The Discovery of Potential Repellent Compounds for Zeugodacus cucuribitae (Coquillett) from Six Non-Favored Hosts

Zeugodacus cucuribitae (Coquillett) (Z. cucuribitae) is a global extremely invasive quarantine pest which has a wide host range of fruits and vegetables. At present, there are a few control measures for Z. cucuribitae, and deltamethrin and avermectin are commonly used. Among the hosts of Z. cucuribitae, Luffa acutangular, Luffa cylindrica, Sechium edule, Brassica oleracea var. botrytis, Musa nana, and Fragaria × ananassa are non-favored hosts. However, it is still not clear why these hosts are non-favored and whether there are any repellent components of Z. cucuribitae in these hosts. In this study, the components of these six hosts were collected from the literature, and the genes of odor and chemical sensation were determined from the genome of Z. cucuribitae. After the potential relationships between these components and genes were determined by molecular docking methods, the KEGG and GO enrichment analysis of these genes was conducted, and a complex network of genes vs. components vs. Kegg pathway vs. GO terms was constructed and used to select the key components for experiments. The results show that oleanolic acid (1 mg/mL, 0.1 mg/mL, and 0.01 mg/mL), rotenone (1 mg/mL, 0.1 mg/mL, and 0.01 mg/mL), and beta-caryophyllene oxide (1 mg/mL, 0.1 mg/mL, and 0.01 mg/mL) had a significant repellent effect on Z. cucuribitae, and three components, rotenone (1 mg/mL and 0.1 mg/mL), echinocystic acid (1 mg/mL, 0.1 mg/mL, and 0.01 mg/mL), and beta-caryophyllene oxide (1 mg/mL, and 0.1 mg/mL) had significant stomach toxicity in Z. cucuribitae. Furthermore, a complex signaling pathway was built and used to predict the effect of these components on Z. cucuribitae. These components probably play roles in the neuroactive ligand–receptor interaction (ko04080) and calcium signaling (ko04020) pathways. This study provides a reference for the prevention and control of Z. cucuribitae and a scientific reference for the rapid screening and development of new pest control drugs. Full article

Cabbage (Brassica oleracea L.) is a globally significant vegetable crop that faces productivity challenges due to fungal and bacterial pathogens. This review highlights the potential of spectral imaging techniques, specifically multispectral and hyperspectral methods, in detecting biotic stress in cabbage, with a particular emphasis on pathogen-induced responses. These non-invasive approaches enable real-time assessment of plant physiological and biochemical changes, providing detailed spectral data to identify pathogens before visible symptoms appear. Hyperspectral imaging, with its high spectral resolution, allows for distinctions among different pathogens and the evaluation of stress responses, whereas multispectral imaging offers broad-scale monitoring suitable for field-level applications. The work synthesizes research in the existing literature while presenting novel experimental findings that validate and extend current knowledge. Significant spectral changes are reported in cabbage leaves infected by Alternaria brassicae and Botrytis cinerea. Early-stage detection was facilitated by alterations in flavonoids (400–450 nm), chlorophyll (430–450, 680–700 nm), carotenoids (470–520 nm), xanthophyll (520–600 nm), anthocyanin (550–560 nm, 700–710 nm, 780–790 nm), phenols/mycotoxins (700–750 nm, 718–722), water/pigments content (800–900 nm), and polyphenols/lignin (900–1000). The findings underscore the importance of targeting specific spectral ranges for early pathogen detection. By integrating these techniques with machine learning, this research demonstrates their applicability in advancing precision agriculture, improving disease management, and promoting sustainable production systems. Structural Variations of Broccoli Polyphenolics and Their Antioxidant Capacity as a Function of Growing Temperature

