Physicochemical and Microbiological Properties of Hazelnuts from Three Varieties Cultivated in Portugal

Hazelnut is an important crop worldwide, and the characteristics of the fruits are quite variable according to a number of factors, including variety and cultivation conditions, which in turn can vary according to harvest year. This study aimed to investigate some physical and chemical characteristics of three hazelnut varieties grown in Portugal (Grada de Viseu, Tonda di Giffoni and Butler) along two different harvesting years (2021 and 2022). Also, the microbial quality was investigated for its relevance to the conservation of the fruits. The physical properties evaluated were biometric characteristics and colour, the chemical components analysed were moisture, lipids, protein, ash and fibre, and the microbial properties investigated were the microorganisms, moulds and yeasts. The results showed that, generically, statistically significant differences were found between the three varieties under study on several properties investigated. The kernel was confirmed as the lighter part of all hazelnuts, and when comparing between varieties, Tonda di Giffoni presented the lighter fruits in both harvesting years. With respect to weight, the Tonda di Giffoni variety was the lightest in both harvest years. Moisture content was observed to be higher than the recommended limits for two of the samples (Grada de Viseu in 2021: 6.01 ± 0.26 g/100 g and Butler in 2022: 6.02 ± 0.37 g/100 g), although the difference was marginal given that the recommended limit is 6%. Not surprisingly, lipids were the major chemical component, ranging from 66.46 ± 1.67 to 70.14 ± 1.75 g/100 g in 2021 and from 64.38 ± 1.67 to 77.77 g/100 g in 2022. It was further observed that the three varieties presented a satisfactory microbiological quality. Finally, applying factor analysis with principal components and Varimax rotation, a solution that explains 92.8% of the variance was obtained. This study provided information that is relevant for the characterisation and evaluation of variability according to the year of hazelnuts of three varieties cultivated in Portugal. Full article

Metagenomic studies have provided deeper insights into the complex interactions between the skin and its microbiota. However, limited research has been conducted on the skin microbiota of Brazilian women. Given that Brazil ranks as the fourth-largest consumer of cosmetics worldwide, the development of new tools to analyze skin microbiota is crucial for formulating cosmetic products that promote a healthy microbiome. Skin samples were analyzed using the Illumina platform. Biometrology assessments were applied. The results showed pH variations were more pronounced in the older age group, along with higher transepidermal water loss values. Metagenomic analysis showed a predominance of Actinobacteria (83%), followed by Proteobacteria (7%), Firmicutes (9%) and Bacteroidetes (1%). In the older group (36–45 years old), an increase in Actinobacteria (87%) was observed and a decrease in Proteobacteria (6%). Moreover, the results differ from the international literature, since an increase in proteobacteria (13.9%) and a decrease in actinobacteria (46.7%) were observe in aged skin. The most abundant genus identified was Propionibacterium (84%), being the dominant species. Interestingly, previous studies have suggested a decline in Cutibacterium abundance with aging; although there is no significant difference, it is possible to observe an increasing trend in this genus in older skin. These studies can clarify many points about the skin microbiota of Brazilian women, and these findings could lead to the development of new cosmetics based on knowledge of the skin microbiome. Background/Objective: The COVID-19 pandemic profoundly transformed social life worldwide, indiscriminately affecting individuals across all age groups. Children have not been exempted from the risk of severe illness and death caused by COVID-19. Objective: This paper sought to describe the clinical findings, laboratory and imaging results, and hospital care provided for severe and critical cases of COVID-19 in unvaccinated children, with or without severe asthma, hospitalized in a public referral service for COVID-19 treatment in the Brazilian state of Pernambuco. Methods: This was a case series study of severe and critical COVID-19 in hospitalized, unvaccinated children, with or without severe asthma, conducted in a public referral hospital between March 2020 and June 2021. Results: The case series included 80 children, aged from 1 month to 11 years, with the highest frequency among those under 2 years old (58.8%) and a predominance of males (65%). Respiratory diseases, including severe asthma, were present in 73.8% of the cases. Pediatric multisystem inflammatory syndrome occurred in 15% of the children, some of whom presented with cardiac involvement. Oxygen therapy was required in 65% of the cases, mechanical ventilation in 15%, and 33.7% of the children required intensive care in a pediatric intensive care unit. Pulmonary infiltrates