Dear Colleagues,
Platelets, historically known for their role in hemostasis and clot formation, additionally play a vital role in inflammation, infection, and immune responses. They actively contribute to the body’s protection mechanisms by means of interacting with immune cells and pathogens. In inflammation, platelets release proinflammatory mediators such as cytokines, chemokines, and growth factors, which recruit immune cells like neutrophils and monocytes to the site of injury or infection. They also amplify the inflammatory response by facilitating leukocyte adhesion and migration. Thrombocytes recognize pathogens via Toll-like receptors during infections and might directly bind to bacteria, viruses, and fungi. They facilitate the engulfment of pathogens in platelet–leukocyte aggregates and consequently enhance their clearance. Additionally, platelets release antimicrobial agents such as platelet factor 4 and β-defensins, which have direct antimicrobial properties. Furthermore, platelets interact with both innate and adaptive immune systems. They influence T-cell responses by supporting dendritic cell maturation and antigen presentation. Moreover, platelets contribute to immune regulation via macrophage activity modulation and tissue repair promotion. However, excessive platelet activation can contribute to chronic inflammation and autoimmune disorders. Understanding platelet involvement in immunity provides insights into new therapeutic approaches for inflammatory and infectious diseases.
