Round 1

Reviewer 1 Report

The manuscript has been greatly improved

Author Response

Dear reviewer,

Thank you for your contribution in reviewing our article.

Reviewer 2 Report

I received the revised manuscript.

Even if improved, some inaccuracies are also present.

Line 131: the sentence is not clear (MTC?).

In the section about RET mutations, probably it should be useful to distinguish the subtypes of MEN syndromes and the genotype-phenotype correlation.

Table 1 can be improved. Please consider to consult the recently published paper:  10.1080/17460441.2020.1674280.

Dear Editors,

Thank you for considering our review article entitled “Novel Targeted Therapies for Metastatic Thyroid Cancer -a Comprehensive Review” for publication in Cancers.

We kindly thank the Reviewer for the critical analysis of the article. We have incorporated the changes, as recommended. The changes have been highlighted through “tracked changes” in the manuscript.  We believe that the paper is now suitable for publication in Cancers.

Here is our point by point response to the reviewer’s comments:

Comment- Line 131: the sentence is not clear (MTC?).

Reply- We apologize for the lack of clarity in this statement. The sentence has been corrected to indicate that RET fusions are observed in papillary thyroid cancer (PTC) (line 131, Page 5)

Comment- In the section about RET mutations, probably it should be useful to distinguish the subtypes of MEN syndromes and the genotype-phenotype correlation.

Reply- Thank you for the comment. As per your suggestion, we have incorporated the information related to the subtypes of MEN syndromes and the genotype-phenotype correlation (line 166, page 6).

Comment- Table 1 can be improved. Please consider to consult the recently published paper:  10.1080/17460441.2020.1674280.

Reply- Thank you for the suggestion. Table 1 has been corrected to include more information on the nature of molecular alterations, estimated frequency of mutations in different types of thyroid cancer (when available), and the biological role of particular mutations in thyroid cancer.