and ground-glass opacities were common findings on chest X-rays and CT scans; inflammatory markers were elevated, and the most commonly used medications were antibiotics, bronchodilators, and corticosteroids. Conclusions: This case series has identified key characteristics of children with severe and critical COVID-19 during a period when vaccines were not yet available in Brazil for the study age group. However, the persistence of low vaccination coverage, largely due to parental vaccine hesitancy, continues to leave children vulnerable to potentially severe illness from COVID-19. These findings may inform the development of public health emergency contingency plans, as well as clinical protocols and care pathways, which can guide decision-making in pediatric care and ensure appropriate clinical management, ultimately improving the quality of care provided. Combined trabeculectomy–phacoemulsification is known to provoke more inflammation and yield a poorer long-term efficacy than trabeculectomy alone. This study evaluates whether a similar trend exists for Ahmed glaucoma valve implantation when performed with or without concurrent phacoemulsification. We retrospectively analyzed 51 eyes from patients who underwent either Ahmed-Alone (n = 25) or PhacoAhmed (n = 26) surgery over a 5-year period. The primary outcomes included intraocular pressure (IOP), the use of IOP-lowering medications, and the need for further surgical intervention. Absolute success was defined as IOP reduction > 20% and IOP < 21 mmHg without medication; relative success allowed for continued pharmacologic therapy. Both groups showed a significant IOP reduction, with similar final mean IOP values (Ahmed-Alone: 14.02 ± 4.76 mmHg; PhacoAhmed: 13.89 ± 4.17 mmHg; p = 0.99) and comparable reductions in medication use (p = 0.52). Reinterventions occurred less frequently and later in the PhacoAhmed group (12% vs. 27.3%; median time: 27.1 vs. 12 months). Absolute success was not achieved in any PhacoAhmed case but occurred in 9.3% of Ahmed-Alone cases; relative success rates were similar (83.3% vs. 81.4%; p = 0.291). These findings suggest that combining phacoemulsification with Ahmed valve implantation does not significantly alter efficacy or safety profiles. Additional prospective studies are warranted to assess long-term outcomes. Full article

Surveillance of swine influenza A virus (swIAV) traditionally focuses on respiratory matrices, yet emerging evidence suggests that fecal shedding and secondary environmental contamination may also contribute to viral dissemination. In this study, we collected and analyzed nasal, rectal, environmental, milk, and colostrum samples from naturally infected pigs in a commercial farm in Minas Gerais, Brazil. IAV RNA was detected in 25% of samples, including 42% from asymptomatic animals, with nasal swabs showing higher detection rates (30%) than rectal swabs (20%), though rectal Ct values were consistently higher, indicative of lower viral loads. We successfully isolated viable viruses from feces and effluent samples. Whole-genome sequencing revealed co-circulation of enzootic pH1N1 clade #2 (HA) and pN1 clade #4 (NA), alongside human-origin H3N2 sequences clustering within clade 3C.2a1b.2a.2a.1, and N2 segments related to pre-3C human lineages from 2001 to 2002. Phylogenetic and p-distance analyses support both recent reverse zoonosis and historical transmission events. Detection of complete HA/NA sequences from rectal swabs and treated effluent further emphasizes the surveillance value of non-respiratory matrices. The integration of respiratory and fecal/environmental sampling appears important to achieve more comprehensive IAV monitoring in swine herds and may have significant implications for One Health strategies in Brazil and beyond. Background: Type 2 diabetes (T2D) is a growing public health concern, particularly in low- and middle-income countries. Although prediabetes is a major risk factor for T2D, it remains largely underdiagnosed and untreated. Structured lifestyle interventions have proven effective in preventing diabetes, but their feasibility within the Brazilian public health system remains unclear. Methods: This multicenter pilot randomized controlled trial assessed the feasibility of a culturally adapted lifestyle intervention (PROVEN-DIA) across the five regions of Brazil. A total of 220 adults at high risk for T2D were randomized to an intervention group or a control group (usual care) and followed for three months. Both groups received similar educational content on healthy eating and physical activity, but the intervention group participated in a structured and personalized lifestyle program with regular follow-up sessions. The primary outcome was adherence to dietary recommendations, assessed using the BALANCE Index—a validated dietary score (range: 0–40) based on the Brazilian Cardioprotective Diet that classifies foods into color-coded groups according to nutritional quality—along with engagement in moderate-to-vigorous physical activity (MVPA). Secondary outcomes included diet quality (DQIR), anthropometric and