The revised manuscript has addressed all concerns raised in the comments. No more comments in this version.
However, there is a suggestion. As additional new figures make the overall number of figures increases to 13. However, some of the figures can be presented in supplementary because of containing same information. Figure 7 is a summarized graph for figure 6, 8 and 9. Therefore, figure 6, 8 and 9 can be moved to supplementary. For figure 12 and 13, only presented the quantitation graphs (figures 12B & 13B) as main figure. For images in figures 12A & 13A can also be moved to supplementary. Additionally, figure12B & 13B can be combined to form a single figure.

We have moved previous Figures 6, 8, and 9 to Supplementary Figures (now S8, S9, and S10), as suggested by Reviewer #1. We have kept the previous Supplementary Figures S8 and S9 (now S11 and S12), since they show larger fields than Figure S10, and in addition, they show the control constructs eGFP-ER and eGFP-MITO, which are not shown in Figure S10.
Further, we have reorganized previous Figures 12 and 13. Figure 9 now shows the quantification of both previous Figures, as suggested by the Reviewer. The representative immunofluorescence images are now presented as Supplementary Figures S13 and S14.
All Figure legends and citations to the new Figures were updated accordingly.
