Dear Colleagues,

The gut microbiome serves as a master regulator of immune homeostasis, intricately balancing inflammatory responses and immune tolerance through constant dialogue with the host. Its composition and functional output are critical for educating the immune system, maintaining epithelial integrity and providing colonization resistance. Dysbiosis, or the disruption of this delicate ecosystem, is a central feature in the pathogenesis of chronic inflammatory diseases and heightened susceptibility to infection. This is powerfully illustrated by the interplay between enteric infection and Inflammatory Bowel Disease. Pathogens can exploit and exacerbate dysbiosis to drive acute inflammation, while in Inflammatory Bowel Disease, a disrupted microbiome perpetuates inappropriate immune activation and chronic tissue injury. Understanding these dynamics underscores the microbiome's pivotal role in immune regulation. Deciphering the mechanisms by which microbial signals calibrate immune responses—in health, during infection and in chronic states like Inflammatory Bowel Disease—is essential for developing novel microbiota-targeted therapies to restore equilibrium and treat disease.