metabolic parameters. Results: Feasibility was demonstrated by a 93.2% retention rate (n = 205). There was no significant difference in the primary outcome (simultaneous improvement in diet and MVPA). However, the PROVEN-DIA group exhibited significantly greater improvements in diet quality, with a 2.8-point increase in the BALANCE Index (vs. 0.5 in the control, p = 0.03), and a significant improvement in the DQIR (p < 0.001). No significant differences between groups were observed in MVPA, HbA1C, glycaemia, or body weight. Conclusions: The PROVEN-DIA intervention proved feasible within the Brazilian public health context, resulting in significant improvements in dietary quality among individuals at high risk for T2D. A larger trial with longer follow-up is warranted to evaluate its effectiveness in preventing the progression to diabetes. However, to enhance physical activity outcomes, specific adaptations and targeted strategies may be required to better support participant engagement in exercise. Full article

Olive mill wastewaters (OMWWs) are an environmental problem in the Mediterranean region, and it is crucial to explore strategies for their treatment and repurposing. The chemical precipitation technique (CPT) has been presented as a cost-effective wastewater treatment solution that might be applied to OMWW. The CPT-resulting precipitant subproducts (sludge) may be reprocessed (e.g., agricultural fertilizer and/or soil amendment), while the treated wastewater may be repurposed or reused (e.g., irrigation, aquaponic, or industrial processes). This study aimed to evaluate the efficacy of CPT in treating wastewater from the olive oil industry from an ecotoxicological perspective. Additionally, to assess the safe use of the obtained sludge in CPT treatment, its effects on soil biota were assessed. For this, a set of ecotoxicological assays using freshwater (Raphidocelis subcapitata, Daphnia magna and Danio rerio), terrestrial invertebrates (Folsomia candida and Enchytraeus crypticus), and plants (Brassica oleracea and Lolium perenne) were used as model organisms. Results demonstrated that CPT reduced OMWW toxicity to freshwater organisms, offering a favorable outlook on CPT’s potential as a wastewater treatment method. Increasing application rates of sludge in soil reduced the shoot biomass and the hydric content of both plants compared to the control. Survival of F. candida and E. crypticus was not affected by sludge in soil at any tested application rate, yet sludge application negatively affected the reproduction of both species, even at relevant sludge application rates (2%) of sludge in soils. Overall, the applicability of this sludge obtained by the CPT treatment in soils should be carefully evaluated due to the observed adverse effects on soil biota. Although the results of CPT were promising in reducing the toxicity of OMWW for these aquatic species, some adjustments/improvements should be performed to improve this technique and use all the obtained resources (treated water and sludge) in a fully circular perspective. Background: Blackberry seed oil (BSO), obtained from Rubus spp. Xavante cultivar via supercritical CO2 extraction, contains bioactive lipids and antioxidants, but its cosmetic application is limited by poor solubility and stability. Nanoencapsulation with poly(ε-caprolactone) (PCL) can overcome these limitations. Methods: BSO was characterized by Ultra-High-Performance Liquid Chromatography coupled with electrospray ionization quadrupole time-of-flight mass spectrometry and incorporated into PCL nanocapsules (NCBSO) using the preformed polymer deposition method. Physicochemical properties, stability (at 4 °C, room temperature, and 37 °C for 90 days), cytotoxicity, and collagen production were assessed in human fibroblasts. Additionally, a predictive in silico analysis using PASS Online, Molinspiration, and SEA platforms was performed to identify the bioactivities of major BSO compounds related to collagen synthesis, antioxidant potential, and anti-aging effects. Results: NCBSO showed a nanometric size of ~267 nm, low polydispersity (PDI < 0.2), negative zeta potential (−28 mV), and spherical morphology confirmed by FE-SEM. The dispersion remained stable across all tested temperatures, preserving pH and colloidal properties. In particular, BSO and NCBSO at 100 µg.mL−1 significantly enhanced in vitro collagen production by 170% and 200%, respectively, compared to untreated cells (p < 0.01). Superior bioactivity was observed for NCBSO. The in silico results support the role of key compounds in promoting collagen biosynthesis and protecting skin structure. No cytotoxic effects were achieved. Conclusions: The nanoencapsulation of BSO into PCL nanocapsules ensured formulation stability and potentiated collagen production. These findings support the potential of NCBSO as a promising candidate for future development as a collagen-boosting cosmeceutical. Full article

