Huangshui (HS) is a key byproduct of Chinese strong-flavor Baijiu (a traditional distilled spirit) fermentation. This study investigated microbial succession and flavor formation across five key fermentation stages using high-throughput sequencing, physicochemical analysis, and untargeted GC-MS. The results show that Lactobacillus dominated the bacterial communities, while Thermoascus, Aspergillus, and Candida were core fungal genera. Redundancy analysis (RDA) identified acidity, available phosphorus, and ammonia nitrogen as the primary physicochemical drivers of microbial succession. Untargeted metabolomics detected 300 volatile compounds, with 29 discriminant volatile metabolites (VIP > 1.0 or p < 0.05), mainly enriched in carbohydrate and amino acid metabolism pathways associated with Lactobacillus, Thermomyces, Wickerhamomyces, and Kazachstania. These findings establish a link among physicochemical properties, microbiota assembly, and flavor metabolism, providing potential targets for optimizing Baijiu fermentation quality. Full article

Baijiu distillers’ grains (BDG), a major fermented cereal by-product of baijiu production, represent an underutilized source of structurally modified dietary fiber with potential value for functional food development. Here, we found that BDG-derived dietary fiber (BDG-DF), mainly composed of mannose (34.83 ± 0.38%) and xylose (35.14 ± 0.25%), promoted short-chain fatty acid production during in vitro fermentation, and its fermentation supernatants reduced IL-1β and TNF-α levels and modestly decreased IL-6 production in a Caco-2/HepG2 co-culture model. In T2D mice, BDG-DF improved glucose tolerance, with high-dose BDG-DF reducing the OGTT area under the curve by 12.4% compared with the T2D group, and alleviated hepatic steatosis. These effects were accompanied by enrichment of Akkermansia and Bifidobacterium and remodeling of bile acid profiles. High-dose BDG-DF was also associated with elevated CA and CDCA levels, altered TGR5/GLP-1 signaling, increased hepatic FXR expression, and reduced CYP7A1 expression. Integrated hepatic proteomics and metabolomics further indicated that BDG-DF was associated with changes in unsaturated fatty acid biosynthesis and PPAR-γ-related metabolic signaling. Overall, these findings suggest that BDG-DF may improve glucose–lipid homeostasis in association with gut microbiota and bile acid remodeling and hepatic PPAR-γ-related metabolic signaling. Low-alcohol base baijiu, a by-product of Jiang-flavor baijiu production, contains abundant flavor compounds, yet its flavor structure remains insufficiently understood, limiting its high-value utilization. This study integrated flavoromics with interpretable machine learning to characterize the flavor structure of Jiang-flavor low-alcohol base baijiu and identify its key driving factors. During distillation, ethanol decreased from 38.67% to 6.50% (v/v), whereas total acidity increased from 2.74 to 7.53 g/L and pH decreased from 3.01 to 2.59, accompanied by a sensory transition from harmonious and full-bodied notes to sour and salty attributes. Based on sensory and flavor component analysis, the samples can be divided into three gradients according to ethanol volume fraction (>20%, 10–20%, <10%), and their flavor profiles exhibit a systematic evolution: as alcohol content decreases, the relative content of esters and alcohols decreases, while the relative content of acids increases. Random Forest identified 3-methyl-1-butanol, furfuryl ethyl ether, and tetramethyl pyrazine as key markers positively associated with harmonious and smooth attributes (r > 0.93) and negatively associated with sour and salty notes. Odor activity value analysis further quantified stage-dependent sensory contributions. The XGBoost model accurately predicted flavor stages (accuracy = 0.955), and SHAP analysis highlighted low-threshold esters and acids as critical regulators of flavor quality. These findings provide a quantitative and interpretable framework for precision blending, quality grading, and the valorization of low-alcohol base baijiu. Full article

Chinese Baijiu distiller’s grains are by-products of the Chinese Baijiu brewing process, characterized by high water content, high acidity, and high fiber content, which make them unsuitable for animal feed, especially for monogastric animals. This study investigated the possibility of increasing the feed value of Nong-flavor Baijiu distiller’s grains (NFBDGs) for monogastric animals via solid-state fermentation by microbial-enzyme synergy. Experiments evaluated microbial growth, pH variation, improvement of crude protein (CP), true protein (TP), and acid-soluble protein (ASP), degradation of crude fiber (CF), acid detergent fiber (ADF), and neutral detergent fiber (NDF). The results indicated that Ligilactobacillus salivarius CRS23, Bacillus subtilis YLZ7, Saccharomyces cerevisiae CJM26, and xylanase were identified for the fermentation of NFBDGs. When the initial moisture content of NFBDGs was 60% and the initial pH was 3.4, under the conditions of aerobic fermentation at 37 °C for 4 days, the pH of NFBDGs increased from 3.49 to 6.04, the contents of CP and TP increased by 33.59% and 31.21%,,, respectively, while the contents of CF, ADF, and NDF decrease by 35.44%, 20.53%, and 25.02% respectively. The nutritional value of NFBDGs was significantly improved after microbial-enzyme synergistic fermentation, providing a new approach for their application as feed. Full article

