Aging and neurodegenerative diseases are characterized by common features involving bioenergetics deficiencies, oxidative stress and alterations of calcium buffering. Mechanisms of mitochondrial-targeted drugs include the modulation of electron transport chain and oxidative phosphorylation, the binding to mitochondrial lipids, free-radical scavenging, calcium signaling, and possible effects on mitochondrial biogenesis and dynamics and on the regulation of mitophagic pathways. One of the main sites of action of mitochondria-targeted drugs is the interaction with respiratory chain components. Mitochondrial-targeted compounds such as Mito-Q, and Mito-apocynin have been developed by conjugating triphenylphosphonium (TPP+) lipophilic cation group with natural molecules, therefore obtaining promising drugs for reestablishing the correct functioning of the mitochondrial respiratory chain. Stabilization of cardiolipin at the inner mitochondrial membrane by elamipretide or SkQ1 and mitochondria-targeted ROS scavengers can also offer a therapeutic approach to prevent bioenergetic impairment associated with several diseases. In addition, the modulation of calcium signaling can be achieved using both MCU agonists and antagonists representing another mitochondrial target for drug therapies development. Finally, potential strategies for treating neurodegenerative diseases based on the modulation of mitochondrial biogenesis, dynamics and/or mitophagic pathways are discussed. Full article

Berries are an excellent source of bioactive compounds, particularly polyphenols, and have been widely used in folk medicine by the Mapuche people of southern Chile. In this study, a hydroalcoholic extract of Berberis congestiflora Gay (BE) was analyzed to determine its phytochemical composition and to evaluate its antioxidant capacity, vasorelaxant effects in rat aortas, and inhibitory activity on enzymes related to chronic non-communicable diseases, including exploration of a possible vasodilatory mechanism in isolated rat aortas. Antioxidant activity was assessed using Oxygen Radical Absorbance Capacity (ORAC), DPPH (2,2-diphenyl-1-picrylhydrazyl), and ABTS (2,2-azinobis-(3-ethylbenzothiazolin-6-sulfonic acid)) radical scavenging assays, as well as ferric reducing antioxidant power (FRAP). Vascular responses to the Berberis extract were studied using isometric tension recordings in an ex vivo rat thoracic aortic ring model, and the chemical constituents of BE were identified for the first time by HPLC-DAD-MS. The extract itself produced a dose-dependent contraction at 100 and 1000 µg/mL and induced relaxation in phenylephrine-precontracted aortas at the same concentrations, with a maximum contraction of 71% and maximum relaxation of 70% at 1000 µg/mL. Mechanistically, the extract triggered calcium-mediated contraction primarily through calcium release from the sarcoplasmic reticulum and, to a lesser degree, via extracellular Ca2+ influx, while its relaxant effect depended on an intact endothelium and activation of the NO/cGMP pathway. In addition, the extract showed inhibitory activity against cholinesterase, glucosidase, and amylase, with IC50 values of 7.33 ± 0.32, 243.23 ± 0.3, and 27.21 ± 0.03 µg/mL, respectively, and docking studies were carried out for selected berry compounds. Overall, these findings indicate that these berries are a rich source of bioactive constituents with antioxidant properties and endothelium-dependent vasodilator effects, supporting their traditional use and highlighting their potential as enzyme inhibitors and as promising candidates for the development of phytotherapeutic products, particularly as supplements for chronic disease management. This study reports the synthesis and detailed characterization of pullulan-isononanoate (Pull-Iso), as well as the preparation and characterization of Pull-Iso films incorporating liposomes loaded with silibinin (SB) and smoke tree (Cotinus coggygria) extract (STExt), to explore the physicochemical and functional properties of pullulan-based biomaterials for potential biomedical applications. Pullulan was successfully esterified with isononanoic acid chloride, as confirmed by 1H and 13C NMR (Nuclear Magnetic Resonance) and Fourier Transform Infrared (FTIR) spectroscopy. Modification significantly reduced the glass transition temperature (Tg), indicating enhanced chain mobility due to the introduction of bulky side chains. Prepared liposomes, embedding SB and extracted smoke tree compounds, exhibited particle sizes ~2000 nm with moderate polydispersity (~0.340) and zeta potential values around –20 mV, demonstrating lower colloidal stability over 60 days, thereby justifying their encapsulation within films. Optical microscopy revealed uniform liposome dispersion in Pull-Iso film with 0.5 g of liposomes, while higher liposome loading (0.75 g of liposomes) induced aggregation and microstructural irregularities. Mechanical analysis showed