Polyphenolics in plants exist in free, soluble-bound, and insoluble-bound structural forms. The concentration of these structural forms depends on the plant’s developmental stage, tissue type, soil water availability, and food preparation methods. In this study, for the first time, the effects of growth temperature (RT—room temperature—23 °C day/18 °C night, HT—high temperature—38 °C day/33 °C night, LT—low temperature—12 °C day/7 °C night) on variations of polyphenolic structural forms—free, soluble-bound (esterified and glycosylated), and insoluble-bound—in broccoli (Brassica oleracea L. convar. botrytis (L.) Alef. var. cymosa Duch.) microgreens were investigated. Using spectrophotometric, RP-HPLC, and statistical analyses, it was found that the highest amount of total phenolics (TP) in broccoli microgreens was present in the esterified form, regardless of the temperature at which they were grown (63.21 ± 3.49 mg GAE/g dw in RT, 65.55 ± 8.33 mg GAE/g dw in HT, 77.44 ± 7.82 mg GAE/g dw in LT). LT significantly increased the amount of free (from 13.30 ± 2.22 mg GAE/g dw in RT to 18.33 ± 3.85 mg GAE/g dw) and esterified soluble TP (from 63.21 ± 3.49 mg GAE/g dw in RT to 77.44 ± 7.82 mg GAE/g dw), while HT significantly increased the amount of TP glycosylated forms (from 14.85 ± 1.45 mg GAE/g dw in RT to 17.84 ± 1.20 mg GAE/g dw). LT also enhanced free and esterified forms of total flavonoids, tannins, hydroxycinnamic acids, and flavonols. HT, on the other hand, increased glycosylated forms of TP, flavonoids, tannins, hydroxycinnamic acids, flavonols, and phenolic acids, and decreased insoluble-bound tannins. According to the ABTS method, HT induced antioxidant potential of free and glycosylated forms, while LT increased antioxidant capacity of free forms only. According to the FRAP method, LT increased antioxidant potential of free and esterified polyphenolic forms. Also, based on ABTS and FRAP assays, esterified polyphenolics showed significantly higher antioxidant capacity than any other form. Principal component analysis showed that structural form had a greater impact than temperature. Hierarchical clustering showed that RT-, HT- and LT-broccoli microgreens were most similar in their glycosylated polyphenolics, but differed the most in esterified forms, which were also the most distinct overall. In conclusion, HT and LT induced specific shifts in the structural forms of broccoli polyphenolics and their antioxidant capacity. Based on the results, we recommend applying LT to increase the amount of free and esterified polyphenolics in broccoli microgreens, while HT may be used to enhance glycosylated forms. Full article

This is a pioneering study on the main drainage system in Gujranwala District, where untreated mixed wastewater is discharged and subsequently used for vegetable irrigation, leading to potential health and environmental risks. This study seeks to develop the spatial pattern of toxic metal accumulation in soil across an 11 km stretch of land used for vegetable cultivation. By using 90 samples of mixed wastewater and sludge, as well as 10 quadruplicate samples of rhizospheric soils and crops from ten vegetable fields, it was observed that the concentrations of Cr, Cu, Cd, Zn, Fe, Pb, Mg, and Ni in cauliflower (Brassica oleracea var. botrytis L.), coriander (Coriandrum sativum L.), radish (Raphanus sativus L.), mustard (Brassica juncea L.), spinach (Spinacia oleracea L.), meadow clover (Trifolium sp. L.), sorghum (Sorghum bicolour L.), garlic (Allium sativum L.), brinjal (Solanum melongena L.), and mint (Mentha L.) were beyond the permissible limits set by the FAO/WHO, 2001. The declining trend of the toxic metal concentrations in the effluent was Mg > Cr > Ni > Zn > Pb > Cd > Cu > Fe, and in sludge, soil, and plants, it varied in the order of Mg > Fe > Cr > Ni > Zn > Pb > Cd > Cu. Radish, mint, and brinjal had the highest quantities of toxic metals. The spatial pattern of toxic metals was determined by using proximity interpolation, Inverse Distance Weighted (IDW), the fine tuning of the interpolation characteristics, and the kriging of selected sample variograms. Toxic metals were found in the following order: plants > soil > sludge > effluents. The most prevalent cause of metal pollution was soil irrigation with polluted water. This study provides crucial information about the extent of contamination, which could help in the identification of public health risk, the assessment of environmental impacts, and also sustainable water management. The use of nitrification inhibitors (NIs) with nitrogen fertilizers represents an effective strategy to reduce nitrogen loss. In addition, nitrification inhibitors are widely applied to improve agricultural yield. However, it is necessary to continue investigating the crop-specific agricultural practice. In this study, a nitrapyrin-based nitrification inhibitor was used to assess its effects on Brassica oleracea L. var. botrytis growth and on the environment. In a pot experiment, cauliflower plants were grown in fertilized soils based on calcium nitrate (SF) and SF + nitrapyrin. At the end of the experiment, the content of nitrogen compounds in soil and percolation water and the cauliflower yield were determined, and the plant tissues were characterized by Fourier-transform infrared spectroscopy. The application of the NI significantly reduced nitrogen losses, increasing nutrient availability in the soil and the element’s absorption in the plant. Co-application of fertilizers and NIs reduced NO3 leaching from 925 to 294 mg/L. Plant tissue characterization by FTIR spectroscopy highlighted variations in the functional groups in response to the application of NIs. These results suggest that applying nitrogen fertilizer in combination with nitrapyrin can mitigate nitrate pollution and improve element absorption and plant growth. Our research has shown that application methods and cropping systems need to be studied to maximize the effectiveness of nitrapyrin-based NIs. Full article