The production of Chi-aroma Baijiu generates free radicals, which may pose potential health risks. This study aimed to scavenge free radicals in Chi-aroma Baijiu and assess the impact on the composition of volatile compounds (VOCs) by using distillers’ grains-derived homologous peptides. Five peptides—Pro-Ala (PA), Pro-Val (PV), Gly-Leu (GL), Asp-Phe (DF), and Pro-Leu (PL)—were isolated from the distillers’ grain filtrate. These peptides are present in trace amounts in Chi-aroma Baijiu. By supplementing five peptides to Chi-aroma Baijiu, they could completely scavenge hydroxyl radicals. The rate of scavenging alkoxy radicals ranged from 10.84% to 35.66%. Furthermore, these peptides had an impact on ethyl nonanoate, diethyl acetal, and octanal, which were recognized as characteristic compounds. Ultraviolet spectroscopy simulation revealed that the highest absorption peaks of certain peptides shift when they interact with various esters. Those peptides interact with ethyl acetate and ethyl lactate via hydrogen bonding and Van der Waals forces, demonstrating their influence on VOCs. These findings offer new insights into free radical scavenging, flavor compound regulation, and the valorization of Baijiu by-products in Chi-aroma Baijiu production. Full article

Prolamins, proteins derived from plants, have extensive applications in pharmaceutics and food science. Jiuzao is a byproduct of the Baijiu brewing industry, and is a great source of prolamin. Despite its importance, knowledge regarding the extraction techniques and the properties of prolamin derived from Baijiu Jiuzao (PBJ) remains limited. Reverse micelles (RMs) extraction offers an efficient and cost-effective method for purifying proteins. In the present study, prolamin was extracted from Baijiu Jiuzao using RMs extraction and subsequently characterized in terms of its secondary structure, morphology, and particle size distribution. Our findings indicate that the purified prolamin extracted using further RMs extraction possessed higher α-helix content (+13.25%), forming a large-scale protein network, and narrower particle size distributions compared to the crude prolamin obtained by NaOH-ethanol method. This research suggests that RMs extraction has potential applications in extracting prolamin from brewing industry byproducts, offering an environmentally friendly approach to Baijiu Jiuzao recycling. Baijiu is a renowned Chinese distilled liquor, notable for its distinctive flavor profile and intricate production process, which prominently involves fermentation and distillation. Ethyl carbamate (EC), a probable human carcinogen, can be potentially formed during these procedures, thus prompting significant health concerns. Consequently, the contamination of EC during Baijiu production has become an increasingly pressing issue. In this study, we developed a rapid and easily operable immunoassay for determining EC in the fermented materials used in Baijiu production. The development of a high-quality antibody specific to EC facilitated a streamlined analytical procedure and heightened method sensitivity. Furthermore, we systematically evaluated other essential parameters. Following optimization, the method achieved an IC50 value of 11.83 μg/kg, with negligible cross-reactivity against EC analogs. The recovery study demonstrated the method’s good accuracy and precision, with mean recovery rates ranging from 86.0% to 105.5% and coefficients of variation all below 10%. To validate the feasibility of the technique, we collected and analyzed 39 samples simultaneously using both the proposed immunoassay and confirmatory gas chromatography–mass spectrometry (GC-MS). A robust correlation was observed between the results obtained from the two methods (R2 > 0.99). The detected EC levels ranged from 2.36 μg/kg to 7.08 μg/kg, indicating an increase during the fermentation process. Full article

Methanol, also known as wood alcohol, is a common hazardous by-product of alcoholic beverage fermentation and serves as a crucial indicator for assessing the safety of alcoholic beverages. However, the metabolic mechanisms of methanol production during the solid-state fermentation of Chinese Baijiu remain unclear. In this study, we sought to determine the primary stage of methanol production in Chinese Baijiu by measuring the methanol content at different stages of fermentation. High-throughput multi-omics sequencing techniques were employed to elucidate methanol metabolic pathways and associated microorganisms. In addition, a comprehensive analysis incorporating environmental factors and microbial interactions was conducted to explore their combined effects on methanol production. Methanol was predominantly produced during pit fermentation, with the most significant increase observed within the first seven days. Microorganisms such as Pichia kudriavzevii, Byssochlamys spectabilis, Penicillium, and Aspergillus played a regulatory role in methanol content during the first seven days through their involvement in butyrate and methane metabolic pathways and pectin degradation modules. During Baijiu production, various types of molds and yeasts participate in methanol production. Differences in their abundance within fermentation cycles may contribute to variations in methanol content between stages. Lactobacillus accumulated abundantly in the first seven days in each stage, suppressing methanol-metabolizing microorganisms. In addition, the increased acidity resulting from Lactobacillus metabolism may indirectly promote methanol generation. Full article

