Pedro Henrique Dias Garcia et al. reviewed the anti-cancerous properties of L-asparaginase, focusing on its mechanism of action and therapeutic applications, as well as the side effects and issues associated with E. Coli L-asparaginase, a commercially available anticancer agent. The potential for using filamentous fungi with superior protein synthesis capabilities as cell factories for the commercial and therapeutic production of this important enzyme has been discussed by the authors. The review has been carefully planned out and is well explained. Additionally, the data and supporting literature are strong and convincing. After addressing the following points, the manuscript can be accepted for publication with minor revisions:
1. Line 159: “However…” is unclear and may need rephrasing.
2. Line 253: What is ‘Perspective I’ is it a typographical error or it is required?
3. There are several acronyms used in the manuscript, and a table with details will assist readers.
4. Grammatical mistakes should be corrected throughout the manuscript.

After carefully reading, I found this work useful and important in its field. The authors collected a set of information, analyzed the data, discussed the weak points, and suggested ways to close the current gaps in producing and using L-asparaginase in pharmaceuticals, making this work very comprehensive.
The manuscript is well-prepared and documented. However, the authors are asked to check the scientific name. It should be written italic.
After correction, I think that this work can be accepted for publication.

This manuscript entitled as "Anticancer asparaginases: Perspectives in using filamentous fungi as cell factories" by Pedro Henrique Dias Garcia et al. reviews the using of filamentous fungi for asparaginases production. Overall, the manuscript has the merits, and the summarizes are interesting to the related research field. However, this manuscript needs minor revision before acceptance for publication in Catalysts. Some comments are useful:
1. Keywords, "Enzyme" is not accurate for the manuscript, which should be changed or deleted. Besides, "Protein Engineering" or "Cell Factory" should be added into the manuscript.
2. Introduction, Paragraph 3, the introduction of acrylamide in food preparation seems irrelevant to the topic of the review. Please change and modify it or give essential explanations.
3. There are some misspellings in the manuscript. e.g. Page 4, Table 1, U/ml should be U/mL.
4. Page 7, Line 251, the combination of the different strategies should be extended.
5. The conclusions and perspectives are too brief, which should be expended.