Cruciferous vegetables of the plant order Brassicales are an attractive dietary component and a valuable source of fiber. However, the nutritional–physiological properties are different when comparing soluble and insoluble fibers. Another significant impact is the transformation of fibers by different influencing factors during food preparation. Cruciferous vegetables, especially, are dominantly processed to soften the matrix. As a result, during cooking, the polysaccharides are dissolved, swelled, or degraded to a certain extent, influencing the composition and the nutritional–physiological properties. The aim of the present study was to analyze the impact of different cooking procedures on changes in the dietary fiber content profile of three different plants: white cauliflower (Brassica oleracea L. var. botrytis), broccoli (B. oleracea L. var. italica), and Brussels sprouts (B. oleracea L. var. gemmifera). The sample material was subjected to direct (“in the water”) and steam cooking. The dietary fiber content and the content of its fractions were determined using an enzymatic analysis method. The results of the research show that the cooking process had a significant influence on the content of dietary fiber fractions in cruciferous vegetables. The concentration of insoluble dietary fiber decreased, whereas the content of soluble dietary fiber increased. When considering the average influence of each process, both steam cooking and direct cooking had a similar impact on changes in the concentrations of dietary fiber fractions. It can therefore be concluded that, when considering dietary fiber content, both processes can be equally well chosen as a thermal treatment for cruciferous vegetables. Full article

Drought and flood (water stress) alter plant metabolism, impacting the phytochemical content and biological effects. Using spectrophotometric, HPLC, and electrophoretic methods, we analyze the effects of water stress on broccoli (Brassica oleracea L. convar. botrytis (L.) Alef. var. cymosa Duch.) sprouts. Drought and flood differently influenced chlorophylls, carotenoids, and porphyrins, with drought having a stronger inhibitory effect on chlorophyll a, total chlorophyll, and porphyrins. Carotenoids and glucosinolates increased under drought but decreased with flooding, suggesting that these compounds play a crucial role in drought tolerance. Nitrate increased with drought from 13.11 ± 1.05 mg/g dw to 22.41 ± 1.20 mg/g dw but decreased under flooding to 5.17 ± 1.03 mg/g dw, and oxalic acid was reduced by drought only (from 48.94 ± 1.30 mg/g dw to 46.43 ± 0.64 mg/g dw). Flood reduced proteins by 29%, phenolics by 15%, flavonoids by 10%, flavonols by 11%, tannins by 36%, and proanthocyanidins by 19%, while drought decreased flavonoids by 23%. Total phenolics and proanthocyanidins were increased by drought by 29% and 7%, respectively, while flooding decreased hydroxycinnamic acids by 13%. Both stress types influenced individual polyphenols differently: drought diminished ferulic acid by 17% and increased sinapic acid by 30%, while flooding reversed these effects and enhanced kaempferol by 22%. These compounds, along with proline (which increased by 139% under drought), emerged as biomarkers of water stress. Flood impacted antioxidant capacity more significantly, while drought-stressed broccoli extracts better protected plasmid DNA against oxidative damage. These findings underline the metabolic plasticity of broccoli sprouts and their potential in targeted crop management for water stress resilience. Full article