a reduction in tensile strength and strain at higher liposome content. The incorporation of liposomes encapsulating STExt and SB significantly enhanced the antioxidant activity of Pull-Iso-based films in a concentration-dependent manner, as demonstrated by DPPH and ABTS radical scavenging assays. These preliminary findings suggest that pullulan esterification and controlled liposome incorporation may enable the development of flexible, bioactive-loaded films, which could represent a promising platform for advanced wound dressing applications, warranting further investigation. Cannabidiol (CBD), the primary bioactive element of cannabis, has shown promise in alleviating pain and inflammation, although mechanisms in periodontal inflammation are not fully understood. To improve its limited solubility and mucosal permeability, the developed chitosan-based mucoadhesive hydrogel incorporating CBD-loaded PLGA nanospheres (CPN hydrogel) was characterized by FT-IR, SEM, particle size, rheological, swelling, and diffusion analyses, followed by biological evaluations, including wound-healing and RT-qPCR-based anti-inflammatory assays. The improved CPN hydrogel had a homogeneous shape, better viscoelastic behavior, and sustained drug release. Over 90% of CBD was released within 96 h, and Franz cell experiments showed improved permeability (124.1 μg/cm2 after 72 h). The gellan gum-based mucosal substrate significantly increased adhesion (1137.33 ± 142.25 s) compared to the control groups. Antioxidant studies indicated 73.65% DPPH radical scavenging, whereas antibacterial tests showed more than 99% suppression of Staphylococcus aureus. Furthermore, in vitro studies validated its wound healing and the downregulation of the inflammatory cytokines IL-6 and TNF-α. The results indicate that the CPN-loaded chitosan hydrogel has extended mucosal retention, strong antibacterial activity, and steady release of CBD. This underscores its significant potential as a targeted treatment for inflammatory oral diseases such as gingivitis and periodontitis. Full article

Potato (Solanum tuberosum L.) is one of the most widely cultivated crops in the world; however, drought is a major constraint to its productivity. Arbuscular mycorrhizal fungi (AMF) have been shown to improve plant resistance under conditions of water stress. However, their effects on potato plants are poorly studied. The purpose of this study was to evaluate the potential of two AMF inocula (two different strains of the AMF species Rhizophagus irregularis with different origin: Southern Spain MI1 and Tunisia MI2) on potato tolerance to drought stress through the determination of growth parameters, photosynthetic parameters, and antioxidant systems, under well-watered (WW; field capacity) and drought stress (DS; 50% of field capacity) conditions. Therefore, the experiment consisted of two factors: AMF strain and watering regime. The results showed that under drought stress conditions, AMF inoculation considerably stimulated photosynthetic performance as compared with non-inoculated controls. Moreover, leaf superoxide dismutase (SOD) and catalase (CAT) activities of inoculated plants were higher in WW conditions, but unchanged in DS conditions. Inoculated plants had significantly higher ascorbate peroxidase (APX) and glutathione reductase (GR) activities than non-inoculated plants under DS conditions. Also, expression of some antioxidant enzyme genes were upregulated by inoculation. Lipid peroxidation content of inoculated plants was lower than that of non-inoculated. Furthermore, there was a high positive correlation between mycorrhizal root colonization (RC) and almost all the measured parameters. The results of this study indicated that AMF inoculation could enhance potato plant tolerance to water stress through the induction of antioxidant mechanisms implicated in scavenging oxygen-free radicals. This study examined whether co-fermentation with Lactobacillus casei CGMCC 15956 and Lactobacillus delbrueckii CGMCC 21287 could enhance the bioactivity of peptides derived from pecan nut cake (PNC) and clarify the underlying mechanisms. The fermented hydrolysate (PNCH) was compared with an unfermented control. PNCH showed higher antioxidant and α-glucosidase inhibitory activities. Total antioxidant capacity increased from 3.17 to 4.81 mM Trolox, and DPPH radical scavenging activity increased from 62.69% to 84.12%. In addition, the IC50 value for α-glucosidase inhibition decreased from 7.549 to 4.509 mg/mL. In a mouse model of acute alcohol-induced liver injury, PNCH significantly alleviated liver damage through the synergistic enhancement of antioxidant and α-glucosidase inhibitory activities. Peptidomic analysis identified two representative bioactive peptides, FAGDDAPR (from actin) and LAGNPDDEFRPQ (from cupin domain–containing protein 1), both of which exhibited antioxidant and α-glucosidase inhibitory activities. Additionally, these peptides alleviated H2O2-induced oxidative stress in Caco-2 cells, significantly improving GSH and MDA levels, as well as SOD activity. Molecular docking suggested potential interactions of these peptides with superoxide dismutase, Keap1, and α-glucosidase. These findings support the high-value utilization of PNC and the development of functional peptide-based ingredients. Full article

