The study aims to propose a useful phylogenetic marker (instead of 16S rDNA) for a rapid classification of Thiothrix genus. It is a very interesting and valuable study for experts on this bacterial genus. Aims, methods and conclusions are actual and correct. Therefore, I recommend accepting this manuscript in its current form.

Question: Is there a possibility to use this marker also in a mixture sample, mixed bacterial cultures?

Response 1: Yes, our study was primarily designed to screen specifically for mixed cultures.

Testing new genetic markers that allow separating species within bacterial genera, provides important tools to further study specific groups of interest, such as the case of Thiothrix. The search for this type of genes seems very relevant to me and in this case very practical.