Cauliflower (Brassica oleraceae L. var. Botrytis Linnaeus) has various health benefits due to its rich bioactive compound content. However, this fresh vegetable faces challenges related to its perishability and short shelf life. This study explores the effect of five drying methods, namely vacuum drying (VD), convective drying (CD), infrared drying (IRD), low-temperature vacuum drying (LTVD) and vacuum freeze-drying (VFD), on the bioactive compounds and health-promoting properties of cauliflower. Analyses of amino acids, hydroxycinnamic acid and its derivatives, glucosinolates, and isothiocyanates, as well as evaluations of their anti-inflammatory, antiproliferative, and neuroprotective properties, were conducted based on these five drying methods. The results revealed that samples treated with VFD and IRD had a higher content of amino acids involved in GSL anabolism. Moreover, VFD samples retained hydroxycinnamic acid derivatives and glucosinolates to a greater extent than other methods. Nonetheless, the CD and VD samples exhibited higher antiproliferative and neuroprotective effects, which were correlated with their high sulforaphane content. Overall, considering the retention of most bioactive compounds from cauliflower and the topical inflammation amelioration induced in mice, VFD emerges as a more satisfactory option. Full article

Research has been increasingly focusing on the preservation of the biodiversity of vegetable crops under sustainable farming management. An experiment was carried out in southern Italy on Brassica oleracea L. var. botrytis, landrace Gigante di Napoli, to assess the effects of two transplanting times (9 September and 7 October), in factorial combination with five nitrogen–potassium ratios (0.6; 0.8; 1.0; 1.2; and 1.4) on plant growth, yield, and quality of cauliflower heads. A split-plot design was used for the treatment distribution in the field, with three replications. The earlier transplant and the 1.2 N:K ratio led to the highest yield, mean weight, and firmness of cauliflower heads which were not significantly affected by both transplanting time and N:K ratio in terms of colour components. The 1.2 N:K ratio led to the highest head diameter with the earlier transplant, whereas the 1.0 ratio was the most effective on this parameter in the later crop cycle. The highest nitrate, nitrogen, and potassium concentrations in the heads were recorded with the earlier transplanting time. Antioxidant activity, ascorbic acid, and polyphenol content increased with the rise of the N:K ratio. The element use efficiency was constantly negative with the N:K increase for nitrogen and was augmented until the 1.2 ratio for potassium. The results of our investigation showed that the optimal combination between transplanting time and N:K ratio is a key aspect to improve head yield and quality of the cauliflower landrace Gigante di Napoli, under the perspective of biodiversity safeguarding and valorisation. Climate change causes shifts in temperature patterns, and plants adapt their chemical content in order to survive. We compared the effect of low (LT) and high (HT) growing temperatures on the phytochemical content of broccoli (Brassica oleracea L. convar. botrytis (L.) Alef. var. cymosa Duch.) microgreens and the bioactivity of their extracts. Using different spectrophotometric, LC-MS/MS, GC-MS, and statistical methods, we found that LT increased the total phenolics and tannins in broccoli. The total glucosinolates were also increased by LT; however, they were decreased by HT. Soluble sugars, known osmoprotectants, were increased by both types of stress, considerably more by HT than LT, suggesting that HT causes a more intense osmotic imbalance. Both temperatures were detrimental for chlorophyll, with HT being more impactful than LT. HT increased hormone indole-3-acetic acid, implying an important role in broccoli’s defense. Ferulic and sinapic acid showed a trade-off scheme: HT increased ferulic while LT increased sinapic acid. Both stresses decreased the potential of broccoli to act against H2O2 damage in mouse embryonal fibroblasts (MEF), human keratinocytes, and liver cancer cells. Among the tested cell types treated by H2O2, the most significant reduction in ROS (36.61%) was recorded in MEF cells treated with RT extracts. The potential of broccoli extracts to inhibit α-amylase increased following both temperature stresses; however, the inhibition of pancreatic lipase was increased by LT only. From the perspective of nutritional value, and based on the obtained results, we conclude that LT conditions result in more nutritious broccoli microgreens than HT. Full article