Oxidative stress contributes significantly to chronic disease burden, necessitating identification of accessible dietary antioxidant sources. Pigeon peas (Cajanus cajan L.) contain substantial bioactive compounds, yet most exist in bound forms with limited bioavailability. This study evaluated wild fermentation combined with systematic extraction optimization to enhance antioxidant recovery from pigeon peas. Seeds underwent wild fermentation in brine solution, followed by extraction under varying conditions (seven solvent systems, three temperatures, and three-time durations). Multiple complementary assays assessed antioxidant capacity (total phenolic content, DPPH radical scavenging, ferric reducing power, and ABTS activity). Fermentation substantially improved antioxidant properties across all parameters, with particularly pronounced effects on radical scavenging activities. Extraction optimization identified 70% methanol at 40 °C for 24 h as optimal, demonstrating marked improvements over conventional protocols. Strong intercorrelations among assays confirmed coordinated enhancement of multiple antioxidant mechanisms rather than isolated changes. The findings demonstrate that both biotechnological processing and analytical methodology critically influence antioxidant characterization in pigeon peas. This integrated approach offers practical guidance for developing antioxidant-rich functional foods, particularly relevant for resource-limited settings where pigeon peas serve as dietary staples. The study establishes foundation for translating fermentation technology into nutritional interventions, though further research addressing bioavailability, microbiological characterization, and bioactive compound identification remains essential. Fucoidan, a complex sulfated polysaccharide derived from marine brown seaweeds, exhibits broad biological activities, including anticoagulant, antitumor, antiviral, anti-inflammatory and lipid-lowering effects. Fucoidan confers neuroprotection in animal models of a broad spectrum of brain disorders such as Parkinson’s disease (PD) and depression. However, the effect of fucoidan on gut-derived neuroinflammation and associated behavioral changes has been scarcely investigated. In comparison to fucoidan from other brown seaweeds, that from Fucus vesiculosus exhibited a better neuroprotective effect in vivo and more potent radical scavenging activity in vitro. Fucoidan from Laminaria japonica ameliorates behavioral disorders related to acute ulcerative colitis (UC) in aged mice. It is of interest to assess the effects of fucoidan administration on intestinal and brain inflammation in the acute colitis mouse model. Fucoidan treatment ameliorated DSS-induced intestinal pathology, reduced the inflammatory mediator expression in the gut and brain, and activated intestinal macrophages and cortical microglia in the UC mice. It also protected the intestinal mucosal barrier and blood–brain barrier as well as prevented neuronal damage, while alleviating anxiety-like behavior in UC mice. These results suggest fucoidan supplementation may help prevent brain disorders, such as depression and PD, potentially involving gut–brain axis-related mechanisms, as fucoidan suppresses gut-derived neuroinflammation. Background: Agri-food by-products are increasingly recognized as valuable sources of tannins, whose antioxidant properties represent the primary driver of their biological activity across human and animal health. The strong redox-modulating capacity of condensed and hydrolysable tannins provides a unifying mechanistic explanation for their effects on inflammation, metabolism, gut integrity and neuroprotection. Methods: This narrative review synthesizes evidence obtained through a structured literature search across major databases, selecting studies that investigated antioxidant mechanisms of tannin-rich matrices from plant- and processing-derived residues. Results: Condensed tannins, particularly proanthocyanidins, consistently display potent antioxidant activity through radical scavenging, metal chelation and activation of endogenous defenses, thereby underpinning their anti-inflammatory, anti-ischemic, neuroprotective and metabolic actions. Hydrolysable tannins similarly exert strong antioxidative effects that support antimicrobial activity, enzyme modulation and protection against neuroinflammation. In animals, the antioxidant capacity of tannins translates into improved oxidative balance, enhanced immune status, reduced tissue damage, better feed efficiency and mitigation of