The liver is critical in alcohol metabolism, and excessive consumption heightens the risk of hepatic damage, potentially escalating to hepatitis and cirrhosis. Jiuzao, a by-product of Baijiu production, contains a rich concentration of naturally active polysaccharides known for their antioxidative properties. This study investigated the influence of Laowuzeng Jiuzao polysaccharide (LJP) on the development of ethanol-induced alcoholic fatty liver. Zebrafish larvae served as the model organisms for examining the LJPs hepatic impact via liver phenotypic and biochemical assays. Additionally, this study evaluated the LJPs effects on gene expression associated with alcoholic fatty liver and the composition of the intestinal microbiota through transcriptomic and 16 S rRNA gene sequencing analyses, respectively. Our findings revealed that LJP markedly mitigated morphological liver damage and reduced oxidative stress and lipid peroxidation in larvae. Transcriptome data indicated that LJP ameliorated hepatic fat accumulation and liver injury by enhancing gene expression involved in alcohol and lipid metabolism. Furthermore, LJP modulated the development of alcoholic fatty liver by altering the prevalence of intestinal Actinobacteriota and Firmicutes, specifically augmenting Acinetobacter while diminishing Chryseobacterium levels. Ultimately, LJP mitigated alcohol-induced hepatic injury by modulating gene expression related to ethanol metabolism, lipid metabolism, and inflammation and by orchestrating alterations in the intestinal microbiota. A novel polysaccharide, HSP80-2, with an average molecular weight of 13.8 kDa, was successfully isolated by the gradient ethanol precipitation (GEP) method from Huangshui (HS), the by-product of Chinese Baijiu. It was mainly composed of arabinose, xylose, and glucose with a molar ratio of 4.0:3.1:2.4, which was completely different from the previous reported HS polysaccharides (HSPs). Morphological observations indicated that HSP80-2 exhibited a smooth but uneven fragmented structure. Moreover, HSP80-2 exerted prebiotic activity evaluated by in vitro fermentation. Specifically, HSP80-2 was utilized by gut microbiota, and significantly regulated the composition and abundance of beneficial microbiota such as Phascolarctobacterium, Parabacteroides, and Bacteroides. Notably, KEGG pathway enrichment analysis illustrated that HSP80-2 enriched the pathways of amino sugar and nucleotide sugar metabolism (Ko00520), galactose metabolism (ko00052), and the citrate cycle (TCA cycle) (ko00020). Meanwhile, the contents of short-chain fatty acids (SCFAs) mainly including acetic acid, propionic acid, and butyric acid in the HSP80-2 group were remarkably increased, which was closely associated with the growth of Lachnoclostridium and Parabacteroides. These results showed that HSP80-2 might be used as a potential functional factor to promote human gut health, which further extended the high value utilization of HS. Full article

Baijiu is a traditional spirit with high reputation in the Chinese community, and whisky, on the other hand, is a renowned spirit in Western culture, with both contributing a major proportion to the consumption and revenue in the global spirit market. Interestingly, starting with similar raw materials, such as grains, diverse production methods lead to different organoleptic profiles. In addition, such enormous attention they attract renders them as a crucial part in food and the related industry. Therefore, great efforts are made in improving product quality and optimizing production processes, such as flavor enhancement, facility development, and deep utilization of byproducts. Given the huge impacts and great involvements of these spirits in the general food industry, research focusing on either spirit is of referential significance for other relevant fields. With the aim of facilitating such collaboration, this review discusses the current research status, in a comparative manner, of both spirits in respect to key production processes-oriented sensory and flavor analysis, deep utilization of byproducts, and spirit-derived functional food investigations. Finally, the internal correlations based on the abovementioned criteria are identified, with research prospects proposed. Full article

Distilled spent grain (DSG), as the major by-product of baijiu making, contains melanoidins generated via the Maillard reaction. In this study, four melanoidin fractions (RF1‒RF4) were isolated successively from dried DSG (DDSG) using sodium hydroxide solution and water as extractants, and the content, preliminary structure, and ACE-inhibitory activities in vitro of melanoidins were first investigated. The antioxidant activity was also evaluated. The results indicated that the total content of melanoidins was 268.60 mg/g DDSG dry weight (dw) using a model system of glucose and serine as standard, and RF4 showed the highest content of melanoidins (174.30 mg/g DDSG dw). Functional groups like C=O, N‒H, C‒N, O‒H, C‒H, C‒O, C-C, and ‒C‒CO‒C‒ were present in the structure of melanoidins from RF4, as determined by Fourier transform infrared (FT-IR) assay. The highest antioxidant activities, as assessed by 2,2′-azino-bis (3-ethylbenzothiazoline-6-sulfonic acid) (ABTS•+), ferric-reducing antioxidant power (FRAP), and 1,1-diphenyl-2-picrylhydrazyl radical (DPPH) assays, and the highest angiotensin-converting enzyme (ACE) inhibitory activity (95.92% at 2 mg RF4/mL) were also exhibited by RF4. The RF4 was further fractionated by ultrafiltration based on molecular weight (MW). The more than 100 kDa melanoidins (RF4-6) exhibited the highest yield and antioxidant activity.
