We report the green synthesis of silver nanoparticles (AgNPs) by using daisy petals (Bellis perennis), leek (Allium porrum) and garlic skin (Allium sativum) as reducing agents and water as solvent. AgNPs are obtained with high monodispersity, spherical shapes and size ranging from 5 to 35 nm and characterized by UV-Vis and TEM techniques. The obtained yields in AgNPs are in concordance with the total phenolic content of each plant. We also study the incorporation of AgNPs in combination with the red algae Pyropia columbina extracts (PCE) into cosmetic formulations and analyze their combined effect as photoprotective agents. Moreover, we carry out the inclusion of the PCE containing mycosporine-like amino acids (MAAs), which are strong UV-absorbing and antioxidant compounds, into β-cyclodextrin (βCD) and pNIPAM nanoparticles and analyze stability and release. The thermoresponsive polymer is grown by free radical polymerization using N-isopropylacrylamide (NIPAM) as the monomer, N,N′-methylenebisacrylamide (BIS) as the cross-linker, and 2,2′-azobis(2-methylpropionamidene) (V50) as the initiator, while βCD complex is prepared by heating in water. We evaluate the nanoparticle and βCD complex formation by UV-Vis and FT-IR, and NMR spectroscopies, respectively, and the nanoparticles’ morphology, including particle size, by TEM. The cosmetic formulations are subsequently subjected to accelerated stability tests and photoprotective analyses: a synergistic effect in the combination of AgNPs and PCE in photoprotection was found. It is not related to a UV screen effect but to the antioxidant activity, having potential against photoaging. Full article

Saponins, a diverse group of natural compounds, offer an interesting pool of derivatives with biomedical application. In this study, three structurally related spirostanol saponins were isolated and identified from the leek flowers of Allium porrum L. (garden leek). Two of them were identical with the already known leek plant constituents: aginoside (1) and 6-deoxyaginoside (2). The third one was identified as new component of A. porrum; however, it was found identical with yayoisaponin A (3) obtained earlier from a mutant of elephant garlic Allium ampeloprasun L. It is a derivative of the aginoside (1) with additional glucose in its glycosidic chain, identified by MS and NMR analysis as (2α, 3β, 6β, 25R)-2,6-dihydroxyspirostan-3-yl β-D-glucopyranosyl-(1 → 3)-β-D-glucopranosyl-(1 → 2)-[β-D-xylopyranosyl-(1 → 3)]-β-D-glucopyranosyl]-(1 → 4)-β-D-galactopyranoside, previously reported also under the name alliporin. The leek native saponins were tested together with other known and structurally related saponins (tomatonin and digitonin) and with their related aglycones (agigenin and diosgenin) for in vitro cytotoxicity and for effects on NO production in mouse peritoneal cells. The highest inhibitory effects were exhibited by 6-deoxyaginoside. The obtained toxicity data, however, closely correlated with the suppression of NO production. Therefore, an unambiguous linking of obtained bioactivities of saponins with their expected immunobiological properties remained uncertain. Full article

This study aimed to analyze the antioxidant activity, phenolic acid profile, color changes, and chemical composition of dried and powdered leek (Allium porrum). Leek was divided into white shaft (WH) and green shaft (GR) and subjected to drying by different methods—convection drying, vacuum drying, and freeze-drying (FD)—at a temperature of 60 °C. A sample freeze-dried at a temperature of 20 °C was used as control. Analyses of the dried leek samples revealed that GR contained a higher amount of ash, protein, fat, fiber, phenolic acids, and flavonoids, and exhibited higher antioxidant capacity compared to WH. The dominant phenolic acid in WH was p-cumaric acid followed by synapic and protocatechuic acids. GR had a several-fold higher content of phenolic acids than WH, with ferulic acid being dominant (about 85% of the total phenolic content). It was also observed that a higher drying temperature resulted in the degradation of phenolic compounds and reduced the antioxidant properties of leek shafts. Most importantly, FD under a temperature of 60 °C caused a similar degree of degradation of biologically active compounds as air drying. An increase in drying temperature was associated with a slight decrease in the lightness of GR, whereas in the case of WH no significant change in this parameter was observed. Full article

