In this review manuscript, Atlante and Valenti have presented a comprehensive overview of mitochondrion biology and highlighted several key discoveries in the field, with an emphasis on some carriers/transporters and their roles in metabolism and diseases, and tissue and cellular context related to their function. Overall, the manuscript is well written.

Other comments:

1.     The manuscript could be much more concise and focused. The English is too complex and distracting in some cases, often extremely long sentence with possible grammatical error (e.g. Line126-135; 371-385;442-446;749-755 et al.).

2.     Typos: e.g. Line 114: “…the electron gradient: should be “electrochemical, electrical or proton gradient"?

It is an interesting theme about the implications of the mitochondria functions in the medical research area, in specialty to bring new information about the mechanisms which are implied into various affections to observe the biological processes such as oxidative stress and tumorigenesis or to study the important biomarkers to assure the quality of life, which have a very good conserved origin in our genome. Is interesting to study the roles of mitochondria in various disorders, and I suggest improving the quality of the manuscript by following the instructions used in the revision.

Minor revision:

-Please modify the title to be in correlation with the manuscript.

-Please revise the abstract because the used terms are very confusing because exists the possibility to decrease the quality and the originality of the manuscript.

-For the entire manuscript please remove or replace the confusing terms, correct the grammatical errors, to bring the new information to establish a valuable knowledge status in the medical sciences.

-Is possible to have in the same cell, many different types of mitochondria, and this may still be a functional cell? Please explain if exist this possibility. Also, please add more references about the implications of the mitochondria dysfunction implied in various cancers.

 

-Please revise the conclusions to be correlated with the manuscript.