The mosquito Aedes aegypti is the vector responsible for the transmission of important arboviruses such as dengue fever, Chikungunya, Zika virus, and yellow fever. These diseases affect millions of people and exert impacts on healthcare systems throughout the world. Given the increasing resistance to synthetic insecticides, essential oils from plants constitute an ecologically viable alternative for the control of this vector. The aim of the present study was to investigate the larvicidal activity of the essential oil (EO), aqueous extract, rutin, and hydrolate from the leaves of Myrciaria floribunda against Aedes aegypti larvae in the initial L4 stage. The yield of EO was 0.47%. Thirty-seven chemical constituents were identified and quantified using chromatographic methods. The major constituents were (E)-caryophyllene (27.35%), 1,8-cineole (11.25%), β-selinene (4.92%), and α-muurolene (4.92%). In the larvicidal tests, the lethal concentration (LC50) was 201.73 ppm for the essential oil, 15.85% for the aqueous extract, and 22.46 ppm for rutin. The hydrolate had no larvicidal activity. The compounds that exhibited larvicidal activity against Aedes aegypti constitute a promising option for the development of natural formulations to diminish the propagation of this vector. Adult entertainment work may be associated with increased vulnerability to sexually transmitted infections, particularly HIV. In Brazil, pre-exposure prophylaxis (PrEP) for HIV infection has been available through the Brazilian Unified Health System (SUS) since November 2017, representing a significant advancement in public sexual health policy. The objective of this study was to understand the individual and social determinants that promote PrEP use among adult entertainment workers. This was a cross-sectional, analytical, and quantitative study. A multivariate modeling approach was employed to identify factors independently associated with PrEP use. The study included 254 adult entertainment workers using oral PrEP through the SUS, predominantly young adults (141; 55.5%), SUS users (248; 97.6%), single (213; 83.9%), non-white (142; 55.9%), cisgender (148; 58.3%), and heterosexual (152; 59.8%). Factors independently associated with greater PrEP use included having adult entertainment as the main source of income (aPR: 2.69; 95% CI: 1.86–3.95), prior use of PEP (aPR: 2.49; 95% CI: 1.63–3.81), undergoing any type of health treatment (aPR: 1.56; 95% CI: 1.15–2.12), and having a history of STIs (aPR: 1.51; 95% CI: 1.10–2.08). Conclusion: PrEP use in this population was strongly influenced by structural and contextual factors, indicating that the availability of the technology alone does not ensure its effectiveness. In this study, Sr0.95Bi0.05TiO3 was synthesized via solid state reaction, characterized, and applied as a visible-light-active photocatalyst for the degradation of oxytetracycline, imidacloprid, and their mixture. To evaluate the influence of the aqueous matrix on pollutant degradation, photocatalytic experiments were carried out in both distilled water and a real environmental sample (surface water). The Sr0.95Bi0.05TiO3 perovskite showed high photocatalytic performance under visible light, achieving nearly complete degradation of oxytetracycline after 2 h, and significant removal of imidacloprid in river water (60% after 3 h). Enhanced degradation in surface water was attributed to favorable ionic composition and pH. The perovskite oxide maintained its photocatalytic performance over five consecutive cycles, with no significant loss in photocatalytic activity or structural and morphological stability. Ecotoxicological assessment using Daphnia magna confirmed that the treated water was non-toxic, indicating that no harmful byproducts were formed. Complementary Density Functional Theory calculations were conducted to complement experimental findings, providing insights into the structural, electronic, and optical properties of the photocatalyst, enhancing the understanding of the degradation mechanisms involved. This integrated approach, combining experimental photocatalytic performance evaluation in different matrices, ecotoxicity testing, and theoretical modeling, highlights Sr0.95Bi0.05TiO3 as a promising, stable, and environmentally safe photocatalyst for practical wastewater treatment applications. Background/Objectives: Mpox re-emerged in 2022 as a global health concern. Between 2022 and 2025, Portugal experienced three distinct outbreak waves, highlighting the critical role of laboratory surveillance and public health interventions. This study describes the epidemiological trends, diagnostic performance, and key lessons learned to improve outbreak preparedness. Methods: A total of 5610 clinical samples from 2802 suspected cases were analyzed at the National Institute of Health Doutor Ricardo Jorge using real-time PCR methods. Positivity rates and viral loads (Ct values) were assessed across different clinical specimen types, including lesion, anal, oropharyngeal swabs, and urine samples. Results: Mpox was confirmed in 1202 patients. The first outbreak accounted for 79.3% of cases (n = 953), followed by a significant reduction in transmission during subsequent waves. Lesion and rectal swabs provided the highest diagnostic sensitivity (95.1% and 87.9%, respectively). Oropharyngeal swabs contributed to diagnosis in cases without visible lesions, while urine samples showed limited utility. Conclusions: This study underscores the importance of sustained laboratory surveillance and adaptive public health strategies in controlling mpox outbreaks. Optimizing specimen collection enhances diagnostic accuracy, supporting early detection. Continuous monitoring, combined with targeted vaccination and effective risk communication, is essential to prevent resurgence and ensure rapid response in non-endemic regions. Full article