Cabbage maggot, Delia radicum (L.) (Diptera: Anthomyiidae) is a serious pest of Brassica such as broccoli (Brassica oleracea var. italica Plenck) and cauliflower (B. oleracea L. var. botrytis) in California’s Central Coast. Since there are limited non-chemical options available for growers to manage D. radicum, there is an urgent need to develop alternative tactics. The objective of this study was to determine the effects of side-by-side plantings of turnip (Brassica rapa var. rapa L.), lettuce (Lactuca sativa L.), cauliflower, and cabbage (B. oleracea L. var. capitata) with broccoli on D. radicum infestation. In 2013 and 2014, the experiments were conducted in Salinas, California. Significantly greater numbers of eggs and larval feeding damage were found on turnip compared with broccoli. Lettuce (Asteraceae), a non-Brassica crop, was compared with broccoli; however, lettuce did not reduce oviposition or larval feeding damage on broccoli. The larval feeding damage on cauliflower was significantly lower than on broccoli when planted side-by-side. The effects on cabbage were not significantly different from broccoli in terms of oviposition and larval feeding damage. This new information generated from the Central Coast of California will be further utilized to develop a trap crop to effectively tackle the D. radicum problem in Brassica fields. Full article

The goal of this work was to define resistant and susceptible variables of young broccoli (Brassica oleracea L. convar. botrytis (L.) Alef. var. cymosa Duch.) plants treated with cold and hot water. Additionally, we wanted to single out variables that could potentially be used as biomarkers of cold/hot-water stress in broccoli. Hot water changed more variables (72%) of young broccoli than cold water (24%) treatment. Hot water increased the concentration of vitamin C for 33%, hydrogen peroxide for 10%, malondialdehyde for 28%, and proline for 147%. Extracts of broccoli stressed with hot water were significantly more efficient in the inhibition of α-glucosidase (65.85 ± 4.85% compared to 52.00 ± 5.16% of control plants), while those of cold-water-stressed broccoli were more efficient in the inhibition of α-amylase (19.85 ± 2.70% compared to 13.26 ± 2.36% of control plants). Total glucosinolates and soluble sugars were affected by hot and cold water in an opposite way, which is why they could be used as biomarkers of hot/cold-water stress in broccoli. The possibility of using temperature stress to grow broccoli enriched with compounds of interest to human health should be further investigated. Full article

