In this manuscript (marinedrugs-934630), authors have developed and evaluated a new QSI (rhodamine isothiocyanate analogue, RIA) as quorum sensing inhibitor to show antibiofilm ability in the natural seawater. While gone through the manuscript, I have found that this study is well designed and processed with scientific soundness. Also, the manuscript is well written and evaluated. In my opinion, this manuscript can be accpeted in its current form.

The manuscript "The rhodamine isothiocyanate analogue as a quorum sensing inhibitor has the potential to control microbe-induced biofouling" by Yu Song et al. is a well structured, elegant, clear, scientifically sound presentation of a very interesting study about the anti-biofouling properties against multispecies biofilms by a proposed new active principle functioning in natural seawater environment.

The authors planned the experimental se-tup very well, adopting a multi-approach strategy that allowed to highlight the effectiveness of the quorum sensing inhibitor rhodamine isothiocyanate analogue both in quantitative and qualitative terms.

It is therefore an original contribution of great appeal to possible readers thanks either to the fundamental information concerning the impact of rhodamine isothiocyanate analogue on the microbial ecology of the biofilms considered or to the useful indications for the transfer of the results obtained to a biotechnological and practical scale.
