Virulence Genes and Antimicrobial Resistance Profiles in Aeromonas hydrophila and Aeromonas dhakensis Isolated from the Brazilian Food Chain

Aeromonas hydrophila and A. dhakensis are ubiquitous microorganisms, widespread in aquatic environments, and can cause severe infections in humans and animals. This study aimed to determine the diversity of virulence genes aerA, hlyA, act, and alt through polymerase chain reaction and the antimicrobial resistance through disk diffusion test of 101 A. hydrophila and 34 A. dhakensis strains from environmental, animal, and human sources gathered between 2016 and 2019 at the National Reference Laboratory for Enteric Diseases. Overall, the virulence gene distribution was act in 35.5% of the samples, alt in 40.7%, aerA in 42.2%, and hlyA in 44.5%. Our results revealed that 76.3% of the 135 isolated Aeromonas exhibited at least one of the genes above. 76.3% of A. hydrophila and 76.5% of A. dhakensis exhibited virulence genes distributed among 15 and 12 virulence profiles, respectively. Antimicrobial resistance was observed in 86% of the strains (87.1% in A. hydrophila and 82.4% in A. dhakensis), with higher rates of resistance to Nalidixic acid (69.3%), Imipenem (31.1%), and Sulfamethoxazole-trimethoprim (15.5%). The occurrence of virulence genes and antimicrobial resistance in A. hydrophila and A. dhakensis from different sources indicates their diversity and pathogenicity, reinforcing that they can be a potential health risk source. To minimize the deleterious effects of oxidative stress and improve oocyte competence, we assessed the impact of melatonin during in vitro pre-maturation (pre-IVM) in bovine cumulus–oocyte complexes (COCs). We compared three groups: control (conventional IVM), pre-IVM control (without melatonin), and pre-IVM + MTn (with melatonin). The analyses included levels of reactive oxygen species (ROS), mitochondrial activity, oocyte lipid content, and the expression of genes related to oxidative stress and lipid metabolism in oocytes and cumulus cells. We also examined embryo quality by evaluating kinetics of development and gene expression. The pre-IVM + MTn group exhibited an increase (p ≤ 0.05) in ROS levels and a decrease (p ≤ 0.05) in lipid content, while maintaining mitochondrial activity similar (p > 0.05) to that of the control group. Regarding gene expression, the effect of pre-IVM, independent of melatonin, was characterized by a decrease in FABP3 transcripts in cumulus cells and reductions in GSS and NFE2L2 transcripts in oocytes (p ≤ 0.05). The pre-IVM + MTn group also displayed a decrease (p ≤ 0.05) in CAT and SOD2 transcript levels. In terms of embryonic development, the pre-IVM + MTn group achieved a higher blastocyst rate on D7 (p ≤ 0.05) compared to the control group (30.8% versus 25.8%), but with similar rates (p > 0.05) to the pre-IVM control group (30.8% versus 35.9%). However, there was a decrease in the levels of the PLAC8 transcript. This study indicates that, under the conditions tested, melatonin did not significantly benefit oocyte competence. Full article