High temperature (HT) causes physiological and biochemical changes in plants, which may influence their nutritional potential. This study aimed to evaluate the nutritional value of broccoli seedlings grown at HT on the level of phytochemicals, macro- and microelements, antioxidant capacity, and their extracts’ in vitro cytotoxicity. Total phenols, soluble sugars, carotenoids, quercetin, sinapic, ferulic, p-coumaric, and gallic acid were induced by HT. Contrarily, total flavonoids, flavonols, phenolic acids, hydroxycinnamic acids, proteins, glucosinolates, chlorophyll a and b, and porphyrins were reduced. Minerals As, Co, Cr, Hg, K, Na, Ni, Pb, Se, and Sn increased at HT, while Ca, Cd, Cu, Mg, Mn, and P decreased. ABTS, FRAP, and β-carotene bleaching assay showed higher antioxidant potential of seedlings grown at HT, while DPPH showed the opposite. Hepatocellular carcinoma cells were the most sensitive toward broccoli seedling extracts. The significant difference between control and HT-grown broccoli seedling extracts was recorded in mouse embryonal fibroblasts and colorectal carcinoma cells. These results show that the temperature of seedling growth is a critical factor for their nutritional value and the biological effects of their extracts and should definitely be taken into account when growing seedlings for food purposes. The gene flow from Brassica oleracea L. wild relatives to B. oleracea vegetable crops have occurred and continue to occur ordinarily in several Mediterranean countries, such as Sicily, representing an important hot spot of diversity for some of them, such as broccoli, cauliflower and kale. For detecting and for exploiting the forgotten alleles lost during the domestication processes of the B. oleracea crops, attention has been pointed to the individuation of specific markers for individuating genotypes characterized by hypertrophic inflorescence traits by the marker assisted selection (MAS) during the first plant growing phases after the crosses between broccoli (B. oleracea var. italica)/cauliflower (B. oleracea var. botrytis) with B. oleracea wild relatives (n = 9), reducing the cultivation and evaluation costs. The desired traits often found in several B. oleracea wild relatives are mainly addressed to improve the plant resistance to biotic and abiotic stresses and to increase the organoleptic, nutritive and nutraceutical traits of the products. One of the targeted traits for broccoli and cauliflower breeding is represented by the inflorescences size as is documented by the domestication processes of these two crops. Based on the previous results achieved, the numerical matrix, obtained utilizing five simple sequence repeats (SSRs), was analyzed to assess the relationship among the main inflorescence characteristics and the allelic variation of the SSRs loci analyzed (BoABI1, BoAP1, BoPLD1, BoTHL1 and PBCGSSRBo39), both for the Brassica oleracea and B. oleracea wild relatives (n = 9) accessions set. The main inflorescence morphometric characteristics, such as weight, height, diameter, shape, inflorescence curvature angle and its stem diameter, were registered before the flower anthesis. We analyzed the correlations among the allelic variation of the SSRs primers utilized and the inflorescence morphometric characteristics to individuate genomic regions stimulating the hypertrophy of the reproductive organ. The relationships found explain the diversity among B. oleracea crops and the B. oleracea complex species (n = 9) for the inflorescence size and structure. The individuated markers allow important time reduction during the breeding programs after crossing wild species for transferring useful biotic and abiotic resistances and organoleptic and nutraceutical traits to the B. oleracea crops by MAS. Studies on the diversity within and among cabbage (Brassica oleracea L. var. capitata L.), cauliflower (Brassica oleracea var. botrytis), and Chinese cabbage (Brassica rapa L. ssp. pekinensis) variants are essential for the development of healthy diets. However, most studies on them have been limited to a single species, with little integrated analysis between them. In this study, the diversity within and among these species and varieties is assessed by determining the contents of 15 major characteristic nutrients, antioxidants, and minerals in 12 varieties of cabbage, 9 varieties of cauliflower, and 12 varieties of Chinese cabbage cultivated under the same conditions. The results show that there are significant differences in the compositional distributions of cabbage, cauliflower, and Chinese cabbage. Cabbage has the highest contents of soluble sugars (27.73 mg·kg−1 FW), flavonoids (5.90 mg·g−1 FW), and Fe (46.90 mg·kg−1 DW). Cauliflower is an ideal source of soluble protein (603.04 mg·kg−1 FW), polyphenols (1.53 mg·g−1 FW), glucosinolates (25.27 μmol·g−1 FW), and Cu (4.25 mg·kg−1 DW). Chinese cabbage is rich in vitamin C (0.45 mg·g−1 FW) and minerals (K, Ca, Mg, P, Mn, and Zn, at 9206.67 mg·kg−1 DW, 3297.00 mg·kg−1 DW, 3322.79 mg·kg−1 DW, 5614.78 mg·kg−1 DW, 15.36 mg·kg−1 DW, and 21.87 mg·kg−1 DW, respectively). There is a correlation between the quality, antioxidant properties, and minerals of the three species. In principal component analysis, a wide distribution of cabbage varieties and a high degree of overlap with the confidence ellipse of cauliflower are observed, indicating that certain cabbage varieties share compositional characteristics with cauliflower. These findings provide a reference for selecting varieties with higher nutritional value and antioxidant properties, as well as breeding new varieties. Full article
