica, and C. laufoshanensis have high oil contents, and their oils are suitable for extraction via the popular mechanical pressing method, with oil yields comparable to or higher than those of the commercialized Camellia species. The volatile components of the oils were collected using MonoTrap adsorbents and analyzed with a thermal desorption system coupled with gas chromatography–mass spectrometry (ATD-GC/MS), primarily consisting of alcohols, ketones, and aldehydes. The results of oxidative stability tests reveal that the seed oils from C. japonica, C. furfuracea, and C. laufoshanensis are higher than or equally stable to those from the commercialized Camellia species. After six months of storage, the stability of these three Camellia seed oils remained relatively high, demonstrating that the seed oils from C. japonica, C. furfuracea, and C. laufoshanensis can withstand high temperatures and can be easily preserved for future applications. One of the main reasons for the low yield of Camellia oleifera Abel is the large number of flowers and fruits that fall off before ripening. The aim of this study was to investigate the effect of foliar spraying of potassium dihydrogen phosphate (KH2PO4) on the fruiting characteristics of C. oleifera, and to provide technical support for its flower and fruit preservation and yield increasing. Three C. oleifera cultivars, ‘Changlin 18’, ‘Changlin 166’, and ‘Changlin 40’, were used as experimental materials to conduct foliar spraying experiments with different water concentrations of KH2PO4. The effects of KH2PO4 on the fruit retention rate, fruit properties, and seed oil quality of each cultivar were measured and analyzed. The application of the optimal concentration of KH2PO4 significantly enhanced various fruit quality metrics of three C. oleifera cultivars. Specifically, the total fruit retention rate was increased by 30.02~87.53%, the transverse diameter by 7.36~21.21%, and the longitudinal diameter by 18.56%, and the fruit weight of ‘Changlin 40’ could increase by 83.63%. It also increased dry seed yield by 27.87~80.81%, dry kernel rate by 10.29~30.12%, dry seed oil content by 28.00~29.77%, total unsaturated fatty acids (TUFAs) by 0.63~5.3%, monounsaturated fatty acids (MUFAs) by 0.30~5.37%, and squalene by 0.09~0.14% during the maturing stage. However, camellia cultivars had the different responses to KH2PO4 concentrations. To promote the fruiting of C. oleifera, improve the economic traits of fruits, and enhance the quality of tea oil, the recommended concentrations of KH2PO4 solution are 4.50 g·L−1, 1.50 g·L−1, and 1.50 g·L−1, for mist spraying on the trees of ‘Changlin 18’, ‘Changlin 166’, and ‘Changlin 40’, respectively. For mixed cultivars of C. oleifera which planted randomly, the recommended concentration range of KH2PO4 solution for spraying is 1.50 to 4.50 g·L−1. Full article

Camellia oil is valuable as an edible oil and serves as a base material for a range of high-value products. Camellia plants of significant economic importance, such as Camellia sinensis and Camellia oleifera, have been classified into sect. Thea and sect. Oleifera, respectively. Fatty acid desaturases play a crucial role in catalyzing the formation of double bonds at specific positions of fatty acid chains, leading to the production of unsaturated fatty acids and contributing to lipid synthesis. Comparative genomics results have revealed that expanded gene families in oil tea are enriched in functions related to lipid, fatty acid, and seed processes. To explore the function of the FAD gene family, a total of 82 FAD genes were identified in tea and oil tea. Transcriptome data showed the differential expression of the FAD gene family in mature seeds of tea tree and oil tea tree. Furthermore, the structural analysis and clustering of FAD proteins provided insights for the further exploration of the function of the FAD gene family and its role in lipid synthesis. Overall, these findings shed light on the role of the FAD gene family in Camellia plants and their involvement in lipid metabolism, as well as provide a reference for understanding their function in oil synthesis. Camellia seed oil (CSO), as a nutrient-rich edible oil, is widely used in foods, cosmetics, and other fields. In this work, the extraction, deacidification, decolorization, and deodorization processes of CSO were respectively optimized for meeting injectable oil standards. The results showed that the CSO extraction rate reached the highest level of 94% at optimized conditions (ultrasonic time, 31.2 min; reaction pH, 9.2; and reaction time, 3.5 h). The physicochemical indexes of CSO and 10 other vegetable oils were evaluated by the principal component analysis method, and the overall scores of vegetable oils were ranked as camellia seed oil > olive oil > rice oil > peanut oil > sesame oil > corn oil > soybean oil > sunflower oil > rapeseed oil > walnut oil > flaxseed oil. The physicochemical indicators of CSO were the most ideal among the 11 vegetable oils, which means that CSO is suitable as an injectable oil. Through the optimized processes of the deacidification, decolorization, and deodorization, the CSO acid value was reduced to 0.0515 mg KOH/g, the decolorization rate reached a maximum of 93.86%, and the OD430 was 0.015, meeting the requirement (≤0.045 of OD430) of injectable oil. After the deodorization process, these parameters of the refractive index, acid value, saponification value, iodine value, absorbance, unsaponifiable, moisture and volatiles, fatty acid composition, and heavy metal limits all met the pharmacopoeia standards of injectable oil in many countries and regions. The possibility of CSO as an injectable oil was first verified through refining-process optimization and nutritional index analysis, providing an important technical reference for the high-value utilization of vegetable oil. Variation in Fruit Traits and Seed Nutrient Compositions of Wild Camellia oleifera: Implications for Camellia oleifera Domestication