Illegal logging is a major threat to tropical forests; however, control mechanisms and efforts to combat illegal logging have not effectively curbed fraud in the production chain, highlighting the need for effective methods to verify the geographic origin of timber. This study investigates the application of multi-elemental analysis combined with Principal Component Analysis (PCA) to discriminate the provenance of tropical timber in the Brazilian Legal Amazon. Wood samples of Hymenaea courbaril L. (Jatobá), Handroanthus sp. (Ipê), and Manilkara huberi (Ducke) A. Chevalier. (Maçaranduba) were taken from multiple sites. Elemental concentrations were determined via Inductively Coupled Plasma Mass Spectrometry (ICP-MS), and CA was applied to evaluate geographic differentiation. Significant differences in elemental profiles were found among locations, particularly when using the intermediate disk portions (25% to 75%), and especially the average of all five sampled portions, which proved most effective in geographic discrimination of the trunk. Elements such as Ca, Sr, Cr, Cu, Zn, and B were especially important for spatial discrimination. These findings underscore the forensic potential of multi-elemental wood profiling as a tool to support law enforcement and environmental monitoring by providing scientifically grounded evidence of timber origin. A brief review of various existing mathematical formulations of the uncertainty relations in quantum mechanics, containing variances of two or more non-commuting operators, is given. In particular, inequalities for the products of higher-order moments of a coordinate and a momentum are considered, as well as inequalities making the uncertainty relations more accurate when additional information about a quantum system is available (for example, the correlation coefficient or the degree of mixing of a quantum state characterized by the trace of the squared statistical operator). We investigate whether a belief in trend continuation, often associated with the so-called “hot-hand effect,” can be endogenously triggered by personal performance feedback in a controlled financial experiment. Participants allocated funds across assets with randomly generated prices, under conditions of known probabilities and varying levels of risk. In a two-stage setup, participants were first exposed to random price sequences to learn the task and potentially develop perceptions of personal success. They then faced additional price paths under incentivized conditions. Our findings show that participants initially increased purchases following gains—consistent with a feedback-driven belief in momentum—but this pattern faded over time. When facing sustained losses, loss aversion dominated decision-making, overriding early optimism. These results highlight how cognitive heuristics and emotional biases interact dynamically, suggesting that belief in trend continuation is context-sensitive and constrained by the reluctance to realize losses. Background/Objectives: Actinobacteria are one of the largest bacterial phyla. These microbes produce bioactive compounds, such as antifungals, antibiotics, immunological modulators, and anti-tumor agents. Studies on actinobacteria isolated from the Brazilian Savannah biome (Cerrado) are scarce and mostly address metagenomics or the search for hydrolytic enzyme-producing microbes. Solanum lycocarpum (lobeira) is a tree widely employed in regional gastronomy and pharmacopeia in Central Brazil. Methods: In this work, 60 actinobacteria isolates were purified from the rhizosphere of S. lycocarpum. Eight Streptomyces spp. isolates were selected for in vitro antifungal activity against Cryptococcus neoformans H99, the C. neoformans 89-610 fluconazole-tolerant strain, C. gattii NIH198, Candida albicans, C. glabrata, and C. parapsilosis. The ability of the aqueous extracts of the isolates to induce the in vitro secretion of tumor necrosis factor (TNF-α), nitric oxide (NO), interleukin-6 (IL-6), and IL-10 by murine macrophages was also evaluated. Results: All extracts showed antifungal activity against at least two yeast species. Streptomyces spp. LAP11, LDB2, and LDB17 inhibited C. neoformans growth by 40–93%. Most extracts (except LAP2) also inhibited C. gattii. None inhibited C. albicans, but all inhibited C. glabrata (40–90%). Streptomyces sp. LAP8 extract increased nitric oxide production by approximately 347-fold in murine macrophages, while LDB11 extract suppressed LPS-induced TNF-α production by 70% and simultaneously increased IL-10 secretion, suggesting immunosuppressive potential. Conclusions: The results revealed that Cerrado actinobacteria-derived aqueous extracts are potential sources of antifungal and immunomodulatory biocompounds. Full article

The soil-borne fungi, Setophoma terrestris and Fusarium spp., are often associated with pink root, although the etiology of the disease remains doubtful. While recognized as the primary inoculum, studies show conflicting views on the formation of chlamydospores and microsclerotia in Setophoma. Therefore, this study aims to clarify the etiology of the pink root of garlic and onion and the formation of chlamydospores and microsclerotia in Setophoma. The isolates were obtained from symptomatic tissues of garlic, leeks, brachiaria, onions, chives, and maize collected from seven different states in Brazil. Representative isolates were selected for pathogenicity tests. Sequence comparison of the tubulin gene showed Setophoma (n = 50) and Fusarium clades (n = 25). Garlic and onion plants inoculated with Setophoma showed pink root symptoms, while plants inoculated with different Fusarium isolates remained asymptomatic. Multigene analysis of pathogenic isolates confirms that only Setophoma terrestris causes pink root in garlic and onion. In addition, brachiaria, chives, and leeks are newly identified hosts of this pathogen in Brazil. To our knowledge, the main sources of primary inoculum of the disease are chlamydospores, pycnidia, colonized roots of garlic, onion, and plant debris of susceptible crops. The new information obtained in this study will be fundamental for researchers in the development of genotypes that are resistant to pink root and will help the efficient management of the disease. This study presents a comprehensive inventory of bryophytes in Serra dos Órgãos National Park (PARNASO), aiming to evaluate species richness, floristic composition and threatened taxa. Despite the state of Rio de Janeiro being one of the most extensively sampled regions for bryophytes in Brazil, detailed surveys of its conservation units remain scarce. Data were obtained through bibliographic review, herbarium specimen analysis, and new field collections. A total of 504 species were recorded, belonging to 202 genera and 76 families. The park harbors three locally endemic species, eight endemic to Rio de Janeiro, and sixty-nine species endemic to Brazil. Additionally, eleven species were identified as threatened, comprising seven Endangered (EN), two Critically Endangered (CR), and two Vulnerable (VU) according to the IUCN guidelines. PARNASO includes four distinct ecosystems along an altitudinal gradient: sub-montane forest (up to 500 m), montane forest (500–1500 m), upper-montane forest (1500–2000 m), and high-altitude fields (above 2000 m). Montane Forest showed the highest species richness, followed by high-altitude fields, upper-montane forest, and sub-montane forest. The findings highlight PARNASO’s importance in preserving bryophyte diversity in a highly diverse yet understudied region. This work contributes valuable baseline data to support conservation strategies and future ecological studies in Atlantic Forest remnants. Full article