oxidative stress-linked methane emission pathways. Conclusions: Antioxidant activity emerges as the central, cross-species mechanism through which tannins mediate diverse health benefits. Tannin-rich agri-food by-products therefore represent promising sustainable antioxidant resources, although their efficacy remains influenced by tannin class, degree of polymerization and dosage, warranting further mechanistic and translational research. Athamanta L. is a small genus of the Apiaceae family, comprising only sixteen species and subspecies, which are distributed in the Canary Islands, Central Europe, and the Mediterranean basin. Background/Objectives: Since the time of Dioscurides, the species of this genus have been reported to have had several ethnopharmacological activities, and some of them are also used currently. Athamanta sicula L., growing in Italy, Tunisia, Algeria, and Morocco, is the only species of this genus present in Sicily. To further explore the phytochemical profile and biological properties of this species, the present study focused on the essential oil (EO) extracted from the aerial parts of wild A. sicula populations collected in central Sicily. Methods: The chemical composition of the EO, obtained by hydrodistillation, was determined by GC–MS analysis. The presence of myristicin was confirmed by isolation and by 1H-NMR spectroscopic characterization. Results: The EO and its main constituents have been tested for possible antimicrobial properties against several bacterial strains, showing MIC values in the of 15–30 mg/mL range, and the mechanism of action was further investigated, revealing membrane-targeting effects consistent with outer membrane permeabilization. In addition, antibiofilm activity (with up to ~80% inhibition of biofilm formation at sub-MICs), antioxidant potential (demonstrating dose-dependent radical scavenging activity), and biocompatibility with eukaryotic cells were assessed to provide a comprehensive pharmacological profile of A. sicula EO. Specifically, the most abundant constituent was myristicin (62.2%), the principal representative of the phenylpropanoid class (64.4%). Hydrocarbon monoterpenes represented the second class of the EO (27.4%), with β-phellandrene (12.2%) as the main compound. Conclusions: Myristicin emerged as the key contributor to the antimicrobial and antibiofilm activity of the EO. The obtained results highlight the relevance of A. sicula EO as a myristicin-rich essential oil with notable in vitro biological activity. Full article

With the growing demand for eco-friendly food packaging, poly(butylene adipate-co-terephthalate) (PBAT)/polylactic acid (PLA) composite films have emerged as promising biodegradable alternatives, but their inherent limitations (e.g., poor antioxidant capacity, weak UV stability, and insufficient antimicrobial activity) hinder practical applications. This study aimed to address these drawbacks by incorporating Rosmarinus officinalis L. extract (RM) as a natural multifunctional additive. PBAT/PLA/RM blend films with RM concentrations of 0.1%, 0.3%, 0.5%, and 1% were fabricated via melt extrusion and blown film processing. Key characterizations were conducted to evaluate thermal stability, mechanical properties, morphology, antioxidant activity, UV-shielding performance, antimicrobial efficacy, and biodegradability. The results showed that RM significantly enhanced the antioxidant capacity of the films, with the highest DPPH radical scavenging activity achieved at 0.3% RM. UV-blocking performance improved incrementally with increasing RM concentration, and films containing ≥0.5% RM filtered over 90% of UVA and UVB radiation. All composite films met biodegradability standards, with over 90% degradation observed after 240 days of composting, though RM prolonged the initial degradation stage by inhibiting early microbial activity. However, the antimicrobial effect of RM was limited, and concentrations exceeding 1% caused film stickiness, impeding processing. This work demonstrates that RM is a viable natural additive for functionalizing PBAT/PLA films, offering enhanced antioxidant and UV-shielding properties while maintaining biodegradability, thus providing a promising solution for sustainable food packaging. To reduce the environmental impact of plastic packaging in the edible fungi supply chain, this study developed an edible natural chitosan composite film loaded with chlorogenic acid–chitosan oligosaccharide nanoparticles (CGA/COS NPs). The effects of CGA/COS NPs as additives on the structure and overall performances of chitosan-based films were systematically studied, and the application effect of nanoparticles/chitosan (NPs/CS) composite films in the preservation of Pleurotus geesteranus was explored. The results showed that the NPs had good compatibility with the film matrix, filled the voids of the chitosan matrix, enhanced the comprehensive performance of the film, and significantly