Camellia oleifera is a woody oil crop with the highest oil yield and the largest cultivation area in China, and C. oleifera seed oil is a high-quality edible oil recommended by the Food and Agriculture Organization of the United Nations. The objectives of this study were to investigate the variation in fruit yield traits and seed chemical compositions of wild C. oleifera in China and to identify the differences between wild C. oleifera and cultivated varieties. In this study, we collected wild C. oleifera samples from 13 sites covering the main distribution areas of wild C. oleifera to comprehensively evaluate 25 quantitative traits of wild C. oleifera fruit and seed chemical compositions and collected data of 10 quantitative traits from 434 cultivated varieties for a comparative analysis of the differences between wild and cultivars. The results showed that the coefficients of variation of the 25 quantitative traits of wild C. oleifera ranged from 2.605% to 156.641%, with an average of 38.569%. The phenotypic differentiation coefficients ranged from 25.003% to 99.911%, with an average of 77.894%. The Shannon–Wiener index (H’) ranged from 0.195 to 1.681. Based on the results of principal component analysis (PCA) and phenotypic differentiation coefficients, 10 traits differed significantly between wild C. oleifera and cultivated varieties, while the differentiation coefficients (VST) for fresh fruit weight, oleic acid, unsaturated fatty acids, stearic acid, and saturated fatty acids were more than 95%, of which fresh fruit weight and oleic acid content were potential domestication traits of C. oleifera. The results of this study can contribute to the efficient excavation and utilization of wild C. oleifera genetic resources for C. oleifera breeding. Full article

Camellia oleifera oil (CO oil) extracted from C. oleifera seeds has a 2300-year consumption history in China. However, there is relatively little research regarding its non-edible uses. This study determined the physicochemical properties of CO oil extracted via direct pressing, identified its main components using GC-MS, and evaluated its antioxidant, moisturizing, and anti-inflammatory activities. The results revealed that CO oil’s acid, peroxide, iodine, and saponification values were 1.06 ± 0.031 mg/g, 0.24 ± 0.01 g/100 g, 65.14 ± 8.22 g/100 g, and 180.41 ± 5.60 mg/g, respectively. CO oil’s tocopherol, polyphenol, and squalene contents were 82.21 ± 9.07 mg/kg, 181.37 ± 3.76 mg/kg, and 53.39 ± 6.58 mg/kg, respectively; its unsaturated fatty acid (UFA) content was 87.44%, and its saturated fatty acid (SFA) content was 12.56%. CO oil also demonstrated excellent moisture retention properties, anti-inflammatory effects, and certain free radical scavenging. A highly stable CO oil emulsion with competent microbiological detection was developed using formulation optimization. Using CO oil in the emulsion significantly improved the formulation’s antioxidant and moisturizing properties compared with those of the emulsion formulation that did not include CO oil. The prepared emulsion was not cytotoxic to cells and could reduce cells’ NO content; therefore, it may have potential nutritional value in medicine and cosmetics. Full article

Camellia oleifera Abel is rich in oil, which is beneficial to human health. Seasonal drought and uneven canopy microclimate distributions result in low yields and picking efficiencies. To reveal the features of fruit development in the canopy and evaluate the impact of pruning on the canopy microclimate and fruit parameters, a field experiment was carried out in Zhuzhou, Hunan. The C. oleifera tree canopy was divided into four positions: inside the upper canopy (UI); inside the lower canopy (LI); outside the upper canopy (UO); and outside the lower canopy (LO). A combination of high-light efficiency tree pruning and mulching with camellia shells and weedproof film (CPM) was tested and compared with mulching (M). Compared with temperature and humidity, canopy light intensity is a key climate factor that affects the development of Camellia oleifera fruits. CPM improved the canopy light distribution and increased the light intensity inside the canopy of C. oleifera. There was no significant difference in the fruit cracking rate among the four canopy positions in the CPM treatment, which indicated that the fruit maturation period tended to be consistent across the canopy positions. Treatment with CPM resulted in 24.14% and 4.11% greater yield and oil content ratios, respectively, than the treatment with M. The fresh weight of the fruits from LO was the lowest, and the fatty acid accumulation was the lowest due to the poor light availability. CMP increased fruit quality, particularly in LI, which was 41.92%, 36.36%, 13.21%, and 12.79% greater in terms of fresh weight, yield, fresh seed rate, and oil content, respectively, than M. Unfortunately, there was no discernible difference in fatty acid components between CPM and M. Our research suggested that pruning combined with the mulching of organic material is an effective way to alleviate seasonal drought and uneven fruit maturation in C. oleifera trees, further increasing yield and fruit quality.