Work Engagement and Compassion Fatigue Among Nursing Professionals During the COVID-19 Pandemic: A Cross-Sectional and Single-Center Study Using the ProQOL-BR and UWES-9 Scales

Objectives: This study investigated levels of work engagement and the occurrence of compassion fatigue among nursing professionals during the COVID-19 pandemic. Methods: A cross-sectional, descriptive, and correlational study was conducted at a Brazilian university hospital between February and April 2022. The Brazilian versions of the Utrecht Work Engagement Scale (UWES-9) and the Professional Quality of Life Scale (ProQOL-BR) were administered. Results: High levels of compassion satisfaction (44.9 points), low levels of burnout (21.0 points), and low levels of secondary traumatic stress (22.8 points) were observed. No professional demonstrated a profile consistent with compassion fatigue. Engagement levels were high for dedication (5.3) and moderate for vigor (4.9), absorption (4.5), and overall engagement (4.9). Burnout showed moderate negative correlations with vigor (r = −0.611, p = 0.005) and dedication (r = −0.599, p = 0.019). Compassion satisfaction showed moderate positive correlations with vigor (r = 0.522, p < 0.001) and dedication (r = 0.572, p < 0.001). The overall engagement score was moderately and positively correlated with compassion satisfaction (r = 0.532, p < 0.001). Conclusions: This study identified high levels of work engagement, especially regarding dedication, and low levels of compassion fatigue among nursing professionals. The data suggest that even amid the emotional and physical demands imposed by the pandemic, participants preserved their emotional well-being and maintained a positive relationship with their work. Wet areas, which are locations in the landscape that consistently retain moisture, and channel networks are important landscape compartments, with key hydrological and ecological functions. Hence, defining their spatial boundaries is an important step towards sustainable watershed management. In catchments of developing countries, wet areas and small order channels of river networks are rarely mapped, although they represent a crucial component of local livelihoods and ecosystems. In this study, topographic attributes generated with a 30 m SRTM DEM were used to map wet areas and stream networks of two tropical catchments in Central Brazil. The topographic attributes for wet areas were the local slope and the slope curvature, and the Topographic Wetness Index (TWI) was used to delineate the stream networks. Threshold values of the selected topographic attributes were calibrated in the Santa Maria catchment, comparing the synthetically generated wet areas and drainage networks with corresponding reference (map) features, and validated in the nearby Santa Maria basin. Drainage network and wet area delineation accuracies were estimated using random basin transects and multi-criteria and confusion matrix methods. The drainage network accuracies were 67.2% and 70.7%, and wet area accuracies were 72.7% and 73.8%, for the Santa Maria and Gama catchments, respectively, being equivalent or higher than previous studies. The mapping errors resulted from model incompleteness, DEM vertical inaccuracy, and cartographic misrepresentation of the reference topographic maps. The study’s novelty is the use of readily available information to map, with simplicity and robustness, wet areas and channel initiation in data-scarce, tropical environments. Full article