improved the antioxidant activity of the film (DPPH free radical scavenging activity increased from 16.95% to 76.47%). Among all the films, the 5%NPs/CS composite film performed the best, not only having stronger barrier properties against moisture, oxygen, and ultraviolet rays, but also having the best thermal stability and mechanical properties, which can effectively extend the shelf life of Pleurotus geesteranus. This study developed a high-performance edible composite film, which provides a new path of great value for solving the preservation problem of perishable agricultural products such as Pleurotus geesteranus and promoting the innovative development of the green food packaging industry. Cadmium sulfide is an important II-VI semiconductor known for its valuable photocatalytic properties ascribable to its band gap energy, which allows light absorption in the visible domain. Nonetheless, the application of cadmium sulfide in wastewater organic pollutant degradation is restricted due to its high toxicity to humans, soil, and marine life. To address this issue, we developed new composite materials by depositing CdS on a bentonite support in a 1:9 mass ratio to develop a photocatalyst with lower toxicity. In the first step, bentonite was activated using an aqueous HCl solution; for the deposition of CdS powder, we proposed the trituration method and compared it with chemical precipitation and hydrothermal synthesis, using thioacetamide as a sulfide ion source. The modified bentonite underwent characterization using X-ray diffraction, scanning electron microscopy, X-ray fluorescence, UV-Vis, and FTIR spectroscopy. The photocatalytic activity was tested in the degradation of Congo red (CR), a persistent diazo dye. The efficiency of removing CR with CdS–bentonite composites depended on the deposition method of CdS, and it was higher than that of pristine CdS and of only adsorption onto acid-activated bentonite. The photocatalytic degradation mechanism was estimated by the scavenger test using ethylenediaminetetraacetic acid disodium salt, ascorbic acid, ethanol, and silver nitrate as radical scavengers. Fermentation is an effective method to enhance the bioactivity of plant proteins, yet the link between the functionality and conformational state of fermented soybean 11S protein (F11S) requires clarification. This study first evaluated the antioxidative efficacy of F11S and its application in emulsion systems, followed by a mechanistic investigation into its structural evolution. Results showed that the bioactivity of F11S was strictly fermentation-time-dependent, reaching its peak at 16 h. At this stage, F11S exhibited maximal scavenging capacities for ·OH (84.51 ± 2.53%) and DPPH radicals (93.84 ± 2.62%). Crucially, in a Tween 20 emulsion system, the F11S-16h fraction demonstrated superior oxidative stability, maintaining the lowest peroxide value (4.33 ± 0.53 mmol/kg) after 15 days of storage. To elucidate the mechanism behind this enhanced functionality, structural analysis was conducted. It revealed that while surface hydrophobicity peaked at 12 h due to protein unfolding, extended fermentation to 16 h induced a refolding process, guiding the protein into a thermodynamically stable conformation. These findings indicate that the stable refolded structure formed at 16 h, rather than maximal hydrophobicity, is the key determinant for the superior antioxidant performance and emulsion stabilizing ability of F11S. Natural biodegradable polymers such as chitosan are gaining increasing importance due to their favorable mechanical properties. Conversely, their limited antimicrobial and antioxidant activity requires enhancement with bioactive components. This study investigated the effect of Teucrium montanum L. hydrolate on the functional properties of chitosan films. The hydrolate was obtained as a by-product of hydrodistillation, and films were prepared with 0.6% (CH-TMh1), 0.8% (CH-TMh2), and 1.2% (CH-TMh3) hydrolate, along with a control film without hydrolate (CH). Hydrolate-enriched films exhibited greater thickness and elongation at break, with the highest values observed in CH-TMh3. The addition of hydrolate reduced moisture content (from 30.09% in CH to 12.25% in CH-TMh3), solubility, and swelling degree. Antioxidant activity increased significantly, with CH-TMh2 showing the highest free radical scavenging activity (92.9%) and total polyphenol content (38.78 mg GAE/g). Films containing hydrolate also displayed pronounced antimicrobial activity, with the largest inhibition zones against S. aureus ATCC 25923 (16.33 mm). Moderate activity was observed against B. subtilis, while there was no activity against C. albicans ATCC 2091. These results confirm that chitosan films enriched with T. montanum L. hydrolate possess improved mechanical, antioxidant, and antimicrobial properties, making them promising for potential application in the packaging of specific food products.