The floating system is a successful strategy for producing baby leaf vegetables. Moreover, compost from agricultural and agri-food industry wastes is an alternative to peat that can be used as a component of growing media in this cultivation system. In this study, we experimented with three composts containing tomato (Solanum lycopersicum L.), leek (Allium porrum L.), grape (Vitis vinifera L.), and/or olive (Olea europaea L.) mill cake residues, which were used as the main component (75/25 volume/volume) of three growing media (GM1, GM2 and GM3) to evaluate their effect on the growth and quality of red baby leaf lettuce (Lactuca sativa L.). We used a commercial peat substrate as a control treatment (100% volume) and in mixtures (25% volume) with the composts. The plants were cultivated over two growing cycles, in spring and summer, and harvested twice in each cycle when the plants had four to five leaves. We found that the percentage of seed germination was significantly higher in plants grown in peat than in those grown in compost growing media. The yield was affected by the growing media in the summer cycle, and we obtained the highest value with GM1. Furthermore, the second cut was more productive than the first one for all the growing media in both cycles. The lettuce quality was also affected by the growing media. In general, the total phenolic content and antioxidant capacity in the leaves was higher in plants grown in the compost growing media, particularly in the second cut, but the nitrate content in the leaves was greater in some of the compost treatments compared with the peat treatment. In addition, an in vitro suppressive activity study demonstrated that the interaction between different fungi and bacteria observed through metagenomics analysis could contribute to the effectiveness of the compost in controlling Pythium irregulare. The use of compost as a component of the growing media in the production of baby leaf vegetables in a floating system does not only favor the crop yield and product quality, but also shows suppressive effects against P. irregulare. Allium genus plants, such as leek (Allium porrum), are rich sources of anti-inflammatory and anti-oxidant secondary metabolites; this is of interest because it demonstrates their suitability as pharmacological alternatives for inflammatory processes, including allergy treatment. The composition of methanolic leek extract (LE) was analyzed by GC–MS and LC–IT/MS, and the total phenolic content and antioxidant capacity were quantified by colorimetric methods. Its pharmacological potential was analyzed in human bronchial epithelial Calu-3 cells, human mast cells LAD2, and humanized rat basophiles RBL-2H3. LE exhibited a cytotoxic effect on Calu-3 cells and HumRBL-2H3 cells only at high concentrations and in a dose-dependent manner. Moreover, LE decreased the degranulation of LAD2 and HumRBL-2H3 cells. LE treatment also significantly prevented alterations in transepithelial electrical resistance values and mRNA levels of glutathione-S-transferase (GST), c-Jun, and NFκB after treatment with H2O2 in ALI-cultured Calu-3 cells. Finally, ALI-cultured Calu-3 cells treated with LE showed lower permeability to Ole e 1 compared to untreated cells. A reduction in IL-6 secretion in ALI-cultured Calu-3 cells treated with LE was also observed. In summary, the results obtained in this work suggest that A. porrum extract may have potential anti-allergic effects due to its antioxidant and anti-inflammatory properties. This study provides several important insights into how LE can protect against allergy. Full article

Leek (Allium porrum) is known for its high antioxidant activity and the ability to accumulate significant amounts of potassium and iron. We assessed yield, quality indicators, antioxidants and elemental composition of nine leek cultivars grown in greenhouses under organic or conventional systems in the Moscow region. The management system did not affect yield, which attained the highest value with the cultivar Giraffe and the lowest with Premier and Cazimir. Pseudo-stem dry matter and sugars were higher with organic management, whereas nitrate concentration was higher with conventional management. The cultivars Vesta and Summer Breeze showed the highest dry matter and total sugar content, whereas Goliath had the highest antioxidant, selenium and potassium concentrations. Among the antioxidants, ascorbic acid attained higher values with organic management. The antioxidant system of leek was characterized by highly significant positive correlations between: Se and polyphenols, Se and ascorbic acid, Se and K, ascorbic acid and polyphenols, ascorbic acid and K, polyphenols and K (r = 0.94, 0.94, 0.95, 0.94, 0.95, 0.96, respectively, at P ≤ 0.001). Negative correlations were recorded between leaf and pseudo-stem Se and between leaf and pseudo-stem polyphenols (r = −0.922 and −0.976, respectively, at P ≤ 0.001). Among the mineral elements, only K was significantly affected by the management system, showing a higher content in organically grown pseudo-stems. Varietal differences in pseudo-stem element composition showed strong positive correlations of: Al with As, Co, Li, Pb and V; Cr with I, Mg, Si, Ca; V with As, Co and Fe; negative correlations of Se with Cr and I. Compared to related species such as garlic (A. sativum), leek accumulated levels equal to garlic of K, Mg, P, Cd, Cu, Mn, Se, Zn, lower levels of Si and significantly higher amounts of Ca, Na, Al, As, Cr, Ni, Pb, Sr, V, Sn, B, Co, Fe, I, Li. The strong relationships between quality, antioxidant and mineral components in leek plants may give wide possibilities in breeding programs for both conventional and organic management systems in greenhouses. Allium ampeloprasum var. porrum has been recognized as a rich source of secondary metabolites, including phenolic acids, flavonoids and flavonoid polymers (proanthocyanidins or condensed tannins), with related health benefits. Both parts of Allium ampeloprasum var. porrum (white bulb and pseudostem) are traditionally consumed either as a vegetable or as a condiment in many Mediterranean countries. The aim of the present study was to optimize the extraction conditions of polyphenols from white leek stem and green leek leaf by implementing a Box-Behnken design (BBD). The optimization considered basic factors affecting extraction efficiency, including extraction time, solvent to plant material ratio and solvent mixture composition. Maximum polyphenol yield was achieved at an extraction time of 80 and 100 min for white leek stem and green leek leaf extracts respectively, solvent to plant material ratio of 5:1 (v/w) and methanol to water ratio of 40:60 (v/v), for both leek extracts. Interestingly, higher total phenolic content was found in green leek leaf extracts compared to white leek stem extracts, due to a possible relationship between polyphenol production and sunlight radiation. High correlation values were also observed between total phenolic content and antioxidant-antiradical activity of optimized leek extracts. Full article