Class IV laser therapy has emerged as a promising non-invasive tool to promote tissue repair, but its effects on post-surgical wound healing in small animals remain underexplored. This preliminary study investigated the impact of class IV laser therapy on surgical wound healing in 49 dogs and cats. Each surgical incision was divided into a Laser Zone (LZ) and a Control Zone (CZ). Wound healing was assessed at three timepoints (T0, T1, T2) using standardized clinical parameters: skin thickness, skin color, presence of hematoma, regional temperature, skin elasticity, and presence of fluids. The LZ consistently showed reduced skin thickness, more vivid pinkish skin color, faster hematoma resolution, increased regional temperature, improved skin elasticity, and decreased fluid accumulation compared to the CZ. These effects were observed across different species, ages, sexes, and body condition scores, indicating robust biological consistency. The results suggest that class IV laser therapy accelerates post-surgical wound healing by modulating inflammation, enhancing vascularization, and supporting extracellular matrix remodeling. This technique may serve as a valuable adjunct to conventional wound management in veterinary practice. Further studies are needed to confirm these findings and explore long-term outcomes. This study evaluated diagnostic trends and the overall utility of cytology in feline patients through the analysis of a large, multicentric dataset from Portugal. A retrospective review of 3068 cytological cases from 130 veterinary practices was conducted, with samples categorised by anatomical location and lesion type. Diagnostic outcomes were statistically assessed, revealing an overall success rate of 66.20%. The highest diagnostic yields occurred in fluid samples (83.48%), glandular tissues (76.67%), and mucous membranes (75.81%), followed by organ-based samples (67.79%), miscellaneous tissues (66.98%), cutaneous/subcutaneous nodules (62.16%), and lymph nodes (57.93%). Neoplastic lesions showed age-associated prevalence, being more common in older cats, with epithelial and melanocytic lesions more frequent in females and round cell/mesenchymal lesions predominating in males. Non-diagnostic samples (33.80%) primarily resulted from insufficient cellularity or suboptimal quality, though no significant correlation existed between diagnostic success and clinical setting. This study underscores that cytology remains a fundamental diagnostic tool in feline medicine, particularly when combined with proper sampling techniques and complementary diagnostic methods, and reinforces its value in clinical decision-making, thereby supporting its broader utilisation in routine veterinary practice. As the global demand for eco-friendly food ingredients grows, marine macroalgae emerge as a valuable resource for multiple applications using a circular bioeconomy approach. In this study, green macroalgae Ulva flexuosa, naturally accumulated in aquaculture ponds as a residual biomass (by-product) of shrimp and oyster farming, were investigated regarding their bioactivity, chemical composition, and antioxidant properties. The use of aquaculture by-products as raw materials not only reduces waste accumulation but also makes better use of natural resources and adds value to underutilized biomass, contributing to sustainable production systems. For this, a comprehensive approach including the evaluation of its composition and environmentally friendly extraction of bioactive compounds was conducted and discussed. Green macroalgae exhibited high fiber (37.63% dry weight, DW) and mineral (30.45% DW) contents. Among the identified compounds, palmitic acid and linoleic acid (ω-6) were identified in the highest concentrations. Pigment analysis revealed a high concentration of chlorophylls (73.95 mg/g) and carotenoids (17.75 mg/g). To evaluate the bioactivity of Ulva flexuosa, ultrasound-assisted solid–liquid extraction was performed using water, ethanol, and methanol. Methanolic extracts showed the highest flavonoid content (59.33 mg QE/100 g), while aqueous extracts had the highest total phenolic content (41.50 mg GAE/100 g). Ethanolic and methanolic extracts had the most potent DPPH scavenging activity, whereas aqueous and ethanolic extracts performed best at the ABTS assay. Overall, we show the upcycling of Ulva flexuosa, an underexplored aquaculture by-product, as a sustainable and sensible strategy for multiple value-added applications. Full article