Coffee beverage quality is determined by a complex interaction of genetic and environmental factors, including specific biochemical characteristics. In this context, the present study aimed to estimate the genetic parameters of elite irrigated Conilon coffee genotypes in the Cerrado over two consecutive years based on the biochemical characteristics of the beans, assessed by near-infrared spectroscopy (NIRS). The research was conducted at the Embrapa Cerrados experimental field, using the unit’s elite collection. Levels of chlorogenic acid (5-ACQ), caffeine, sucrose, citric acid and trigonelline were analyzed in the raw beans of 18 genotypes harvested in two consecutive years. Data were subjected to analysis of variance in a time-subdivided plot design, considering genotypes as plots and years as subplots, with means grouped by the Scott-Knott test at 5% significance. Results showed significant genetic variability for caffeine, sucrose and trigonelline, while chlorogenic and citric acid levels did not differ significantly among genotypes. A significant genotype × year interaction was observed for caffeine, sucrose, and 5-ACQ. Estimated heritabilities were high for caffeine (85.5%), trigonelline (80.1%), sucrose (62%) and citric acid (60%). Selection gains were positive for sucrose (5.58%), citric acid (10.01%) and trigonelline (8.27%), and negative for caffeine (−6.87%) and 5-ACQ (−0.47%). It is concluded that among the compounds evaluated, caffeine shows the greatest potential for selection, enabling effective gains in raw bean composition, while sucrose and trigonelline present moderate potential for genetic improvement. In Brazil, Plasmodium infections in non-human primates (NHPs) have been associated with P. simium and P. brasilianum, which are morphologically and genetically similar to the human-infecting species P. vivax and P. malariae, respectively. Surveillance and monitoring of wild NHPs are crucial for understanding the distribution of these parasites and assessing the risk of zoonotic transmission. This study aimed to detect the presence of Plasmodium spp. genetic material in Platyrrhini primates from 47 municipalities in the states of Bahia and Minas Gerais. The animals were captured using Tomahawk-type live traps baited with fruit or immobilized with tranquilizer darts. Free-ranging individuals were chemically restrained via inhalation anesthesia using VetBag® or intramuscular anesthesia injection. Blood samples were collected from the femoral vein. A total of 298 blood and tissue samples were collected from 10 primate species across five genera: Alouatta caraya (25), Alouatta guariba clamitans (1), Callicebus melanochir (1), Callithrix geoffroyi (28), Callithrix jacchus (4), Callithrix kuhlii (31), Callithrix penicillata (175), Callithrix spp. hybrids (15), Leontopithecus chrysomelas (16), Sapajus robustus (1), and Sapajus xanthosthernos (1). Molecular diagnosis was performed using a nested PCR targeting the 18S small subunit ribosomal RNA (18S SSU rRNA) gene, followed by sequencing. Of the 298 samples analyzed, only one (0.3%) from Bahia tested positive for Plasmodium brasilianum/P. malariae. This represents the first detection of this parasite in a free-living C. geoffroyi in Brazil. These findings highlight the importance of continued surveillance of Plasmodium infections in NHPs to identify regions at risk for zoonotic transmission. Full article

The European Union Emissions Trading System (EU ETS) and its underlying market structure play a central role in the EU’s climate policy. This study analyses how the network of trading relationships within the EU ETS has evolved from a hub-dominated architecture to one marked by structural change and the emergence of new trading dynamics. Using transaction data from Phases I–IV, we apply complex network analysis to assess changes in connectivity, centrality, and community structure. We then construct a Digital Twin of the EU ETS, integrating graph neural networks and logistic regression models to simulate the entry of new participants and predict future trading links. The results indicate shifts in network composition and connectivity, especially in Phase IV, where regulatory innovations and institutional mechanisms appear to play a key role. While our analysis focuses on structural dynamics, these patterns may have broader implications for market performance and policy effectiveness. These findings underscore the importance of monitoring the evolving trading network alongside price signals to support a resilient, efficient, and environmentally credible carbon market. Full article

This study examines the impact of reproductive inequality on the long-term survival of Homo sapiens by comparing two reproductive models: the Pareto (power-law) distribution of unequal reproduction and the Gaussian (normal) distribution of equal reproduction. We conducted simulations to explore how genetic diversity, measured by heterozygosity, evolves over time. The results predict population crashes due to genetic bottlenecks under both models, but with large differences in timing. We refer to Pareto reproductive inequality as the Genghis Khan effect. This effect accelerates the loss of genetic diversity, increasing the species’ vulnerability to environmental stressors, resource depletion, and genetic drift, and thereby raising the risk of an earlier population collapse. Our findings showcase the importance of reproductive balance for the prolonged presence of Homo sapiens on this planet. Full article