Growing vegetables economically in the use of constructed wetland for wastewater treatment can play a role in overcoming water and food scarcity. Allium porrum L., Solanum melongena L., Ipomoea aquatica Forsk., and Capsicum annuum L. plants were selected to grow in hybrid constructed wetland (CW) under natural conditions. The impact of the ratio of nitrate to ammonium nitrogen on ammonium and nitrate nitrogen removal and on total nitrogen were studied in wastewater. Constructed wetland planted with Ipomoea aquatica Forsk. and Solanum melongena L. showed higher removal efficiency for ammonium nitrogen under higher ammonium concentration, whereas Allium porrum L.-planted CW showed higher nitrate nitrogen removal when NO3–N concentration was high in wastewater. Capsicum annuum L.-planted CW showed little efficiency for both nitrogen sources compared to other vegetables. Full article

Six essential oils (EOs) from the Alliaceae family, namely garlic (Allium sativum), onion (Allium cepa), leek (Allium porrum), Chinese chive (Allium tuberosum), shallot (Allium ascalonicum) and chive (Allium schoenoprasum) were characterized by GC and GC-MS and evaluated for their functional food properties. Antibacterial properties were tested on five food-borne pathogens: Two Gram-positive Staphylococcus aureus (ATCC 25923), Listeria monocytogenes (ATCC 19115) and three Gram-negative Salmonella Typhimurium (ATCC 14028), Escherichia coli (ATCC 8739) and Campylobacter jejuni (ATCC 33291) bacteria. Antioxidant and radical-scavenging properties were tested by means of Folin-Ciocalteu and 2,2-diphenyl-1-picrylhydrazyl (DPPH) assays. Garlic, Chinese chive and onion EOs had the highest antibacterial activity whereas shallot and leek EOs were the strongest antioxidants. Heating caused a decrease in the antioxidant activity of these Eos, as shown in the Total Polar Materials (TPM) test. Suggestions on relationships between chemical composition and biological activities are presented. Results show that the EOs could be of value in the food industry as alternatives to synthetic antioxidants. Full article

Laser scanning is a non-invasive method for collecting and parameterizing 3D data of well reflecting objects. These systems have been used for 3D imaging of plant growth and structure analysis. A prerequisite is that the recorded signals originate from the true plant surface. In this paper we studied the effects of species, leaf chlorophyll content and sensor settings on the suitability and accuracy of a commercial 660 nm active laser triangulation scanning device. We found that surface images of Ficus benjamina leaves were inaccurate at low chlorophyll concentrations and a long sensor exposure time. Imaging of the rough waxy leaf surface of leek (Allium porrum) was possible using very low exposure times, whereas at higher exposure times penetration and multiple refraction prevented the correct imaging of the surface. A comparison of scans with varying exposure time enabled the target-oriented analysis to identify chlorotic, necrotic and healthy leaf areas or mildew infestations. We found plant properties and sensor settings to have a strong influence on the accuracy of measurements. These interactions have to be further elucidated before laser imaging of plants is possible with the high accuracy required for e.g., the observation of plant growth or reactions to water stress.
